[Objective] The aim of this study is to evaluate the safety and bacteriostasis of Yandureqing(AEE)and its microemulsion(AEE-ME).[Method]The acute toxicity was tested in mice by intragastric administration,and median l...[Objective] The aim of this study is to evaluate the safety and bacteriostasis of Yandureqing(AEE)and its microemulsion(AEE-ME).[Method]The acute toxicity was tested in mice by intragastric administration,and median lethal dose(LD50)as well as its 95% confidence interval was calculated by modified Karber method;the bacteriostasis was investigated by cylinder plate method.[Result]LD50 of AEE in mice was 10.937 g/kg with the 95% confidence interval of 9.309-12.850 g/kg;and LD50 of AEE-ME in mice was 5.357 g/kg with the 95% confidence interval of 4.388-6.566 g/kg.The MICs of AEE to Escherichia coli O149 from swine,Staphylococcus aureus,Salmonella pullorum and Streptococcus agalactiae were 10.00,20.00,20.00 and 10.00 mg/ml,respectively;while the MICs of AEE-ME to the 4 kinds of bacteria mentioned above were 5.00,10.00,5.00 and 5.00 mg/ml in turn,and that to Pseudomonas aeruginosa is 20.00 mg/ml.[Conclusion]AEE is an actually nontoxic drug and AEE-ME belongs to the low toxic preparation.AEE and AEE-ME have obvious bacteriostasis,in which AEE-ME is superior to AEE.展开更多

The MIC and MBC was 3.91 mg/ml and 31.25 mg/ml on S. aureus and E. coil [ Conclusion] The result showed that the different extract from Ilex Kudingcha C. J. When combined with folium isatidis or flos poprli, the MIC of rhizoma coptidis was 1/8 extracting liq-uid or 1/16 extracting liquid compared with single use. When combined with rhizoma coptidis, the MIC of folium isatidis and flos poprli were 1/8 extracting liquid and 1/16 extracting liquid.[Conclusion] When rhizoma coptidis was combined with folium isatidis or flos poprli, the FIC values were 2 and 0.625, performing independent action and additive effect, respectively.展开更多
At present,effective fixation and anti-infection implant materials represent the mainstay for the treatment of open fractures.However,external fixation can cause nail tract infections and is ineffective for fixing sma...At present,effective fixation and anti-infection implant materials represent the mainstay for the treatment of open fractures.However,external fixation can cause nail tract infections and is ineffective for fixing small fracture fragments.Moreover,closed reduction and internal fixation during the early stage of injury can lead to potential bone infection,conducive to bone nonunion and delayed healing.Herein,we designed a bone adhesive with anti-infection,osteogenic and bone adhesion fixation properties to promote reduction and fixation of open fractures and subsequent soft tissue repair.It was prepared by the reaction of gelatin(Gel)and oxidized starch(OS)with vancomycin(VAN)-loaded mesoporous bioactive glass nanoparticles(MBGNs)covalently cross-linked with Schiff bases.Characterization and adhesion experiments were conducted to validate the successful preparation of the Gel-OS/VAN@MBGNs(GOVM-gel)adhesive.Meanwhile,in vitro cell experiments demonstrated its good antibacterial effects with the ability to stimulate bone marrow mesenchymal stem cell(BMSCs)proliferation,upregulate the expression of alkaline phosphatase(ALP)and osteogenic proteins(RunX2 and OPN)and enhance the deposition of calcium nodules.Additionally,we established a rat skull fracture model and a subcutaneous infection model.The histological analysis showed that bone adhesive enhanced osteogenesis,and in vivo experiments demonstrated that the number of inflammatory cells and bacteria was significantly reduced.Overall,the adhesive could promote early reduction of fractures and antibacterial and osteogenic effects,providing the foothold for treatment of this patient population.展开更多

