AIM To investigate gut microbial diversity and the interventional effect of Xiaoyaosan(XYS) in a rat model of functional dyspepsia(FD) with liver depression-spleen deficiency syndrome. METHODS The FD with liver depression-spleen deficiency syndrome rat model was established through classic chronic mild unpredictable stimulation every day. XYS group rats received XYS 1 h before the stimulation. The models were assessed by parameters including state ofthe rat, weight, sucrose test result and open-field test result. After 3 wk, the stools of rats were collected and genomic DNA was extracted. PCR products of the V4 region of 16 S rD NA were sequenced using a barcoded Illumina paired-end sequencing technique. The primary composition of the microbiome in the stool samples was determined and analyzed by cluster analysis.RESULTS Rat models were successfully established, per data from rat state, weight and open-field test. The microbiomes contained 20 phyla from all samples. Firmicutes, Bacteroidetes, Proteobacteria, Cyanobacteria and Tenericutes were the most abundant taxonomic groups. The relative abundance of Firmicutes, Proteobacteria and Cyanobacteria in the model group was higher than that in the normal group. On the contrary, the relative abundance of Bacteroidetes in the model group was lower than that in the normal group. Upon XYS treatment, the relative abundance of all dysregulated phyla was restored to levels similar to those observed in the normal group. Abundance clustering heat map of phyla corroborated the taxonomic distribution. CONCLUSION The microbiome relative abundance of FD rats with liver depression-spleen deficiency syndrome was significantly different from the normal cohort. XYS intervention may effectively adjust the gut dysbacteriosis in FD. 展开更多

Objective:To observe changes in the molecular expression of the NLR Family Pyrin Domain Containing Protein 3(NLRP3)pathway in depressed rats after treatment with Xiaoyaosan,and identify the regulatory mechanism of this compound.Methods:Male SpragueeD awley rats were randomly divided into five groups with 12 rats in each group,including the control group,model group,Fluoxetine group,Xiaoyaosan group,and MCC950 group.A depression model was generated by chronic immobilization stress(induced by 3 h of restraint immobilization every day),and the drugs were administered at the same time in each group for 21 days.The effects of Xiaoyaosan on behavioral changes of depressed rats were observed through macroscopic characterization,body mass,open field experiments,and a sucrose preference test.The m RNA and protein expression of the NLRP3 signaling pathway was examined by fluorescence real-time quantitative PCR and Western blot assays.Results:The Xiaoyaosan group,Fluoxetine group,and MCC950 group rats showed improved depressive behavior and an increased weight of sucrose water consumption.The protein and mRNA expression levels of NLRP3,Caspase-1,and IL-1 b were also decreased in the Fluoxetine,Xiaoyaosan,and MCC950 groups.Conclusion:NLRP3,Caspase-1,and IL-1 b protein and mRNA expression levels were increased in the cortex of depressed rats,while Xiaoyaosan protected cortical tissue in these rats by decreasing NLRP3,Caspase-1,and IL-1 b protein and mRNA expression. 展开更多

OBJECTIVE Depression is one of the prevalent and prominent complex psychiatric diseases and the number of depressed patients has been on the rise globally during the recent decades.Xiaoyaosan,as a famous Chinese herbal formula,has been widely used in depression patients for a long time. However,the therapeutic mechanisms remain uncertain because of difficulty of depression pathophysiology and the lack of bioinformatic approach to understand the molecular connection. METHODS In this thesis,we applied a network pharmacology approach to explain the potential mechanisms between Xiaoyaosan and depression involved in oral bioavailability screening,drug-likeness assessment,caco-2 permeability,blood-brain barrier target recognition and network analysis. RESULTS 66 active compounds in Xiaoyaosan formula with favorable pharmacokinetic profiles are predicted as active compounds for anti-depression treatment. Network analyses show that these 66 compounds target 40depression-associated proteins including especially HTR2A,NR 3C1,MAOB,XDH and CNR2. These proteins are mainly involved in the neuroactive ligand-receptor interaction,serotonergic synapse,cAMP signaling pathways and calcium signaling pathways. CONCLUSION The integrated network pharmacology method is an effective approach to illustrate the anti-depression mechanisms of herbs,and this in silico approach can be applied in drug discovery. 展开更多

