Wuji Town,which is located by the east of Mt.Hengshan(one of the Five Mountains in China),and the side of Mishui River(a principal east-west water transport in ancient central and southern regions of Hunan),once was an important commercial town and a political,economic and cultural center in the lower and middle reaches of Mishui River due to the particular historical opportunities.Its tourist resources are characterized by "integrated natural scenery and ancient town,highly concentrated scenic spots,prosperous business in the history,row upon row of stores and workshops,numerous temples and ancestral halls,and various festivals".It was proposed that the protection and rural tourism development of Wuji Town should be based on consciousness raising,unification of concepts,scientific planning,application of integrated protection means,and development of regional clusters on the basis of famous mountain and city. 展开更多

Theory and mechanism analysis of Wuji Pill in Anti-lung Cancer:A study based on network pharmacology and molecular docking technology

Background:To explore the effective components and mechanism of Classic ancient prescription Wuji pill on lung cancer(LC)with the help of network pharmacology and molecular docking technology.Methods:TCMSP database,Chinese Pharmacopoeia(2020 Edition)and related literature were searched to identify the main chemical composition and targets of constituent drugs in Wuji pill,including Houpo(Magnoliae Officinalis Cortex),Huangqin(Scutellariae Radix),Badou(Crotonis Fructus),Huanglian(Coptidis Rhizoma)and Renshen(Ginseng Radix et Rhizoma).Meanwhile,Genecards,OMIM,TTD,Drugbank and PharmGkb databases were used to obtain the main targets of LC.The"chemical compound-target"network was constructed by using Cytoscape 3.8.2 software with the disease drug intersection gene as the research target.The PPI network was constructed by using the string database for protein interaction analysis,and the core target genes were sorted out.The Gene ontology go function enrichment analysis and KEGG signal pathway enrichment analysis were carried out based on the ClusterProfiler of R software.Finally,the Autodock Vina software was used to conduct molecular docking between the core target with high value and the key components.Results:A total of 60 active components,190 corresponding targets and 2516 GO and 163 KEGG pathways of Wuji pill were screened out,including 179 potential targets of LC.The core targets were ESR1,CDKN1A,TP53,NR3C1,MAPK1,AKT1,MAPK8,JUN,RELA,the core components were quercetin,kaempferol,β-sitosterol,stigmasterol,tetrahedron,protoopioid,etc,and the core pathways were PI3K-Akt,MAPK,IL-17,etc.Conclusion:This study preliminarily revealed the mechanism of action of Wuji pill on multi component-multi target-multi pathway of LC,which provided reference for the further clinical development and research of Wuji pill. 展开更多

Elucidating the mechanism of Wuji powder in polycystic ovary syndrome treatment:a network pharmacology approach

To elucidate the potential mechanism of Wuji powder in treating polycystic ovary syndrome(PCOS),this study utilized TCMSP database to screen the active ingredients of each constituent drug in Wuji powder.Subsequently,we predicted the target proteins associated with these active ingredients.In parallel,we employed the GeneCards database to identify genes related to PCOS and determined the intersection of drug targets and disease targets using the online tool available at http://bioinformatics.psb.ugent.be/webtools/Venn/.The shared genes were considered as the target proteins of Wuji powder in the treatment of PCOS.Utilizing the String website,we constructed a protein-protein interaction(PPI)network diagram and identified key protein modules and hub genes within the PPI network using Cytoscape 3.7.1 software.Further analysis in the DAVID database involved GO and KEGG pathway enrichment analyses of the genes within the identified key modules.Our investigation revealed a total of 63 active ingredients in Wuji powder with potential therapeutic effects on PCOS,corresponding to 266 drug targets.Intersection with PCOS-related disease targets yielded 174 shared targets.Ten key modules and ten hub genes(TNF,MMP9,AKT1,ECG,VEGFA,PTGS2,IL-6,MAPK3,STAT3,and CXCL8)were identified through network analysis.KEGG pathway enrichment analysis uncovered 58 signaling pathways,including TNF,MAPK,and PI3K-Akt signaling pathways.GO functional annotation delineated five cellular components(CC),nine molecular functions(MF),and 58 biological processes(BP).Noteworthy findings included extracellular space,enzyme binding,and drug response among CC,MF,and BP categories,respectively.These results collectively suggested that Wuji powder might exert its therapeutic effects on PCOS by modulating the TNF/PI3K/AKT signaling pathway. 展开更多

Blood deficiency syndrome(BDS)refers to a pathological state with blood dysfunction and organ dystrophy in traditional Chinese medicine.Danggui Wuji granules(DWG)was developed from a formula containing Angelicae Sinensis Radix and Musculus et Os Galli Domestici.Herein,we investigated the mechanism of DWG in treating BDS by modulating gut microbiota.We found that DWG protected mice from BDS by elevating the levels of red blood cell count,hemoglobin,and hematocrit in peripheral blood and increasing the erythrocyte membrane Na+-K+-ATPase activity.Danggui Wuji granules changed the composition and metabolites of colonic flora.Notably,Lactobacillus,Muribaculaceae,and Alistipes were the main genera showing changes after DWG treatment.Our findings revealed that DWG presented a positive therapeutic effect on BDS in mice by regulating the gut microbiota and metabolites.The protective mechanism of DWG was associated with pathways such as metabolic pathways,biosynthesis of secondary metabolites,ABC transporters,ribosome,thyroid hormone synthesis,lysine degradation,galactose metabolism,tyrosine metabolism,etc. 展开更多

A qualitative and quantitative test method of fingerprint of Wuji gastric floating sustained-release tablets was established. High performance liquid chromatography (HPLC) was adopted, using Agilent ZORBAX SB-C18 column (250 mm×4.6 mm, 5 μm) as the chromatographic column, and acetonitrile-0.05 mol/L potassium dihydrogen phosphate solution as the mobile phase in a gradient elution with the flow rate of 1.0 mL/min. Sample solution (10 μL) was injected and was tested at the wavelength of 225 nm for 75 min at the column temperature of 30 ℃, Fingerprint similarity software (2004A version) was used to conduct data analysis. A total of 11 batches of Wuji gastric floating sustained-release tablets were tested and analyzed with HPLC fingerprint. Seventeen common peaks were found and the similarity of the 11 batches of agents was greater than 0.9, indicating that the production process of the agent is stable and feasible. The method is operable and could effectively control the quality of Wuji gastric floating sustained-release tablets. 展开更多
