The CD4 binding site(CD4bs) of envelope glycoprotein(Env) is an important conserved target for anti-human immunodeficiency virus type 1(HIV-1) neutralizing antibodies. Neutralizing monoclonal antibodies IgG1 b12(b12) ...The CD4 binding site(CD4bs) of envelope glycoprotein(Env) is an important conserved target for anti-human immunodeficiency virus type 1(HIV-1) neutralizing antibodies. Neutralizing monoclonal antibodies IgG1 b12(b12) could recognize conformational epitopes that overlap the CD4 bs of Env. Different virus strains, even derived from the same individual, showed distinct neutralization susceptibility to b12. We examined the key amino acid residues affecting b12 neutralization susceptibility using single genome amplification and pseudovirus neutralization assay. Eleven amino acid residues were identified that affect the sensitivity of Env to b12. Through site-directed mutagenesis, an amino acid substitution at position 182 in the V2 region of Env was confirmed to play a key role in regulating the b12 neutralization susceptibility. The introduction of V182 L to a resistant strain enhanced its sensitivity to b12 more than twofold. Correspondingly, the introduction of L182 V to a sensitive strain reduced its sensitivity to b12 more than tenfold. Amino acid substitution at positions 267 and 346 could both enhance the sensitivity to b12 more than twofold. However, no additive effect was observed when the three site mutageneses were introduced into the same strain, and the sensitivity was equivalent to the single V182 L mutation. CRF07_BC is a major circulating recombinant form of HIV-1 prevalent in China. Our data may provide important information for understanding the molecular mechanism regulating the neutralization susceptibility of CRF07_BC viruses to b12 and may be helpful for a vaccine design targeting the CD4 bs epitopes.展开更多

Hematopoietic traits of red and white blood cells(like erythrocytosis and myeloproliferative disease), autoimmune disorders(like type 1 diabetes, coeliac disease, juvenile idiopathic arthritis, rheumatoid arthritis, thrombotic antiphospholipid syndrome, lupus erythematosus, multiple sclerosis, hypothyroidism and vitiligo), also vascular pathology(like kidney glomerular filtration rate deficits, serum urate levels, plasma beta-2-microglobulin levels, retinal microcirculation problems, diastolic and systolic blood pressure and hypertension, cardiovascular infarction), furthermore obesity, neurodegenerative conditions(like the polyglutamine-expansion disorder spinocerebellar ataxia type 2, Parkinson's disease, the motor-neuron disease amyotrophic lateral sclerosis, and progressive supranuclear palsy), andfinally longevity were reported. Now it is important to clarify, in which ways the loss or gain of function of the locally encoded proteins SH2B3/LNK and ataxin-2, respectively, contribute to these polygenic health problems. SH2B3/LNK is known to repress the JAK2/ABL1 dependent proliferation of white blood cells. Its null mutations in human and mouse are triggers of autoimmune traits and leukemia(acute lymphoblastic leukemia or chronic myeloid leukemia-like), while missense mutations were found in erythrocytosis-1 patients. Ataxin-2 is known to act on RNA-processing and trophic receptor internalization. While its polyglutamine-expansion mediated gain-of-function causes neuronal atrophy in human and mouse, its deletion leads to obesity and insulin resistance in mice. Thus, it is conceivable that the polygenic pathogenesis of type 1 diabetes is enhanced by an SH2B3-dysregulation-mediated predisposition to autoimmune diseases that conspires with an ATXN2-deficiency-mediated predisposition to lipid and glucose metabolism pathology.展开更多
Objective:To investigate influence of folic acid and vitamin B12 combined therapy on plasma homocysteine (Hcy) level, blood vessels endothelial function and inflammatory factors in patients with vascular dementia and ...Objective:To investigate influence of folic acid and vitamin B12 combined therapy on plasma homocysteine (Hcy) level, blood vessels endothelial function and inflammatory factors in patients with vascular dementia and type H hypertension.Methods:100 cases of patients with vascular dementia and type H hypertension accorded with the inclusion criteria were selected as research objects. They were randomly divided as the control group and the therapeutic group, 50 cases each. For control group, Enalapril tablets were administered by mouth for treatment. For therapeutic group, folic acid and vitamin B12 treatment were provided on the basis of treatment for control group. Treatments were continued for 12 weeks. Plasma Hcy levels, inflammatory factors [(interleukin-6 (IL-6), interleukin-8 (IL-8) and hypersensitive C reaction protein (hs-CRP)], blood vessels endothelial function indexes variation in patients before and after treatment were observed and detected.Results:Plasma Hcy, IL-6, IL-8 and hs-CRP levels in two groups of patients after treatment were significantly decreased comparing with the same group before treatment, and the above index levels in therapeutic group after treatment were significantly lower than control group (P<0.05);For comparison of blood vessels endothelial function indexes in the patients, NO levels in two groups after treatment were increased in various degrees, and endothelin-1 (ET-1) were decreased. The differences between levels of the two indexes in therapeutic group before and after treatment were significant, and levels after treatment in therapeutic group were significantly better than in control group (P<0.05). While variations of the differences in control group before and after treatment were not significant (P>0.05);After treatment, diastolic pressure and systolic pressure in the two groups of patients were significantly improved comparing with before treatment (P<0.05). However, after treatment, the differences of levels between therapeutic group and control group were not significant (P>0.05). MMSE score in therapeutic group after treatment was significantly higher than before treatment, and significantly higher than in control group (P<0.05).Conclusions: Combined therapy of folic acid and vitamin B12 for treating vascular dementia with type H hypertension could effectively decrease plasma Hcy and inflammatory factor levels, and improve blood vessels endothelial function and dementia degree on patients. It has certain clinical value which deserves to be promoted.展开更多

