The results showed an obvious increase in soluble protein content. Compared with control, Chl a, Chl b, total Chl contents and the ratio of Chl a to Chl b all decreased to some extent. ATP content increased in some varieties and decreased in others, but the stronger the drought resistance of the variety , the higher the ATP content. The correlation coefficient(r)of the soluble protein content, ratio of Chl a to Chl b and ATP content as a percentage of the drought-resistant sweet potato control variety are 0. 8968, - 0. 8509 and 0. 8200, respectively, P<0. 01. The water-soluble molecules with low molecular weights are the major determinants of the palatability of diets. The present study was conducted to investigate the palatability of water-soluble extracts from single protein source(single extract pellets) and the mixture of these extracts with different proportions(blended extract pellets) in juvenile turbot(Scophthalmus maximus). Then according to the palatability of blended extract pellets, an optimal mixture proportion was selected, and a new protein source made from raw protein materials with the selected proportion was formulated to replace fishmeal. Summarily, the palatability of single extract pellets for turbot was descendent from fishmeal to pet-food grade poultry by-product meal, wheat gluten meal, soybean meal, peanut meal, meat and bone meal, and corn gluten meal. Subsequently, according to the palatability of single extract pellets, 52 kinds of blended extract pellets were designed to test their palatability. The results showed that the pellets presented remarkably different palatability, and the optimal one was diet 52(wheat gluten meal: pet-food grade poultry by-product meal: meat and bone meal: corn gluten meal = 1:6:1:2). The highest ingestion ratio(the number of pellets ingested/the number of pellets fed) was 0.73 ± 0.03, which was observed in Diet 52. Then five isonitrogenous(52% crude protein) and isocaloric(20 k J g^(-1) gross energy) diets were formulated by replacing 0(control), 35%, 50%, 65% and 80% of fishmeal with No.52 blending proportion. After a 10-weeks feeding trial, a consistent feed intake was found among all replacement treatments. Replacement level of fishmeal up to 35% did not significantly influence final body weight, specific growth rate, feed efficiency ratio, and protein efficiency ratio of turbot. Therefore, the water-soluble extracts of protein sources play an important role in improving the palatability of non-fishmeal protein sources in aquafeed.展开更多

We analyzed the amino acid residues present in the water-soluble and transmembrane proteins of 6 thermophilic and 6 mesophilic species of the domains Archaea and Eubacteria, and characterized them as favorable or unfa...We analyzed the amino acid residues present in the water-soluble and transmembrane proteins of 6 thermophilic and 6 mesophilic species of the domains Archaea and Eubacteria, and characterized them as favorable or unfavorable. The characterization was performed by comparing the observed number of each amino acid residue to the expected number calculated from the percentage of nucleotides present in each gene. Amino acids that were more or less abundant than expected were considered as favorable or unfavorable, respectively. Comparisons of amino acid compositions indicated that the water-soluble proteins were rich in charged residues such as Glu, Asp, Lys, and His, whereas hydrophobic residues such as Trp, Phe, and Leu were abundant in transmembrane proteins. Interestingly, our results found that although the Trp residue was abundant in transmembrane proteins, it was not defined as favorable by our calculations, indicating that increased numbers of a particular amino acid does not necessary indicate it is a favorable residue. Amino acids with high G + C content such as Ala, Gly, and Pro were frequently observed as favorable in species with low G + C content. Comparatively, amino acids with low G + C content such as Phe, Tyr, Lys, Ile, and Met were frequently observed as favorable in species with high G + C content. These are the examples to increase the supply of amino acids than expected. Amino acids with neutral G + C content, i.e., Glu and Asp were favorable in water-soluble proteins from all species analyzed, and Cys was unfavorable both in water-soluble and transmembrane proteins. These results indicate that amino acid compositions are essentially determined by the nucleotide sequence of the genes, and the amino acid content is altered by a deviation from expectation.展开更多

