Payenapara lleloneura Kurz.(Kan-zaw),an endemic medicinal plant only found in Tanintharyi Region of Myanmar,is widely used in the treatment of various cancer and different ailments.In the present research,the seeds we...Payenapara lleloneura Kurz.(Kan-zaw),an endemic medicinal plant only found in Tanintharyi Region of Myanmar,is widely used in the treatment of various cancer and different ailments.In the present research,the seeds were phytochemical investigated for their nutritional potential for their use as functional foods or novel diet oil resources.Nutritional evaluation showed that the seeds are rich in fats and carbohydrates(soluble sugars and starch).Fatty acid analyses showed that the seeds accumulate very richα-eleostearic acid(α-ESA,18:3^(Δ9 cis,11-trans),13 trans),an important conjugated fatty acid,up to more than 70% of total fatty acids.The seed oil derived from the Kan-zaw tree contains approximately 3.25% β-eleostearic acid(18:3^(Δ9 trans,11 trans,13 trans)),an unusual conjugated fatty acid that imparts a potent anticancer application and industrially important drying qualities to Kan-zaw oil.Physicochemical properties of the Kan-zaw seeds were examined;petroleum ether(60–90℃)extract of seed oils were also investigated for the saponification value,iodine value and estimation of acid value.Further,the present study investigated cytotoxic potential of ethanol,methanol,acetone,chloroform Kan-zaw seed extracts and commercial Kan-zaw oil against human cervical cancer cell line(HeLa).The Kan-zaw extracts and oil have shown significant anticancer activity on HeLa cells.展开更多

After integrating and proofreading 140 original QTLs ...The composition and quantity of amino acids influence the protein content and nutritional value of soybeans and also have an important impact upon soybean quality. After integrating and proofreading 140 original QTLs associated with amino acid contentfrom soybase(http://www.soybase.org/), 138 QTLs were further analyzed to determine high-confidence QTL regions. Meta-analysis was first carried out using the Bio Mercator ver. 2.1 software, yielding 33 consensus QTLs. The consensus QTL confidence intervals(CIs) ranged from 0.07 to 19.85 Mb. Next, the overview method was used to optimize the CIs, and 57 "real" QTLs were mapped. Candidate genes in the consensus QTL regions were obtained from Phytozome and were annotated using the Gene Ontology(GO), Kyoto Encyclopedia of Genes and Genomes(KEGG), Swissprot, and gene annotation databases. Finally, 16 unpublished candidate genes controlling the content of five types of amino acids were identified with Blast. These results laid the foundation for fine mapping of soybean amino acid-related QTLs and marker-assisted selection.展开更多

Amino acids of the 30 cases were detected by high speed chromatography.RESULTS The quantity of amino acids in black stones was the highest (2269mg/g) and that of pure cholesterol stones was the lowest, only 14mg/g in a case. In five groups of gallstones, the contents of amino acids were: black stone > mixed bilirubin stone and glucoprotein stone > mixed cholesterol stone > pure cholesterol stone. The proportions were: 9595∶2902 and 2805∶578∶1. Aliphatic amino acids accounted for about 50% of the total amino acids in gallstones and glycine 153% of the total amount of 17 kinds of amino acids.CONCLUSION In mixed stones, the higher level of bilirubin, the higher contents of amino acids. The acidic amino acids of bilirubin stones were relatively higher than those of cholesterol stones.展开更多