Don under near-natural conditions using the thermal-desorption cold trap gas chromatography/mass spectrometer technique. The effects of the VOCs on airborne microorganisms were investigated using the method of natural sedimentation. Results showed that the major VOCs were as follows: limonene, β-pinene, α-pinene, and α-caryophyllene in Pinus tabulaeformis and P. bungeana; limonene, borneol acetate, β-pinene, myrcene, and tricylene in S. chinensis; limonene, α-pinene, myrcene, camphene, and β-pinene in Picea koraiensis; and limonene, 2, (10)-pinene, α-pinene, and myrcene in C. deodara. These VOCs and the corresponding foliar extracts inhibited the growth of bacteria and stimulated the growth of fungi. Experimental data using monomers of the VOCs demonstrated that limonene, β-pinene, and three aldehydes could significantly inhibit bacterial growth, suggesting an inhibitory effect of VOCs on the growth of airborne microorganisms in the five conifer species. The bacteriostasis and air-decontaminating effects of plant VOCs are further discussed in terms of their chemical composition.展开更多

Result: The results of drug sensitivity test of Shuanghuanglian combined with Cefoperazone-Sulbactam sodium against the resistant strains of Pseudomonas aeruginosa were compared with the results of drug sensitivity test of the two separately used, and the difference was statistically significant (P 〈 0.05) [The drug sensitivity test results of Shuanghuanglian and cefoperazone sulbactam sodium to Pseudomonas aeruginosa resistant strains were statistically significant compared with the drug sensitivity test results of Shuanghuanglian and Cefoperazone Sulbactam Sodium used separately (P 〈 0.05)];There was a dependence between strains and concentration in the effect of the combination of the two drugs. Conclusion: The combination of Shuanghuanglian and cefoperazone sulbactam sodium has synergistic antibacterial or bactericidal effect on Pseudomonas aeruginosa resistant strains. .展开更多

[Method...[Objective] This study aimed to reveal the therapeutic mechanism of compound sarcandra aerosol, which was exclusively owned by the Second Affiliated Hospital of Guiyang College of Traditional Chinese Medicine. [Method] An inflammation model was established by xylene-induced inflammation test and carrageenan- induced inflammation test to analyze the anti-inflammatory effect of compound sarcandra aerosol. By bacteriostasis test in vitro, the antibacterial effect of compound sarcandra aerosol against five common pathogens of pharyngitis was investigated. Blood samples were collected from Wistar rats in different compound sarcandra aerosol groups and control group to compare blood routine indicators and interleukin-1 (IL-1) levels. [Result] Three different concentrations of compound sarcandra aerosol could reduce degrees of xylene-induced ear edema in mice and carrageenan- induced paw edema in rats, but the anti-inflammatory effect of compound sarcandra aerosol was reduced as the concentration declined. In bacteriostasis test in vitro, the minimum inhibitory concentration of compound sarcandra aerosol against Streptococcus pneumoniae, Streptococcus hemolytis, Corynebacterium diphtheriae, Staphylococcus aureus and Salmonella typhimurium was 76, 105 38, 65 and 30 mg/ml, respectively. Compound sarcandra aerosol reduced white blood cell count, neutrophil count and lymphocyte percentage in pharyngitis model rats. Moreover, interleukin-1 level in watermelon frost lozenge group and different compound sarcandra aerosol groups was lower compared with control group. [Conclusion] Compound sarcandra aerosol can effectively treat pharyngitis by exerting anti-inflammatory and antibacterial effect and reducing interleukin-1 level. This study laid a solid foundation for clinical application of compound sarcandra aerosol.展开更多

