Biliverdin Reductase-A correlates with inducible nitric oxide synthasein in atorvastatin treated aged canine brain 被引量:2

Alzheimer’s disease is a neurodegenerative disorder characterized by progressive cognitive impairment and neuropathology. Recent preclinical and epidemiological studies proposed statins as a possible therapeutic drug for Alzheimer’s disease, but the exact mechanisms of action are still unknown. Biliverdin reductase-A is a pleiotropic enzyme involved in cellular stress responses. It not only transforms biliverdin-IX alpha into the antioxidant bilirubin-IX alpha but its serine/threonine/ tyrosine kinase activity is able to modulate cell signaling networks. We previously reported the beneficial effects of atorvastatin treatment on biliverdin reductase-A and heme oxygenase-1 in the brains of a well characterized pre-clinical model of Alzheimer’s disease, aged beagles, together with observed improvement in cognition. Here we extend our knowledge of the effects of atorvastatin on inducible nitric oxide synthase in parietal cortex, cerebellum and liver of the same animals. We demonstrated that atorvastatin treatment (80 mg/day for 14.5 months) to aged beagles selectively increased inducible nitric oxide synthase in the parietal cortex but not in the cerebellum. In contrast, inducible nitric oxide synthase protein levels were significantly decreased in the liver. Significant positive correlations were found between biliverdin reductase-A and inducible nitric oxide synthase as well as heme oxygenase-1 protein levels in the parietal cortex. The opposite was observed in the liver. Inducible nitric oxide synthase up-regulation in the parietal cortex was positively associated with improved biliverdin reductase-A functions, whereas the oxidative-induced impairment of biliverdin reductase-A in the liver negatively affected inducible nitric oxide synthase expression, thus suggesting a role for biliverdin reductase-A in atorvastatin-dependent inducible nitric oxide synthase changes. Interestingly, increased inducible nitric oxide synthase levels in the parietal cortex were not associated with higher oxidative/nitrosative stress levels. We hypothesize that biliverdin reductase-A-dependent inducible nitric oxide synthase regulation strongly contributes to the cognitive improvement observed following atorvastatin treatment. 展开更多

It has also been demonstrated to have important cytoprotective functions during oxidative stress. The present study aimed to examine the cytoprotective effect of BV on NRK-52 E cells, a proximal tubular cell line derived from rat kidney. Cells were treated with 50 μmol/L cisplatin for 24 h(cisplatin group) or pre-treated with BV for 30 min, then with 50 μmol/L cisplatin for 24 h(cisplatin+BV group). Those given no treatment served as a control. Cell apoptosis was evaluated by flow cytometry and cell viability by Cell Counting Kit-8(CCK-8). The protein expressions of cleaved caspase3, Bax and Bcl-2 were assessed by Western blotting. Reactive oxygen species(ROS) levels were measured using carboxydichlorodihydrofluorescein diacetate(H2DCF). The results showed that cisplatin induced the apoptosis of NRK-52 E cells, decreased cell viability, and increased the formation of ROS by upregulating the expression of cleaved caspase3 and Bax and decreasing Bcl-2 protein expression. These effects could be significantly reversed by pretreatment with BV. It was concluded that BV can protect against cisplatin-induced cell apoptosis through the anti-oxidative effects. 展开更多

As one of the biological endogenous pigments,biliverdin(BV)and its dimethyl ester(BVE)have extremely weak uorescence in solution with quantum yield less than 0.01%.However,the situation reverses with the addition of zinc ions.The strength for uorescence of BVE-Zn^2+ complex is greatly enhanced and uorescence quantum yield can increase to5%.Herein,we studied ultrafast excited state dynamics of BVE-Zn^2+ complex in ethanol,npropanol,and DMSO solutions in order to reveal the mechanism of uorescence quantum yield enhancement.The results show that BVE can form a stable coordination complex with zinc with 1:1 stoichiometry in solution.BVE is structurally and energetically more stable in the complex.Using picosecond time-resolve uorescence and femtosecond transient absorption spectroscopy,we show that smaller non-radiative rate constant of BVE-Zn^2+ complex in DMSO is the key to increasing its uorescence quantum yield and the excited state decay mechanism is also revealed.These results provide valuable information about the uorescence property change after BVE binding to metal ions and may provide a guidance for the study of phytochromes or other uorescence proteins in which BV/BVE acts as chromophores. 展开更多

