HPLC was used for the component analysis of the polysaccharide. The results indicate that Physalis alkekengi L. polysaccharide(PAP) was composed of rhamnose, xylose, arabinose, galactose, and glucose. Free radicals scavenging activity of PAP was studied through 3 free radicals scavenging tests. PAP exhibited high scavenging effects on OH and DPPH radicals, and both the scavenging rates were about 80%. The scavenging rate of O2^- radical was about 22%. 展开更多

The extracts from leaves of Manglietia insignis (Wall) Blume, Manglietia chingii Dandy and Manglietia yuyuanensis Law were prepared by organic solvent extraction and their components were analyzed by GC/MS and quant... The extracts from leaves of Manglietia insignis (Wall) Blume, Manglietia chingii Dandy and Manglietia yuyuanensis Law were prepared by organic solvent extraction and their components were analyzed by GC/MS and quantified. Meanwhile, the free radicals restraining activities were detected. The 21 compounds in M. insignis, 36 compounds in M. chigii and 20 compounds in M. yuyuanensis were identified. There were 11 common components in the extracts from three Manglietia species, and 12 components in two Manglietia species. The results of relative contents of every component in three extracts showed that the main constituents of M. insignis were terpenoids and alkene, amounting to 38.93%, followed by alkane (28.18%), the nitrogen containing compounds (15.73%) and aromatic compounds (7.23 %). The main constituents of leaf extract from M. chingii were the terpenoids and alkene, carboxylic acid, alkane and aromatic compounds, amounting to 30.22%, 14.17%, 13.87% and 13.29%, respectively, The main constituents of M. yuyuanensis were alcohol compounds, the terpenoids and alkene, and aromatic compounds, amounting to 28.00%, 25.38% and 18.00% respectively. The results showed that the three extracts had strong function of restraining oxygen free radicals. The ultra oxygen anions activity was restrained at the highest level, when the three extracts were diluted by hundred-fold, whereas the restraining capacity of hydroxyl free radicals reached maximum, when the three extracts were diluted by twenty-fold. The above results provide scientific evidences for further approaching the ecological healthy function of three MangUetia species 展开更多

The chemical activation of various precursors is effective for creating additional closed pores in hard carbons for sodium storage.However,the formation mechanism of closed pores under the influence of pore-forming agents is not well understood.Herein,an effective chemical activation followed by a high-temperature self-healing strategy is employed to generate interconnected closed pores in lignin-derived hard carbon(HCs).By systematic experimental design combined with electron paramagnetic res-onance spectroscopy,it can be found that the content of free radicals in the carbon matrix influences the closure of open pores at high temperatures.Excessively high activation temperature(>700 C)leads to a low free radical concentration,making it difficult to achieve self-healing of open pores at high tempera-tures.By activation at 700°C,a balance between pore making and self-healing is achieved in the final hard carbon.A large number of free radicals triggers rapid growth and aggregation of carbon microcrys-tals,blocking pre-formed open micropores and creating additional interconnected closed pores in as-obtained hard carbons.As a result,the optimized carbon anode(LK-700-1300)delivers a high reversible capacity of 330.8 mA h g^(-1) at 0.03 A g^(-1),which is an increase of 86 mA h g^(-1) compared to the pristine lignin-derived carbon anode(L-700-1300),and exhibits a good rate performance(202.1 mA h g^(-1) at 1 A g^(-1)).This work provides a universal and effective guidance for tuning closed pores of hard carbons from otherprecursors. 展开更多

