Subtype B aMPV has recently become the dominant epidemic strain in China. We developed an attenuated aMPV subtype B strain by serial passaging in Vero cells and evaluated its safety and efficacy as a vaccine candidate. The safety test showed that after the 30th passage, the LN16-A strain was fully attenuated, as clinical signs of infection and histological lesions were absent after inoculation. The LN16-A strain did not revert to a virulent strain after five serial passages in chickens. The genomic sequence of LN16-A differed from that of the parent wild-type LN16(wtLN16) strain and had nine amino acid mutations. In chickens, a single immunization with LN16-A induced robust humoral and cellular immune responses, including the abundant production of neutralizing antibodies, CD4^(+) T lymphocytes, and the Th1(IFN-γ) and Th2(IL-4 and IL-6)cytokines. We also confirmed that LN16-A provided 100% protection against subtype B aMPV and significantly reduced viral shedding and turbinate inflammation. Our findings suggest that the LN16-A strain is a promising live attenuated vaccine candidate that can prevent infection with subtype B aMPV.展开更多

A widely held hypothesis is that brain size is positively correlated with behavioral plasticity. Methods: A total of 71 Chestnut Thrushes(Turdus rubrocanus) were caught in the wild population. We quantified behavior plasticity of activity of individuals measured in the same cage across two contexts(common and with a novel object stimulation), using a random regression analysis. We then investigated whether head volume(a proxy for brain size) was associated with behavioral plasticity in activity level using Spearman rank-order correlation. Results: We found no significant evidence that activity plasticity was associated with relative head volume. There was no sex difference in head volume or in variance in head volume. Conclusions: We speculate that the absence of an association between brain volume and activity behavior plasticity may result from the inaccuracy of using external skull measurements to estimate brain size, or from a particular part of the brain being responsible for plasticity in activity level.展开更多

However, it was nearly eliminated by chestnut blight caused by the Ascomycetous fungus <em>Cryphonectria parasitica</em>. Identification of compounds specific to species and backcross hybrids may help further refine disease resistance breeding and testing. Phenolic compounds produced by plants are significant to their defense mechanisms against fungal pathogens. Therefore, an analytical platform has been developed to estimate the total phenolic content in leaf tissues of the American chestnut, Chinese chestnut (<em>Castanea mollissima</em>), and their backcross breeding generations (B3F2 and B3F3) using the Folin-Ciocalteu reagent assay with UV/Vis spectrophotometry which may be used to predict blight resistance. Adsorption (765 nm) results from leaf tissue extraction in methanol/water (95%:5% v/v) and pH 2, show that the variations among these four tree species are significant (ANOVA p = 2.3 × 10-7). The kinetics of phenolic compound solid-liquid extraction was elaborated using Peleg, second order, Elovich, and power law models. In addition, extensive analysis using headspace solid phase microextraction (SPME) gas chromatography and mass spectrometry was conducted to identify volatile organic compounds (VOCs) from the leaf of American chestnut, Chinese chestnut, and their backcross hybrids B3F2 and B3F3. A total of 67 VOCs were identified among all chestnut types. Many of the metabolites associated with the Chinese chestnut have been reported to have antifungal properties, whereas the native and hybrid American chestnut metabolites have not. Most of the antifungal metabolites showed the strongest efficacy towards the Ascomycota phylum. A partial least squares discriminant analysis (PLS-DA) model (R2X = 0.884, R2Y = 0.917, Q2 = 0.584) differentiated chestnut species and hybrids within the first five principal component (PCs).展开更多