In addition, the shelf lives of various grass carp products were compared after disinfection, so as to provide certain basis for the processing of fresh-cut grass carp products. The re- sults showed that the optimal disinfection conditions of sodium hypochlorite were as follows: concentration 300 mg/L, soak time 5 min and solid-liquid ratio 1 g : 5 ml. Under the optimal disinfection conditions, the inhibition rates of total bacteria and Pseudomonas reached 83% and 81%, respectively. The shelf life of refrigerated fresh-cut grass carp belly disinfected by sodium hypochlorite and packed in bag filled with gases could be extended as long as 11 d, which was increased by 5 d (83%) compared with that in the control group. The disinfection by sodium hypochlorite could significantly reduce the initial bacterial colony number and improve the sensory quality of fresh-cut grass carp belly, as well as extend the shelf life of refrigerated and modified gas-packed grass carp belly.展开更多
Objective To study the anti-inflammatory effect of a poly-prescription of traditional Chinese medicine GK001.Methods 1.Inhibitory effect on pain in mice:The pain was induced by i.p.0.2 ml of 0.6% HAc per mouse 1 h pos...Objective To study the anti-inflammatory effect of a poly-prescription of traditional Chinese medicine GK001.Methods 1.Inhibitory effect on pain in mice:The pain was induced by i.p.0.2 ml of 0.6% HAc per mouse 1 h post dosing GK001.The writhing numbers of mice were recorded in 10 minutes and the inhibitory rate of pain was calculated compared with the control group.2.Antipyretic effect In single dose experiment 15 healthy rabbits weighing 1.7-2.8 kg with body temperature(BT)measured in the experiment day meeting to the requirements were selected for the experiment and divided into 5 groups(3 in each group),which were dosed orally with GK001 and 1 h later followed by i.p.injection of 40 EU bacterial endotoxin standard·kg-1.Then,the BT of rabbits was measured every 30 min during 1-3 h after administration.The difference between the highest BT post-dose and the average BT pre-dose was calculated.In multi-dose experiment rabbits were selected and grouped as well as received i.p.endotoxin in the same way as above,but were administered with GK001 for consecutive 5 day.3.Bacteriostatic effect.The antibacterial activities of GK001 on Bacillus Pumilus,Bacillus Subtilis and Micrococcus Luteus were measured in vitro at concentrations of 0.125-1.0 g·mL-1.Results 1.The GK001 showed a significant and dose-dependent pain-suppressant effect,with inhibitory rate being 45.2%,31.2% and 20.8% at high,medium and low dose,respectively(P<0.05).2.Both single and multiple administration of GK001 had no effect on rabbit pyrogen response caused by endotoxin.3.GK001 had bacteriostatic effects on the aforementioned 3 bacteria significantly and in dose-dependent fashion.Conclusions GK001 has analgesic and in vitro antibacterial but no antipyretic effects.展开更多

CSFL caused by spinal dural defect is a common complication of spinal surgery,which need repair such as suture or sealants.However,low intracranial pressure symptoms,wound infection and prolonged hospital associated w...CSFL caused by spinal dural defect is a common complication of spinal surgery,which need repair such as suture or sealants.However,low intracranial pressure symptoms,wound infection and prolonged hospital associated with pin-hole leakage or loose seal effect were often occurred after surgical suture or sealants repair.Stable,pressure resistance and high viscosity spinal dural repair patch in wet environment without suture or sealants was highly needed.Herein,a bioactive patch composed of alginate and polyacrylamide hydrogel matrix cross-linked by calcium ions,and chitosan adhesive was proposed.This fabricated patch exhibits the capabilities of promoting defect closure and good tight seal ability with the bursting pressure is more than 790 mm H2O in wet environment.In addition,the chitosan adhesive layer of the patch could inhibit the growth of bacterial in vitro,which is meaningful for the postoperative infection.Furthermore,the patch also significantly reduced the expression of GFAP,IBA-1,MBP,TNF-α,and COX-2 in early postoperative period in vivo study,exerting the effects of anti-inflammatory,analgesic and adhesion prevention.Thus,the bioactive patch expected to be applied in spinal dural repair with the good properties of withstanding high pressure,promoting defect closure and inhibiting postoperative infection.展开更多
The effects of melittin on growth and bacteriostasis of four pathogens were extensively investigated using scanning electron microscopy(SEM) and transmission electron microscopy(TEM).The results indicated that the mel...The effects of melittin on growth and bacteriostasis of four pathogens were extensively investigated using scanning electron microscopy(SEM) and transmission electron microscopy(TEM).The results indicated that the melittin had a marked bacteriostatic effect on the four pathogenic bacteria.Among these,E.cacotowora was influenced most powerfully and quickly,the yeast and F.fulva were the second,and the S.aureus was inhibited by a low concentration but was killed by a high concentration.It was observed in the experiments that melittin killed pathogenic bacteria in three ways.One was by pore formation.After integrating melittin into the plasma membrane,a vacuole was formed then penetrated,resulting in bacterial content leakage.The vacuole also experienced plasmolysis and the growing cavity destroyed the membrane.A third effect was the formation of vacuoles in the cells which induced the pycnosis of the cytoplasm resulting in a cell death.The mechanism of melittin bacteriostasis was the result of integrating melittin with phospholipod double layers of the plasmalemma and the endomembranes.展开更多

