Effects of ultrasonic-assisted extraction on bioactive compounds,volatile flavors and antioxidant activities of vine tea water extracts

Background:Ampelopsis grossedentata,vine tea,which is the tea alternative beverages in China.In vine tea processing,a large amount of broken tea is produced,which has low commercial value.Methods:This study investigates the influence of different extraction methods(room temperature water extraction,boiling water extraction,ultrasonic-assisted room temperature water extraction,and ultrasonic-assisted boiling water extraction,referred to as room temperature water extraction(RE),boiling water extraction(BE),ultrasonic assistance at room temperature water extraction(URE),and ultrasonic assistance in boiling water extraction(UBE))on the yield,dihydromyricetin(DMY)content,free amino acid composition,volatile aroma components,and antioxidant properties of vine tea extracts.Results:A notable influence of extraction temperature on the yield of vine tea extracts(P<0.05),with BE yielding the highest at 43.13±0.26%,higher than that of RE(34.29±0.81%).Ultrasound-assisted extraction significantly increased the DMY content of the extracts(P<0.05),whereas DMY content in the RE extracts was 59.94±1.70%,that of URE reached 66.14±2.78%.Analysis revealed 17 amino acids,with L-serine and aspartic acid being the most abundant in the extracts,nevertheless ultrasound-assisted extraction reduced total free amino acid content.Gas chromatography-mass spectrometry analysis demonstrated an increase in the diversity and quantity of compounds in the vine tea water extracts obtained through ultrasonic-assisted extraction.Specifically,69 and 68 volatile compounds were found in URE and UBE extracts,which were higher than the number found in RE and BE extracts.In vitro,antioxidant activity assessments revealed varying antioxidant capacities among different extraction methods,with RE exhibiting the highest DPPH scavenging rate,URE leading in ABTS•+free radical scavenging,and BE demonstrating superior ferric ion reducing antioxidant activity.Conclusion:The findings suggest that extraction methods significantly influence the chemical composition and antioxidant properties of vine tea extracts.Ultrasonic-assisted extraction proved instrumental in elevating the DMY content in vine tea extracts,thereby enriching its flavor profile while maintaining its antioxidant properties. 展开更多

[Objectives]To establish a TLC and content determination method of Pileostegia tomentellal,with umbelliferone as the indicator component.[Methods]TLC identification was performed by silica gel G thin layer plate with n-hexane-ethyl acetate(4:3)as the developing agent,and the plate was examined by UV lamp(365 nm).The umbelliferone content was determined by HPLC:Inertsil ODS-3 C 18 column(4.60 mm×250 mm,5μm);mobile phase acetonitrile-0.2%phosphoric acid gradient elution;detection wavelength 320 nm,flow rate 1.0 mL/min,column temperature 30℃,injection volume 10μL.[Results]The chromatogram of P.tomentellal showed the same color spot in the same position as that of reference medicinal material,and the spot was clear with good specificity.Umbelliferone showed a good linear relationship when the injection volume was 2.63-131.27μg/mL(R^(2)=0.9997).The average recovery of umbelliferone in the low,middle and high adding groups of P.tomentellal was 99.57%and the RSD was 2.15%.[Conclusions]The method can effectively identify Yao medicine P.tomentellal and accurately determine the content of umbelliferone in medicinal materials,which will provide a scientific basis for the development and utilization of medicinal resources of Yao medicine P.tomentellal. 展开更多

Jujube contains abundant cyclic adenosine monophosphate(cAMP)and the ultrasonic-assisted pectinase extraction(UAPE)conditions for obtaining the maximum cAMP yield from jujube were optimized.Orthogonal array design was applied to evaluate the effects of 4 variables by UAPE on cAMP yield.The results showed that the optimal cAMP yield(783.0μg/g)was derived at ratio of liquid to solid 5 mL/g,ratio of pectinase to raw material 1.5%,time 60 min and temperature 40℃.Moreover,the effect of cAMP on the anti-allergic function of action induced by immunoglobulin E(IgE)and its meschanism was investigated through establishing the sensitized cell model in rat basophilic leukemia(RBL-2 H3)cells using dinitrophenylated(DNP)-bovine serum albumin(BSA)-IgE.The results showed that cAMP interfered with sensitized cells,effectively inhibited the occurrence of basophil degranulation in dose dependence,and significantly reduced the activity ofβ-hexosamindase(β-hex),at the optimal concentration of 50μg/mL.The level of anti-inflammatory factor interleukin-10(IL-10)was promoted and the content of pro-inflammatory factor tumor necrosis factor-α(TNF-α)was suppressed by cAMP.In addition,influx of intracellular Ca^(2+) was repressed effectively.Our results demonstrate that jujube cAMP regulated the cytokine balance in the allergy pathway through blocking the influx of extracellular Ca^(2+),with the prevention of allergy symptoms. 展开更多