[Methods] The ‘Yanshanzaofeng’ chestnuts in four orchards located in Qianxi, Qianan, Funing, and Qinglong of Hebei Science and Technology Normal University were selected. The climate and soil nutrient conditions of the four orchards were investigated. Growth indicators such as branch length and diameter, nut weight, and internal quality indicators such as starch, fat, and protein content were analyzed. Principal component analysis was conducted on nine climate factors and twelve soil factors in the four chestnut orchards to identify the most closely related ecological factors influencing chestnut growth and quality. [Results] (i) Different orchards had varying climate conditions, with Qianan orchard having higher rainfall than the others, Funing orchard having the highest number of sunshine hours in the growing season, and Qinglong orchard experiencing the greatest temperature difference during the growing season. (ii) Significant differences were found in soil nutrient content among the orchards, with the coefficient of variation for organic matter and mineral elements ranging from 19.1% (S) to 80.3% (available phosphorus). (iii) The main ecological factor influencing chestnut growth was the photosynthetic factor, while fruit quality was influenced by a combination of climatic factors, photosynthetic factors, and nutrient factors. Key ecological factor indicators included: annual precipitation, annual sunshine hours, growing season precipitation, growing season sunshine hours, soil organic matter, available phosphorus, available potassium, alkali nitrogen, copper, zinc, available boron, and sulfur. Soluble solids were significantly positively correlated with growing season sunshine hours, and fat was significantly positively correlated with available potassium and alkali nitrogen, and significantly positively correlated with available phosphorus. [Conclusions] Orchards with high growing season precipitation, long sunshine hours, and high organic matter and nitrogen-phosphorus-potassium content in the soil are more conducive to promoting the growth of ‘Yanshanzaofeng’ chestnuts and improving fruit quality.展开更多

The battery cages measured 99 × 66 × 25 cm that can be sufficient for 5 birds. Day old Chicken broilers totaling 180 were assigned to dietary treatments comprising of 3 concentrations of Lysine (0.85, 1.14, and 1.42) each in combination with 4 concentrations of Glutamine (0, 1, 2, and 3). Each dietary treatment was replicated 3 times and each replication had 5 birds. The birds were given feed and water ad libitum with a 23-hour light regimen for a period of 4 weeks. Then, the experimental birds were evaluated for body weight gain, feed consumption, and feed conversion in order to determine their optimum requirement for dietary Lysine and Glutamine. Based on the findings of this study, the highest performance was observed in birds fed the diet supplemented with 1.42 lysine and 1% glutamine, but the highest improvement in feed conversion was observed in diet contain 1.14 and 1.42 with 1% and 3% glutamine, respectively. Birds fed 1.42 lysine and 1% glutamine had the highest total body weight gain and feed consumption. The lysine requirements in the diet for Chicken are between 1.14 and 1.42 with glutamine level of 1%.展开更多

文摘Avian metapneumovirus(aMPV) is a highly contagious pathogen that causes acute upper respiratory tract diseases in chickens and turkeys, resulting in serious economic losses. Our findings suggest that the LN16-A strain is a promising live attenuated vaccine candidate that can prevent infection with subtype B aMPV.

文摘Background Chicken is one of the most numerous and widely distributed species around the world, and many studies support the multiple ancestral origins of domestic chickens. The research regarding the yellow skin phenotype in domestic chickens(regulated by BCO2)likely originating from the grey junglefowl serves as crucial evidence for demonstrating the multiple origins of chickens. However, beyond the BCO2 gene region, much remains unknown about the introgression from the grey junglefowl into domestic chickens. Therefore,in this study, based on wholegenome data of 149 samples including 4 species of wild junglefowls and 13 local domestic chicken breeds,we explored the introgression events from the grey junglefowl to domestic chickens. Results We successfully detected introgression regions besides BCO2,including two associated with growth trait(IGFBP2 and TKT),one associated with angiogenesis(TIMP3)and two members of the heat shock protein family(HSPB2 and CRYAB).Our findings suggest that the introgression from the grey junglefowl may impact the growth performance of chickens.Furthermore,we revealed introgression events from grey junglefowl at the BCO2 region in multiple domestic chicken breeds, indicating a phenomenon where the yellow skin phenotype likely underwent strong selection and was retained.Additionally, our haplotype analysis shed light on BCO2 introgression event from different sources of grey junglefowl into domestic chickens, possibly suggesting multiple genetic flows between the grey junglefowl and domestic chickens. Conclusions In summary, our findings provide evidences of the grey junglefowl contributing to the genetic diversity of domestic chickens, laying the foundation for a deeper understanding of the genetic composition within domestic chickens, and offering new perspectives on the impact of introgression on domestic chickens.

文摘Background: The drivers of intraspecific variation in behavioral plasticity are poorly known. Conclusions: We speculate that the absence of an association between brain volume and activity behavior plasticity may result from the inaccuracy of using external skull measurements to estimate brain size, or from a particular part of the brain being responsible for plasticity in activity level.

