The study aims to reveal phylogenetic and evolutionary relationship between aerobic anoxygenic phototrophic bacteria(AAnPB) and their relatives,anaerobic anoxygenic phototrophic bacteria(AnAnPB) and nonphototrophic bacteria(NPB,which had high homology of 16S rDNA gene with AAnPB and fell into the same genus),and validate reliability and usefulness of farnesyl pyrophosphate synthase(FPPS) gene for the phylogenetic determination.FPPS genes with our modified primers and 16S rDNA genes with general primers,were amplified and sequenced or retrieved from GenBank database.In contrast to 16S rDNA gene phylogenetic tree,AAnPB were grouped into two clusters and one branch alone with no intermingling with NPB and AnAnPB in the tree constructed on FPPS.One branch of AAnPB,in both trees,was located closer to outgroup species than AnAnPB,which implicated that some AAnPB would be diverged earlier in FPPS evolutionary history than AnAnPB and NPB.Some AAnPB and NPB were closer located in both trees and this suggested that they were the closer relatives than AnAnPB.Combination codon usage in FPPS with phylogenetic analysis,the results indicates that FPPS gene and 16S rRNA gene have similar evolutionary pattern but the former seems to be more reliable and useful in determining the phylogenic and evolutionary relationship between AAnPB and their relatives.This is the first attempt to use a molecular marker beside 16S rRNA gene for studying the phylogeny of AAnPB,and the study may also be helpful in understanding the evolutionary relationship among phototrophic microbes and the trends of photosynthetic genes transfer. 展开更多

Aerobic anoxygenic phototrophic bacteria(AAPB)repre sent a major group of bacterioplankton assemblages in many water systems and some are assumed to be closely associated with phytoplankton.However,studies on relationships between AAPB and cyanobacterial blooms are in scarcity.The dynamics of the abundance and diversity of AAPB was compared based on pufM gene in Meiliang Bay(featured by cyanobacterial blooms)and East Bay(featured by macrophyte)of Taihu Lake,a shallow subtropical lake in the East China plain.AAPB abundance was not significantly different between the two sites,and they were positively correlated with dissolved organic carbon(DOC)concentration.The ratios of AAPB to total bacteria varied from 3.4%to 11.5%and peaked in winter in both site s.No significant differences of AAPB community compositions were detected between the two sites,but there was a separation between warm seasons(June,August,and October)and cold seasons(December,February,and April).Rhizobiales and Limnohabitans-like pufM sequences were significantly contributors for the difference between two seasons,and specially enriched in cold seasons.Chlorophyll a(Ch1 a)and DOC were the most significant variables influencing the AAPB community structure.Furthermore,Porphyrobacter and Rhodospirillales-like pufM sequences were positively correlated with Ch1 a,indicating potential influence of cyanobacterial blooms on these AAPB taxa.These results suggested that diverse AAPB ecotypes coexisted in Taihu Lake,and their ecological role in carbon cycling in the lake may not be ignored. 展开更多

Anaerobic, aerobic, and facultative bacteria are all present in corrosive environments. However, as previous studies to address corrosion in the marine environment have largely focused on anaerobic bacteria, limited attention has been paid to the composition and function of aerobic and facultative bacteria in this process. For analysis in this study, ten samples were collected from rust layers on steel plates that had been immersed in seawater for diff erent periods (i.e., six months and eight years) at Sanya and Xiamen, China. The cultivable aerobic bacterial community structure as well as the number of sulfate-reducing bacteria (SRB) were analyzed in both cases, while the proportion of facultative SRB among the isolated aerobic bacteria in each sample was also evaluated using a novel approach. Bacterial abundance results show that the proportions are related to sea location and immersion time;abundances of culturable aerobic bacteria (CAB) and SRB from Sanya were greater in most corrosion samples than those from Xiamen, and abundances of both bacterial groups were greater in samples immersed for six months than for eight years. A total of 213 isolates were obtained from all samples in terms of CAB community composition, and a phylogenetic analysis revealed that the taxa comprised four phyla and 31 genera. Bacterial species composition is related to marine location;the results show that Firmicutes and Proteobacteria were the dominant phyla, accounting for 98.13% of the total, while Bacillus and Vibrio were the dominant genera, accounting for 53.06% of the total. An additional sixfacultative SRB strains were also screened from the isolates obtained and were found to encompass the genus Vibrio (four strains), Staphylococcus (one strain), and Photobacterium (one strain). It is noteworthy that mentions of Photobacterium species have so far been absent from the literature, both in terms of its membership of the SRB group and its relationship to corrosion. 展开更多