Objective To understand the development status of traditional Chinese medicine extraction technology,so as to provide a reference for enterprises and scientific research institutions to determine the direction of technological innovation.Besides,the government can formulate scientific and technological innovation strategies.Methods Based on patent information,statistical methods were applied to analyze the development of traditional Chinese medicine extraction technology.Then,the core technology was identified by constructing a patent citation network.Finally,the main path analysis method was applied to discover the technological evolution path.Results and Conclusion The extraction technology of traditional Chinese medicine is still in the growth stage.At present,its R&D is based on enterprises,supplemented by scientific research institutions.The core technologies are ultrasonic extraction and enzymatic extraction.The main path represented by enzymatic extraction technology reveals the evolution process of traditional Chinese medicine extraction technology.Based on this,relevant suggestions are put forward.Firstly,technological innovation cooperation should be strengthened to explore new models of achievement transformation and cooperation.Secondly,key enterprises of technology clusters should be cultivated to accelerate breakthroughs in core technologies.Thirdly,combined technologies can achieve high-efficiency enzyme-assisted extraction and open up new ways of extracting traditional Chinese medicine.Fourthly,traditional techniques should be improved to develop innovative modern techniques. 展开更多

In order to optimize the microwave-assisted extraction technology of polyphenols from Loropetalum chinense (R. Br.) Oliv., the effects of microwave power, ethanol concentration, solid to liquid ratio and extraction time on polyphenols extraction rate were investigated. On the basis of single-factor test, a four-factor and three-level orthogonal test was designed by response surface method to establish a mathematical model between the response value and various factors. The results showed that the intensity of effect of different influencing factor on polyphenols ex- traction rate ranked as microwave power's〉solid to liquid ratio's〉extraction time's〉 ethanol concentration's. The optimum microwave-assisted extraction conditions for polyphenols from L. chinense were as follows: extraction power 254 W, ethanol concentration 60%, extraction time 12.5 rain and solid to liquid ratio 1:17. Under the optimum extraction conditions, the extraction rate of polyphenols from L. chinense was 19.17%. 展开更多

[Method] With ellagic acid yield as an indicator, ultrasound extraction method was adopted to extract the ellagic acid from pomegranate peels, and the concentration of ellagic acid was measured by using ul- traviolet spectrophotometry; L9(34) orthogonal experiment was designed with four fac- tors at three levels, to investigate the effect of extraction temperature, extraction du- ration, solid-liquid ratio and ultrasound power on extraction rate of ellagic acid. [Re- sult] The optimal extraction condition for ellagic acid is extraction temperature of 30 ~C, solid-liquid ratio of 1:200, extraction duration of 20 min, and ultrasound power of 50 W. The relationship between primary and secondary factors affecting experimental indicators was solid-liquid ratio 〉 extraction duration 〉 ultrasound power 〉 extraction temperature. [ Method] The effective ingredients of medicinal plants were extracted by water, ethanol and acetone. The minimum inhibitory concentration (MIC) and antifungal rate of extracts from medicinal plants were determined using plate diluting method. [Resets] The optimum medicinal plants and the extraction methods inhibited two plant pathogens were as follows: clove extracted by ethanol and acetone, and cinnamon extracted by ethanol could be adopted to inhibit Fusarium oxysporum Schl., MIC at that mo- ment reached 4 mg/ml. Clove extracted by ethanol and acetone, and cinnamon extracted by ethanol could be adopted to inhibit the pathogen of Cistanche deserticola, MIC at that moment reached 4 mg/ml. Ultrasonic-assisted extraction (UAE) of American ginseng polysaccharides (AGP) was investigated using response surface methodology. Three-factor-three-level Box-Behnken design was employed to optimize the ultrasonic power, extraction time and ratio of water to raw material to obtain a high AGP yield. The analysis of variance and response surface plots indicated that ultrasonic power was the most important factor affecting the extraction yield. The optimal conditions were ultrasonic power 400 W, extraction time 71 min, and ratio of water to raw material 33 mL g-1. Under these conditions, the yield of AGP was 8.09%, which was agreed closely to the predicted value. Gas chromatography (GC) analysis showed that AGP was composed of arabinose, rhamnose, galactose, glucose, and galacturonic acid. Fourier transform infrared spectra revealed the general characteristic absorption peaks of AGP. In addition, AGP exhibited good immunostimulating activities by up-regulating the production of nitric oxide and cytokines. Compared with hot water extraction, UAE required shorter extraction time and gave a higher extraction yield, without changing the structure and immunostimulating activity of AGP. The results indicated that UAE could be an effective and advisable technique for the large scale production of plant polysaccharides. 展开更多