On the basis of fatty acid composition, all strains could be differentiated as to species. There are five fatty acids present at 1.0% or greater with linoleic 71% of the total. Oleic, palmitic, stearic, vaccenic plus traces of palmitoleic and linolenic comprise the remainder. Knowledge of different effects of various organic materials on soil humic substance is important for both environmental safety and sustainable agriculture.A pot experiment was conducted at Jilin Agricultural Universit...Knowledge of different effects of various organic materials on soil humic substance is important for both environmental safety and sustainable agriculture.A pot experiment was conducted at Jilin Agricultural University,Jilin Province in northeast China to discover the influence of herb residue,animal excrement,woody residue,animal remnant on fulvic acid(FA) composition and structure using differential thermal analysis-thermogravimetric(DTA-TG),fourier transform infrared spectroscopy(FTIR) and elemental analysis.DTA-TG showed the range of peak temperature in the first exothermic reaction increased following the trend: CK〉herb residue〉animal excrement〉woody residue=animal remnant,and the most weight loss was observed in animal excrement.Moreover,the second exothermic reaction of CK-and animal excrement-FA was presented as double peaks,the order of weight loss in that area was animal remnant〉CK〉woody residue〉animal excrement〉herb residue.According to FTIR,herb residue displayed higher adsorption intensity at 2 950,1 420,1 240 and 1 030 cm-1,animal excrement was in reverse.At the same time,herb residue-and animal excrement-FA had an absorption peak at 1 720 cm-1,while other organic materials didn't have this peak.As elemental analysis showed,FA isolated from various treatments was significantly distinct.It was clearly shown from our results that FA composition and structure in amended soils may be affected in different ways and at various extents on dependence of the nature and origin of amendment.展开更多

Naringin (20, 40 and 80 mg/kg) was given orally for 7 days to Wistar rats before induction...The aim of this study was to evaluate the effect of naringin on experimentally induced inflammatory bowel dis- ease in rats. Naringin (20, 40 and 80 mg/kg) was given orally for 7 days to Wistar rats before induction of colitis by intrarectal instillation of 2 mL of 4% (v/v) acetic acid solution. The degree of colonic mucosal damage was analyzed by examining mucosal damage, ulcer area, ulcer index and stool consistency. Intrarectal administration of 4% acetic acid resulted in significant modulation of serum alkaline phosphatase, lactate dehydrogenase, superoxide dismutase (SOD), glutathione (GSH), malondialdehyde (MDA) and myeloperoxidase (MPO) content along with colonic nitric oxide (NO), xanthine oxidase (XO) level and protein carbonyl content in the colonic tissue as well as in blood. Naringin (40 and 80 mg/kg) exerted a dose dependent (P 〈 0.05) ameliorative effect, as it significantly increased hematological parameter as well as colonic SOD and GSH. There was a significant (P 〈 0.05) and dose dependant inhibition of macroscopical score, ulcer area along with colonic MDA, MPO activity by the 7 days of pretreatment of naringin (40 and 80 mg/kg). Biochemical studies revealed a significant (P 〈 0.05) dose dependant inhibition in serum alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) levels by pretreatment of naringin. Increased levels of colonic NO, XO, protein carbonyl content and DNA damage were also sig- nificantly decreased by naringin pretreatment. The findings of the present investigation propose that naringin has an anti-inflammatory, anti-oxidant and anti-apoptotic potential effect at colorectal sites as it modulates the production and expression of oxidative mediators such as MDA, MPO, NO and XO, thus reducing DNA damage.展开更多