Activities of sucrose synthase, sucrose phosphate synthase and acid invertase increased till flowering stage in leaves and mid-milky stage(14 d after flowering) in grains and thereafter declined in concomitant with the contents of reducing sugar. Under aerobic conditions, the activities of acid invertase and sucrose synthase(cleavage) significantly decreased in conjunction with the decrease in non-reducing sugars and starch content in all the varieties. Disruption of starch biosynthesis under the influence of aerobic conditions in both leaves and grains and the higher build up of sugars possibly resulted in their favoured utilization in nitrogen metabolism. Feng Ai Zan, PR115 and PR120 maintained higher levels of sucrose synthase enzymes in grains and leaves and contents of metabolites(amino acid, protein and non-reducing sugar) under aerobic conditions, while PR116, Punjab Mehak 1 and PAU201 performed better under transplanting conditions, thus showing their adaptation to environmental stress. Yield gap between aerobic and transplanting rice is attributed primarily to the difference in sink activity and strength. Overall, it appear that up-regulation of sucrose synthase(synthesis) and sucrose phosphate synthase under aerobic conditions might be responsible in enhancing growth and productivity of rice varieties.展开更多

The genes encoding water soluble proteins w...The occurrence frequencies of the dinucleotides of genes of three thermophilic and three mesophilic species from both archaea and eubacteria were investigated in this study. The genes encoding water soluble proteins were rich in the dinucleotides of purine dimers, whereas the genes encoding membrane proteins were rich in pyrimidine dimers. The dinucleotides of purine dimers are the counterparts of pyrimidine dimers in a double-stranded DNA. The purine/pyrimidine dimers were favored in the thermophiles but not in the mesophiles, based on comparisons of observed and expected frequencies. This finding is in agreement with our previous study which showed that purine/pyrimidine dimers are positive factors that increase the thermal stability of DNA. The dinucleotides AA, AG, and GA are components of the codons of charged residues of Glu, Asp, Lys, and Arg, and the dinucleotides TT, CT, and TC are components of the codons of hydrophobic residues of Leu, Ile, and Phe. This is consistent with the suitabilities of the different amino acid residues for water soluble and membrane proteins. Our analysis provides a picture of how thermophilic species produce water soluble and membrane proteins with distinctive characters: the genes encoding water soluble proteins use DNA sequences rich in purine dimers, and the genes encoding membrane proteins use DNA sequences rich in pyrimidine dimers on the opposite strand.展开更多

The aim of the present work is to investigate the potential utility of water-soluble chitosan (WSC) as vehicles to load and deliver proteins. WSC nanoparticles (WSC NP) with various formations were prepared based on ionic gelation of WSC with pentasodium tripolyphosphate (TPP) anions. Bovine serum albumin (BSA) was used as a model protein drug incorporated into the WSC nanoparticles. Blank and BSA-loaded WSC nanoparticles were examined and determined to have a spherical shape with diameters between 35―190 nm, and zeta potential between 35―42 mV. FTIR confirmed that the tripolyphosphoric groups of TPP linked to the ammonium groups of WSC in the nanoparticles. Some factors affecting delivery properties of BSA have been investigated. Altering the concentration of BSA from 0.05 to 1 mg/mL enhanced the loading capacity of BSA but decreased loading efficiency simultaneously. Also, with the introduction of poly ethylene glycol (PEG), BSA release accelerated. Nanoparticle preparation from WSC with various deacetylation degrees (DDs) from 72.6% to 90% and MWs ranging from 3.5 to 15.8 kDa promoted loading efficiency and decreased the release rate. These results indicate that WSC nanoparticles are promising carriers for protein delivery.展开更多
To understand the transfer process of soluble reactive phosphorus (SRP) on the lake sediment-water interface in a mesotrophic shallow lake in South China, the SRP concentrations and the oxidation-reduction potential...To understand the transfer process of soluble reactive phosphorus (SRP) on the lake sediment-water interface in a mesotrophic shallow lake in South China, the SRP concentrations and the oxidation-reduction potential (ORP) across the sediment-water interfaces were continually monitored. Sediment samples were collected from Xinghu Lake in Guangdong Province. The ORP dynamics at different layers of overlying water was similar for six experimental systems, whereas those in porewater were significantly different. The ORP in overlying water was 200-300 mV higher than those in sediments. The oxygen penetration depth ranged from 2 to 10 mm in Xiannu Lake sediments. The variation amplitudes of ORP increased with sediment depth, but the mean ORP values were all about 218 mV. The SRP concentrations in porewater maintained at a low level of about 0.049 mg/L because of high atom ratio of total iron and total manganese to total phosphorus. The SRP concentrations and variation amplitudes in porewater increased with sediment depth. The SRP in overlying water mainly originated from S RP transference of the porewater of middle and bottom sediments (3-15 cm). The ORP variation and SRP transfer in porewater played important roles in changing SRP concentrations. A distinct SRP concentration gradient appeared in overlying water when intense exchange occurred at the sediment-water interface; therefore, it was necessary to monitor the SRP concentration profiles to accurately estimate the internal loading.展开更多

