Objective:To explore the mechanism of Radix Astragali-Lithospermum Erythrorhizon on the treatment of diabetic ulcer through the method of network pharmacology.Methods:This study included 32 compounds and 81 key targets.100 GO functional items and 116 KEGG signal pathways were obtained by enrichment analysis.Quercetin,kaempferol,isorhamnetin,mononetin,sitosterol,ivy sapogenin and other components of astragalus-purple herb play a key role in the targets of interleukin-6,cystatin 3,vascular endothelial growth factor,epidermal growth factor receptor and mitogen-activated protein kinase 8 in diabetic ulcer,and are mainly concentrated in AGE-RAGE,TNF and other signal pathways.Results:There were 32 compounds and 81 key targets.100 GO functional items and 116 KEGG signal pathways were obtained by enrichment analysis.Quercetin,kaempferol,isorhamnetin,mononetin,sitosterol,ivy sapogenin and other components of astragalus-purple herb play a key role in the targets of interleukin-6,cystatin 3,vascular endothelial growth factor,epidermal growth factor receptor and mitogen-activated protein kinase 8 in diabetic ulcer,and are mainly concentrated in AGE-RAGE,TNF and other signal pathways.Conclusion:Radix Astragali-Lithospermum Erythrorhizon may play the role of inhibiting inflammation,anti-apoptosis,promoting cell proliferation,angiogenesis and immune regulation through multi-components and multitargets,and play a role in the treatment of diabetic ulcer. 展开更多

目的:通过网络药理学方法探讨黄芪-紫草药对治疗糖尿病溃疡的作用机制。方法:使用中药系统药理学平台筛选黄芪-紫草药对的有效成分和作用靶点;通过检索人类基因信息数据库和人类孟德尔遗传研究平台选取糖尿病溃疡的疾病靶点;筛选药物与... 目的:通过网络药理学方法探讨黄芪-紫草药对治疗糖尿病溃疡的作用机制。方法:使用中药系统药理学平台筛选黄芪-紫草药对的有效成分和作用靶点;通过检索人类基因信息数据库和人类孟德尔遗传研究平台选取糖尿病溃疡的疾病靶点;筛选药物与疾病共同靶点,应用Cytoscape软件构建药物-化合物-作用靶点-疾病互作网络;使用STRING平台构建蛋白质间相互作用网络(PPI);使用Bioconductor进行GO功能富集分析和KEGG通路分析。结果:本次研究包含了32个化合物和81个关键作用靶点。富集分析得到100个GO功能条目和116条KEGG信号通路。黄芪-紫草药对中的槲皮素、山柰酚、异鼠李素、刺芒柄花素、谷甾醇、常春藤皂甙元等成分作用于糖尿病溃疡中的白细胞介素-6、胱天蛋白酶3、血管内皮生长因子、表皮生长因子受体、丝裂原激活蛋白激酶8等靶点起着关键作用,并主要富集在AGE-RAGE、TNF等多条信号通路上。结论:黄芪-紫草药对可能通过多成分多靶点发挥抑制炎症反应、抗细胞凋亡、促进细胞增殖、血管生成、免疫调节等作用,起到治疗糖尿病溃疡的目的。 展开更多