The crystal structure has been established by single-crystal X-ray diffraction, and characterized by FT-IR. Fluorescent property was investigated in this work. Serum bile acids and biliary lipids were also studied in 90 matched samples..RESULTS The serum DCA was 0955μmol/L±0078μmol/L in CS group, which was more than that of control group (0696μmol/L±0047μmol/L), P<001. The ratio of DCA/chenodeoxycholic acids (CDCA) was 176±030 in CS group, about two times that in control group (092±014). The mole percent of DCA in bile was positively related to cholesterol saturation index (CSI) (P<001) and the mole percent of CDCA in bile negatively to CSI (P=001). There was correlation between the mole percent of DCA, CDCA and cholic acid in bile and in serum.CONCLUSION It is suggested that DCA is lithogenic and the increased amount of DCA or the ratio of DCA/CDCA in serum may be one of the features of cholesterol gallstone patients.展开更多
Background: Basal ileal endogenous amino acid(AA) losses(IAAend) and standardized ileal digestibility(SID) values of cereal grains, such as barley, are apparently underestimated when determined according to the...Background: Basal ileal endogenous amino acid(AA) losses(IAAend) and standardized ileal digestibility(SID) values of cereal grains, such as barley, are apparently underestimated when determined according to the nitrogen(N)-free method. Regression analysis between the dietary apparent ileal digestible content(c AID) and total crude protein(CP) and AA can be considered as alternative approach to obtain more accurate values for IAAendand SID of AA in cereal grains.Methods: Eight hulled barley genotypes were used, with barley being the only source of CP and AA in the assay diets. The diets contained 95 % as-fed of these eight barley genotypes each, ranging in CP content between 109.1 and 123.8 g/kg dry matter(DM). Nine ileally T-cannulated barrows, average body weight(BW) 30 ± 2 kg, were allotted to a row-column design comprising eight periods with 6 d each and nine pigs. On d 5 and the night of d 6 of every period, ileal digesta were collected for a total of 12 h. The IAAend and the SID were determined by linear regression analysis between c AID and total dietary CP and AA.Results: There exist linear relationships between cA ID and total CP and AA(P 〈 0.001). The IAAend of CP, Lys, Met, Thr and Trp amounted to 35.34, 1.08, 0.25, 1.02 and 0.38 g/kg DM intake(DMI), respectively, which are greater compared to average IAAend determined previously under N-free feeding conditions. The SID of CP, Lys, Met, Thr and Trp was 90,79, 85, 79 and 86 %, respectively, and was greater when compared to tabulated values. Moreover, these SID values were greater than those reported in literature, based on correction of apparent ileal digestibility(AID) of CP and AA for their IAAendvalues. Summarized, the results of the present regression analysis indicate greater IAAendin barley-based diets compared to those obtained by N-free feeding.Conclusions: For low-protein feed ingredients like barley the regression method may be preferred over correction of AID values for their IAAenddetermined under N-free feeding conditions, as intercepts and slopes of the linear regression equations between cA ID and total dietary CP and AA provide direct estimates of IAAendand SID of CP and AA in the presence of the assay feed ingredient.展开更多
The development of a method for the quantitative determination of the sericin content ratio(SCR)is urgently needed for silk refining and the purification of silk fibroin for biomedical applications.In this work,a seri...The development of a method for the quantitative determination of the sericin content ratio(SCR)is urgently needed for silk refining and the purification of silk fibroin for biomedical applications.In this work,a series of sericin/fibroin mixed samples with known SCRs were prepared by mixing initial samples of extracted sericin and fibroin from Bombyx mori silk.Significant differences were found in the contents of characteristic hydrophilic amino acids abundant in sericin and hydrophobic amino acids abundant in fibroin,and several linear relation-ships of SCR associated with the content ratios of Ser/Ala,Asp/Ala,Lys/Ala,Asp/Gly and Ser/Gly were estab-lished by amino acid analysis.Subsequently,the linear equation expressing SCR(%)as a function of the Asp to Ala content ratio X(%)was established as SCR=2.5634X−12.5587(R 2=0.9972).The results indicated that the SCR of degummed silks calculated by the equation is more objective and effective than the results obtained by the traditional weight loss method.Our study provides a novel approach for the sensitive and quantitative detection of the sericin content within the detection limit in unknown silks,which can contribute to quality control in the silk production process.展开更多

The first product from a porous scaffold of zein, a resorbable bone substitute, has passed the biological evaluation of medical devices (ISO 10993) by the China Food and Drug Administration. However, Class III medical devices need quality monitoring before being placed on the market, and such monitoring includes quality control of raw materials, choice of sterilization method, and evaluation of biocompatibility. In this paper, we investigated four sources of zein through amino acid analysis (AAA) and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) in order to monitor the composition and purity, and control the quality of raw materials. We studied the effect of three kinds of sterilization method on a porous zein scaffold by SDS-PAGE. We also compared the changes in SDS-PAGE patterns when irradiated with different doses of gamma radiation. We found that polymerization or breakage did not occur on peptide chains of zein during gamma-ray (y-ray) sterilization in the range of 20-30 kGy, which suggested that γ-ray sterilization is suitable for porous zein scaffolds, Regarding cell compatibility, we found a difference between using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay and a cell-counting kit-8 (CCK-8) assay to assess cell proliferation on zein film, and concluded that the CCK-8 assay is more suitable, due to its low background optical density.展开更多

