After "the Belt" concept comes into being, the anthropologists must upgrade their research from previously established fixed-point perspective to a more extensive observation over the "Belt" or a wider range, while the concept of "the Road" encourages anthropologists to study the culture of the other half of the world, namely, to read the world history as a whole new picture from a Chinese or eastern point of view. AIM:To evaluate the differences between human lacrimal gland adenoid cystic carcinoma with high-grade transformation(LACC-HGT)primar y cells cultured by high-grade transformation tissue and non-high-grade transformati...AIM:To evaluate the differences between human lacrimal gland adenoid cystic carcinoma with high-grade transformation(LACC-HGT)primar y cells cultured by high-grade transformation tissue and non-high-grade transformation(non-HGT)primary cells cultured by non-highgrade transformation tissue in proliferation,metastasis,drug susceptibility,and genes.METHODS:LACC-HGT primary cells were established by tissue block culture,and the 4^(th)to 10^(th)generation primary cells were selected as research objects.The cells were preliminarily identified by immunofluorescent staining.The differences between non-HGT and LACC-HGT primary cells in terms of proliferation,metastasis,and drug susceptibility were compared by cell counting kit-8(CCK-8)assay,wound healing,and drug sensitivity experiments.Differentially expressed genes were screened using mRNA array.Gene expression was analyzed using real-time quantitative polymerase chain reaction(RT-qPCR).RESULTS:LACC-HGT primary cells were successfully cultured by tissue block culture.Immunofluorescence staining results showed that cytokeratin(CK)and CK7 expression levels were positive in LACC-HGT primary cells.CCK-8 results showed that the proliferation ability of LACCHGT cells was significantly higher than that of non-HGT cells.Wound healing experiment showed that the migration ability of LACC-HGT cells was significantly higher than that of non-HGT cells.LACC-HGT cells were also less sensitive to cisplatin and paclitaxel than non-HGT cells.Compared with non-HGT cells,9566 differentially expressed genes were found in LACC-HGT primary cells,of which 5162 were upregulated and 4404 were down-regulated.The expression of N-acetylneuraminate pyruvate lyase(NPL),MARVEL domain containing 3(MARVELD3),syntabulin(SYBU),and allograft inflammatory factor 1(AIF1)was higher in LACCHGT cells than in non-HGT cells,whereas that of periostin(POSTN)was lower.CONCLUSION:LACC-HGT primary cells have faster proliferation,stronger migration ability,and poorer sensitivity to chemotherapy drugs than non-HGT primary cells.The expression of mRNAs in non-HGT and LACC-HGT primary cells are significantly different.These features are speculated to be the reasons why high-grade transformation tissues exhibit higher malignant degree and poorer prognosis than their counterparts.展开更多

Immature inflorescence was cultured on im...The plants of hybrid wheatgrass (A. cristatum×A. desertorum cv. Hycrest-Mengnong) were directly induced from embryogenic callus regenerated from immature inflorescence. Immature inflorescence was cultured on improved MS medium containing 2.0-3.0 mg L^-1 2,4-D to regenerate callus. The calli were then transferred to hormone-free MS medium for differentiation and 1/2 MS medium for rooting. Results showed that callus initiation frequency was 83.4% and plant regeneration frequency was 59.6%. Phosphinothricin acetyltransferase (bar) gene was transformed into the hybrid wheatgrass by particle bombardment. Resistant callus was obtained using selecting agent, herbicide glufosinate of 0.5 mg L^-1, and some transgenic plants were recovered in vitro. The transgenic plants were identified by PCR and Southern blot analysis and these plants developed normally in the glufosinate medium, whereas the nontransgenic plants did not. The results demonstrated that bar cDNA integrated into the genomic DNA of the transgenic plants. The transgenic frequencies of bar gene were 1.1%.展开更多