In this study, we proposed a practical ion substitution method to synthesize in situ silver phosphate on the surface of a two-level, threedimensional chitosan/nano-hydroxyapatite scaffold. A release test of silver ions in a phosphate buffered saline(PBS) solution was performed to demonstrate that silver ions were released continuously from the silver phosphate during the initial 6 days of the study. The antibacterial property and cytocompatibility of the scaffolds treated with different concentrations of silver nitrate solution were assessed by in vitro assays with Escherichia coli and MC3T3-E1, respectively. The ability of the silver-containing scaffolds to induce bacteriostasis was confirmed by the inhibition zone(15 mm) and high bactericidal rate([99 %). Cell proliferation, morphology and the alkaline phosphatase activity of MC3T3-E1 cultured on the scaffold with low silver phosphate contents were comparable with those cultured on control samples.展开更多

Methods: Clinical observation on 447 cases of asthma was done in double- or single-blinded control method, and the therapeutic effect and safety of JXT were analysed. The effect of JXT in anti-asthma, expectorant, antisepsis, hypoxia tolerance was studied, and its acute and chronic toxicity was also examined. Results: The clinical control rate in patients treated with JXT was 38.2%, the markedly effective rate 31.2% and the total effective rate 93.4%, whereas in patients treated with Oleum Vitex Negundo, the respective rates were 13.3%, 20.0% and 68.5%. Results of pulmonary function and IgE measurement were consistent with clinical effect. Experimental study showed that JXT has the actions of anti-asthma, expectorant, antisepsis and hypoxia tolerance enhancement, with its LD50 (40.48±5.17) g/kg. Conclusion: JXT is a new Chinese herbal preparation of good effect, with less toxic-side effect for asthma treatment.展开更多

When combined with rhizoma coptidis, the MIC of folium isatidis and flos poprli were 1/8 extracting liquid and 1/16 extracting liquid.[Conclusion] When rhizoma coptidis was combined with folium isatidis or flos poprli, the FIC values were 2 and 0.625, performing independent action and additive effect, respectively.

文摘At present,effective fixation and anti-infection implant materials represent the mainstay for the treatment of open fractures.However,external fixation can cause nail tract infections and is ineffective for fixing small fracture fragments.Moreover,closed reduction and internal fixation during the early stage of injury can lead to potential bone infection,conducive to bone nonunion and delayed healing.Herein,we designed a bone adhesive with anti-infection,osteogenic and bone adhesion fixation properties to promote reduction and fixation of open fractures and subsequent soft tissue repair.It was prepared by the reaction of gelatin(Gel)and oxidized starch(OS)with vancomycin(VAN)-loaded mesoporous bioactive glass nanoparticles(MBGNs)covalently cross-linked with Schiff bases.Characterization and adhesion experiments were conducted to validate the successful preparation of the Gel-OS/VAN@MBGNs(GOVM-gel)adhesive.Meanwhile,in vitro cell experiments demonstrated its good antibacterial effects with the ability to stimulate bone marrow mesenchymal stem cell(BMSCs)proliferation,upregulate the expression of alkaline phosphatase(ALP)and osteogenic proteins(RunX2 and OPN)and enhance the deposition of calcium nodules.Additionally,we established a rat skull fracture model and a subcutaneous infection model.The histological analysis showed that bone adhesive enhanced osteogenesis,and in vivo experiments demonstrated that the number of inflammatory cells and bacteria was significantly reduced.Overall,the adhesive could promote early reduction of fractures and antibacterial and osteogenic effects,providing the foothold for treatment of this patient population.
文摘: In order to make clear the functions of plant volatile organic compounds (VOCs) on bacteriostasis and air decontamination, we analyzed the composition and content of VOCs in Pinus tabulaeformis Carr., P. bungeana Zucc., Sabina chinensis Antoine, Picea koraiensis Nakai, and Cedrus deodara G. Don under near-natural conditions using the thermal-desorption cold trap gas chromatography/mass spectrometer technique. The effects of the VOCs on airborne microorganisms were investigated using the method of natural sedimentation. Results showed that the major VOCs were as follows: limonene, β-pinene, α-pinene, and α-caryophyllene in Pinus tabulaeformis and P. bungeana; limonene, borneol acetate, β-pinene, myrcene, and tricylene in S. chinensis; limonene, α-pinene, myrcene, camphene, and β-pinene in Picea koraiensis; and limonene, 2, (10)-pinene, α-pinene, and myrcene in C. deodara. These VOCs and the corresponding foliar extracts inhibited the growth of bacteria and stimulated the growth of fungi. Experimental data using monomers of the VOCs demonstrated that limonene, β-pinene, and three aldehydes could significantly inhibit bacterial growth, suggesting an inhibitory effect of VOCs on the growth of airborne microorganisms in the five conifer species. The bacteriostasis and air-decontaminating effects of plant VOCs are further discussed in terms of their chemical composition.