The results showed that the extent of hydrolysis and the concentration of Gln peptides in the hydrolysate were 22.02%and 6.05 mmol/L, respectively. The hydrolysates were fractionated by size-exclusion chromatography on a Sephadex G-15 into five major fractions(GelF1–GelF5). The peptide(GelF1) fraction with the highest glutamine peptide content(51.8%) was further evaluated to determine its molecular weight distribution. Most(92.37%) peptides were less than 1 000 Da. Glutamic acid and glutamine were the most abundant amino acids, accounting for up to 12.98% of the total amino acid content. In addition, the total amino acid content in GelF1 was higher than that in GelF2 and GelF3.展开更多

The representative strategies for the development of new nucleic acids analysis in this field are summarized, and key advantages and possible limitations are discussed. This study aimed to determine the characteristics of soil microbial community composition and its relationship with soil chemical properties during natural recovery in the Loess Plateau.The soil microbial community co...This study aimed to determine the characteristics of soil microbial community composition and its relationship with soil chemical properties during natural recovery in the Loess Plateau.The soil microbial community composition was analyzed by comparing the soil microbial phospholipid fatty acids(PLFAs) of eight croplands abandoned for 1,3,5,10,13,15,20,and 30 yr in the Dunshan watershed,northern Loess Plateau,China.The results showed that soil organic carbon,total nitrogen,soil microbial biomass carbon,and soil microbial biomass nitrogen significantly increased with the abandonment duration,whereas the metabolic quotient significantly decreased.The Shannon richness and Shannon evenness of PLFAs significantly increased after 10 yr of abandonment.Gram-negative,Gram-positive,bacterial,fungal,and total PLFAs linearly increased with increased abandonment duration.Redundancy analysis showed that the abandonment duration was the most important environmental factor in determining the PLFA microbial community composition.The soil microbial PLFAs changed from anteiso-to iso-,unsaturated to saturated,and short-to long-chain during natural recovery.Therefore,in the Loess Plateau,cropland abandonment for natural recovery resulted in the increase of the soil microbial PLFA biomass and microbial PLFA species and changed the microbial from chemolithotrophic to a more heterotrophic community.展开更多

Amino acids of the 30 cases were detected by high speed chromatography.RESULTS The quantity of amino acids in black stones was the highest (2269mg/g) and that of pure cholesterol stones was the lowest, only 14mg/g in a case. In five groups of gallstones, the contents of amino acids were: black stone > mixed bilirubin stone and glucoprotein stone > mixed cholesterol stone > pure cholesterol stone. The proportions were: 9595∶2902 and 2805∶578∶1. Aliphatic amino acids accounted for about 50% of the total amino acids in gallstones and glycine 153% of the total amount of 17 kinds of amino acids.CONCLUSION In mixed stones, the higher level of bilirubin, the higher contents of amino acids. The acidic amino acids of bilirubin stones were relatively higher than those of cholesterol stones.
文摘A sensitive and selective method has been developed for the detection of free ethacrynic acid in human urine. Using 4-(bromomethyl)-7-methoxycoumarin, ethacrynic acid was transformed into a fluorescent derivative, and was analysed by HPLC-fluorescence detector. The detection limit is 0.1ug/ml urine. The method is suitable for screening ethacrynic acid in doping control and studying its metabolism.

文摘pH-fixed titration method for the determination of weak acids and bases has been studied in this paper.It is not necessary to know the ionization constant of weak acid or base and the concentration of titrant. This method had been applied to determine phenol,4-aminoantipyrine and glycine,whose ionization constants range from 10^(-10)to 10^(-12).The results were satisfactory.
文摘The cellular fatty acids from a total of 62 strains of Torulopsis glabrata (T. glabrata), Saccharomyces cerevisiae (S. cerevisiae), Rhodotorula rubra (R. rubra), Candida krusei (C. krusei), Candida albicans (C. albicans) and Candida tropicalis (C. tropicalis) were examined by capillary gas chromatography. On the basis of fatty acid composition, all strains could be differentiated as to species. These results indicate that capillary gas chromatographic analysis of cellular fatty acids is likely to be useful for rapid identification or grouping of newer isolates of yeast species.
文摘The fatty acid composition of the low water-use shrub Fremontodendron californicum was examined by high temperature capillary gas chromatography. The ground seeds were extracted by supercritical fluid extraction (SFE) to obtain the oil (25.6% w/w) and for subsequent determination of the fatty acid composition. There are five fatty acids present at 1.0% or greater with linoleic 71% of the total. Oleic, palmitic, stearic, vaccenic plus traces of palmitoleic and linolenic comprise the remainder. The fatty acid methyl ester composition would make the oil suitable for biodiesel production.