Whether tranilast had antagonistic effect on proliferation inhibition and collagen synthesis promotion induced by TGF-β 2 in cultured human trabecular meshwork cells was investigated. Suspension of 1×104 cultur...Whether tranilast had antagonistic effect on proliferation inhibition and collagen synthesis promotion induced by TGF-β 2 in cultured human trabecular meshwork cells was investigated. Suspension of 1×104 cultured human trabecular meshwork cells of 3—5 passage was distributed in each well of a 96-well disk and divided into control group and experimental group. After 24 h, 0 μg/ml (control), 12.5 μg/ml, 25 μg/ml, 50 μg/ml tranilast with 3.2 ng/ml TGF-β 2 were added into the incubation medium. Another 24 h later, proliferation and collagen synthesis in cultured human trabecular meshwork cells were examined respectively by using tetrazolium-based semiautomated colormetric (MTT) assay and 3H-proline incorporation with liquid scintillation technique. The results showed absorbance (A) values of the experimental groups were 0.9036±0.3017, 1.1361± 0.1352, 1.2457±0.1524 according to the different concentrations of tranilast, and 0.8956± 0.1903 of the control group. In comparison with the control group, 25 μg/ml (q'=3.23, P< 0.05), 50 μg/ml (q'=4.70, P<0.01) tranilast significantly antagonized the decrease of the A values induced by TGF-β 2 in the cultured human trabecular meshwork cells. In comparison with the control group [817.37±124.21 cpm/104 cells], 12.5 μg/ml (620.33±80.46 cpm/104 cells, q'= 4.26, P< 0.05), 25 μg/ml (594.58±88.13 cpm/104 cells, q'=4.81, P<0.01), 50 μg/ml (418.64±67.90 cpm/104 cells, q'=8.62, P<0.01) tranilast significantly inhibited the incorporation of 3H-proline into the cultured human trabecular meshwork cells promoted by TGF-β 2 in a dose-dependent manner. It was concluded that tranilast had the antagonistic effect on the proliferation inhibition and collagen synthesis promotion induced by TGF-β 2 in the cultured human trabecular meshwork cells.展开更多

Genetic transformation is one of the key steps in the molecular breeding of chrysanthemum,which relies on an optimal regeneration and transformation system.However,the regeneration system of different chrysanthemum cu...Genetic transformation is one of the key steps in the molecular breeding of chrysanthemum,which relies on an optimal regeneration and transformation system.However,the regeneration system of different chrysanthemum cultivars varies,and the regeneration time of most cultivars is long.To screen cultivars with highly efficient regeneration,leaves and shoot tip thin cell layers(tTCL)from eight chrysanthemum cultivars with different flower colors and flower types were cultured on Murashige and Skoog media(MS)supplemented with 1.0-5.0 mg L^(−1)6-benzylaminopurine(6-BA)and 0.1-1.0 mg L^(−1)α-naphthaleneacetic(NAA).The results showed that the most efficient regeneration media were MS+6-BA 1.0 mg L^(−1)+NAA 0.5 mg L^(−1)for leaf explants and MS+6-BA 5.0 mg L^(−1)+NAA 0.1 mg L^(−1)for tTCL explants.Subsequently,another 13 chrysanthemum cultivars were screened by using the media,and finally,three cultivars with high regeneration efficiency were obtained from 21 cultivars.Among these,C1 had the highest regeneration efficiency:the regeneration rate of leaf explants reached 80.0%after 42 days of culture,and the regeneration rate of tTCL explants reached 100%after 31 days of culture.Furthermore,we also established the transformation system for C1 as follows:preculturing for one day,infecting with Agrobacterium suspension(OD_(600)=0.6)for 10 min,and cultivating in the regeneration medium with 350 mg L^(−1)carbenicillin and 10 mg L^(−1)kanamycin,thus ultimately achieving a transformation rate of 4.0%.In this study,a new chrysanthemum cultivar with an efficient regeneration and transformation system was screened,which is beneficial to enrich the flower color of chrysanthemum transgenic plant recipients and to the functional research of flower color or type-related genes.展开更多

The regenerated plants with both the three target genes and the selectable marker gene hpt were selected for anther culture. RT-PCR analysis indicated that target genes were inserted in rice genomic DNA and successfully transcribed. It took only one year to obtain double haploid selectable marker-free transgenic plants containing the three target genes with co-transformation followed by anther culture technique, and the efficiency was 12.2%. It was also noted that one or two target genes derived from the binary vector were lost in some transgenic rice plants.展开更多