文摘The Chantecler chicken,a unique Canadian indigenous breed,is well adapted to extremely cold environments. However, its genetic characteristics have not been well studied. Here,we analyzed the whole genomes of 10 Chantecler chickens and 121 worldwide chickens, which indicated that Chantecler chickens were derived from commercial chickens and exhibit a high level of inbreeding.Based on a genome-wide scan,we identified two vital candidate regions containing ME3 and ZNF536,which are related to fat metabolism and nervous system in cold adaptation, respectively. We also found that the genetic mechanism of cold adaptation in Chantecler chickens differed from that of chickens from other cold regions, such as northern China. Our study indicated that specialized commercial chickens in the early 20th century contained sufficient genetic diversity to adapt to extreme cold environments over a very short time. These findings enrich our understanding of the adaptive potential of commercial species.

文摘Background:Egg production is economically important in the meat-type chicken industry. To better understand the molecular genetic mechanism of egg production in meat-type chicken, genetic parameter estimation,genome-wide association analyses combined with meta-analyses,Bayesian analyses, and selective sweep analyses were performed to screen single nucleotide polymorphisms(SNPs)and other genetic loci that were significantly associated with egg number traits in 11,279 chickens from seven material lines.Results:Yellow-feathered meat-type chickens laid 115 eggs at 43 weeks of age and white-feathered chickens laid 143 eggs at 60 weeks of age, with heritability ranging from 0.034–0.258.Based on meta-analyses and selective sweep analyses, one region(10.81–13.05 Mb)on chromosome Z was associated with egg number in all lines.Further analyses using the W2 line was also associated with the same region, and 29 SNPs were identified that significantly affected estimation of breeding value of egg numbers. The 29 SNPs were identified as having a significant effect on the egg number EBV in 3194 birds in line W2.There are 36 genes in the region, with glial cell derived neurotrophic factor,DAB adaptor protein 2,protein kinase AMP-activated catalytic subunit alpha 1,NAD kinase 2,mitochondrial,WD repeat domain 70,leukemia inhibitory factor receptor alpha, complement C6,and complement C7 identified as being potentially affecting to egg number. In addition, three SNPs(rs318154184,rs13769886,and rs313325646)associated with egg number were located on or near the prolactin receptor gene. Conclusion:Our study used genomic information from different chicken lines and populations to identify a genomic region(spanning 2.24 Mb)associated with egg number.Nine genes and 29 SNPs were identified as the most likely candidate genes and variations for egg production. These results contribute to the identification of candidate genes and variants for egg traits in poultry.
文摘BACKGROUND Chicken skin mucosa(CSM)surrounding colon polyps is a common endoscopic finding with pale yellow-speckled mucosa during a colonoscopy screening.Although reports about CSM surrounding small colorectal cancer are scarce, and its clinical significance in intramucosal and submucosal cancers is unclear, previous studies have suggested it could be an endoscopic predictive marker for colonic neoplastic and advanced polyps.Currently, because of the inaccurate preoperative evaluation by endoscopists, many small colorectal cancers, particularly lesions with a diameter<2 cm,are improperly treated. Therefore, more effective methods are required to better assess the depth of the lesion before treatment.AIM To explore potential markers of small colorectal cancer early invasion under white light endoscopy, providing patients with better treatment alternatives.METHODS This retrospective cross-sectional study included 198 consecutive patients[233 early colorectal cancers(ECCs)]who underwent endoscopy or surgical procedures at the Digestive Endoscopy Center of Chengdu Second People’s Hospital between January 2021 and August 2022.The participants had pathologically confirmed colorectal cancer with a lesion diameter<2 cm and received endoscopic or surgical treatment, including endoscopic mucosal resection and submucosal dissection.Clinical pathology and endoscopy parameters, including tumor size, invasion depth, anatomical position, and morphology, were reviewed.Fisher’s exact test,theχ2 test, and Student’s t-test were used to analyze the patient’s basic characteristics.Logistic regression analysis was used to examine the relationship between morphological characteristics, size,CSM prevalence, and ECC invasion depth under white light endoscopy.Statistical significance was set at P<0.05.RESULTS The submucosal carcinoma(SM stage)was larger than the mucosal carcinoma(M stage)with a significant difference(17.2±4.1 vs 13.4±4.6 mm,P<0.01).M-and SM-stage cancers were common in the left colon;however,no significant differences were found between them(151/196,77%and 32/37,86.5%,respectively,P=0.199).The endoscopic features of colorectal cancer revealed that CSM,depressed areas with clear boundaries, and erosion or ulcer bleeding were more common in the SM-stage cancer group than in the M-stage cancer group(59.5%vs 26.2%,46%vs 8.7%,and 27.3%vs 4.1%,respectively,P<0.05).CSM prevalence in this study was 31.3%(73/233).The positive rates of CSM in flat, protruded, and sessile lesions were 18%(11/61),30.6%(30/98),and 43.2%(32/74),respectively,with significant differences(P=0.007).CONCLUSION CSM-related small colorectal cancer was primarily located in the left colon and could be a predictive marker of submucosal invasion in the left colon.