The anti-depressant effects of Xiaoyaosan (Free and Easy Wanderer's Powder) may inhibit apoptosis by down regulating the [Ca^2+]i overload. PC12 cells were containing with corticosterone to simulate neuronal injury, dosed with Xiaoyaosan-containing serum, and the following parameters were measured: apoptosis,[Ca^2+]i overload, and changes in mitochondrial membrane potential (△ψm) and bioenergetics. High-dose Xiaoyaosan-containing serum (the dose of intragastric administration was 2.633 g herb/mL) not only inhibited morphological injury and cell apoptosis, but also suppressed [Ca^2+]i overload induced by corticosterone. Xiaoyaosan also blocked the decrease in mitochondrial membrane potential and adenosine triphosphate levels induced by corticosterone. Xiaoyaosan dose-dependently suppresses corticosterone-induced apoptosis, potentially via inhibiting [Ca^2+]ioverload, and prevents impaired mitochondrial bioenergetics by maintaining mitochondrial membrane potential and adenosine triphosphate production, providing a putative mechanism for its antidepressant effects. 展开更多

In order to explain the antidepressive effect of Xiaoyaosan, all of the recent reports were sought out on the current situation of the antidepressive monomer compositions of Xi- aoyaosan and their mechanism. By exploring the relationship between the effects and the active ingredients, the mechanism and the active ingredients, the antidepressive active ingredients such as quercetin, isorhamnetin, isoliquiritigenin, saikosaponinA, saikosaponinD, glycyrrhiz- kaempferol, liquiritin, isoliquiritin, liquiritigenin, inate, paeoniflorin, albiflorin, ferulic acid, curcumin and their mechanism of antidepressive such as regulating the monoamine neurotransmitter systems, regulating the neuroendocrine system, affecting the neural plasticity and neu- affecting the cytokines level, anti-oxidative stress were rotrophic, affecting the cellular and molecular mechanisms , summarized. By the overview in the level of monomer rather than formulae, we uncovered the antidepressive mech- anism of Xiaoyaosan and further confirmed that Xiaoyaosan, as a classic antidepressive Chinese prescription, had many characteristics, such as multi-level , multi-channel, multi-target and so on . 展开更多

Aim The study focused on identification and antidepression active verification of constituents from An- gelica Sinensis Radix contrubuting to Xiaoyaosan based on UPLC-PDA guided isolation technique. Methods The UPLC-PDA chromatogram of Xiaoyaosan was compared with that of Angelica Sinensis Radix. The relative retention time of each peak and the Uhraviolet spectra providing by PDA were used in the analyses. Constituents were isola- ted from Angelica Sinensis Radix under the guidance of UPLC-PDA investigation. Structures of the isolates were elu- cidated by NMR techniques. Anti-depression effect was evaluated on glutamate-induced neurons. Results Five marker peaks of Xiaoyaosan fingerprint were belong to Angelica Sinensis Radix, and they were determined as conife- ryl ferulate( I), E-butylidenephthalide ( II), ligustilide (III), Z-butylidenephthalide ( IV ), 14-Acetoxy-12-sene- cioyloxytetradeca-2E,8E,10E-trien-4,6-diyn-l-ol(V). Compound V was isolated from the plants of Umbelliferae for the first time. Treatment with compound I, III, IV can protect PC12 and SH-SY5Y cell from glutamate-induced cytotoxicity. Antidepression bioactivity of compound I was first investigated. Conclusion UPLC-PDA guided iso-lation technique was confirmed to be a rapid and accurate method to identify the main active constituents from An- gelica Sinensis Radix contrubuting to Xiaoyaosan. 展开更多