Research was conducted to optim...Bacopa monnieri (L.) Wettst. (Scrophulariaceae) is a highly sought after medicinal plant with therapeutic properties as cognition enhancer as well as for other brain and body functions. Research was conducted to optimize a thin cell layer explant based micropropagation system to assist mass propagation. Thin cell layers (TCL) derived from leaf and internode segments were used as explants. Murashige and Skoog medium was used to formulate shoot induction, elongation, and rooting media. Shoot induction media were prepared by supplementing three concentrations (0.1, 1.0, and 10.0 μM) of four cytokinins 6-benzylaminopurine, 2-isopentenyl-adenine, 6-3-Hydroxybenzylaminopurine, and thidiazuron to study adventitious shoot bud induction response. An optimum shoot bud induction response was observed on MS medium supplemented with 10.0 μM 6-benzylaminopurine for both leaf and stem transverse thin cell layer (tTCL) explants. The average number of shoot buds from leaf tTCL explants was 59, whereas, on an average, 33 shoot buds were regenerated from internode tTCL explants. Elongation of adventitious shoot buds was achieved best in a liquid medium using Liquid Lab Rocker<sup>®</sup> system. Elongated shoots recorded 100% rooting in MS medium supplemented with 5 μM indole butyric acid. Bacopa micropropagation employing tTCL explants for initial shoot bud induction and using LLR<sup>®</sup> boxes in subsequent elongation step can achieve cost effective way to regenerate high volume of plantlets and biomass required for herbal industry. Leaf and stem tTCL explants both were suitable for Agrobacterium tumefaciens (EHA105) mediated genetic transformation. Successful transformation was scored within three days of co-cultivation with Agrobacterium suspension on the basis of Enhanced Green Fluorescent Protein (EGFP) expression as an early and non-destructible screening device. Transformation frequencies of 83% and 76% were accomplished for leaf and stem tTCL explants, respectively. Greenhouse grown Bacopa plants were analyzed as fresh and dry methanolic extracts for total polyphenol content (811.93 ± 7.98 and 814 ± 17.64 GAE mg g-1) and the Trolox Equivalent Antioxidant Capacity values were 1918.25 ± 173.12 and 3163.14 ± 403.25 μmol/g, respectively.展开更多

The three parties of the state, mar...It has been over ten years since the Intangible Cultural Heritage(ICH) protection movement was launched in China. During this period, a unique discourse system has been established. The three parties of the state, market and academic circles have respectively created administrative, industrial and knowledge discourses about the ICH. First, the representation of folk culture was incorporated into both the ICH protection system with list as the core and the cultural reconstruction strategy in the transition period, subsequently into national narrative through implementation of policies and administration by the government. Second, market operations have transformed heritage resources into cultural capital to boost the culture sector and tourism, and thus reinvigorate folk culture and brought about qualitative changes. Third, academics have made ICH discourses integrated with multidiscipline and extend academic discourse from universities and research institutions to the public sphere and cultural practice. The formation of the above discourse types reflects the articulation and game playing of ICH discourses in the folk culture field, and therefore they combine to form an ideal way to approach the cultural transformation and cultural politics of contemporary China.展开更多

Many approaches are verified for stable introduction of foreign genes into the plant genome. The review examined the different constraints that limit the success of rice genetic transformation via Agrobacterium-mediated approach and suggested possible solutions. Explant identification, gene transfer technique and construct to tailor the integration, transgene expression without collateral to genetic damage and transformant selection are among the technical challenges affecting the rice transformation. Despite the contests, Agrobacteriummediated transformation system has been a better option for producing transgenic rice varieties because of its exact T-DNA processing and simple integration of low copy-number transgene. This information is necessary for improving the transformation system for recalcitrant rice varieties.展开更多