文摘Extra-pair copulation(EPC)can potentially maximize individual reproductive fitness, and this process may involve sexual selection of male and female traits that reflect individual quality.Previous studies have implied that adult characteristics are associated with the probability of extra-pair paternity(EPP),but it differs between species. Moreover, there are relatively few examples of the adaptive rationale for females’engagement in EPCs based on an examination of these more traditionally recorded traits,in combination with female flight-mediated traits such as wing length. We investigated whether EPP existed in the wild Chestnut Thrush(Turdus rubrocanus)population during three breeding seasons(2019–2021),and whether paternity was related to morphological traits of males and females.Eight highly variable microsatellite loci were used to identify paternity, and generalized linear mixed models were used to analyze the relationship between paternity and morphological traits. We found that EPP existed in the Chestnut Thrush.53.3%(N=41/77)of the broods contained at least one extra-pair offspring(EPO),and 34.6%(N=72/208)of the nestlings were EPO.We also found that male wing length was negatively associated with the probability of EPP and the proportion of EPO.Female body length was positively related to the probability of EPP.Both female body condition and mass were negatively associated with the proportion of EPO.While other traits of male and female did not relate to the probability of EPP or the proportion of EPO.Extra-pair males had better body condition compared to the males they cuckolded.EPO did not differ from their half-siblings in terms of body size or body condition. The results suggest that body size and body condition were associated with EPP in the Chestnut Thrush. This study provides fundamental information for further studies on the evolution and maintenance of EPP in the Chestnut Thrush, and it is also useful for the comparison of EPP among Turdus species.
文摘The American chestnut (<em>Castanea dentata</em>) was once a dominant tree species in the Appalachian Mountains and played a critical role in the ecological system. A partial least squares discriminant analysis (PLS-DA) model (R2X = 0.884, R2Y = 0.917, Q2 = 0.584) differentiated chestnut species and hybrids within the first five principal component (PCs).

文摘Fasting is typically used before feeding metabolizable energy assessment in broilers.Previous studies have shown that fasting cause atrophy of the intestinal villus.Whether fasting affects intestinal permeability during refeeding by altering barrier function and nutrient absorption is of concern.Here,23-d-old broilers were randomly assigned to 5 treatments, fasted for 0,12,24,36,and 48 h,respectively, and then refed for 2 d,to study the impact of different duration of fasting on the intestinal regeneration and barrier function during refeeding. Results showed that the intestinal morphology in fasted birds was recovered in 2 d of refeeding at most.As fasting durations increased, enterocytes per intestinal villus were linearly and quadratically increased(both P<0.05),whereas goblet cells per intestinal villus was linearly decreased(both P<0.05).Besides,the mRNA level of lysozyme was linearly decreased as fasting durations increased during refeeding(both P<0.05),while quadratically increased mucin 2 was observed only after 1 d of refeeding(P<0.05).Linear increase effects were observed for claudin 2 and zonula occludens-1with increased fasting durations after 1 d of refeeding(all P<0.05),and linear and quadratical effects were observed for claudin 2 at 2 d of refeeding(both P<0.05).Besides,we found that intestinal permeability to creatinine,4 and 70 kD dextran were linearly and quadratically decreased with increased fasting durations at 6 h and 1 d of refeeding(all P<0.05).Furthermore,jejunum proteomic from birds refed for 6 h showed that birds fasted for 36 h showed increased antimicrobial peptides and upregulated retinol metabolism when compared to the nonfasted birds(P<0.05).Further study showed that retinyl ester catabolism was inhibited during fasting and enhanced during refeeding. Results of intestinal organoid culture showed that retinol benefits the cell proliferation and enterocyte differentiation. In conclusion, the intestinal permeability to small and large molecules was decreased during refeeding by strengthening the intestinal barrier function, and the activated retinol metabolism during refeeding is involved in the intestinal regeneration and strengthens the intestinal barrier.