Objective:To investigate the antioxidant activity of soil-borne aetinobacteria.Methods:The total phenolic contents,the level of antioxidant potential by DPPH radical scavenging activity,MO scavenging activity,and ABTS radical scavenging activity in ethyl acelale extract were determined.Results:The 16 S rDNA sequencing analysis revealed that Streptomyces sp.strain MJM 10778.which was isolated from Hambak Mountain.Korea,has 99.9% similarity to Streptomyces misionensis(S.misionenis) NBRC 13063.The physiological and the morphological test revealed that the strain MJM 10778 has different characteristics from the strain NBRC.13063.The entire antioxidant assay with the ethyl acelale extract displayed good radical scavenging activity.The IC_(50) values of the strain MJM 10778 extract on DPPH,.NO.and ABTS radicals were identified to he 92.8 μg/mL,0.02 μg/ml,and 134.9 μg/mL,respectively.The ethyl acetate extract of the strain MJM 10778 showed an 81.500% of cell viability at 100 μg/mL in Raw264.7cell viability assay.Conclusions:The results obtained suggesl that the ethyl acetate extract of Streptomyces sp.strain MJM 10778 could be considered as a potential source of drug for the diseases that is caused by free radicals with its anti-oxidant activities and low cytotoxicity. 展开更多

The present study was aimed to investigate the in vitro free radical scavenging activity of various leaf extracts(aqueous,methanol,acetone chloroform and petroleum ether)of Kedrostis foetidissima.In vitro free radical scavenging activities of the extracts were assessed against DPPH and hydroxyl radicals.The metal chelating activity and reducing power ability of the extracts were also determined.The free radical scavenging activity was found to be high in methanolic extract for DPPH and hydroxyl radicals in a concentration dependent manner followed by chloroform,aqueous,acetone and petroleum ether extracts.The metal chelating activity and reducing power ability was also found to be high in methanolic extract.The difference in scavenging potential of the extracts may be due to variation in the percentage of phytoconstituents extracted in various solvents.Thus the result suggests that the methanolic leaf extract of K.foetidissima could serve as a potential source of antioxidants and can be explored as a therapeutic agent in free radical induced diseases.©2015 Beijing Academy of Food Sciences.Production and hosting by Elsevier B.V.All rights reserved. 展开更多

Ascorbic acid, for having synergic effect with tea extracts, could enhance the scavenging activity of tea extracts markedly, and the contrary was cupric ion. Objective:To evaluate in vitro antioxidant and antibacterial activity of methanolic extract of Arnebia benthamii(A.benthamii) whole plant.Methods:Plasmid damage was analyzed by agarose gell electrophoresis.Calf thymus DNA was monitored by TBARS formation.DPPH, reducing power and lipid peroxidation was evaluated by using standard procedures.Antibacterial assay was monitored by disc diffusion method.Results:DPPH radical scavenging and hydroxyl radical scavenging potential of the plant revealed that the extract to be active radical scavenger.Reducing(Fe<sup>3+</sup>-Fe<sup>2+</sup>) power and lipid peroxidation inhibition efficiency(TBARS assay) of the extract was also evaluated and the extract showed promising activity in preventing lipid peroxidation and might prevent oxidative damages to biomolecules.The extract offered a significant protection against plasmid and calf thymus UNA damage induced by hydroxyl radicals. The extract was also evaluated on different bacterial strains and the maximum antibacterial activity was exhibited against Escherichia coli(E.coli) when compared with standard drug. Conclusions:These findings demonstrate that the methanol extract of A.benthamii has excellent anti-oxidant activities and could be considered as a potential source of lead molecules for pharmaceutical industries. 展开更多

Gelatin from the sea cucumber(Paracaudina chinens var.) was hydrolyzed by bromelain and the hydrolysate was found to have a high free radical scavenging activity. The hydrolysate was fractionated through an ultrafiltration membrane with 5 kDa molecular weight cutoff(MWCO). The portion(less than 5 kDa) was further separated by Sephadex G-25. The active peak was col-lected and assayed for free radical scavenging activity. The scavenging rates for superoxide anion radicals(O2·-) and hydroxyl radi-cals(·OH) of the fraction with the highest activity were 29.02% and 75.41%,respectively. A rabbit liver mitochondrial free radical damage model was adopted to study the free radical scavenging activity of the fraction. The results showed that the sea cucumber gelatin hydrolysate can prevent the damage of rabbit liver and mitochondria. 展开更多