Abundance of general aerobic heterotrophic bacteria in the Bering Sea and Chukchi Sea and their adaptation to temperature 被引量:4

The abundance of general aerobic heterotrophic bacteria(GAB) from the water and sediment in the Bering Sea and the Chukchi Sea was determined by using petri dish cultivation and counting method. The abundance of GAB among the different sea areas, sampling sites, layers of sediments surveyed and adaptability to differential temperatures was studied. The result obtained showed that: the occurrence percentage of GAB in the surface water was higher than that in sediment, but the abundance was only 0.17% of sediment. The occurrence percentage of GAB in surficial layer of sediment was higher than that in the other layers. The occurrence percentage of GAB in surficial layer of sediment was higher than that in the other layers. The occurrence percentage, abundance and its variation of GAB in the Bering Sea were higher than that in the Chukchi Sea respectively. The average value of the abundance in the all sediment surveyed was 3166.3×10 2 CFU·g -1(wet.). The abundance of GAB in sediment showed a trend: roughly higher in the lower latitudinal area than higher latitude. The results from temperature test mean that: the majority of bacteria tested were cold-adapted ones, minority might be mesophilic bacteria. The results indicated that, Arctic ocean bacteria had a stronger adaptability to environmental temperature. 展开更多

Distribution of general aerobic heterotrophic bacteria in sediment core taken from the Canadian basin and the Chukchi Sea 被引量:1

The occurrence percentage and abundance of General Aerobic Hetero- trophic Bacteria (GAB) were determined by using the method of MPN for 182 sub- samples from 10 sediment cores taken from the Canadian basin and the Chukchi Sea at two different culturing temperatures. The results showed that the general occurrence percentage of GAB was quite high, average abundanees of GAB at cultured temperatares of 4℃ and 25℃ were 4.46 ×10^7 and 5.47×10^7 cells·g^-1(wt), respectively. The highest abundance of GAB occurred at 20 -22 cm section in the sediment. GAB abundances changed among the section of sediments, but there is a trend : the a-bundances at the middle or lower sections were lower than those at upper section. Cultivation at 25℃ could improve the occurrence percentage and abundances of GAB, which suggests that the increasing of temperature may change the living circum-stances of GAB. The differences of GAB among the latitudes areas indicated that occurrence percentage and abundances of GAB in middle latitude areas were higher than those in the higher or lower latitude areas, and were more obvious at 4℃ than those at 25℃. The GAB abundances in sediment under the shallower water seemed to be low- er than those in sediments under the deeper water and this status was more obvious at 25℃ than that at 4℃. 展开更多

This paper determined the abundance of General Aerobic heterotrophic Bacteria (GAB) in surficial sediments from the Chukchi Sea and the Canadian basin by using MPN and discussed their geographical distribution. The result shows that the determination percentages of the GAB were high, even till 100 percentage. The abundance range and averages of GAB for 4℃and 25℃ were from 4.00 × 10^2 to 2.40× 10^6 , 1.71×10^6 ind.·g^-1 (wet sample ) and from 2.40 ×10^5 to 2.40×10^7 , 1.10×10^7 ind. ·g^-1 (wet sample ) respectively. Not only the abundance range but also the averages of GAB in 25℃ were higher than that in 4℃. The abundance of GAB in sediments show a tendency that it is roughly greater in the lower latitudinal area than in the higher latitudinal area. The abundance of GAB increased from east to west as for the longitudinal distribution. With the water depth increasing, the abundance of GAB at 4℃ decreased, but GBA at 25℃ is not changed obviously with water depth. It seems that warmer circumstantial temperature is more suitable for some GAB. 展开更多

Available information on the microbial mechanisms associated with heavy metal(HM)passivation during co-composting amended with phosphate rock(PR)remains limited.Thus,this study investigated the dynamic changes in bacterial communities and HM-fractions(Zn,Cu,Cd,Cr and Pb)during swine manure composting with maize straw,and ascertained the bacterial influence on HM-passivation.The results demonstrated that the addition of PR improved HM-passivation,especially for Zn and Cd,with their bioavailability factors(BFs)reduced by 247.41 and 176.25%,respectively.As for bacterial communities,the proportion of Firmicutes decreased,while the proportions of Proteobacteria,Bacteroidetes,DeinococcusThermus and Gemmatimonadetes increased in all treatments.PR significantly changed the primary bacterial phyla in the thermophilic phase.Bacteroidetes were the main bacterial component controlling the passivation of Zn,Cu and Cr,while Deinococcus-Thermus mainly regulated the mobility of Zn and Pb,and Proteobacteria only dominated the transformation among Cd-fractions.These results may provide a reference for the use of HM-passivation techniques during composting. 展开更多