文摘Knowledge of different effects of various organic materials on soil humic substance is important for both environmental safety and sustainable agriculture.A pot experiment was conducted at Jilin Agricultural University,Jilin Province in northeast China to discover the influence of herb residue,animal excrement,woody residue,animal remnant on fulvic acid(FA) composition and structure using differential thermal analysis-thermogravimetric(DTA-TG),fourier transform infrared spectroscopy(FTIR) and elemental analysis.DTA-TG showed the range of peak temperature in the first exothermic reaction increased following the trend: CK〉herb residue〉animal excrement〉woody residue=animal remnant,and the most weight loss was observed in animal excrement.Moreover,the second exothermic reaction of CK-and animal excrement-FA was presented as double peaks,the order of weight loss in that area was animal remnant〉CK〉woody residue〉animal excrement〉herb residue.According to FTIR,herb residue displayed higher adsorption intensity at 2 950,1 420,1 240 and 1 030 cm-1,animal excrement was in reverse.At the same time,herb residue-and animal excrement-FA had an absorption peak at 1 720 cm-1,while other organic materials didn't have this peak.As elemental analysis showed,FA isolated from various treatments was significantly distinct.It was clearly shown from our results that FA composition and structure in amended soils may be affected in different ways and at various extents on dependence of the nature and origin of amendment.
文摘The aim of this study was to evaluate the effect of naringin on experimentally induced inflammatory bowel dis- ease in rats. Naringin (20, 40 and 80 mg/kg) was given orally for 7 days to Wistar rats before induction of colitis by intrarectal instillation of 2 mL of 4% (v/v) acetic acid solution. The degree of colonic mucosal damage was analyzed by examining mucosal damage, ulcer area, ulcer index and stool consistency. Intrarectal administration of 4% acetic acid resulted in significant modulation of serum alkaline phosphatase, lactate dehydrogenase, superoxide dismutase (SOD), glutathione (GSH), malondialdehyde (MDA) and myeloperoxidase (MPO) content along with colonic nitric oxide (NO), xanthine oxidase (XO) level and protein carbonyl content in the colonic tissue as well as in blood. Naringin (40 and 80 mg/kg) exerted a dose dependent (P 〈 0.05) ameliorative effect, as it significantly increased hematological parameter as well as colonic SOD and GSH. There was a significant (P 〈 0.05) and dose dependant inhibition of macroscopical score, ulcer area along with colonic MDA, MPO activity by the 7 days of pretreatment of naringin (40 and 80 mg/kg). Biochemical studies revealed a significant (P 〈 0.05) dose dependant inhibition in serum alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) levels by pretreatment of naringin. Increased levels of colonic NO, XO, protein carbonyl content and DNA damage were also sig- nificantly decreased by naringin pretreatment. The findings of the present investigation propose that naringin has an anti-inflammatory, anti-oxidant and anti-apoptotic potential effect at colorectal sites as it modulates the production and expression of oxidative mediators such as MDA, MPO, NO and XO, thus reducing DNA damage.

文摘One new polymer [Co(L)(H2O)2]n(1) was synthesized by 4-(ethoxycarbonyl)-5-methyl-1H-1,2,3-triazole-1-carboxylic acid(Emtc) under the in situ solvent thermal reaction(H2L = 1-(carboxymethyl)-5-methyl-1H-1,2,3-triazole-4-carboxylic acid). The title complex performs a wave-like 2D framework and the ligand H2L demonstrates the coordination mode as μ4-η-2:η-1η-1:η-1. The crystal structure has been established by single-crystal X-ray diffraction, and characterized by FT-IR. Fluorescent property was investigated in this work. Hirshfeld surface analysis has also been carried out on 1, and obvious main intermolecular interactions are observed.
文摘IM To analyze serum bile acids and biliary lipids of patients with cholesterol gallstone(CS) and explore the relationship between deoxycholic acid (DCA) and CS disease.METHODS Analysis of bile acids in serum was done with gaschromatography in two groups: CS group (n=151) and control group (n=256). Serum bile acids and biliary lipids were also studied in 90 matched samples..RESULTS The serum DCA was 0955μmol/L±0078μmol/L in CS group, which was more than that of control group (0696μmol/L±0047μmol/L), P<001. The ratio of DCA/chenodeoxycholic acids (CDCA) was 176±030 in CS group, about two times that in control group (092±014). The mole percent of DCA in bile was positively related to cholesterol saturation index (CSI) (P<001) and the mole percent of CDCA in bile negatively to CSI (P=001). There was correlation between the mole percent of DCA, CDCA and cholic acid in bile and in serum.CONCLUSION It is suggested that DCA is lithogenic and the increased amount of DCA or the ratio of DCA/CDCA in serum may be one of the features of cholesterol gallstone patients.