The formation of·CCl3 radicals in liver nuclei was suggested by spin trapping of them with N-t-butyl-α-phenylnitrone followed by GC/MS detection of the resulting adduct. Comparison of its formation in microsomal biotransformation of CCl4 was made. In aerobic nuclear activation mixtures containing NADPH and CCl4, significant decrease in the arachidonic acid content of nuclear lipids was observed (27. 8%, compared to control), the intensity of this decrease was lower than that occurring in the corresponding microsomal incubation mixtures (29.1%). Significant decreases in arachidonic acid content of nuclear and endoplasmic reticulum lipids were also observed in animals at 6 hours of poisoning with the haloalkane. During aerobic nuclear metabolism of CCl4 or CBrCl3, cholesterol oxidation products were detected: a ketocholesterol, an epoxide like structure and 7-ketocholesterol. Nuclear protein carbonyl formation was not promoted during nuclear CCl4 biotransformation. NADPH by itself may lead to protein carbonyl formation during prolonged periods of incubation. CBrCl3 in contrast, led to decreased protein carbonyl formation. No increase in nuclear protein carbonyl formation was observed in CCl4 intoxicated animals during periods of time between 1 to 6 hours after treatment. The results indicate that during nuclear biotransformation of CCl4 or CBrCl3 reactive free radicals, PUFA degradation, reactive aldehydes and cholesterol oxidation products are formed, nearby DNA and regulatory proteins. 展开更多

Blood plasma of 39 cancer patients who had not previously undergone anti-tumor treatment, and 21 patients with recurrent epithelial tissues cancer before treatment and after the first course of chemotherapy were investigated. The treatment effectiveness of patients with relapse was evaluated throughout the year. The control group was the blood plasma of 14 healthy people. Free radical activity and oxidative modification of proteins were assessed in blood plasma. In malignant neoplasms of epithelial tissues, free radical activity and oxidative modification of plasma proteins are significantly higher. At the initial stages of carcinogenesis, the oxidative modification level of plasma proteins increases both due to aldehyde and ketone carbonyl derivatives. In the terminal stage of malignant tumors, an increase in the oxidative modification of proteins occurs due to aliphatic ketone dinitrophenyl hydrazones, which are the markers of protein aggregation characteristic of the late stages of oxidative stress. In case of an objective response to polychemotherapy, there is a significant decrease in free radical plasma activity after the first course of therapy and a significant increase in carbonyl derivatives recorded at a wavelength of 230 nm, which can serve as a marker of its effectiveness. 展开更多

The aim of the present work is to assess the value of Detarium Senegalense by determining the content of total phenols, total flavonoids and total anthocyanins, and by evaluating the free radical scavenging activity of Detarium Senegalense extracts. For this purpose, sequential extraction using solvents of increasing polarity was essential. The various extracts obtained underwent phytochemical and biochemical analyses. Phytochemical screening revealed the presence of flavonoids, alkaloids, tannins, polyphenols, anthocyanins and steroids/terpenes. Quantitative analysis of total polyphenols, total flavonoids and total anthocyanins yielded the following results: total flavonoids (0.803 ± 0029 mg EQ/100g P for acetone extract of roots and 0.871 ± 0.401 mg EQ/100g P for methanol extract of leaves);total polyphenols (23.298 ± 12.68 mg EAG/100g P for acetone extract of roots and 24.69 ± 0.49 401 mg EAG/100g P for methanol extract of leaves);total monomeric anthocyanins (44.697 ± 0.939 mg EC3G/100g P and 16.699 ± 0.193 mg EC3G/100g P respectively for acetone and methanol extracts of stem bark). DPPH free radical scavenging activity was 1.674 ± 0.023 mg/mL for the acetone extract and 0.934 ± 0.24 mg/mL for the methanol extract of roots. . 展开更多