Objective:To highlight the presence of aerobic bacteria in live ticks infested sheep,in Diyala Governorate,Iraq.Methods:One hundred and thirty adult alive ticks were picked up from sheep which were reared in different farms in Diyala Governorate,Iraq,during the period from November 2012 to May 2013.Ticks were classified in the Natural History Museum in Baghdad.They were dissected aseptically for extraction of the salivary gland and midgut.The removed tissue from each organ was inoculated in buffer peptone water(1%)and incubated for 2 h at 37 °C,to maintain weak and/or injured bacterial cells,then transmitted to nutrient broth incubated at 37 °C for 18 h.Culturing was done on three solid bacteriological media(nutrient,blood and McConkey agars),and then incubated at37 ℃ for 24 h.Bacterial identification was performed by using multiple biochemical tests and API-20 strips.Data were analyzed by using Statistical Analysis System version 9.1,2010.Chi-square test was used for comparison at significant level of P ≤ 0.05.Results:Two species of ticks were identified[Rhipicephalus(Boophilus) annulatus and Hyalomma turanicum].High bacterial isolation rate was observed(483 isolates).A significant high isolation rate was recorded from Rhipicephalus annulatus(63.14%).Six bacterial species were identified[Escherichia coli(28.36%),Pseudomonas aeruginosa(18.01%),Bacillus cereus(14.69%),Staphylococcus aureus(13.66%),Citrobacter freundii(13.04%),and Enterobacter species(12.21%)].Also the high bacterial isolation rates were recorded in the temperate months(November,March and April);these coincided with high reproductive performance of ticks.Conclusions:The high isolation rate of aerobic pathogens from ticks might reflect the active contribution of this arthropod in environmental contamination and increase the probability of transmitting bacterial pathogens to their hosts. 展开更多

Surface water sources in the oil producing Niger Delta region of Nigeria are highly susceptible to pollution by petroleum hydrocarbons and so it is important to understand the microbial diversity of such ecosystems. Water and sediment samples were collected between April-August, 2013 from Bodo creeks and taken to Environmental Microbiology laboratory of University of Portharcourt for analysis. A total of thirty aerobic heterotrophic bacterial strains isolated ranged from 3.0 - 7.0 × 104 cfu for surface water and 1.6 - 5.6 × 104 cfu for sediment samples of Bodo creek using serial dilution and spread plate technique. Pure cultures of bacteria were obtained on the basis of their morphological characteristics and subjected to biochemical tests and further classified on the basis of 16S rRNA gene sequence analysis. The DNA was isolated from size fractionated samples and the diversity of bacteria in each fraction was studied using PCR amplification of partial 16S rRNA. The sequences were submitted to NCBIGen bank for identification and assigning of accession numbers. The isolated aerobic heterotrophic bacteria belong to the families of Enterobacteriaceae, Bacilliceae, Alcaligenaceae, Pseudomonadaceae, Flavobactericeae and Planococcaceae. 展开更多

Three new bacteria HS-03, HS-043 and HS-047 isolated from different ecosystems were found capable of aerobic denitrification. The potential application of these strains in wastewater treatment under aerobic conditions was investigated, These three bacteria all presented high nitrogen removal from wastewater that more than 98% of 10 mmol/L nitrate could be removed in 12--24 h by adding cheap external carbon source and low concentration of iron as well as molybdate. The mechanism at molecular level was analyzed. The success of this aerobic denitrification applied to wastewater treatment may serve as an alternative to enhance the practical nitrogen removal from wastewater. Main biochemical and physiological features of these strains were characterized. The 16S rDNA sequences were compared with the published data in GenBank by using BLAST. The results of phenotype and genotype proved that strain HS-03 and HS-047 belonged to Pseudomonas stutzeri and Pseudomonas pseudoalcaligenes respectively. Strain HS-043 was identified as Delftia clcidovorans of which denitrifying activity has not previously been explored. 展开更多