文摘Background: Basal ileal endogenous amino acid(AA) losses(IAAend) and standardized ileal digestibility(SID) values of cereal grains, such as barley, are apparently underestimated when determined according to the nitrogen(N)-free method. Regression analysis between the dietary apparent ileal digestible content(c AID) and total crude protein(CP) and AA can be considered as alternative approach to obtain more accurate values for IAAendand SID of AA in cereal grains.Methods: Eight hulled barley genotypes were used, with barley being the only source of CP and AA in the assay diets. The diets contained 95 % as-fed of these eight barley genotypes each, ranging in CP content between 109.1 and 123.8 g/kg dry matter(DM). Nine ileally T-cannulated barrows, average body weight(BW) 30 ± 2 kg, were allotted to a row-column design comprising eight periods with 6 d each and nine pigs. On d 5 and the night of d 6 of every period, ileal digesta were collected for a total of 12 h. The IAAend and the SID were determined by linear regression analysis between c AID and total dietary CP and AA.Results: There exist linear relationships between cA ID and total CP and AA(P 〈 0.001). The IAAend of CP, Lys, Met, Thr and Trp amounted to 35.34, 1.08, 0.25, 1.02 and 0.38 g/kg DM intake(DMI), respectively, which are greater compared to average IAAend determined previously under N-free feeding conditions. The SID of CP, Lys, Met, Thr and Trp was 90,79, 85, 79 and 86 %, respectively, and was greater when compared to tabulated values. Moreover, these SID values were greater than those reported in literature, based on correction of apparent ileal digestibility(AID) of CP and AA for their IAAendvalues. Summarized, the results of the present regression analysis indicate greater IAAendin barley-based diets compared to those obtained by N-free feeding.Conclusions: For low-protein feed ingredients like barley the regression method may be preferred over correction of AID values for their IAAenddetermined under N-free feeding conditions, as intercepts and slopes of the linear regression equations between cA ID and total dietary CP and AA provide direct estimates of IAAendand SID of CP and AA in the presence of the assay feed ingredient.

文摘The development of a method for the quantitative determination of the sericin content ratio(SCR)is urgently needed for silk refining and the purification of silk fibroin for biomedical applications.In this work,a series of sericin/fibroin mixed samples with known SCRs were prepared by mixing initial samples of extracted sericin and fibroin from Bombyx mori silk.Significant differences were found in the contents of characteristic hydrophilic amino acids abundant in sericin and hydrophobic amino acids abundant in fibroin,and several linear relation-ships of SCR associated with the content ratios of Ser/Ala,Asp/Ala,Lys/Ala,Asp/Gly and Ser/Gly were estab-lished by amino acid analysis.Subsequently,the linear equation expressing SCR(%)as a function of the Asp to Ala content ratio X(%)was established as SCR=2.5634X−12.5587(R 2=0.9972).The results indicated that the SCR of degummed silks calculated by the equation is more objective and effective than the results obtained by the traditional weight loss method.Our study provides a novel approach for the sensitive and quantitative detection of the sericin content within the detection limit in unknown silks,which can contribute to quality control in the silk production process.
文摘Touch deoxyribonucleic acid(DNA)refers to the DNA that is left behind from skin cells when a person touches or comes into contact with an object.In crimes where the identification of suspects becomes a challenge,touch DNA has been a proven investigative tool.The present study aims to provide a systematic review of the role of touch DNAin criminal cases which discusses the nature and importance of touch DNA evidence at crime scenes;various phenomena including the transfer and persistence of touched samples;different factors affecting the touch sample deposition and DNA shedding;the best recovery methods and collection of samples from different substrates;and the interpretation of profiles through advanced techniques that identify the suspects.The present study also aims to optimize standard protocols in the laboratories for touched samples appropriate to the substrates that improve the success rate of profiles from touched items.
文摘Our previous studies have shown that zein has good biocompatibility and good mechanical properties. The first product from a porous scaffold of zein, a resorbable bone substitute, has passed the biological evaluation of medical devices (ISO 10993) by the China Food and Drug Administration. However, Class III medical devices need quality monitoring before being placed on the market, and such monitoring includes quality control of raw materials, choice of sterilization method, and evaluation of biocompatibility. In this paper, we investigated four sources of zein through amino acid analysis (AAA) and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) in order to monitor the composition and purity, and control the quality of raw materials. We studied the effect of three kinds of sterilization method on a porous zein scaffold by SDS-PAGE. We also compared the changes in SDS-PAGE patterns when irradiated with different doses of gamma radiation. We found that polymerization or breakage did not occur on peptide chains of zein during gamma-ray (y-ray) sterilization in the range of 20-30 kGy, which suggested that γ-ray sterilization is suitable for porous zein scaffolds, Regarding cell compatibility, we found a difference between using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay and a cell-counting kit-8 (CCK-8) assay to assess cell proliferation on zein film, and concluded that the CCK-8 assay is more suitable, due to its low background optical density.

