目的 探讨血小板活化因子乙酰水解酶基因突变 (Val2 79-Phe)与急性脑出血的关系。方法 对 10 2例急性脑出血患者、174例原发性高血压患者及 2 2 5例健康对照者 ,分别用多聚酶链式反应分析基因组DNA的突变等位基因 ,用Stafforini等的... 目的 探讨血小板活化因子乙酰水解酶基因突变 (Val2 79-Phe)与急性脑出血的关系。方法 对 10 2例急性脑出血患者、174例原发性高血压患者及 2 2 5例健康对照者 ,分别用多聚酶链式反应分析基因组DNA的突变等位基因 ,用Stafforini等的方法测定血浆PAF乙酰水解酶的活性。结果 脑出血组突变的发生率 (4 7.1% )显著高于对照组 (P <0 .0 1)。高血压组突变的发生率(32 .7% )与对照组 (2 4 .4 % )之间无显著差异 (P >0 .0 5 )。结论 血浆中血小板活化因子乙酰水解酶基因突变所致的酶缺乏可能是急性脑出血的独立危险因素。 展开更多

The norepinephrine transporter(NET) is a member of the Na+/Cl- dependent neurotransmitter transporter family and constitutes the target of several clinically important antidepressants. To delineate the critical amino acid residues and the function of C-terminal in regulating transport activity of NET, here we constructed two site mutants (V70F, F72V; V70I, F72V) and one C-terminal truncated mutant (△611-617). The wild type and mutants of NET were expressed in Xenopus oocytes by injection of their cRNA. We found that all of these mutants lost their transport activity. These results indicate that the amino acid residues of V70 and F72 3 and the last seven amino acids of C-terminal are essential to the transport activity of NET. 展开更多

Study groups were matched to control groups by race and age. The prevalence of these polymorphisms did not differ between patients with RM and controls or between Caucasian and blacks, suggesting that these polymorphisms cannot be considered a risk factor for RM. 展开更多

Lipoprotein-associated phospholipase A2(Lp-PLA2),a member of the phospholipase A2 superfamily,is an enzyme that hydrolyses phospholipids,is found in blood circulation as a sign of inflammation,and takes a role in atherogenesis.There is an epidemiologic relation between increased Lp-PLA2 levels and coronary heart disease.Lp-PLA2 is an enzyme that is produced by macrophages and takes a role as an independent predictor of a coronary event.A genetic variant of Val279Phe on the Lp-PLA2 gene has been reported with various results in Japan,China,Korea,and Caucasian populations.This study aims to analyse the influence of the Val279Phe genetic variant on acute myocardial infarction(AMI)at Saiful Anwar Hospital,Indonesia.This study was conducted on 151 patients(111 AMI patients and 40 non-AMI patients).The genetic variant of Val279Phe was identified through a genotyping method.There were no significant differences in age,total cholesterol level,LDL-C(low-density lipoprotein cholesterol)level,and family history data between AMI and non-AMI patients.However,AMI patients had low HDL-C(high-density lipoprotein cholesterol),triglyceride levels,dyslipidaemia,and hypertension risk factors compared to non-AMI patients.The frequency of the GG genotype(279Val)was dominant in both AMI and non-AMI groups.Further analysis suggested that the GG genotype has a 2.9 times greater risk of AMI compared to the GT/TT genotype(279Phe).This study concluded that the Val279Phe genetic variant undoubtedly influenced AMI risk,which is a warrant for further development of early detection and improving strategy to prevent AMI in patients. 展开更多

Summary: Integrins such as αvβ3, α5β31 play a key role in angiogenesis regulation, invasion and metastasis, inflammation, wound healing, etc. The up-regulation of integrin αvβ3 after cerebral ischemic stroke can promote angiogenesis, which in turn improves functional recovery. In addition, the integrin αvβ3 inhibitor can block the blood-brain barrier (BBB) leakage induced by vascular endothelial growth factor (VEGF) and also can reduce inflammatory reaction, decrease the deposition of fibrinogen. Other studies showed that integrin αvβ3 is not essential in revascularization. Therefore, the effect of integrin αvβ3 in the whole process of brain function recovery merits further study. 展开更多