The hippocampus is one of the most commonly studied brain regions in the context of depression.The volume of the hippocampus is significantly reduced in patients with depression,which severely disrupts hippocampal neuroplasticity.However,antidepressant therapies that target hippocampal neuroplasticity have not been identified as yet.Chinese medicine(CM)can slow the progression of depression,potentially by modulating hippocampal neuroplasticity.Xiaoyaosan(XYS)is a CM formula that has been clinically used for the treatment of depression.It is known to protect Gan(Liver)and Pi(Spleen)function,and may exert its antidepressant effects by regulating hippocampal neuroplasticity.In this review,we have summarized the association between depression and aberrant hippocampal neuroplasticity.Furthermore,we have discussed the researches published in the last 30 years on the effects of XYS on hippocampal neuroplasticity in order to elucidate the possible mechanisms underlying its therapeutic action against depression.The results of this review can aid future research on XYS for the treatment of depression. 展开更多

目的基于TGF-β1/CD147信号探讨慢性束缚应激促小鼠乳腺癌进展及逍遥散调节机制。方法40只BABL/c小鼠随机分为移植瘤组(Tumor)、模型组(Model)、逍遥散组(Xiaoyaosan)和米非司酮组(Mifepristone),将4T1细胞株接种于各组小鼠腋下,待成瘤... 目的基于TGF-β1/CD147信号探讨慢性束缚应激促小鼠乳腺癌进展及逍遥散调节机制。方法40只BABL/c小鼠随机分为移植瘤组(Tumor)、模型组(Model)、逍遥散组(Xiaoyaosan)和米非司酮组(Mifepristone),将4T1细胞株接种于各组小鼠腋下,待成瘤后,除Tumor组外其余各组小鼠均进行慢性束缚应激21天,同时Xiaoyaosan组和Mifepristone组小鼠给予相对应的药物灌胃,Tumor组和Model组小鼠灌胃生理盐水。造模结束后,小鼠麻醉断头处死,测量小鼠瘤体重量和体积、内脏指数;采用ELISA方法检测各组小鼠血清肿瘤标志物糖类抗原ca199(Carbohydrate antigen199,CA199)、癌胚抗原(Carcino-embryonic antigen,CEA)、血管内皮生长因子(Vascular endothelial growth factor,VEGF)的含量,血清多巴胺(Dopamine,DA)和皮质酮(Corticosterone,CORT)的含量,以及肿瘤组织转化生长因子β1(Transforming growth factor-β1,TGF-β1)和白介素10(Interleukin 10,IL-10)的含量。采用免疫组化和Western blot方法检测各组小鼠肿瘤组织巨噬细胞极化标志物诱导型一氧化氮合酶(Inducible nitric oxide synthase,iNOS)和精氨酸酶1(Arginase-1,Arg-1)的表达,以及肿瘤组织中细胞外基质金属蛋白酶诱导剂(Extracellular matrix metalloproteinase inducer,EMMPRIN,CD147)及其下游信号分子基质金属蛋白酶2(Matrix metalloproteinases 2),MMP2、基质金属蛋白酶9(MMP9)和VEGF的表达。结果与Tumor组比较,Model组小鼠肿瘤重量和体积,血清CA199、CEA、VEGF、CORT含量,肿瘤TGF-β1和IL-10含量均显著增加;内脏指数和血清DA含量显著减少;肿瘤巨噬细胞M2型极化标志物Arg-1的表达显著增加,M1型极化标志物iNOS的表达显著降低;肿瘤CD147及其下游信号分子MMP2、MMP9和VEGF蛋白表达显著增加,逍遥散和米非司酮均可有效逆转以上改变。结论慢性束缚应激促小鼠乳腺癌进展的机制与肿瘤相关巨噬细胞M2型极化释放TGF-β1增加、激活CD147及其下游相关信号有关,而逍遥散可缓解应激条件下皮质酮增高引起的巨噬细胞M2型极化,减少TGF-β1的生成,抑制CD147及其下游信号,从而抑制慢性应激引起的小鼠乳腺癌进展。 展开更多