This is consistent with the suitabilities of the different amino acid residues for water soluble and membrane proteins. Our analysis provides a picture of how thermophilic species produce water soluble and membrane proteins with distinctive characters: the genes encoding water soluble proteins use DNA sequences rich in purine dimers, and the genes encoding membrane proteins use DNA sequences rich in pyrimidine dimers on the opposite strand.
文摘High MW chitosan (CS) solutions have already been proposed as vehicles for protein delivery. The aim of the present work is to investigate the potential utility of water-soluble chitosan (WSC) as vehicles to load and deliver proteins. WSC nanoparticles (WSC NP) with various formations were prepared based on ionic gelation of WSC with pentasodium tripolyphosphate (TPP) anions. Bovine serum albumin (BSA) was used as a model protein drug incorporated into the WSC nanoparticles. Blank and BSA-loaded WSC nanoparticles were examined and determined to have a spherical shape with diameters between 35―190 nm, and zeta potential between 35―42 mV. FTIR confirmed that the tripolyphosphoric groups of TPP linked to the ammonium groups of WSC in the nanoparticles. Some factors affecting delivery properties of BSA have been investigated. Altering the concentration of BSA from 0.05 to 1 mg/mL enhanced the loading capacity of BSA but decreased loading efficiency simultaneously. Also, with the introduction of poly ethylene glycol (PEG), BSA release accelerated. Nanoparticle preparation from WSC with various deacetylation degrees (DDs) from 72.6% to 90% and MWs ranging from 3.5 to 15.8 kDa promoted loading efficiency and decreased the release rate. These results indicate that WSC nanoparticles are promising carriers for protein delivery.

文摘To understand the transfer process of soluble reactive phosphorus (SRP) on the lake sediment-water interface in a mesotrophic shallow lake in South China, the SRP concentrations and the oxidation-reduction potential (ORP) across the sediment-water interfaces were continually monitored. Sediment samples were collected from Xinghu Lake in Guangdong Province. The ORP dynamics at different layers of overlying water was similar for six experimental systems, whereas those in porewater were significantly different. The ORP in overlying water was 200-300 mV higher than those in sediments. The oxygen penetration depth ranged from 2 to 10 mm in Xiannu Lake sediments. The variation amplitudes of ORP increased with sediment depth, but the mean ORP values were all about 218 mV. The SRP concentrations in porewater maintained at a low level of about 0.049 mg/L because of high atom ratio of total iron and total manganese to total phosphorus. The SRP concentrations and variation amplitudes in porewater increased with sediment depth. The SRP in overlying water mainly originated from S RP transference of the porewater of middle and bottom sediments (3-15 cm). The ORP variation and SRP transfer in porewater played important roles in changing SRP concentrations. A distinct SRP concentration gradient appeared in overlying water when intense exchange occurred at the sediment-water interface; therefore, it was necessary to monitor the SRP concentration profiles to accurately estimate the internal loading.