文摘Madecassoside is a natural active component extracted from Centella asiatica.In recent years,a large number of studies have reported that madecassoside has a variety of biological activities,such as anticancer,anti-inflammatory and antibacterial effects,prevention and treatment of cardiovascular diseases,nerve damage,visceral damage and arthritis,and other pharmacological effects.In this paper,the pharmacological action and mechanism of madecassoside were reviewed to provide a theoretical basis for further research of madecassoside and drug development.
文摘Objective: To explore the antibacterial activity of combined use of Shuanghuanglian and cefoperazone sulbactam sodium on resistant strains of Pseudomonas aeruginosa. Methods: The Pseudomonas aeruginosa strains which were sensitive and resistant to cefoperazone sulbactam sodium were selected to prepare different test bacterial solutions respectively;The experimental liquid of Shuanghuanglian and Cefoperazone Sulbactam Sodium were prepared separately and set as different test groups and control groups;The Drug Sensitivity Tests of Shuanghuanglian and cefoperazone sulbactam sodium at different concentration gradients which were used alone or used in combination were carried out for different strains with sensitivity and resistance, And use standard entry as a reference control. Result: The results of drug sensitivity test of Shuanghuanglian combined with Cefoperazone-Sulbactam sodium against the resistant strains of Pseudomonas aeruginosa were compared with the results of drug sensitivity test of the two separately used, and the difference was statistically significant (P 〈 0.05) [The drug sensitivity test results of Shuanghuanglian and cefoperazone sulbactam sodium to Pseudomonas aeruginosa resistant strains were statistically significant compared with the drug sensitivity test results of Shuanghuanglian and Cefoperazone Sulbactam Sodium used separately (P 〈 0.05)];There was a dependence between strains and concentration in the effect of the combination of the two drugs. Conclusion: The combination of Shuanghuanglian and cefoperazone sulbactam sodium has synergistic antibacterial or bactericidal effect on Pseudomonas aeruginosa resistant strains. .

文摘[Objective] This study aimed to reveal the therapeutic mechanism of compound sarcandra aerosol, which was exclusively owned by the Second Affiliated Hospital of Guiyang College of Traditional Chinese Medicine. [Method] An inflammation model was established by xylene-induced inflammation test and carrageenan- induced inflammation test to analyze the anti-inflammatory effect of compound sarcandra aerosol. By bacteriostasis test in vitro, the antibacterial effect of compound sarcandra aerosol against five common pathogens of pharyngitis was investigated. Blood samples were collected from Wistar rats in different compound sarcandra aerosol groups and control group to compare blood routine indicators and interleukin-1 (IL-1) levels. [Result] Three different concentrations of compound sarcandra aerosol could reduce degrees of xylene-induced ear edema in mice and carrageenan- induced paw edema in rats, but the anti-inflammatory effect of compound sarcandra aerosol was reduced as the concentration declined. In bacteriostasis test in vitro, the minimum inhibitory concentration of compound sarcandra aerosol against Streptococcus pneumoniae, Streptococcus hemolytis, Corynebacterium diphtheriae, Staphylococcus aureus and Salmonella typhimurium was 76, 105 38, 65 and 30 mg/ml, respectively. Compound sarcandra aerosol reduced white blood cell count, neutrophil count and lymphocyte percentage in pharyngitis model rats. Moreover, interleukin-1 level in watermelon frost lozenge group and different compound sarcandra aerosol groups was lower compared with control group. [Conclusion] Compound sarcandra aerosol can effectively treat pharyngitis by exerting anti-inflammatory and antibacterial effect and reducing interleukin-1 level. This study laid a solid foundation for clinical application of compound sarcandra aerosol.