Background:Biliverdin (BV) has a protective role against ischemia-reperfusion injury (IRI).However,the protective role and potential mechanisms of BV on lung IRI (LIRI) remain to be elucidated.Thus,we aimed to investigate the protective role and potential mechanisms of BV on LIRI.Methods:Lungs were isolated from Sprague-Dawley rats to establish an ex vivo LIRI model.After an initial 15 min stabilization period,the isolated lungs were subjected to ischemia for 60 min,followed by 90 min ofreperfusion with or without BV treatment.Results:Lungs in the I/R group exhibited significant decrease in tidal volume (1.44 ± 0.23 ml/min in I/R group vs.2.41 ± 0.31 ml/min in sham group;P 〈 0.001),lung compliance (0.27 ± 0.06 ml/cmH2O in I/R group vs.0.44 ± 0.09 ml/cmH2O in sham group;P 〈 0.001;1 cmH2O=0.098 kPa),and oxygen partial pressure (PaO2) levels (64.12 ± 12 mmHg in FR group vs.114 ± 8.0 mmHg in sham group;P 〈 0.001;1 mmHg =0.133 kPa).In contrast,these parameters in the BV group (2.27 ± 0.37 ml/min of tidal volume,0.41 ± 0.10 ml/ cmH2O of compliance,and 98.7 ± 9.7 mmHg of PaO2) were significantly higher compared with the I/R group (P =0.004,P 〈 0.001,and P 〈 0.001,respectively).Compared to the I/R group,the contents of superoxide dismutase were significantly higher (47.07 ± 7.91 U/mg protein vs.33.84 ± 10.15 U/mg protein;P =0.005) while the wet/dry weight ratio (P 〈 0.01),methane dicarboxylic aldehyde (1.92 ± 0.25 nmol/mg protein vs.2.67 ± 0.46 nmol/mg protein;P 〈 0.001),and adenosine triphosphate contents (297.05 ± 47.45 nmol/mg protein vs.208.09 ± 29.11 nmol/mg protein;P =0.005) were markedly lower in BV-treated lungs.Histological analysis revealed that BV alleviated LIRI.Furthermore,the expression of inflammatory cytokines (interleukin-1 β,interleukin-6,and tumor necrosis factor-α) was downregulated and the expression of cyclooxygenase-2,inducible nitric oxide synthase,and Jun N-terminal kinase was significantly reduced in BV group (all P 〈 0.01 compared to I/R group).Finally,the apoptosis index in the BV group was significantly decreased (P 〈 0.01 compared to I/R group).Conclusion:BV protects lung IRI through its antioxidative,anti-inflammatory,and anti-apoptotic effects. 展开更多

The electrochemical redox behavior of bilirubin(BR Ⅳ<sub>α</sub>),biliverdin(BV Ⅳ<sub>α</sub>)and theiroxidized product bile-purpurin(Bi-Pu)have been studied by in situ spectroelectrochemical techniques,whichreveals that the transformation of BR Ⅳ<sub>α</sub>■BV Ⅳ<sub>α</sub>■Bi-Pu can be achieved by controlling poten-tials.The kinetic investigation has shown that the heterogeneous electron transfer reactions of the couples ofⅠ/Ⅱ and Ⅳ/Ⅲ were quasi-reversible and irreversible at a clean platinum electrode with the formal heteroge-neous electron transfer rate constants 1.5×10<sup>-4</sup>cm·s<sup>-1</sup>and 4.8×10<sup>-5</sup>cm·s<sup>-1</sup>,respectively. 展开更多

目的制备载抑制性胆绿素还原酶A(BVR)多肽及全氟戊烷(PFP)的聚乳酸-羟基乙酸(PLGA)纳米微球(PPB NBs),评估其体外超声成像效果及与低强度聚焦超声(LIFU)联合使用抑制乳腺癌增殖及转移的能力。材料与方法采用双乳法及碳二亚胺催化法制备P... 目的制备载抑制性胆绿素还原酶A(BVR)多肽及全氟戊烷(PFP)的聚乳酸-羟基乙酸(PLGA)纳米微球(PPB NBs),评估其体外超声成像效果及与低强度聚焦超声(LIFU)联合使用抑制乳腺癌增殖及转移的能力。材料与方法采用双乳法及碳二亚胺催化法制备PPB NBs,观察其形态大小、粒径、稳定性、表面电位及多肽包封率,并观察不同LIFU辐照时间的显影效果、载PFP的PLGA纳米微球(PP NBs)的细胞毒性,检测PPB NBs联合LIFU的抗增殖能力、抑制肿瘤细胞迁移和侵袭的能力。结果PPB NBs乳液呈乳白色,纳米微球为粒径均一的球形,粒径(195.60±8.79)nm,7 d内粒径变化范围在24.50 nm内,多分散系数为0.064±0.030,表面电位为(-7.58±0.50)mV,多肽包封率为(82.94±1.10)%。二维超声及超声造影发现经LIFU辐照120 s为最佳显像时间,可证明PPB NBs成功包载PFP。PP NBs细胞毒性弱,细胞存活率在浓度最高(0.8 mg/ml)时也超过75%。PPB NBs抗肿瘤能力呈浓度依赖性,多肽浓度为1 mg/ml时,细胞抑制率达(65.40±0.02)%。与其他组相比,PPB NBs联合LIFU可显著抑制肿瘤细胞迁移及侵袭能力。结论本实验成功制备PPB NBs,其不但可以成功显像,且可以利用LIFU定点释放BVR多肽,从而抑制乳腺癌细胞的增殖与转移,达到有效的抗肿瘤作用。 展开更多

