However, studies on completely stopping the hepatitis B immune globu- lin (HBIG) and nucleos(t)ide analogs (NUC) after LT are few. The aim of the current study was to evaluate the safety of anti- virus prophylaxis withdrawal in liver recipients whose serum hepatitis B e antigen (HBeAg) and HBV DNA are negative. We analyzed 190 patients undergone LT for HBV-related liver dis- ease from 2006 to 2012 and found that 10 patients completely stopped the HBIG and NUC due to poor compliance. These patients were liver biopsied and checked monthly with serum HBV markers, HBV DNA and liver function. Among the 10 patients, 9 did not show the signs of HBV recurrence after a mean follow-up of 51.6 months (range 20-73) after with- drawal of the HBIG and NUC. The average time from LT to the withdrawal of the anti-virus drug was 23.8 (13-42) months; one patient showed hepatitis B surface antigen-positive and detectable HBV DNA after stopping anti-virus drugs and this patient was successfully treated with entecavir. Our data sug- gested that complete withdrawal of anti-virus prophylaxis was safe and feasible for patients whose serum HBeAg and HBV DNA were negative at the time of LT.展开更多

Plant Dicer-like(DCL)and Argonaute(AGO)are the key enzymes involved in anti-virus post-transcriptional gene silencing(AV-PTGS).Here we show that AV-PTGS exhibited nucleotide preference by calculating a relative AV-PTG...Plant Dicer-like(DCL)and Argonaute(AGO)are the key enzymes involved in anti-virus post-transcriptional gene silencing(AV-PTGS).Here we show that AV-PTGS exhibited nucleotide preference by calculating a relative AV-PTGS efficiency on processing viral RNA substrates.In comparison with genome sequences of dicot-infecting Turnip mosaic virus(TuMV)and monocot-infecting Cocksfoot streak virus(CSV),viral-derived small interfering RNAs(vsiRNAs)displayed positive correlations between AV-PTGS efficiency and G+C content(GC%).Further investigations on nucleotide contents revealed that the vsiRNA populations had G-biases.This finding was further supported by our analyses of previously reported vsiRNA populations in diverse plant-virus associations,and AGO associated Arabidopsis endogenous siRNA populations,indicating that plant AGOs operated with G-preference.We further propose a hypothesis that AV-PTGS imposes selection pressure(s)on the evolution of plant viruses.This hypothesis was supported when potyvirus genomes were analysed for evidence of GC elimination,suggesting that plant virus evolution to have low GC%genomes would have a unique function,which is to reduce the host AV-PTGS attack during infections.展开更多

Cytopathic effect assay and MTT method were employe...This study examined the anti-viral effect of ursolic acid on guinea pig cytomegalovirus (GPCMV) and explored the steps of viral replication targeted by ursolic acid. Cytopathic effect assay and MTT method were employed to determine the 50% cellular cytotoxicity (CC50), 50% effective concentration (EC50) and therapeutic index (TI) with GPCMV. To investigate the specific anti-viral effect of ursolic acid at different temperatures and time points, two other medicines, ganciclovir and Jinyebaidu (JYBD), serving as controls, were studied for comparison. Our results showed that the CC50 of ganciclovir, JYBD and ursolic acid were 333.8, 3015.6, 86.7 μg/mL, respectively; EC50 of ganciclovir, JYBD and ursolic acid was 48.1, 325.5 and 6.8 μg/mL, respectively; TI of ganciclovir, JYBD and ursolic acid was 7, 9, 13, respectively. Similar with ganciclovir, ursolic acid could inhibit the viral synthesis, but did not affect the viral adsorption onto and penetration into cells. We are led to conclude that the anti-cytomegalovirus effect of ursolic acid is significantly stronger than ganciclovir or JYBD, and the cytotoxic effect of ursolic acid lies in its ability to inhibit viral synthesis.展开更多