逍遥散对胃癌荷瘤共病抑郁小鼠程序性死亡受体1抑制剂治疗的增敏作用及机制探讨

目的 观察逍遥散对胃癌荷瘤共病抑郁小鼠程序性死亡受体1(programmed cell death protein 1,PD-1)抑制剂治疗的增敏作用,并探讨其作用机制。方法 采用皮下移植胃癌细胞系——MCF细胞构建荷瘤小鼠共60只,随机分为荷瘤对照组、荷瘤共病抑... 目的 观察逍遥散对胃癌荷瘤共病抑郁小鼠程序性死亡受体1(programmed cell death protein 1,PD-1)抑制剂治疗的增敏作用,并探讨其作用机制。方法 采用皮下移植胃癌细胞系——MCF细胞构建荷瘤小鼠共60只,随机分为荷瘤对照组、荷瘤共病抑郁组、PD-1抑制剂组、逍遥散联合PD-1抑制剂组,每组15只。荷瘤对照组不做任何干预,共饲养56天;荷瘤共病抑郁组是在荷瘤对照组的基础上每天以慢性不可预知温和应激干预,每天行适量生理盐水灌胃,分别在第1天、15天、29天、43天小鼠尾静脉注射适量生理盐水;PD-1抑制剂组是在荷瘤共病抑郁组的基础上,将小鼠尾静脉注射生理盐水改为PD-1抑制剂;逍遥散联合PD-1抑制剂组是在PD-1抑制剂组的基础上,将生理盐水灌胃改为逍遥散水提物灌胃。分组后第57天,荷瘤对照组、荷瘤共病抑郁组进行糖水偏好测试、陌生环境摄食实验,分析两组小鼠的行为学变化。计算荷瘤共病抑郁组、PD-1抑制剂组、逍遥散联合PD-1抑制剂组小鼠的生存率并绘制生存曲线;获取小鼠瘤体,并测量瘤体体积及重量,计算抑瘤率;采用ELISA法检测小鼠肿瘤组织吲哚胺-2,3-双加氧酶(indoleamine-2,3-dioxygenase, IDO)、犬尿氨酸(kynurenine, Kyn)、芳香烃受体(aryl hydrocarbon receptor, AhR)的数值;对肿瘤组织通过免疫组化法检测小鼠肿瘤组织叉头样转录因子3(forkhead box P3,Foxp3)表达情况。结果 (1)行为学改变:与荷瘤对照组比较,荷瘤共病抑郁组的糖水偏好率显著降低(P<0.01),摄食潜伏时间显著延长(P<0.01)。(2)生存率差异:荷瘤共病抑郁组小鼠生存率为20%,PD-1抑制剂组小鼠生存率为26.7%,逍遥散联合PD-1抑制剂组小鼠生存率为40%。(3)肿瘤增值差异:逍遥散联合PD-1抑制剂组瘤体体积小于PD-1抑制剂组(P<0.05),PD-1抑制剂组小于荷瘤共病抑郁组(P<0.05);逍遥散联合PD-1抑制剂组瘤体重量显著小于PD-1抑制剂组(P<0.01),PD-1抑制剂组显著小于荷瘤共病抑郁组(P<0.01);逍遥散联合PD-1抑制剂组的抑瘤率为35.4%优于PD-1抑制剂组的22.1%。(4)相关蛋白表达差异:逍遥散联合PD-1抑制剂组肿瘤组织中IDO的表达显著低于PD-1抑制剂组及荷瘤共病抑郁组(P<0.01);逍遥散联合PD-1抑制剂组肿瘤组织中Kyn的表达显著低于PD-1抑制剂组(P<0.01),PD-1抑制剂组小于荷瘤共病抑郁组(P<0.05);逍遥散联合PD-1抑制剂组肿瘤组织中AhR的表达显著低于PD-1抑制剂组及荷瘤共病抑郁组(P<0.01);(5)各组小鼠肿瘤组织Foxp3表达情况:与荷瘤共病抑郁组比较,PD-1抑制剂组、逍遥散联合PD-1抑制剂组肿瘤组织中Foxp3蛋白表达均显著降低(P<0.01);与PD-1抑制剂组比较,逍遥散联合PD-1抑制剂组肿瘤组织中Foxp3蛋白表达显著降低(P<0.01)。结论 逍遥散对PD-1抑制剂的增敏作用可能与通过下调IDO、Kyn、AhR水平,进而有效降低调节性T淋巴细胞的增值与分化而实现的。 展开更多

