The cysteine protease caspase-1(Casp-1)contributes to innate immunity through the assembly of NLRP3,NLRC4,AIM2,and NLRP6 inflammasomes.Here we ask whether caspase-1 activation plays a regulatory role in house dust mite(HDM)-induced experimental allergic airway inflammation.We report enhanced airway inflammation in caspase-1-deficient mice exposed toHDMwith a marked eosinophil recruitment,increased expression of IL-4,IL-5,IL-13,aswell as full-length and bioactive IL-33.Furthermore,mice deficient for NLRP3 failed to control eosinophil influx in the airways and displayed augmented Th2 cytokine and chemokine levels,suggesting that the NLPR3 inflammasome complex controls HDM-induced inflammation.IL-33 neutralization by administration of soluble ST2 receptor inhibited the enhanced allergic inflammation,while administration of recombinant IL-33 during challenge phase enhanced allergic inflammation in caspase-1-deficient mice.Therefore,we show that caspase-1,NLRP3,and ASC,but not NLRC4,contribute to the upregulation of allergic lung inflammation.Moreover,we cannot exclude an effect of caspase-11,because caspase-1-deficient mice are deficient for both caspases.Mechanistically,absence of caspase-1 is associated with increased expression of IL-33,uric acid,and spleen tyrosine kinase(Syk)production.This study highlights acritical role of caspase-1 activation andNLPR3/ASCinflammasomecomplex in the down-modulation of IL-33 in vivo and in vitro,thereby regulating Th2 response in HDM-induced allergic lung inflammation. 展开更多

妊娠高血压综合征患者体内一氧化氮、内皮素-1及血清尿素氮、尿酸水平相关性分析 被引量:3

目的探讨血浆一氧化氮(NO)、内皮素-1(ET-1)及血清尿素氮(BUN)、尿酸(UA)水平与妊娠期高血压综合征相关性。方法选取2017年1月—2018年1月某医院收治的56例妊娠期高血压综合征患者作为研究对象(观察组),同期选取50例住院分娩的健康孕妇... 目的探讨血浆一氧化氮(NO)、内皮素-1(ET-1)及血清尿素氮(BUN)、尿酸(UA)水平与妊娠期高血压综合征相关性。方法选取2017年1月—2018年1月某医院收治的56例妊娠期高血压综合征患者作为研究对象(观察组),同期选取50例住院分娩的健康孕妇作为对照组。采用酶联免疫吸附法进行血浆ET-1检测,硝酸根还原酶法进行NO检测,酶联免疫吸附法进行BUN、UA检测。观察比较两组研究对象血浆NO、ET-1及血清BUN、UA水平具体情况,并观察不同程度妊娠期高血压综合征患者与对照组NO、ET-1、BUN、UA水平的差异。结果观察组NO水平为(17.09±2.04)μmol/L,低于对照组的(28.45±0.78)μmol/L,观察组的ET-1、BUN、UA水平为(61.33±5.05)ng/L,(6.79±1.30)mmol/L,(352.61±76.11)μmol/L,高于对照组的(44.54±6.12)ng/L,(4.17±1.23)mmol/L,(276.14±49.02)μmol/L,差异均具有统计学意义(t=37.019,15.465,10.624,6.068,P值均<0.01);轻度、中度、重度妊娠期高血压综合征患者ET-1、BUN、UA水平较对照组呈梯形上升,NO水平较对照组呈梯形下降,差异均具有统计学意义(P值均<0.01);NO与BUN、UA水平均呈负相关(r=-0.684,P=0.001;r=-0.712,P=0.021),ET-1与BUN、UA水平呈正相关(r=0.804,P=0.003;r=0.635,P=0.005)。结论妊娠期高血压综合征患者病情越重,其血浆ET-1水平越高、NO水平越低。上述指标可作为病情严重程度及疗效监测的指标。 展开更多