Objective: To investigate suitable condition for extraction of the active components from Ajuga nipponensis(A. nipponensis). Methods: Orthogonal experimental design was used to determine the optimal extraction parameters for ecdysterones and flavonoids. Finally, the hepatoprotective abilities of A. nipponensis extracts were evaluated by CCl_4-induced animal models. Results:Maximum yields of flavonoids(7.87±0.10) mg/g and ecdysterones(0.73±0.02) mg/g could be obtained when the extraction time was 50 min, the extraction temperature was 60 ℃, and the ratio of sample to 70%(v/v) ethanol was 1:20(w/w). The antioxidant property of A. nipponensis was correlated to the concentration of its extracts. At 5 mg/m L, A. nipponensisextract scavenged 84.8% of DPPH radical and had absorbance values of 2.43±0.04 reducing power. Upon CCl_4-induced liver injury, glutamic oxaloacetic transaminase and glutamic pyruvic transaminase decreased significantly after the mice were treated with A. nipponensis. Histological researches also explained that A. nipponensis reduced the extent of liver lesions induced by CCl_4. Conclusions: A. nipponensis exhibited potent antioxidant activity in chemical experimental models and hepatoprotective effect against CCl_4-induced liver damage. 展开更多

[ Method ] Using rice bran as raw material, effects of ultrasonicassisted extraction technology and different organic solvents, extraction durations, solid-liquid ratios and extraction temperatures on extraction results of rice bran oil were investigated. Based on the results of single-factor experiment, L9 (34 ) orthngonal experiment with three factors at three levels was conducted to optimize the extraction technological parameters, thus obtaining the optimal technological conditions for extracting rice bran oil. [ Result] Anhydrous ethanol led to the best extraction result; after ultrasonic treatment, the extraction rate of rice bran oil from raw material was not enhanced. The optimized technological parameters were : extraction temperature of 80 ℃, extraction duration of 20 rain and solid-liquid ratio of 1:24 ( g: ml). Under the optimized technological conditions, the extraction rate of rice bran oil reached 19.33%. [ Conclusion] This study laid solid foundation for further investigation and development of rice bran oil. 展开更多

The work described here is based on a comparative study of carotenoids and fatty acids extracted from Synechococcus sp. with (1) pure supercritical CO2, (2) CO2 with 5% (v/v) ethanol as cosolvent and (3) ultrasound-assisted extraction using N, N-dimethylformamide (DMF). The effects of extraction conditions on supercritical CO2 extraction with and within cosolvent were analyzed at different temperatures (40℃, 50℃ and 60℃) and pressures (200, 300 and 400 bars). SFE with CO2 proved to be the most selective method for the extraction of β-carotene, but under these conditions the contents of zeaxanthin and fatty acids were only comparable to or lower than those obtained with techniques that use SFE cosolvent. The SFE technique with CO2 and ethanol simultaneously extracted β-carotene and zeaxanthin and not only increased the concentrations of fatty acids obtained, but also helped to remove fatty acids (palmitoleic and linolenic acid) that were not obtained with pure CO2. Comparison of the supercritical technology with the ultrasound-assisted extraction (UAE) shows that the former technique is the most appropriate due to the fact that ethanol is generally regarded as a safe solvent in comparison to DMF. 展开更多