Depression is one of the most prevalent and serious mental disorders with a significant burden of disease.Xiaoyaosan(XYS),a well-known Chinese formula,has been widely used in the treatment of depression.Both clinical studies and animal experiments have indicated that XYS has an obvious antidepressant activity.How to select objective pharmacodynamic markers for traditional Chinese medicine(TCM)treatments based on clinical metabolites,linking pathogenic genes,and drug targets,is a bottleneck problem in the modernization of TCM.To address this issue,we sorted out clinical metabonomics experiments of XYS in treating depression,constructed a metabolic profile of therapeutic biomarkers,and deduced metabolic biomarker-protein interactions networks.The therapeutic biomarkers found for XYS were involved in neurotransmitter synthesis,energy metabolism,and gut microbial metabolism.This study aims to provide a scientific basis for the clinical diagnosis of depression and evaluate the efficacy of XYS in its treatment. 展开更多

Objective: Depression is one of the prevalent and prominent complex psychiatric diseases, and the number of depressed patients has been on the rise globally during the recent decades.Xiaoyaosan, as a famous Chinese herbal formula, has been widely used in depression patients for a long time.However, the therapeutic mechanisms remain uncertain because of the difficulty of depression pathophysiology and the lack of bioinformatic approach to understand the molecular connection.Materials and Methods: In this thesis, we applied a network pharmacology approach to explain the potential mechanisms between Xiaoyaosan and depression involved in oral bioavailability screening, drug-likeness assessment, caco-2 permeability, blood–brain barrier target recognition, and network analysis.Results: Sixty-six active compounds in Xiaoyaosan formula with favorable pharmacokinetic profiles are predicted as active compounds for antidepression treatment.Network analyses showed that these 66 compounds target 40 depression-associated proteins including especially HTR2A, NR3C1, monoamine oxidase inhibitor B, XDH, and CNR2.These proteins are mainly involved in the neuroactive ligand–receptor interaction, serotonergic synapse, cAMP signaling pathways, and calcium signaling pathways.Conclusion: The integrated network pharmacology method can provide a new approach for understanding the pathogenesis of depression and uncovering the molecular mechanisms of Xiaoyansan, which will also facilitate the application of traditional Chinese herbs in modern medicine. 展开更多

逍遥散对皮质酮诱导的PC12损伤模型保护作用及神经保护成分分析验证研究

目的:通过探究逍遥散的神经保护作用及其化学组成,发现逍遥散中可能发挥神经保护作用的有效活性成分。方法:采用水提法提取逍遥散,通过高分辨质谱-液相色谱质谱联用仪分析提取液的化学成分;制备逍遥散含药血清;构建皮质酮(Corticosteron... 目的:通过探究逍遥散的神经保护作用及其化学组成,发现逍遥散中可能发挥神经保护作用的有效活性成分。方法:采用水提法提取逍遥散,通过高分辨质谱-液相色谱质谱联用仪分析提取液的化学成分;制备逍遥散含药血清;构建皮质酮(Corticosterone,CORT,400μmol/L)诱导PC12细胞损伤模型,检测含药血清对PC12细胞增殖率的影响;用SIMCA软件进行细胞增殖率-成分含量相关性分析,找到逍遥散中可能具有神经保护作用的化合物。结果:与模型组相比,逍遥散含药血清可减轻CORT诱导的细胞损伤,并促进细胞增殖(P=0.000)。液质谱图结合软件分析比对发现,主要药效化合物有:芍药苷、甘草酸、甘草苷、迷迭香酸、没食子酸、2-苯基乙醛、(15Z)-9,12,13-Trihydroxy-15-octadecenoic acid、丹酚酸B、甘草皂苷G2等,单体验证结果显示苯乙醛具有一定的神经保护作用(P=0.000)。结论:证明逍遥散具有神经保护作用,分析并鉴定出55种化合物有可能是其发挥作用的主要成分,发现逍遥散中未曾报道的7种可能具有神经保护作用的化合物,并验证出苯乙醛具有神经保护作用且此前从未报道。本研究成果将为阐明逍遥散神经保护作用的物质基础提供参考,也为拓展逍遥散的临床应用提供数据支撑。 展开更多