The cranberry phenols are good free radi-cal-scavengers, but they were less efficient at inhibiting the lipid peroxidation. Of all the samples tested, the intermediate polarity fraction of extract rich in apolar phenolic compounds of fruit presented the highest antiradical activity while the most hydrophobic fractions of the anthocyanin-rich extract from fruit and pomace appeared to be the most efficient at inhibiting the lipid peroxidation. The antioxidant or pro-oxidant activity of fractions increased with the con-centration. The phenol polarity and the technological process to manufacture cranberry juice can influence the antioxidant and antiradical activities of fractions. 展开更多

[Objective] This study aimed to explore factors influencing the effects of theabrownin in Pu-erh tea on scavenging DPPH radicals. [Method] Taking Pu-erh tea as main research object, theabrownin in Pu-erh tea from tea piles with different times of mixture, stored for different years and processed by both the old and new techniques were extracted and isolated. And DPPH method was used to study the effects of theabrownin solution and tea liquor on scavenging radicals and their antioxidant activity. [Result] The result showed that the theabrownin content in Pu-erh tea from tea piles with one to four times of mixture revealed a first declining and subsequently rising trend. Under different pH conditions, the rates of DPPH radical scavenging of 1 mg/ml theabrownin solution made from tea piles with different times of mixture decreased gradually with the increase of pH, and all the rates were lower than that of the control vitamin C solution; the rates of DPPH radical scavenging of theabrownin from tea samples stored for different years and produced by different techniques dropped slowly within pH range 3.0-6.0; when pH was higher than 6.0, the rates of DPPH radical scavenging of theabrownin from tea samples produced by both new and traditional techniques in 2001 declined obviously, while the declining trend was not obvious in either the black tea or the Pu-erh tea produced by new technique in 2005. Under different theabrownin concentrations, the rates of DPPH radical scavenging of theabrownin solution made from tea piles with different times of mixture ascended as the concentration increased and the highest rate occurred in tea pile with four times of mixture, reaching up to 46.56%±0.88%; for tea samples stored for different years and produced by different techniques, the rates of DPPH radical scavenging of theabrownin rose gradually with the increase of the theabrownin concentration; the rate of radical scavenging of theabrownin in the black tea ascended obviously and that in Pu-erh tea was decreasing within the concentration range of 0.04 -0.06 mg/ml. At pH6.0, the DPPH radical scavenging ability of theabrownin in 0.1 mg/ml theabrownin solution and tea liquor containing 0.1mg/ml theabrownin from tea piles with different times of mixture ascended with the time of mixture; solution made from tea samples produced by different techniques in different years, the DPPH radical scavenging ability of theabrownin solution and tea liquor followed: Pu-erh tea produced by the new technique in 2001 ﹥ Pu-erh tea produced by the old technique in 2001﹥ Pu-erh tea produced by the new technique in 2005. And the DPPH radical scavenging ability of theabrownin in tea liquor containing 0.1 mg/ml theabrownin was higher than that in 0.1 mg/ml theabrownin Solution made from tea samples either with different times of mixture or produced by different techniques in different years. The above results indicated that many factor influenced the effects of theabrownin on eliminating DPPH radicals, including the time of fermentation, fermentation technique, years of storage, concentration and pH all had great influence on the effects of theabrownin on eliminating radicals. To ensure a good effect on scavenging radicals, it is better to prepare theabrownin at pH6.0 with concentration between 0.08 and 0.1 