文摘AIM:To evaluate the differences between human lacrimal gland adenoid cystic carcinoma with high-grade transformation(LACC-HGT)primar y cells cultured by high-grade transformation tissue and non-high-grade transformation(non-HGT)primary cells cultured by non-highgrade transformation tissue in proliferation,metastasis,drug susceptibility,and genes.METHODS:LACC-HGT primary cells were established by tissue block culture,and the 4^(th)to 10^(th)generation primary cells were selected as research objects.The cells were preliminarily identified by immunofluorescent staining.The differences between non-HGT and LACC-HGT primary cells in terms of proliferation,metastasis,and drug susceptibility were compared by cell counting kit-8(CCK-8)assay,wound healing,and drug sensitivity experiments.Differentially expressed genes were screened using mRNA array.Gene expression was analyzed using real-time quantitative polymerase chain reaction(RT-qPCR).RESULTS:LACC-HGT primary cells were successfully cultured by tissue block culture.Immunofluorescence staining results showed that cytokeratin(CK)and CK7 expression levels were positive in LACC-HGT primary cells.CCK-8 results showed that the proliferation ability of LACCHGT cells was significantly higher than that of non-HGT cells.Wound healing experiment showed that the migration ability of LACC-HGT cells was significantly higher than that of non-HGT cells.LACC-HGT cells were also less sensitive to cisplatin and paclitaxel than non-HGT cells.Compared with non-HGT cells,9566 differentially expressed genes were found in LACC-HGT primary cells,of which 5162 were upregulated and 4404 were down-regulated.The expression of N-acetylneuraminate pyruvate lyase(NPL),MARVEL domain containing 3(MARVELD3),syntabulin(SYBU),and allograft inflammatory factor 1(AIF1)was higher in LACCHGT cells than in non-HGT cells,whereas that of periostin(POSTN)was lower.CONCLUSION:LACC-HGT primary cells have faster proliferation,stronger migration ability,and poorer sensitivity to chemotherapy drugs than non-HGT primary cells.The expression of mRNAs in non-HGT and LACC-HGT primary cells are significantly different.These features are speculated to be the reasons why high-grade transformation tissues exhibit higher malignant degree and poorer prognosis than their counterparts.
文摘Summary: In order to investigate whether cultured normal human lens epithelial cells (LEC) express transforming growth factor β (TGF-β), reverse transcriptase polymerase chain reaction (RT-PCR) and immunohistochemical methods were used for detection of TGF-β mRNA and protein in cultured normal human LEC. The results showed that a single RT-PCR amplified product about 310bp was obtained, and the sequence was homologous to the known sequence. TGF-β immunostain was positive in the plasma of LEC. It was suggested that normal human LEC could produce TGF-β, and LEC could be affected by TGF-β through autocrine action.
文摘The plants of hybrid wheatgrass (A. cristatum×A. desertorum cv. Hycrest-Mengnong) were directly induced from embryogenic callus regenerated from immature inflorescence. Immature inflorescence was cultured on improved MS medium containing 2.0-3.0 mg L^-1 2,4-D to regenerate callus. The calli were then transferred to hormone-free MS medium for differentiation and 1/2 MS medium for rooting. Results showed that callus initiation frequency was 83.4% and plant regeneration frequency was 59.6%. Phosphinothricin acetyltransferase (bar) gene was transformed into the hybrid wheatgrass by particle bombardment. Resistant callus was obtained using selecting agent, herbicide glufosinate of 0.5 mg L^-1, and some transgenic plants were recovered in vitro. The transgenic plants were identified by PCR and Southern blot analysis and these plants developed normally in the glufosinate medium, whereas the nontransgenic plants did not. The results demonstrated that bar cDNA integrated into the genomic DNA of the transgenic plants. The transgenic frequencies of bar gene were 1.1%.

文摘Vitis vinifera cv.Chardonnay suspension cultures were established from proembryogenic mass and employed for optimizing Agrobacterium-mediated transformation system.One-factor-at-a-time experiment revealed that OD600 of Agrobacterium,time of inoculation,co-cultivation,and cell-drying before inoculation significantly affected the transformation efficiency which reached maximum 21.5% at the following conditions:0.8 of OD600,25 min of inoculation,2 d of co-cultivation,and 10 min of cell drying.Response surface methodology experiments based on a five-level,four-factor central-composite rotatable design were then used to optimize these selected factors.The optimized conditions for Chardonnay grape transformation were:0.8711 of OD600,28.9 min of inoculation,2.25 d of co-cultivation and 11.76 min of cell drying.After optimization,transformation efficiency was 26.2% and there were no interactions among different factors.

文摘Whether tranilast had antagonistic effect on proliferation inhibition and collagen synthesis promotion induced by TGF-β 2 in cultured human trabecular meshwork cells was investigated. Suspension of 1×104 cultured human trabecular meshwork cells of 3—5 passage was distributed in each well of a 96-well disk and divided into control group and experimental group. After 24 h, 0 μg/ml (control), 12.5 μg/ml, 25 μg/ml, 50 μg/ml tranilast with 3.2 ng/ml TGF-β 2 were added into the incubation medium. Another 24 h later, proliferation and collagen synthesis in cultured human trabecular meshwork cells were examined respectively by using tetrazolium-based semiautomated colormetric (MTT) assay and 3H-proline incorporation with liquid scintillation technique. The results showed absorbance (A) values of the experimental groups were 0.9036±0.3017, 1.1361± 0.1352, 1.2457±0.1524 according to the different concentrations of tranilast, and 0.8956± 0.1903 of the control group. In comparison with the control group, 25 μg/ml (q'=3.23, P< 0.05), 50 μg/ml (q'=4.70, P<0.01) tranilast significantly antagonized the decrease of the A values induced by TGF-β 2 in the cultured human trabecular meshwork cells. In comparison with the control group [817.37±124.21 cpm/104 cells], 12.5 μg/ml (620.33±80.46 cpm/104 cells, q'= 4.26, P< 0.05), 25 μg/ml (594.58±88.13 cpm/104 cells, q'=4.81, P<0.01), 50 μg/ml (418.64±67.90 cpm/104 cells, q'=8.62, P<0.01) tranilast significantly inhibited the incorporation of 3H-proline into the cultured human trabecular meshwork cells promoted by TGF-β 2 in a dose-dependent manner. It was concluded that tranilast had the antagonistic effect on the proliferation inhibition and collagen synthesis promotion induced by TGF-β 2 in the cultured human trabecular meshwork cells.