目的:探讨逍遥散加味联合曲安奈德治疗口腔扁平苔藓(OLP)肝郁血瘀证的疗效及对唾液炎症因子的影响。方法:选取84例OLP肝郁血瘀证患者按照随机数字表法分为观察组(n=42)和对照组(n=42)。对照组予曲安奈德注射液1 ml混合等量2%盐酸利多卡... 目的:探讨逍遥散加味联合曲安奈德治疗口腔扁平苔藓(OLP)肝郁血瘀证的疗效及对唾液炎症因子的影响。方法:选取84例OLP肝郁血瘀证患者按照随机数字表法分为观察组(n=42)和对照组(n=42)。对照组予曲安奈德注射液1 ml混合等量2%盐酸利多卡因注射液于病损边缘局部注射,观察组在对照组治疗基础上予以逍遥散加味治疗,治疗8周后比较两组的临床疗效、症状评分、唾液炎症因子、免疫功能及不良反应发生情况,记录治疗后6个月的复发情况。结果:观察组临床总有效率为92.86%高于对照组的76.19%(P<0.05)。观察组治疗后主观和客观症状评分低于对照组(P<0.05)。观察组治疗后唾液白介素-6(IL-6)、白介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)水平低于对照组(P<0.05)。观察组治疗后CD4^(+)、CD4^(+)/CD8^(+)水平高于对照组,CD8^(+)水平明显低于对照组(P<0.05)。两组不良反应总发生率比较,差异无统计学意义(P>0.05)。观察组复发率明显低于对照组(P<0.05)。结论:逍遥散加味联合曲安奈德可有效提高OLP肝郁血瘀证患者的治疗效果,进一步缓解患者的临床症状,降低复发率及唾液炎症因子表达水平,提高机体免疫功能,安全性较好。 展开更多

【目的】探讨逍遥散加味颗粒联合中药塌渍外敷治疗桥本甲状腺炎患者的临床效果。【方法】选取2020年1月至2021年12月榆林市中医医院与西安市碑林区中医医院收治的66例桥本甲状腺炎患者,按照随机数字表法分为对照组和观察组,每组33例。... 【目的】探讨逍遥散加味颗粒联合中药塌渍外敷治疗桥本甲状腺炎患者的临床效果。【方法】选取2020年1月至2021年12月榆林市中医医院与西安市碑林区中医医院收治的66例桥本甲状腺炎患者,按照随机数字表法分为对照组和观察组,每组33例。观察组患者给予逍遥散加味颗粒内服联合中药塌渍外敷治疗,对照组患者给予硒酵母片口服治疗,比较两组患者临床疗效及治疗前后甲状腺功能指标、甲状腺相关抗体水平以及中医症状体征评分。【结果】观察组治疗总有效率明显高于对照组(P<0.05);治疗后,两组促甲状腺激素(TSH)水平均下降,且观察组TSH水平显著低于对照组(P<0.05);治疗后,两组游离三碘甲状腺原氨酸(FT 3)、游离甲状腺素(FT 4)水平较治疗前均显著升高,且观察组明显高于对照组(P<0.05);治疗后,两组血清甲状腺球蛋白抗体(TGAb)甲状腺过氧化酶抗体(TPOAb)水平较治疗前均显著降低(P<0.05),且观察组上述指标水平均显著低于对照组(P<0.05);治疗后,对照组神疲乏力、气短懒言、腰膝酸软、口干舌燥、消瘦、心悸、失眠、烦热、自汗盗汗、大便溏稀等中医症状体征评分均显著高于观察组(P<0.05)。【结论】逍遥散加味颗粒口服联合中药塌渍外敷治疗桥本甲状腺炎患者,可明显改善患者甲状腺功能并降低患者自身甲状腺抗体水平,同时可有效缓解患者临床症状体征,达到标本兼治的目的。 展开更多