文摘Glutamine peptides were obtained from defatted soybean meal by enzymatic hydrolysis using a combination of Protamex~? and trypsinase. The results showed that the extent of hydrolysis and the concentration of Gln peptides in the hydrolysate were 22.02%and 6.05 mmol/L, respectively. The hydrolysates were fractionated by size-exclusion chromatography on a Sephadex G-15 into five major fractions(GelF1–GelF5). The peptide(GelF1) fraction with the highest glutamine peptide content(51.8%) was further evaluated to determine its molecular weight distribution. Most(92.37%) peptides were less than 1 000 Da. Glutamic acid and glutamine were the most abundant amino acids, accounting for up to 12.98% of the total amino acid content. In addition, the total amino acid content in GelF1 was higher than that in GelF2 and GelF3.
文摘Nucleic acids are natural biopolymers of nucleotides that store, encode, transmit and express genetic information, which play central roles in diverse cellular events and diseases in living things. The analysis of nucleic acids and nucleic acids-based analysis have been widely applied in biological studies, clinical diagnosis, environmental analysis, food safety and forensic analysis.During the past decades, the field of nucleic acids analysis has been rapidly advancing with many technological breakthroughs.In this review, we focus on the methods developed for analyzing nucleic acids, nucleic acids-based analysis, device for nucleic acids analysis, and applications of nucleic acids analysis. The representative strategies for the development of new nucleic acids analysis in this field are summarized, and key advantages and possible limitations are discussed. Finally, a brief perspective on existing challenges and further research development is provided.

文摘This study aimed to determine the characteristics of soil microbial community composition and its relationship with soil chemical properties during natural recovery in the Loess Plateau.The soil microbial community composition was analyzed by comparing the soil microbial phospholipid fatty acids(PLFAs) of eight croplands abandoned for 1,3,5,10,13,15,20,and 30 yr in the Dunshan watershed,northern Loess Plateau,China.The results showed that soil organic carbon,total nitrogen,soil microbial biomass carbon,and soil microbial biomass nitrogen significantly increased with the abandonment duration,whereas the metabolic quotient significantly decreased.The Shannon richness and Shannon evenness of PLFAs significantly increased after 10 yr of abandonment.Gram-negative,Gram-positive,bacterial,fungal,and total PLFAs linearly increased with increased abandonment duration.Redundancy analysis showed that the abandonment duration was the most important environmental factor in determining the PLFA microbial community composition.The soil microbial PLFAs changed from anteiso-to iso-,unsaturated to saturated,and short-to long-chain during natural recovery.Therefore,in the Loess Plateau,cropland abandonment for natural recovery resulted in the increase of the soil microbial PLFA biomass and microbial PLFA species and changed the microbial from chemolithotrophic to a more heterotrophic community.
