Forty-five specimens from patients with cervical cancer (stageⅠ-Ⅲ), 32 specimens from patients with cervical intraepithelial neoplasia (CIN) (Ⅰ, Ⅱ) and 20 normal cervical samples from patients with hysteromyoma were collected in Department of Pathology for comparison. The expression levels of SHP-2 and IFN-β proteins were detected by using immunohistochemistry. The mRNA expression level of SHP-2 was detected by using quantitative real-time polymerase chain reaction (PCR). HPVs were detected by HPV GenoArray Test. The Spearman correlation was used to compare the expression level of SHP-2 in HPV infected cervical cancer vs non-HPV infected normal cervix. The level of SHP-2 protein expression in the cancer tissues (88.8%) was significantly higher than in CIN tissues (62.5%) and normal cervixes (45%) (P<0.05 and P<0.05, respectively). The SHP-2 mRNA levels in the cancer tissues were upregulated as compared with those in the normal cervixes (P<0.05). Twenty-one (46.7%) cervical cancers, 25 (78.1%) CINs and 17 (85%) normal cervixes showed IFN-β positive staining in cytoplasm. There was statistically significant difference in the expression rate of IFN-β between cervical cancer and normal cervix (χ2=8.378, P<0.05) as well as between cervical cancer and CIN (χ2=7.695, P<0.05). HPV16/18 infections could be found in normal cervixs (15%), CINs (68.7%) and cervical cancers (84.4%). There was a correlation between HPV infection and SHP-2 expression in cervical cancer (rs=0.653, P<0.05). SHP-2 may be a useful prognostic and diagnostic indicator for HPV infected cervical cancer. In cervical cancers, SHP-2 mRNA and protein overexpression was associated with IFN-β lower-expression.展开更多

Methods:Antibody against human STAT2 was confirmed by plasmids transient transfection and Western blot Immunohistochemistry was used to delect STAT2 expression in the cervical biopsies by using the confirmed antibody against STAT2 as the primary antibody.Results:It was found that the overall rate of positive STAT2 expression in the cervicitis,dysplasia and cenical cancer groups were 38.5%,69.49%and 76.991,respectively.The STAT2 levels are significantly increased in premalignant dysplasia and cervical cancer,as compared lo cervicitis(P【 0.05). Noticeably,STAT2 signals were mainly found in the cytoplasm,implying that STAT2 was not biologically active.Conclusions:These findings reveal an association between cenical cancer progression and augmented STAT2 expression.In conclusion.STAT2 increase appears to be an early detectable cellular event in cenical cancer development.展开更多

Focke has been traditionally used to treat cancer in Asian countries for centuries. In the present study, transplanted U14 cervical cancer mouse model was used to evaluate the antitumor and immunomodulatory activity of Duchesnea phenolic fraction (DPF). ELISA and RIA assay were employed to measured the serum concentration of Th1/Th2 cytokines (IL-2, IL-4, IFN-γ and TNF-α). Administration with 0.25 g/kg, 0.5 g/kg and 1 g/kg DPF significantly reduced the tumor weight by 34.37%, 43.89% and 56.28%, respectively, as compared to the tumor control group. Furthermore, the serum level of IL-2, IFN-γ and TNF-α increased and IL-4 level decreased in a dose-dependent manner during DPF treatment, indicating that the antitumor activity of DPF may be associated with the decrease of TNF-α level and restoration of the balance of Th1/Th2 cell responses. These data suggested that DPF, a mixture of plant polyphenols, had potent anticancer activity which was in part accomplished by its immunomodulatory ability.展开更多
BACKGROUND Cervical cancer is a gynecological malignancy common in middle-aged and older patients,with a high mortality rate.Spondin-2 is an extracellular matrix protein that involved in innate and acquired immune res...BACKGROUND Cervical cancer is a gynecological malignancy common in middle-aged and older patients,with a high mortality rate.Spondin-2 is an extracellular matrix protein that involved in innate and acquired immune responses.Herein,we investigated the relationship between serum Spondin-2 expression,tumor invasion and infiltration,and immune response in patients with cervical cancer and provided a theoretical basis for clinical practice.AIM To investigate the relationship between serum Spondin-2 expression and cervical cancer-related indicators.METHODS Overall,147 patients with cervical cancer who were admitted to our institution between January 2019 and August 2019 were assigned to the cervical cancer group,and 92 patients with benign uterine lesions and 86 healthy individuals were assigned to the benign and control groups,respectively.In each group,serum Spondin-2 expression was measured,and the receiver operating characteristic(ROC)curve was determined.Patients with cervical cancer were classified into high or low Spondin-2 groups depending on the Spondin-2 threshold value used for diagnosing cervical cancer.Patient’s clinical data were collected to compare the clinicopathologic characteristics,immune cytokine levels,and prognosis of patients with varying Spondin-2 expression levels.RESULTS The expression level of serum Spondin-2 was significantly higher in the cervical cancer group than in the benign and control groups(P<0.05).According to the ROC curve,the cutoff value of Spondin-2 used in the diagnosis of cervical carcinoma was 25.68±7.11μg/L.The proportion of patients with Federation of Gynecology and Obstetrics stage III,nerve invasion,vascular invasion,and lymph node metastasis was higher in the high Spondin-2 group than in the low Spondin-2 group(P<0.05).Interleukin-5(IL-5)and IL-4 Levels were higher in the high Spondin-2 group than in the low Spondin-2 group.In contrast,IL-2 and tumor necrosis factor-αlevels were lower in the high Spondin-2 group than in the low Spondin-2 group(P<0.05).After 3 years of follow-up,progression-free survival and overall survival were significantly shorter in the high Spondin-2 group than in the low Spondin-2 group(P<0.05).CONCLUSION The expression of serum Spondin-2 is upregulated in patients with cervical carcinoma and is related to tumor invasion and infiltration,antitumor immune response,and prognosis.展开更多

Cervical cancer specimens were obtained from 47 pre-menopausal patients. The phase of menstrual cycle was determined by case history and HE staining of uterine endometrium. COX-2 was immunohistochemically stained by SABC staining and the staining intensity was determined with computerized image analysis system. Hela cells were incubated with alcohol, E2, E2+MPA, MPA for 12, 24 and 48 h respectively. The expression of COX-2 in Hela cells was detected by Western blotting and reverse transcriptase-polymerase chain reaction (RT-PCR). Our results showed that the expression of COX-2 was significantly higher during proliferative phase than secretory phase (P〈0.05), but there was no difference in the positive rate between proliferative phase and secretory phase (P〉0.05). Incubation with E2 could significantly enhance the expression of COX-2 continually. On the contrary, E2+MPA and MPA alone could decrease the expression of COX-2 as compared with the control and E2 group (P〈0.05 and P〈0.01 respectively). It is concluded that the expression of COX-2 in cervical cancer of pre-menopausal patients and Hela cells was regulated by estrogen/progestogen.展开更多