目的:从巨噬细胞吞噬功能角度探讨逍遥散缓解情志应激诱导的乳腺癌“易感性”的作用机制。方法:将20只小鼠采用完全随机方法分为对照组、模型组、低剂量逍遥散组、高剂量逍遥散组,每组5只。小鼠连续28 d给予拘束应激或低、高剂量的逍遥... 目的:从巨噬细胞吞噬功能角度探讨逍遥散缓解情志应激诱导的乳腺癌“易感性”的作用机制。方法:将20只小鼠采用完全随机方法分为对照组、模型组、低剂量逍遥散组、高剂量逍遥散组,每组5只。小鼠连续28 d给予拘束应激或低、高剂量的逍遥散(3.25 g/kg,6.50 g/kg),在第7天皮下接种4T1小鼠乳腺癌细胞建立小鼠荷瘤模型,记录小鼠的瘤径、瘤重以及肿瘤拍照。小鼠取材后通过高效液相-紫外分光法检测血浆中皮质酮的含量;免疫组织化学方法观察肿瘤组织中巨噬细胞吞噬现象;采用蛋白质印迹法、实时荧光定量反转录聚合酶链反应(RT-qPCR)、免疫荧光技术检测肿瘤组织中“吃我”信号受体低密度脂蛋白受体相关蛋白(LRP1)、“不吃我”信号受体信号调节蛋白α(SIRPα)和微RNA(miRNA)的表达情况。结果:与模型组比较,低、高剂量逍遥散组显著缓解应激诱导的乳腺癌“易感性”,抑制肿瘤组织中巨噬细胞的吞噬功能,其作用与促进巨噬细胞LRP1的蛋白和基因表达水平,抑制SIRPα蛋白表达有关(P<0.05);逍遥散对巨噬细胞的调节作用,与增加miRNA-4259/6680-5p/7847-3p表达有关(P<0.05)。结论:逍遥散降低情志应激诱导的乳腺癌“易感性”,其作用机制与缓解LRP1-SIRPα信号紊乱并促进巨噬细胞对肿瘤细胞的吞噬作用有关。 展开更多

目的利用液相色谱质谱(liquid chromatography-tandem mass spectrometry,LC-MS)技术进行逍遥散治疗抑郁症的代谢组学实验研究。方法纳入2021年6月—2022年6月在南阳市第四人民医院经逍遥散治疗的36例抑郁症患者作为研究对象,采集患者... 目的利用液相色谱质谱(liquid chromatography-tandem mass spectrometry,LC-MS)技术进行逍遥散治疗抑郁症的代谢组学实验研究。方法纳入2021年6月—2022年6月在南阳市第四人民医院经逍遥散治疗的36例抑郁症患者作为研究对象,采集患者的空腹血液样本。采用高效液相色谱(high performance liquid chromatography,HPLC)和质谱联用技术对代谢物样本进行分析,采用逆向相色谱柱进行分离,质谱采用高分辨质谱测量碎片;通过质谱化合物的浓度和碎片图谱特征,得到代谢物的定性和定量结果。结果逍遥散治疗后抑郁症患者血浆代谢轮廓发生了显著改变,在代谢组学谱图中鉴定出19个差异显著的代谢物。逍遥散治疗后抑郁症患者的代谢物谱主要集中在为甘油磷脂代谢、脂肪酸代谢等9种代谢途径。逍遥散治疗后上述代谢组学指标水平趋于正常组。结论通过LC-MS技术,可以有效地鉴定出逍遥散治疗抑郁症的代谢物谱,为进一步的药物研究和临床应用提供了重要的参考依据。 展开更多

目的:观察海马及杏仁核α-氨基羟甲基恶唑丙酸(AMPA)受体亚基和相关调节蛋白在束缚应激状态下蛋白表达变化及逍遥散的调节作用。方法:使用每天捆绑3 h的方法制作慢性束缚应激动物模型,并用逍遥散进行干预,分别于7 d后和21 d后用Western ... 目的:观察海马及杏仁核α-氨基羟甲基恶唑丙酸(AMPA)受体亚基和相关调节蛋白在束缚应激状态下蛋白表达变化及逍遥散的调节作用。方法:使用每天捆绑3 h的方法制作慢性束缚应激动物模型,并用逍遥散进行干预,分别于7 d后和21 d后用Western blot方法检测各组大鼠海马CA1区、CA3区、齿状回(DG)和杏仁核的AMPA受体亚基GluR2/3及N-乙基顺丁烯二酰亚胺敏感性的融合蛋白(NSF)、PKC作用蛋白1(PICK1)蛋白表达的情况。结果:7 d应激可使DG和杏仁核的GluR2/3、NSF表达显著降低(P均<0.05),使PICK1在CA1区的表达量显著增多(P<0.05),逍遥散对PICK1变化显示出一定调节作用。21 d应激可使CA1区的GluR2/3、NSF表达升高,其中GluR2/3有显著性差异(P<0.01),而在杏仁核表达有降低趋势,逍遥散对其均有显著调节作用(均为P<0.05),21 d应激使杏仁核PICK1表达量出现升高趋势,逍遥散可显著降低其表达(P<0.05)。结论:AMPA受体在短期重复应激和慢性应激状态下反应不同,海马和杏仁核反应相反,逍遥散对慢性应激状态下AMPA受体表达的调节作用较短期重复应激强。 展开更多