文摘With fresh-cut grass carp belly as the test material, the antibacterial con- ditions of a disinfection agent sodium hypochlorite were optimized. In addition, the shelf lives of various grass carp products were compared after disinfection, so as to provide certain basis for the processing of fresh-cut grass carp products. The re- sults showed that the optimal disinfection conditions of sodium hypochlorite were as follows: concentration 300 mg/L, soak time 5 min and solid-liquid ratio 1 g : 5 ml. Under the optimal disinfection conditions, the inhibition rates of total bacteria and Pseudomonas reached 83% and 81%, respectively. The shelf life of refrigerated fresh-cut grass carp belly disinfected by sodium hypochlorite and packed in bag filled with gases could be extended as long as 11 d, which was increased by 5 d (83%) compared with that in the control group. The disinfection by sodium hypochlorite could significantly reduce the initial bacterial colony number and improve the sensory quality of fresh-cut grass carp belly, as well as extend the shelf life of refrigerated and modified gas-packed grass carp belly.
文摘Objective To study the anti-inflammatory effect of a poly-prescription of traditional Chinese medicine GK001.Methods 1.Inhibitory effect on pain in mice:The pain was induced by i.p.0.2 ml of 0.6% HAc per mouse 1 h post dosing GK001.The writhing numbers of mice were recorded in 10 minutes and the inhibitory rate of pain was calculated compared with the control group.2.Antipyretic effect In single dose experiment 15 healthy rabbits weighing 1.7-2.8 kg with body temperature(BT)measured in the experiment day meeting to the requirements were selected for the experiment and divided into 5 groups(3 in each group),which were dosed orally with GK001 and 1 h later followed by i.p.injection of 40 EU bacterial endotoxin standard·kg-1.Then,the BT of rabbits was measured every 30 min during 1-3 h after administration.The difference between the highest BT post-dose and the average BT pre-dose was calculated.In multi-dose experiment rabbits were selected and grouped as well as received i.p.endotoxin in the same way as above,but were administered with GK001 for consecutive 5 day.3.Bacteriostatic effect.The antibacterial activities of GK001 on Bacillus Pumilus,Bacillus Subtilis and Micrococcus Luteus were measured in vitro at concentrations of 0.125-1.0 g·mL-1.Results 1.The GK001 showed a significant and dose-dependent pain-suppressant effect,with inhibitory rate being 45.2%,31.2% and 20.8% at high,medium and low dose,respectively(P<0.05).2.Both single and multiple administration of GK001 had no effect on rabbit pyrogen response caused by endotoxin.3.GK001 had bacteriostatic effects on the aforementioned 3 bacteria significantly and in dose-dependent fashion.Conclusions GK001 has analgesic and in vitro antibacterial but no antipyretic effects.

文摘Objective:To investigate the inhibitory effect of calcium hydroxide on methicillin-resistant Staphylococcus aureus and the related inhibition mechanism.Methods:To determine the minimum inhibitory concentration of calcium hydroxide using microplate dilution method;to compare the effects of calcium hydroxide at 8MIC,MIC,1/4MIC and 0 concentrations on MRSA using growth curve method;to determine the effects of calcium hydroxide on the cell membrane of methicillin-resistant Staphylococcus aureus using calcium xanthophyll and propidium iodide fluorescence staining The effect of calcium hydroxide on the morphology of methicillin-resistant Staphylococcus aureus was observed by scanning electron microscopy;the inhibition mechanism of calcium hydroxide on MRSA was investigated by sodium dodecyl sulfate polyacrylamide gel electrophoresis of protein bands.Results:The MIC of calcium hydroxide on MRSA was 3.125 mg/mL;the fluorescence intensity showed significant changes after co-culture of calcium hydroxide with bacteria;MRSA appeared to be significantly crumpled and broken in the presence of calcium hydroxide;the SDS-PAGE experimental bands indicated that the protein inside the bacteria decreased accordingly with the increase of calcium hydroxide concentration.Conclusion:Calcium hydroxide has a significant inhibitory effect on the growth of MRSA,and its bactericidal mechanism may be related to the destruction of bacterial body structure.