文摘Genetic transformation is one of the key steps in the molecular breeding of chrysanthemum,which relies on an optimal regeneration and transformation system.However,the regeneration system of different chrysanthemum cultivars varies,and the regeneration time of most cultivars is long.To screen cultivars with highly efficient regeneration,leaves and shoot tip thin cell layers(tTCL)from eight chrysanthemum cultivars with different flower colors and flower types were cultured on Murashige and Skoog media(MS)supplemented with 1.0-5.0 mg L^(−1)6-benzylaminopurine(6-BA)and 0.1-1.0 mg L^(−1)α-naphthaleneacetic(NAA).The results showed that the most efficient regeneration media were MS+6-BA 1.0 mg L^(−1)+NAA 0.5 mg L^(−1)for leaf explants and MS+6-BA 5.0 mg L^(−1)+NAA 0.1 mg L^(−1)for tTCL explants.Subsequently,another 13 chrysanthemum cultivars were screened by using the media,and finally,three cultivars with high regeneration efficiency were obtained from 21 cultivars.Among these,C1 had the highest regeneration efficiency:the regeneration rate of leaf explants reached 80.0%after 42 days of culture,and the regeneration rate of tTCL explants reached 100%after 31 days of culture.Furthermore,we also established the transformation system for C1 as follows:preculturing for one day,infecting with Agrobacterium suspension(OD_(600)=0.6)for 10 min,and cultivating in the regeneration medium with 350 mg L^(−1)carbenicillin and 10 mg L^(−1)kanamycin,thus ultimately achieving a transformation rate of 4.0%.In this study,a new chrysanthemum cultivar with an efficient regeneration and transformation system was screened,which is beneficial to enrich the flower color of chrysanthemum transgenic plant recipients and to the functional research of flower color or type-related genes.
文摘The 'double T-DNA' binary vector p13HSR which harbored two independent T-DNAs, containing hygromycin phosphotransferase gene (hpf) in one T-DNA region and three target genes (hLF, SB401, RZ10) in another T-DNA region, was used to generate selectable marker-free transgenic rice by Agrobacterium-mediated transformation. The regenerated plants with both the three target genes and the selectable marker gene hpt were selected for anther culture. RT-PCR analysis indicated that target genes were inserted in rice genomic DNA and successfully transcribed. It took only one year to obtain double haploid selectable marker-free transgenic plants containing the three target genes with co-transformation followed by anther culture technique, and the efficiency was 12.2%. It was also noted that one or two target genes derived from the binary vector were lost in some transgenic rice plants.