Objective In this study, we examined the biodegradation of Dichloroethylene (DCE) by two strains of aerobic bacteria. Methods Using batch experiments, we measured the biodegradation rates of DCE and the residual concentrations of DCE for each bacterial strain. The varying trends in biodegradation rates with different initial concentrations of DCE were fitted to kinetic models.Results The biodegradation kinetics of DCE by the strain DT-X, which uses toluene as co-metabolic substrate, fitted the Monod model (corresponding parameters: vmax=0.O075 h^-1, Ks=2.12 mg/L). The biodegradation kinetics of DCE by the strain DT-M, which uses 1,1-Dichloroethylene as single substrate, fitted the Haldane model (parameters: Vmax =0.0046 hz, Ks=4.25 mg/L, Ki=8.47 mg/L). Conclusion The substrate removal rate constant of 1,1-Dichloroethylene of the co-metabolic strain DT-X was much higher than that of strain DT-M. The substrate removal rates obtained from both bacterial strains in this study were higher than those reported in similar studies. 展开更多

Aerobic anoxygenic phototrophic bacteria(AAPB)are significant bacterial groups in aquatic ecosystems,known for their rapid growth and photoheterotrophic characteristics.However,the distribution and ecological assembly process of AAPB in low irradiation freshwater basins remain unclear,warranting further investigation.In this study,we present the diversity,abundance,spatial variations,ecological process,and community interaction of AAPB in sediment of Three Gorges Reservoir(TGR)under low irradiation.Our findings demonstrate the dominant genera of AAPB community that exist in the TGR area also are appeared in different waters,with some regional preference.Moreover,the concentration of pufM gene,an indicator for AAPB,maintains a consistently high numerical level ranging from(2.21±0.44)×10^(4)to(9.98±0.30)×10^(7)gene copies/g.Although solar irradiation is suggested as the major factor affecting AAPB,it remains unclear whether and how AAPB differ between regions due to varying solar irradiation levels.Our results show spatial differences between total bacteria and AAPB communities,with significant differences observed only in AAPB.Geographical and environmental factor contributed less than 10%to the spatial difference of community,with sediment type and environmental factors being the key factors influencing microbial community structure.The stochastic process plays a dominant role in the aggregation and replacement of AAPB communities,among which the most contribution is dispersal limitation.For AAPB network,Yoonia and Gemmobacter are the hubs for modules.Those results valuable insights into the AAPB communities in TGR with low irradiation. 展开更多

Lead(Pb) and its compounds are common pollutants in industrial wastewaters.To develop appropriate Pb 2+ treatment technologies,aerobic granules(AG) and bacterial alginates(BA) were studied as alternative biosorbents to remove Pb 2+ from aqueous solutions.The biosorption mechanism of AG and BA were further analyzed to determine which functional groups in AG and BA are active in Pb 2+ biosorption.In this paper,the Pb 2+ biosorption behavior of AG and BA was respectively investigated in batch experiments from the perspectives of the initial pH,contact time,and initial Pb 2+ concentration.The results showed that biosorption of Pb 2+ by AG and BA occurred within 60min at the initial Pb 2+ concentrations(0 150 mg L-1).The actual saturated Pb 2+ biosorption capability of AG was 101.97 mg g-1(dry weight of aerobic granular biomass).When the initial pH was 5,the biosorption capability of AG and BA was highest at the initial Pb 2+ concentrations(0 20mg L-1).During the process of Pb 2+ biosorption,K +,Ca 2+,and Mg 2+ were released.The Ion Chromatography(IC) and Fourier Transform Infrared Spectroscopy(FTIR) further highlighted the main role of ion exchange between Ca 2+ and Pb 2+ and sequestration of Pb 2+ with carboxyl(-COO) of AG and BA.This analogical analysis verifies that BA is responsible for biosorption of Pb 2+ by AG.At the same optimal pH,AG cultivated with different carbon source has different Pb 2+ biosorption capacity.The Pb 2+ biosorption by AG with sodium acetate as the sole carbon source is higher than AG with glucose as carbon source. 展开更多

AIM: To evaluate the effect of hormonal status on aerobic conjunctival flora in women. METHODS: One hundred fifty-eight women [reproductive-aged (n=55), pregnant (n=51), and postmenopausal (n=52)] who admitted to outpatient clinic of Obstetrics and Gynecology Department of Denizli State Hospital were enrolled. Age, body-mass index (BMI), obstetric history, cigarette smoking, drug usage, presence of systemic disease, and intraocular pressure (IOP) were recorded for each patient. The samples were taken from the lower fornix with two culture swabs and directly incubated in culture containing 5% sheep blood, eosin-methylene blue and chocolate agar. The other swab specimen was Gram stained. All growths and microscopic results were analyzed. RESULTS: The coagulase-negative Staphylococcus was the predominant organism isolated in the conjunctival samples in both three groups. The aerobic microorganism growth rate for all isolated aerobic organisms revealed no significant change in the three groups (P>0.05). The conjunctival culture positivity rates were similar in the three groups (49% in reproductive-aged, 57% in pregnant and 58% in postmenopausal women) (P>0.05). Age, IOP, BMI, gravidity, parity, cigarette smoking, drug usage, and presence of systemic diseases did not have an effect on culture positivity in three groups. CONCLUSION: Results of this study showed that conjunctival aerobic flora and bacterial colonization did not differ between reproductive-aged, pregnant and postmenopausal women. 展开更多