Introduction: Prolonged immuno-suppressed status promised to induce internal growth of malignant cell and infectious agent, yet, only a small part of affected individuals seek medical attention or berried by commercia...Introduction: Prolonged immuno-suppressed status promised to induce internal growth of malignant cell and infectious agent, yet, only a small part of affected individuals seek medical attention or berried by commercially over-flowed fake information. Several studies have described complementary and alternative medicine as effective strategies for improving anti-infectious agent including malignant cell. The purpose of this study was to investigate the effect of a fermented herbal decoction (FHD) both in vitro and in vivo to malignant cells and microorganism by regulating leukocyte subset proportioning FHD as dietary material. Methods: In this approach of alternative study, selective anti-cancer effect by fermented decoction was tried to show first in vitro system both, cancer cell and virus strain. The fermented herbal decoction consisting of 80 sorts of herbs and fruits. The selective toxicity was set up and then for immunological factors in animal and human. The most important factor is to reduce side effect for a normal cell. Results: First, FHD was proved as safe by animal test. FHD regulated also the proportion of granulocyte and lymphocyte ratio both animal and human. In vitro culture showed selective toxicity by FHD against human melanoma and leukemia cell line but reduced toxicity was showed by normal cell line. As for the anti-virus activity, anti-virus effect was tested on the feeder layer of human fibroblast cell, after 9 days of culture. Second, FHD inhibits colon cancer growth in 3-methylholanthrene induced cancer in rat. Conclusion: The present results suggest that our fermented herbal decoction showed selective anti-cancer activities and anti-virus activities, together with the regulative effect on the immune system.展开更多
To investigate the anti-human papilloma virus (HPV) effect of the external liniment of traditional Chinese medicine “Keyouling”, the efficacy of this drug acting upon the proliferation and growth of the normal epide...To investigate the anti-human papilloma virus (HPV) effect of the external liniment of traditional Chinese medicine “Keyouling”, the efficacy of this drug acting upon the proliferation and growth of the normal epidermis cells of rabbits′ penis prepuce in different concentrations was observed. An experimental model of pathology of histocytes infected with HPV in vitro was established by using the human HPV infected prepuce epidermis cells as virus carrier to infect the sub-cultured epidermis cells of rabbits′ penis prepuce. The direct killing effect and the blocking effect of infections of epidermis cells in rabbits′ penis prepuce induced by HPV with different concentrations of “Keyouling” were determined by 3 kinds of culture methods: (1) the infected epidermis cells of rabbits′ penis prepuce were challenged by co-culture with HPV suspension and with culture medium containing drug; (2) the co-culture with epidermis cells of rabbits′ penis prepuce and HPV suspension was challenged with drug; (3) co-culture with culture medium containing drug and epidermis cells of rabbits′ penis prepuce was challenged with HPV suspension. It was found that “Keyouling” had little effect on the proliferation and growth of the normal epidermis cells of rabbits′ penis prepuce, but it had direct killing effect on HPV, and blocked effectively the infection of the epidermis cells of rabbits′ penis prepuce by HPV. It concludes that “Keyouling” has significant killing effect to HPV, but not normal histocytes in vitro , indicating that it is safe for clinical use. Its blocking effect on the HPV infection of rabbits′ penis prepuce epidermis cells suggested “Keyouling” may be clinically used to prevent the development of tumors or to inhibit the further progression or metastasis of tumors.展开更多