From naming to local color preference, the visual experience accumulation in Chinese red lies not just in the solitary and fossilized historical relics, but in a ceaseless and dynamic process of formation, recognition, conversion and transmission. Moreover, the symbolic constitution and ethnic identi cation of Chinese red exempli es historical accomplishments in local exchanges of artifacts, technologies, systems, and outlooks with foreign partners. Therefore, as part of the country's cultural heritage, Chinese red also encompasses two aspects, cultural integration based on its identification and artistic productivity with a focus on di erentiation.
文摘Bacopa monnieri (L.) Wettst. (Scrophulariaceae) is a highly sought after medicinal plant with therapeutic properties as cognition enhancer as well as for other brain and body functions. Research was conducted to optimize a thin cell layer explant based micropropagation system to assist mass propagation. Thin cell layers (TCL) derived from leaf and internode segments were used as explants. Murashige and Skoog medium was used to formulate shoot induction, elongation, and rooting media. Shoot induction media were prepared by supplementing three concentrations (0.1, 1.0, and 10.0 μM) of four cytokinins 6-benzylaminopurine, 2-isopentenyl-adenine, 6-3-Hydroxybenzylaminopurine, and thidiazuron to study adventitious shoot bud induction response. An optimum shoot bud induction response was observed on MS medium supplemented with 10.0 μM 6-benzylaminopurine for both leaf and stem transverse thin cell layer (tTCL) explants. The average number of shoot buds from leaf tTCL explants was 59, whereas, on an average, 33 shoot buds were regenerated from internode tTCL explants. Elongation of adventitious shoot buds was achieved best in a liquid medium using Liquid Lab Rocker<sup>®</sup> system. Elongated shoots recorded 100% rooting in MS medium supplemented with 5 μM indole butyric acid. Bacopa micropropagation employing tTCL explants for initial shoot bud induction and using LLR<sup>®</sup> boxes in subsequent elongation step can achieve cost effective way to regenerate high volume of plantlets and biomass required for herbal industry. Leaf and stem tTCL explants both were suitable for Agrobacterium tumefaciens (EHA105) mediated genetic transformation. Successful transformation was scored within three days of co-cultivation with Agrobacterium suspension on the basis of Enhanced Green Fluorescent Protein (EGFP) expression as an early and non-destructible screening device. Transformation frequencies of 83% and 76% were accomplished for leaf and stem tTCL explants, respectively. Greenhouse grown Bacopa plants were analyzed as fresh and dry methanolic extracts for total polyphenol content (811.93 ± 7.98 and 814 ± 17.64 GAE mg g-1) and the Trolox Equivalent Antioxidant Capacity values were 1918.25 ± 173.12 and 3163.14 ± 403.25 μmol/g, respectively.

文摘It has been over ten years since the Intangible Cultural Heritage(ICH) protection movement was launched in China. During this period, a unique discourse system has been established. The three parties of the state, market and academic circles have respectively created administrative, industrial and knowledge discourses about the ICH. First, the representation of folk culture was incorporated into both the ICH protection system with list as the core and the cultural reconstruction strategy in the transition period, subsequently into national narrative through implementation of policies and administration by the government. Second, market operations have transformed heritage resources into cultural capital to boost the culture sector and tourism, and thus reinvigorate folk culture and brought about qualitative changes. Third, academics have made ICH discourses integrated with multidiscipline and extend academic discourse from universities and research institutions to the public sphere and cultural practice. The formation of the above discourse types reflects the articulation and game playing of ICH discourses in the folk culture field, and therefore they combine to form an ideal way to approach the cultural transformation and cultural politics of contemporary China.

With the rapid development of the city, the phenomena of“large-scale demolition and construction” and “a thousand cities having the same pattern” are becoming more and more serious, and the urban characteristics containing rich historical information are gradually lost. As an important witness of the historical development of urban industry, industrial heritage contains precious historical and cultural values. Starting urban memory, and taking Taoxichuan Square as the example,through field research and memory interview, the continuation manner of urban memory in industrial heritage space is studied, and the transformation strategy of industrial heritage is put forward from the perspective of urban memory, making industrial heritage have a unique life cycle and cultural precipitation,inherit the urban context and promote the harmonious development of society.
文摘Social transformations have been changed in thinking and observable current religious institutions in contemporary architectural manifestations. Likewise, observed these changes in religious buildings of the past, they were current and contemporary in its own time, which has allowed them to remain in force in its architectural spatiality and its urban relationships, serving as the historical bridge 1;hat allows analyzing the contemporary perspective in Mexico and particularly in Yucatan. The goal of this paper is to carry out a contemporary look to the interior and exterior of the temples in Yucatan colonial architecture to establish the process of change and permanence of spatiality and volumes that have allowed liturgy procedures as well as the way of approach to the sacredness of the Catholic communities from the colonial period to the present day. On the other hand, interest to establish, in particular, as the colonials religious buildings have been maintained in force and have appreciated for religious service to continue and in some cases, as they have given other kinds of services while maintaining its primordial spatiality.
文摘Genetic transformation of rice(Oryza sativa L.) by introducing beneficial traits is now a central research instrument in plant physiology and a practical tool for plant improvement. Many approaches are verified for stable introduction of foreign genes into the plant genome. The review examined the different constraints that limit the success of rice genetic transformation via Agrobacterium-mediated approach and suggested possible solutions. Explant identification, gene transfer technique and construct to tailor the integration, transgene expression without collateral to genetic damage and transformant selection are among the technical challenges affecting the rice transformation. Despite the contests, Agrobacteriummediated transformation system has been a better option for producing transgenic rice varieties because of its exact T-DNA processing and simple integration of low copy-number transgene. This information is necessary for improving the transformation system for recalcitrant rice varieties.