This study was conducted to investigate the effects of cellulase dosage, enzymolysis time, pH and enzymolysis temperature on procyanidin extraction rate by single factor experiment, with tartary buckwheat shell as an experimental material.Main process parameters were optimized to obtain a regression model by response surface methodology. The results of variance analysis indicated that the regression model reflected the relationship between buckwheat shell procyanidin extraction rate with enzyme dosage, enzymolysis time, pH and enzymolysis temperature; and the optimal process parameters were enzyme dosage of 6.5 mg/g, enzymolysis time of 1.5 h, pH at 4.7 and enzymolysis temperature at 46 ℃. Three parallel experiments were conducted under these process parameters. In practice, the highest procyanidin extraction rate was 6.78 g/100 g. The relative error between the predicted value of regression model and the actual value was 1.3%. The regression equation fitted the real situation better. 展开更多

[ Objective] To optimize the ultrasonic-assisted extraction process of chlorogenic acid from sunflower meal. [ Method] Based on the sin-gle-factor test, the extraction factors of ethanol concentration, extraction temperature and extraction time were optimized using the response surface methodology with the extraction rate of chlorogenic acid as the response value. [ Result] The optimum extraction conditions of chlorogenic acid from sunflower meal were 57% of ethanol concentration, 39 min of extraction time, 53 ~C of extraction temperature, 100 W of ultrasonic power, 1 : 14 ( g/ ml) of solid-liquid ratio and one time of extraction. Under these conditions, the extraction rate of chlorogenic acid was 2.25%. [ Conclusion] Ultra-sonic assisted extraction method has high extraction rate but short time, the study provides a theoretical basis for the industrialized production of chlorogenic acid in sunflower meal. 展开更多

[ Methed] Taking Black Tea as materials, the Box-Behnken response surface design method of tea polyphenols ultrasonic-assisted extraction process optimi- zation was applied. The regression optimization model of the ethanol volume fraction, ultrasonic time, and ultrasonic temperature, ethyl acetate extract stalling time and tea polyphenols was established. [ Result] The influence of four factors on the black tea polyphenol solvents affecting size was as follows: ultrasonic time 〉 ethanol volume fraction 〉 stalling extraction time 〉 ultrasound temperature. The best extraction process was ultrasonic time 80 min, volume fraction of 88.99% ethanol, extraction time 89.97 min, ultrasonic temperature of 80℃. At the optimized technical parameters, the black tea phenolic extraction yield can be as high as 73.50%. [Methods]The ultraviolet-visible spectrophotometry was used to determine the content of total flavonoids in Pueraria lobata( Willd.) Ohwi. With the puerarin as index,the reflux extraction and single factor test were employed to investigate the effects of temperature,time,ethanol concentration and solid-liquid ratio on the content of total flavonoids in Pueraria lobata( Willd.) Ohwi,respectively. Under the optimal extraction technology,the content of total flavonoids in Pueraria lobata( Willd.) Ohwi at different altitudes was determined.[Results] The optimum extraction process was as follows: 70%ethanol; solid-liquid ratio of 1∶ 30; 1 h reflux extraction. Under these conditions,the extraction rate of flavonoids in Pueraria lobata( Willd.) Ohwi was 11. 48%,the total flavonoids content of different kudzu parts was in the order of roots > stems > leaves,and the total flavonoids content of the sample at about an altitude of 1000 m was significantly higher than at the altitudes of 1400 m and 1700 m.[Conclusions]It was suggested that the Pueraria lobata( Willd.) Ohwi should not be cultivated as medicinal plant in too high mountains,and the stems and leaves of Pueraria lobata( Willd.) Ohwi could be used as raw materials for extracting total flavonoids. 展开更多

A mathematical model based on Fick's first lawwas established to describe the process of ultrasonic-assisted extraction of arctigenin from acid hydrolyzed FructusArctii.Acid hydrolization with hydrochloric acid promotes the conversion of arctiin to arctigenin in the arctiin-rich active pharmaceutical ingredient, and the hydrolyzed products were further examined to investigate the process setup. By considering the mechanism of the extraction process and experimental data, the effects of parameters including solven to solid ratio, particle size of hydrolyzed samples, ethano volume fraction, ultrasound power, extraction temperature and extraction time on concentration of arctigenin were analyzed in detail. The model was suitable for simulating the process of ultrasonic-assisted extraction of arctigenin. The simulation results of the model agree well with experimental data with the deviation below13%, indicating that the mathematical mode can provide valuable guidance for the extraction of arctigenin from acid hydrolyzed FructusArctii. 展开更多