mg/ml for drinking. LuFeO3 crystallites of different sizes and morphologies were synthesized via a hydrothermal route. The sonocatalytic properties of the as-synthesized samples were investigated by degrading various organic dyes, including acid orange 7 (AOT), rhodamine B (RhB), methyl orange (MO), and methylene blue (MB), under ultrasonic irradiation, revealing that they exhibit excellent sonocatalytic activity toward the degradation of these dyes. Particularly, the synthesized bar-like particles with lengths of-3 μm and widths of-1μm have the highest sonocatalytic activity, and the degradation percentage of AO7 reaches 89% after 30 min of sonocatalysis. The effects of inorganic anions such as CI-, NO3-, SO42-, PO43-, and HCO3- on the sonocatalysis efficiency were investigated. Hydroxyl radicals (·OH) detected by fiuorimetry using terephthalic acid as a probe molecule were found to be produced over the ultrasonic-irradiated LuFeO3 particles. The addition of ethanol, which acts as a· OH scavenger, leads to quenching of "OH radicals and a simultaneous decrease in the dye degrada- tion. Group 2 served as hepatotoxin(CCL_4 treated) group.Group 3 served as positive control(Silyntarin) group,and Group 4.S and ft served as(73,150 and 300 nig/kg bw p.o.)L.racemosa leaf extract treated groups.Moreover,in vitro antioxidant DPPH,hydroxyl radical scavenging activity(HRSA),NO,ferric reducing antioxidant power(FRAP),lipid hydroperoxide(LPO) and super oxide dismutase(SOD) were also analyzed for the leaf extract.Results:The levels of the serum parameters such as serum glutamic oxaloacetic transaminase(SGOT).serum glutamic pyruvic transaminase(SGPT).alkaline phosphatase(ALP),bilirubin,cholesterol(CHL).sugar and lactate dehydrogenase(LDH) were significantly increased in COL_4 treated rats when compared with the control group(P<0.05).But the L.racemosa leaf extract treated rats showed maximum reduction of SGOT[(210.16±19.63)IU/L].SGPT[(82.37±13.87) IU/L].ALP[(197.63±23.4.3)IU/L],bilurubilt[(2.13 ±0.84) mg/dL].cholesterol[(163.83± 13.63) mg/dL].sugar[(93.00±7.63) mg/dL]and LDH[(1134.00) ±285.00)IU/L]were observed with the high dose(300 mg/kg bw) of leaf extract treated rats. Histopathological scores showed that,no visible changes were observed with high dose(300 mg/ kgbw) of leaf extract treated rats except few mild necrosis.The IC_(50) values were observed as(56.37 ±4.87)μg/mL,(57.68±1.98) μg/mL,(64.15±2.90)μg/mL,(61.94±3.98)μg/mL,(94.53± 1.68) μg/mL and(69.7±2.65)μg/mL for DPPH,HRSA,NO,FRAP,LPO and SOL) radical scavenging activities, respectively.Conclusions:In conclusion,the hepatoprotective effect of the L.racemosa leaf extract might be due to the presence of phenolic groups,terpenoids and alkaloids and in vitro antioxidant properties. 展开更多

The anthocyanin pigment extracted from green-wheat-bran was studied to identify its antioxidant activity.The antioxidant activities of the pigment were evaluated by anti-lipid peroxidation,total antioxidant activity (TAA),superoxide anion radical scavenging activity (SARSA),active oxygen scavenging activity (AOSA),and DPPH (1,1-diphenyl-2 picrylhydrazyl free radical) radical scavenging activity.The results showed that the pigment had higher antioxidant activity and TAA,SARSA,AOSA and DPPH.scavenging activities at a certain concentration than Vc (antiscorbutic vitamin,vitamin C),and the capacity increased with the increase of pigment concentration.Its TAA was 51.06 U mL-1,1.73 times of Vc,and SARSA 18 025.21 U mL-1,2.26% higher than Vc,and AOSA 3 776.31 U mL-1,1.24 times of Vc.As to the DPPH.scavenging activity of the pigment,there was a trend that higher concentration performed higher activity significantly improved with the company of Vc.The pigment showed significant antioxidant activities evaluated by different assays.Results will provide a better understanding on antioxidant activity of green wheat and allow the screening or breeding of green wheat varieties with higher antioxidant activity for food processing. 展开更多