目的探讨逍遥散加味方对过敏性结膜炎患者泪膜功能的影响。方法取2013年3月至2015年4月中国中医科学院望京医院眼科门诊60例诊断为过敏性结膜炎患者,中医辨证为肝郁气滞型。将所有患者随机分为观察组和对照组,每组30例。观察组局部点色... 目的探讨逍遥散加味方对过敏性结膜炎患者泪膜功能的影响。方法取2013年3月至2015年4月中国中医科学院望京医院眼科门诊60例诊断为过敏性结膜炎患者,中医辨证为肝郁气滞型。将所有患者随机分为观察组和对照组,每组30例。观察组局部点色甘酸钠滴眼液联合逍遥散加味方干预治疗;对照组单纯局部点色甘酸钠滴眼液,两组干预治疗2周后,观察临床症状积分、基础泪液分泌试验(SIt)、泪膜破裂时间(BUT)、角膜荧光素染色(FL)情况。结果对照组治疗后临床症状积分较治疗前明显下降(P<0.05),其他指标与治疗前比较差异无统计意义(P>0.05)。观察组与本组治疗前比较,治疗后患者临床症状积分显著下降,SIt明显增多,BUT延长,FL积分减少,差异有统计意义(P<0.05)。治疗后两组各项指标间差异均有统计意义(P<0.05),说明观察组疗效显著优于对照组。结论逍遥散加味方联合色甘酸钠滴眼液,能明显改善过敏性结膜炎患者泪膜功能,减少干眼症的发生。 展开更多

目的 :探讨丹栀逍遥散 (DZXYS)防治D 半乳糖 (D galactose ,D gal)致拟阿尔茨海默病 (AD)模型的作用机制。 方法 :大鼠皮下注射D gal 15 0mg/ (kg·d)× 4 9d造模 ,以0 .5 4mg/ (kg·d)盐酸多奈哌齐为阳性对照药物 ,用12 .6... 目的 :探讨丹栀逍遥散 (DZXYS)防治D 半乳糖 (D galactose ,D gal)致拟阿尔茨海默病 (AD)模型的作用机制。 方法 :大鼠皮下注射D gal 15 0mg/ (kg·d)× 4 9d造模 ,以0 .5 4mg/ (kg·d)盐酸多奈哌齐为阳性对照药物 ,用12 .6 4 g/ (kg·d)DZXYS同步灌胃给药防治模型损伤。第 4 2~ 4 6天进行水迷宫实验和跳台实验 ;第 5 0天处死大鼠取脑组织 ,生物化学方法检测脑组织超氧化歧化酶 (SOD)活性和脂质过氧化物 (MDA)含量。结果 :①DZXYS提高AD大鼠主动学习记忆成绩的疗效优于盐酸多奈哌齐。②DZXYS恢复AD大鼠被动学习记忆成绩的疗效相当于盐酸多奈哌齐。③DZXYS使造模大鼠大脑SOD活性从 35 2 .6 7± 37.71回升到 (380 .73± 14 .85 )u/mg pr(P <0 .0 5 )。④DZXYS使造模大鼠大脑MDA含量从 7.88± 0 .70回降到 (6 .6 4± 0 .80 )ng/mg pr(P <0 .0 1)。结论 :减轻自由基损害是DZXYS防治拟AD模型动物的主要机制之一。 展开更多