We questioned the...Objectives: Diabetic retinopathy (DR) is one of the most common microvascular complications of type 2 diabetes mellitus (T2DM). It is multifactorial with the contribution of multiple genetic factors. We questioned the association of polymorphisms in the peroxisome proliferator-activated receptor ?2 (PPAR?2) gene (Pro12Ala and C161T) with DR in an Egyptian population. Methods: This case control study included one hundred healthy individuals and 252 T2DM among them 122 with DR and 130 without DR. Genotyping was done by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP). Results: The Pro12Ala Ala allele was associated with decreased risk of DR with an odds ratio (OR) of 0.484, 95% confidence interval (CI) (0.254 - 0.920), and a p value = 0.024. The C161T T allele was associated with increased risk of DR with OR = 2.593, 95% CI (1.672 - 4.020), p < 0.001. However, when considering other covariates such as glycosylated hemoglobin (HbA<sub>1c</sub>) in multivariate regression analysis only C161T was associated with increased risk of DR with OR = 3.479, 95% CI (1.907 - 6.346), p < 0.001, while the significant association with Pro12Ala was lost. HbA<sub>1c</sub> was higher in Pro/Pro genotype when compared to those with Ala/Ala and Pro/Ala genotypes. Conclusion: We report that T allele of C161T increased risk of DR in the studied population. Further studies are warranted to investigate functional implications of polymorphisms of the PPAR-? gene in DR development.展开更多
Solid precipitation is not only the main supply for glacier mass,but also exerts an important influence on surface albedo and intensifies glacier melting.However,precipitation type observation is very scarce in the hi...Solid precipitation is not only the main supply for glacier mass,but also exerts an important influence on surface albedo and intensifies glacier melting.However,precipitation type observation is very scarce in the high alpine glaciers,which limits the precise simulation of glacier mass balance.This study assessed three discrimination methods of precipitation types including Ding method,Dai method and Froidurot method based on surface albedo observation data on the Laohugou Glacier No.12(LHG Glacier)in western Qilian Mountains.The results showed that Ding method had a best applicability on the LHG Glacier,the other two need to calibrate parameters when they are used in the high elevation glacier region.Then we fitted the relationship between snowfall probability and fresh snow albedo,and put forward a revised formula to simulate fresh snow albedo based on Ding method,which is expected to reduce the uncertainty in glacier mass and energy balance model.Finally,we found a best air temperature threshold of 4℃for discriminating monthly precipitation types.In order to accurately simulate the glacier melt,it is necessary to obtain the threshold temperature appropriately in different glacier region with different elevation and humidity.展开更多
Objective:To study the relationship of interleukin-12(IL-12)and IL-4 levels with macroangiopathy in type 2 diabetes mellitus by comparing the serum IL-12 and IL-4 levels and intima-media thickness(IMT)between the heal...Objective:To study the relationship of interleukin-12(IL-12)and IL-4 levels with macroangiopathy in type 2 diabetes mellitus by comparing the serum IL-12 and IL-4 levels and intima-media thickness(IMT)between the healthy controls and the type 2 diabetic patients with or without macroangiopathy.Methods:30 healthy controls and 120 cases with type 2 diabetes mellitus from November 2011 to November 2012 were selected.In accordance with the carotid intima-media thickness,the type 2 diabetes mellitus patients were divided into group A with IMT below 1.0 mm,group B with IMT between 1.0 mm and 1.2 mm,group C with IMT between 1.2 mm and 1.4 mm,group D with IMT over 1.4 mm.ELISA was used to measure the levels of serum IL-12 and IL-4 in all groups.And the measurement results of serum IL-12 and IL-4 levels in the groups were compared.Results:Type 2 diabetes mellitus groups had significantly higher serum IL-12 levels and much lower IL-4 levels than the healthy control group.In the type 2 diabetes mellitus groups,the thicker the IMT was,the higher the incidence of complications became.IL-12 level was positively correlated with IMT while the IL-4 level was negatively correlated with IMT.Conclusions:IL-12 levels in patients with type 2 diabetes were significantly higher and IL-4 levels were significantly lower than those in the healthy people.In patients with type 2 diabetes,the IL-12 levels increased while the IL-4 levels gradually declined with the advance of macroangiopathy.展开更多

文摘Objective:To study the relationship of interleukin-12(IL-12)and IL-4 levels with macroangiopathy in type 2 diabetes mellitus by comparing the serum IL-12 and IL-4 levels and intima-media thickness(IMT)between the healthy controls and the type 2 diabetic patients with or without macroangiopathy.Methods:30 healthy controls and 120 cases with type 2 diabetes mellitus from November 2011 to November 2012 were selected.In accordance with the carotid intima-media thickness,the type 2 diabetes mellitus patients were divided into group A with IMT below 1.0 mm,group B with IMT between 1.0 mm and 1.2 mm,group C with IMT between 1.2 mm and 1.4 mm,group D with IMT over 1.4 mm.ELISA was used to measure the levels of serum IL-12 and IL-4 in all groups.And the measurement results of serum IL-12 and IL-4 levels in the groups were compared.Results:Type 2 diabetes mellitus groups had significantly higher serum IL-12 levels and much lower IL-4 levels than the healthy control group.In the type 2 diabetes mellitus groups,the thicker the IMT was,the higher the incidence of complications became.IL-12 level was positively correlated with IMT while the IL-4 level was negatively correlated with IMT.Conclusions:IL-12 levels in patients with type 2 diabetes were significantly higher and IL-4 levels were significantly lower than those in the healthy people.In patients with type 2 diabetes,the IL-12 levels increased while the IL-4 levels gradually declined with the advance of macroangiopathy.