FHD regulated also the proportion of granulocyte and lymphocyte ratio both animal and human. In vitro culture showed selective toxicity by FHD against human melanoma and leukemia cell line but reduced toxicity was showed by normal cell line. As for the anti-virus activity, anti-virus effect was tested on the feeder layer of human fibroblast cell, after 9 days of culture. Second, FHD inhibits colon cancer growth in 3-methylholanthrene induced cancer in rat. Conclusion: The present results suggest that our fermented herbal decoction showed selective anti-cancer activities and anti-virus activities, together with the regulative effect on the immune system.
文摘To investigate the anti-human papilloma virus (HPV) effect of the external liniment of traditional Chinese medicine “Keyouling”, the efficacy of this drug acting upon the proliferation and growth of the normal epidermis cells of rabbits′ penis prepuce in different concentrations was observed. An experimental model of pathology of histocytes infected with HPV in vitro was established by using the human HPV infected prepuce epidermis cells as virus carrier to infect the sub-cultured epidermis cells of rabbits′ penis prepuce. The direct killing effect and the blocking effect of infections of epidermis cells in rabbits′ penis prepuce induced by HPV with different concentrations of “Keyouling” were determined by 3 kinds of culture methods: (1) the infected epidermis cells of rabbits′ penis prepuce were challenged by co-culture with HPV suspension and with culture medium containing drug; (2) the co-culture with epidermis cells of rabbits′ penis prepuce and HPV suspension was challenged with drug; (3) co-culture with culture medium containing drug and epidermis cells of rabbits′ penis prepuce was challenged with HPV suspension. It was found that “Keyouling” had little effect on the proliferation and growth of the normal epidermis cells of rabbits′ penis prepuce, but it had direct killing effect on HPV, and blocked effectively the infection of the epidermis cells of rabbits′ penis prepuce by HPV. It concludes that “Keyouling” has significant killing effect to HPV, but not normal histocytes in vitro , indicating that it is safe for clinical use. Its blocking effect on the HPV infection of rabbits′ penis prepuce epidermis cells suggested “Keyouling” may be clinically used to prevent the development of tumors or to inhibit the further progression or metastasis of tumors.

文摘Lycorine is the major active component from the amaryllidaceae family plant Lycoris radiate,a represent traditional Chinese medicinal herb,and is one of the typical alkaloids with pyrrolophenanthridine nucleus core.Lycorine has drawn great interest in medicinal field due to its divergent chemical structures and multiple biological functions,as well as pharmacological effects on various diseases.Accumulated evidence shows that lycorine not only possesses strong pharmacological effects on many diseases,including anti-leukemia,anti-tumor,anti-angiogenesis,anti-virus,anti-bacteria,anti-inflammation,and anti-malaria,but also exerts many other biological functions,such as inhibition of acetylcholinesterase and topoisomerase,suppression of ascorbic acid biosynthesis,and control of circadian period length.Notably,lycorine exhibits its numerous pharmacological effects on various diseases with very low toxicity and mild side effects.The divergent chemical structures,multiple biological functions,and very low toxicity of lycorine imply that the agent is a potential drug candidate that warrants for further preclinical and clinic investigation.

文摘Seven indole alkaloid glycosides containing a 1′-(4″-hydroxy-3″,5″-dimethoxyphenyl)ethyl unit(1-7)were isolated from an aqueous extract of Isatis indigotica leaves(da qing ye).Their structures were determined by spectroscopic data analysis combined with enzymatic hydrolysis as well as comparison of their experimental CD(circular dichroism)and calculated ECD(electrostatic circular dichroism)spectra.Based on analysis of[α]D20 and/or Cotton effect(CE)data of 1-7,two simple roles to assign location and/or configuration ofβ-glycopyranosyloxy and 1′-(phenyl)ethyl units in the indole alkaloid glycosides are proposed.Stereoselectivity in plausible biosynthetic pathways of 1-7 is discussed.Compounds 3 and 4 and their mixture in a 3:2 ratio showed activity against KCNQ2 in CHO cells.The mixture of 5 and 6(3:2)exhibited antiviral activity against influenza virus H1 N1 PR8 with IC5064.7μmol/L(ribavirin,IC5054.3μmol/L),however,the individual 5 or 6 was inactive.Preliminary structure-activity relationships were observed.

文摘Traditionally, the materials which are regarded as antigens recognized by γδ T lymphocytes are protein and carbohydrate, not nucleic acid or lipid. Recently, it has been demonstrated that γδ T cells can recognize lipid A and directly induce immune responses that involve CD1 (cluster of differentiation type 1) family and Toll like receptors (TLRs). This is a review about the interacting-mechanism, immunological effect and clinical application of them.