文摘CSFL caused by spinal dural defect is a common complication of spinal surgery,which need repair such as suture or sealants.However,low intracranial pressure symptoms,wound infection and prolonged hospital associated with pin-hole leakage or loose seal effect were often occurred after surgical suture or sealants repair.Stable,pressure resistance and high viscosity spinal dural repair patch in wet environment without suture or sealants was highly needed.Herein,a bioactive patch composed of alginate and polyacrylamide hydrogel matrix cross-linked by calcium ions,and chitosan adhesive was proposed.This fabricated patch exhibits the capabilities of promoting defect closure and good tight seal ability with the bursting pressure is more than 790 mm H2O in wet environment.In addition,the chitosan adhesive layer of the patch could inhibit the growth of bacterial in vitro,which is meaningful for the postoperative infection.Furthermore,the patch also significantly reduced the expression of GFAP,IBA-1,MBP,TNF-α,and COX-2 in early postoperative period in vivo study,exerting the effects of anti-inflammatory,analgesic and adhesion prevention.Thus,the bioactive patch expected to be applied in spinal dural repair with the good properties of withstanding high pressure,promoting defect closure and inhibiting postoperative infection.
文摘The effects of melittin on growth and bacteriostasis of four pathogens were extensively investigated using scanning electron microscopy(SEM) and transmission electron microscopy(TEM).The results indicated that the melittin had a marked bacteriostatic effect on the four pathogenic bacteria.Among these,E.cacotowora was influenced most powerfully and quickly,the yeast and F.fulva were the second,and the S.aureus was inhibited by a low concentration but was killed by a high concentration.It was observed in the experiments that melittin killed pathogenic bacteria in three ways.One was by pore formation.After integrating melittin into the plasma membrane,a vacuole was formed then penetrated,resulting in bacterial content leakage.The vacuole also experienced plasmolysis and the growing cavity destroyed the membrane.A third effect was the formation of vacuoles in the cells which induced the pycnosis of the cytoplasm resulting in a cell death.The mechanism of melittin bacteriostasis was the result of integrating melittin with phospholipod double layers of the plasmalemma and the endomembranes.

文摘Bone tissue scaffolds based on bioactive polymer–hydroxyapatite composites have caused infections that seriously limit their extended application. In this study, we proposed a practical ion substitution method to synthesize in situ silver phosphate on the surface of a two-level, threedimensional chitosan/nano-hydroxyapatite scaffold. A release test of silver ions in a phosphate buffered saline(PBS) solution was performed to demonstrate that silver ions were released continuously from the silver phosphate during the initial 6 days of the study. The antibacterial property and cytocompatibility of the scaffolds treated with different concentrations of silver nitrate solution were assessed by in vitro assays with Escherichia coli and MC3T3-E1, respectively. The ability of the silver-containing scaffolds to induce bacteriostasis was confirmed by the inhibition zone(15 mm) and high bactericidal rate([99 %). Cell proliferation, morphology and the alkaline phosphatase activity of MC3T3-E1 cultured on the scaffold with low silver phosphate contents were comparable with those cultured on control samples.
文摘Objective: To evaluate the therapeutic effect of Juanxiao Tablet (JXT) in treating asthma. Methods: Clinical observation on 447 cases of asthma was done in double- or single-blinded control method, and the therapeutic effect and safety of JXT were analysed. The effect of JXT in anti-asthma, expectorant, antisepsis, hypoxia tolerance was studied, and its acute and chronic toxicity was also examined. Results: The clinical control rate in patients treated with JXT was 38.2%, the markedly effective rate 31.2% and the total effective rate 93.4%, whereas in patients treated with Oleum Vitex Negundo, the respective rates were 13.3%, 20.0% and 68.5%. Results of pulmonary function and IgE measurement were consistent with clinical effect. Experimental study showed that JXT has the actions of anti-asthma, expectorant, antisepsis and hypoxia tolerance enhancement, with its LD50 (40.48±5.17) g/kg. Conclusion: JXT is a new Chinese herbal preparation of good effect, with less toxic-side effect for asthma treatment.