文摘[Objectives] This study conducted investigations on the climate and soil nutrients of different chestnut orchards in the Yanshan region, selected key ecological factors affecting the growth and fruit quality of chestnuts, and provided a theoretical basis for the cultivation, introduction, and scientific planting of high-quality chestnuts. [Conclusions] Orchards with high growing season precipitation, long sunshine hours, and high organic matter and nitrogen-phosphorus-potassium content in the soil are more conducive to promoting the growth of ‘Yanshanzaofeng’ chestnuts and improving fruit quality.
文摘The optimum levels of Lysine and Glutamine needed for growth performance and maintenance of the chicken broilers were evaluated in a randomized 3 × 4 factorial arrangement of dietary treatments. The lysine requirements in the diet for Chicken are between 1.14 and 1.42 with glutamine level of 1%.

文摘BACKGROUND Lipid metabolism reprogramming is suspected to exist in pre-cancerous lesions, including colorectal adenoma.Screening colonoscopy frequently reveals chicken skin mucosa(CSM;white or yellow-white speckled mucosa)surrounding colo-rectal polyps, caused by macrophages engulfing and accumulating the lipids decomposed by colon cells or adjacent tumors.CSM-positive colorectal polyps are associated with various diseases;however, their prognosis varies greatly.Cold snare polypectomy is commonly used to resect lesions up to 10 to 15 mm in diameter without signs of submucosal invasion but is controversial for CSM-positive colorectal polyps.Improved imaging is required to diagnose and treat CSM-positive colorectal polyps.METHODS This retrospective cohort study included 177 patients with CSM-positive colorectal polyps diagnosed using endoscopy. All patient-related information was extracted from the Goldisc soft-clinic DICOM system or electronic medical record system.Based on the pathological results, patients were classified as non-neoplastic polyps(five juvenile polyps),neoplastic polyps,non-invasive high-grade neoplasia(NHGN),or submucosal invasive carcinoma(SM stage cancer). We analyzed and compared the clinical features, suspected risk factors for malignant transformation of neoplastic polyps, and early infiltration of sub-mucosal carcinoma.RESULTS The diameters of NHGN and SM polyps were much smaller than those of neoplastic polyps. Most NHGN polyps had a deeper red mucosal color. On logistic regression analyses, diameter and deeper red mucosal color were independent risk factors for malignant transformation of neoplastic polyps.Type 1 CSM was more common in high-grade intraepithelial neoplasia and SM;type 2 CSM was more common in neoplastic polyps.Logistic regression analyses revealed no significant differences in the malignant transformation of neoplastic polyps or early submucosal invasion of CSM-positive colorectal cancer.Changes in the CSM mucosa surrounding neoplastic polyps and submucosal invasion of colorectal cancer disappeared within 12 months. No tumor recurrence was found during either partial or complete endoscopic resection of the CSM.CONCLUSION CSM-positive colorectal polyps>1 cm in diameter or with deeper red mucosa may be related to NHGN.Resection of CSM surrounding colorectal adenomas did not affect tumor recurrence.

文摘Inducing animal viruses to adapt to chicken embryos or chicken embryo fibroblasts(CEF) is a common method to develop attenuated live vaccines with full security.Canine distemper virus(CDV) also does this, but the mechanisms and particular receptors remain unclear.Virus overlay protein blot assays were carried out on CEF membrane proteins, which were extracted respectively with a Mem-PER TM kit,a radioimmunoprecipitation assay buffer or a modified co-immunoprecipitation method, and revealed a common 57 kDa positive band that differed from the 42-kDa positive band in Vero cells and also from those receptors reported in lymphocytes and 293 cells, indicating a receptor diversity of CDV and the possibility of the 57-kDa protein acting as a receptor that is involved in adaptive infection of CDV Kunming strain to CEF.