为了提高赤水乌骨鸡的生产效益,并完善赤水乌骨鸡蛋品质研究内容,随机选取314只8月龄赤水乌骨鸡母鸡,对其进行SLCO1B3基因检测,将其分为绿壳纯合型、绿壳杂合型、白壳纯合型,并各收集60个鸡蛋,测定其蛋重、蛋壳颜色、蛋形指数、蛋壳强... 为了提高赤水乌骨鸡的生产效益,并完善赤水乌骨鸡蛋品质研究内容,随机选取314只8月龄赤水乌骨鸡母鸡,对其进行SLCO1B3基因检测,将其分为绿壳纯合型、绿壳杂合型、白壳纯合型,并各收集60个鸡蛋,测定其蛋重、蛋壳颜色、蛋形指数、蛋壳强度、蛋白高度、蛋黄颜色、哈氏单位、蛋黄重量等指标,同时测定蛋壳中胆绿素及原卟啉含量。结果表明:在蛋黄颜色和蛋壳指数上,3种基因型无显著性差异(P>0.05),且蛋壳指数均在1.3~1.35之间;在蛋壳强度上,绿壳纯合型显著低于绿壳杂合型及白壳纯合型(P<0.05);在蛋重上,绿壳杂合型显著高于白壳纯合型和绿壳纯合型;在蛋白高度上,绿壳纯合型与白壳纯合型显著高于绿壳杂合型(P<0.05),绿壳纯合型蛋黄重量显著低于绿壳杂合型(P<0.05),且白壳纯合型的蛋黄重量介于两者之间,与另外两种基因型不存在显著差异(P>0.05);在哈氏单位上,绿壳纯合型及白壳纯合型显著高于绿壳杂合型(P<0.05)。在胆绿素及原卟啉含量上,绿壳纯合型蛋壳胆绿素含量显著高于绿壳杂合型蛋壳胆绿素含量及白壳纯合型胆绿素蛋壳含量(P<0.05),绿壳杂合型蛋壳胆绿素含量显著高于白壳纯合型蛋壳胆绿素含量;白壳纯合型蛋壳原卟啉含量显著高于绿壳杂合型蛋壳原卟啉含量及绿壳纯合型原卟啉蛋壳含量(P<0.05),绿壳杂合型蛋壳原卟啉含量显著高于绿壳纯合型蛋壳原卟啉含量。结果提示:绿壳纯合型赤水乌骨鸡的鸡蛋蛋品质优于绿壳杂合型赤水乌骨鸡,市场价值高于白壳纯合型赤水乌骨鸡,绿壳纯合型赤水乌骨鸡具有更高的开发利用价值。 展开更多

本研究分析比较了武定鸡绿壳蛋系和粉壳蛋系蛋壳色素的种类及含量,发现绿壳蛋系的蛋壳色素可能是由原卟啉和胆绿素组成的,而粉壳蛋系的蛋壳色素可能只由原卟啉组成,并且两种蛋壳色素在绿壳蛋鸡群体内个体间的分布差异极显著(P<0.01)。

目的探讨胆绿素(biliverdin,BV)对脂多糖诱导RAW264.7巨噬细胞NLRP3炎性体激活的影响及其作用机制。方法用不同浓度胆绿素(10、20、30μmol/L)处理小鼠RAW264.7巨噬细胞30min后再用LPS(1μg/ml)处理细胞6h,ELISA法检测培养上清液IL-β和... 目的探讨胆绿素(biliverdin,BV)对脂多糖诱导RAW264.7巨噬细胞NLRP3炎性体激活的影响及其作用机制。方法用不同浓度胆绿素(10、20、30μmol/L)处理小鼠RAW264.7巨噬细胞30min后再用LPS(1μg/ml)处理细胞6h,ELISA法检测培养上清液IL-β和IL-18中的分泌量。30μmol/L胆绿素处理细胞30min后再用LPS处理细胞6h,Western blot法检测胞内NLPR3、caspase-1、ASC和磷酸化IκB(p-IκB)的蛋白表达水平;NF-κB抑制剂BAY11-7082处理细胞1h后,然后给予细胞30μmol/L胆绿素孵育30min,经1μg/ml LPS孵育6h,ELISA法检测细胞培养上清液中IL-β、IL-18的表达水平。结果 (1)在受到LPS作用后,RAW264.7巨噬细胞分泌IL-β、IL-18水平显著升高;而在预先给予胆绿素后,抑制了LPS诱导的IL-β、IL-18表达,并且呈剂量依赖性(P<0.05)。(2)与空白对照组相比,LPS处理巨噬细胞6h后,NLRP3炎性体各组分蛋白和pIκB蛋白表达明显上调(P均<0.05),在同时间点,巨噬细胞在LPS处理前经过胆绿素预处理后,NLRP3炎性体各组分蛋白和p-IκB蛋白表达都显著降低(均P<0.05)。(3)与LPS组相比,BAY+胆绿素明显抑制了LPS诱导的IL-β、IL-18的表达(P<0.05),并且BAY和胆绿素的对于LPS诱导炎性反应对的联合抑制作用显著的高于BAY或者胆绿素的单独作用(P<0.05)。结论在RAW264.7巨噬细胞中,胆绿素可以通过抑制NF-κB的活性进而抑制NLRP3炎性体的形成,从而发挥抗炎作用。 展开更多