Bacteria play critical roles in regulating soil phosphorus(P) cycling. The effects of interactions between crops and soil P-availability on bacterial communities and the feedback regulation of soil P cycling by the bacterial community modifications are poorly understood. Here, six soybean(Glycine max) genotypes with differences in P efficiency were cultivated in acidic soils with long-term sufficient or deficient P-fertilizer treatments. The acid phosphatase(AcP) activities, organic-P concentrations and associated bacterial community compositions were determined in bulk and rhizosphere soils. The results showed that both soybean plant P content and the soil AcP activity were negatively correlated with soil organic-P concentration in P-deficient acidic soils. Soil P-availability affected the ɑ-diversity of bacteria in both bulk and rhizosphere soils. However, soybean had a stronger effect on the bacterial community composition, as reflected by the similar biomarker bacteria in the rhizosphere soils in both P-treatments. The relative abundance of biomarker bacteria Proteobacteria was strongly correlated with soil organic-P concentration and AcP activity in low-P treatments. Further high-throughput sequencing of the phoC gene revealed an obvious shift in Proteobacteria groups between bulk soils and rhizosphere soils, which was emphasized by the higher relative abundances of Cupriavidus and Klebsiella, and lower relative abundance of Xanthomonas in rhizosphere soils. Among them, Cupriavidus was the dominant phoC bacterial genus, and it was negatively correlated with the soil organic-P concentration. These findings suggest that soybean growth relies on organic-P mineralization in P-deficient acidic soils, which might be partially achieved by recruiting specific phoCharboring bacteria, such as Cupriavidus. 展开更多

The aim of our study was to use a biosurfactant produced by Bacillus and Lactobacillus isolates as an antiseptic in the formulation of local soap. A total of 60 isolates were characterized by microbiological techniques (30 Bacillus and 30 Lactobacillus) and the ability to produce biosurfactants was demonstrated by a hydrocarbon emulsification index (E24). The emulsification indexes (E24) varied from 9% to 100% for Bacillus and from 33% to 100% for Lactobacillus as well. The antagonistic assay showed that biosurfactants were able to inhibit the formation of biofilms and growth of pathogens such as Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus, Salmonella typhirium, Shigella boydii and Proteus mirabilis. The biosurfactant consortium (BioC) from Bacillus consortium and from Lactobacillus was able to inhibit biofilm formation and the pathogens growth. The BioC was stable to alkaline pH and the temperatures stability of Biosurfactant was ranging from 50°C to 90°C. The soap was made by the cold saponification process using one biosurfactant consortium formulated. This soap has a pH of 10 and showed good cleaning power and good foam stability. Similarly, the soap showed good antiseptic power and disinfection power against all pathogens tested. Handwashing is critical to preventing disease transmission. The persistence of pathogens in waste water was evaluated. The BioS produced showed good disinfection power against all pathogens tested. The valor of reduction on the hands and in the waste water was significantly more than compared to the control soaps used. This soap could be used in the prevention, fighting, and treatment of bacterial and viral infections. 展开更多

Chilled chicken is inevitably contaminated by microorganisms during slaughtering and processing,resulting in spoilage.Cutting parts of chilled chicken,especially wings,feet,and other skin-on products,are abundant in collagen,which may be the primary target for degradation by spoilage microorganisms.In this work,a total of 17 isolates of spoilage bacteria that could secrete both collagenase and lipase were determined by raw-chicken juice agar(RJA)method,and the results showed that 7 strains of Serratia,Aeromonas,and Pseudomonas could significantly decompose the collagen ingredients.The gelatin zymography showed that Serratia liquefaciens(F5)and(G7)had apparent degradation bands around 50 kDa,and Aeromonas veronii(G8)and Aeromonas salmonicida(H8)had a band around.65 and 95 kDa,respectively.The lipase and collagenase activities were detected isolate-by-isolate,with F5 showing the highest collagenase activity.For spoilage ability on meat in situ,F5 performed strongest in spoilage ability,indicated by the total viable counts,total volatile basic nitrogen content,sensory scores,lipase,and collagenase activity.This study provides a theoretical basis for spoilage heterogeneity of strains with high-producing collagenase in meat. 展开更多
