Mechanism of Yanghe Pingchaun granules on airway remodeling in asthmatic rats based on IL-6/JAK2/STAT3 signaling axis

Objective: To investigate the effects of Yanghe Pingchuan Granules on airway remodeling in asthmatic rats, and to explore the mechanism of Interleukin-6/Janus kinase 2/ Signal transducing activator of transcription 3(IL-6/JAK2/STAT3) signal axis. Methods: We separated 42 healthy male SD rats into two groups, a control group (7) and a model group (35).The model group was sensitized with a combination of ovalbumin (OVA) and aluminum hydroxide for 2 weeks, while the control group was given an equal amount of physiological saline.After 2 weeks, the modeling group was randomly divided into Model group, Yanghe Pingchuan Granules high, medium and low dose groups and Dexamethasone group, each group consisted of 7 animals. After 4 weeks, OVA atomization and gavage were used for stimulation and treatment. Yanghe Pingchuan Granules high, middle and low groups were given 15.48, 7.74, 3.87 g∙kg-1 Yanghe Pingchuan Granules daily, dexamethasone group was given 0.0625 mg∙kg-1 dexamethasone daily, and the other groups were given the same amount of normal saline. HE, PAS and Masson staining were used to observe the lung histopathological changes in rats. The levels of interleukin-6, IL-23 and IL-17A were detected by ELISA. The expression levels of JAK-2, P-JAK2, STAT3 and P-STAT3 in lung tissues were detected by Western blot. Real-time quantitative polymerase chain reaction (qRT-PCR) was used to detect the mRNA expression levels of IL-6, JAK2 and STAT3 in rat lung tissue. Results: The lung tissue structure of the model group was severely damaged compared to the control group, accompanied by a great many of inflammatory cell infiltration, goblet cell hyperplasia, subepithelial collagen fiber deposition and airway epithelial thickening were more obvious. The expressions of IL-6, IL- 23 and IL-17A in serum were significantly increased (P<0.01), the protein expression levels of JAK-2, P-JAK2, STAT3 and P-STAT3 and the mRNA expression levels of IL-6, JAK2 and STAT3 in lung tissue were significantly increased (P<0.01);Compared with the model group, inflammatory cell infiltration, goblet cell proliferation, subepithelial collagen fiber deposition and airway epithelial thickening were significantly reduced in each administration group, and the expressions of IL-6, IL-23 and IL-17A in serum were significantly decreased (P< 0.01). The protein expression levels of JAK-2, P-JAK2, STAT3 and P-STAT3 and mRNA expression levels of IL-6, JAK2 and STAT3 in lung tissue were significantly decreased (P<0.01). Conclusion: Yanghe Pingchuan Granules can significantly alleviate airway remodeling in asthmatic rats, and its mechanism may be through inhibiting the IL-6/JAK2/STAT3 signal axis. 展开更多

We found that mononuclear cells from patients produce spontaneously IL-2 in the absence of exogenous lee-tin stimulation, the proportion of Tac+ cells in mononuclear cells increased. The release of IL-2 and or Tac antigen expression were elevated in almost every patient with AA. The plasma from patients stimulate mitogen-induced blastogenesis and Tac antigen expression of normal human lymphocytes. Immunological 1 abnormalities of patients with AA possibly might represents secondary response to bone marrow depression. 展开更多

Association of Interleukin-6-174G/C Polymorphism with the Risk of Diabetic Nephropathy in Type 2 Diabetes:A Meta-analysis 被引量:6

Previous studies reported the association between interleukin-6(IL-6)-174G/C gene polymorphism and the risk of diabetic nephropathy in type 2 diabetes mellitus(T2DN).However,the results remain controversial.In the present study,we conducted a meta-analysis to further examine this relationship between IL-6-174G/C gene polymorphism and T2DN.Three databases(PubMed,SinoMed and ISI Web of Science)were used to search clinical case-control studies about IL-6-174G/C polymorphism and T2DN published until Apr.14,2018.Fixed-or random-effects n lodels were used to calculate the effect sizes of odds ratio(OR)and 95%confide nee intervals(95%CI).Moreover,subgroup analysis was performed in tenns of the excretion rate of albuminuria.All the statistical analyses were con ducted using Stata 12.0.A total of 11 case-control studies were included in this study,involving 1203 cases of T2DN and 1571 cases of T2DM without DN.Metaanalysis showed that there was an association between IL-6-174G/C polymorphism and increased risk of T2DN under the allelic and recessive genetic models(G vs.C:OR=1.10,95%CI 1.03-1」&P=0.006;GG vs.CC+GC:OR=1.11,95%CI 1.02-1.21,P=0.016).In the subgroup analysis by albuminuria,a significant association of IL-6-174G/C polymorphism with risk of T2DN was noted in the microalbuminuria group under the recessive model(OR=1.54,95%CI 1.02-2.32,P=0.038).In conclusion,this meta-analysis suggests that IL-6-174G/C gene polymorphism is associated with the risk of T2DN. 展开更多

目的探究二甲双胍联合补虚活血方治疗2型糖尿病的疗效及对血清C反应蛋白(CRP)、白细胞介素-6(IL-6)、血清尿酸的影响。方法选择2014年8月-2016年8月收治的120例2型糖尿病患者,随机分为两组,每组60例。对照组采用二甲双胍治疗,治疗组在... 目的探究二甲双胍联合补虚活血方治疗2型糖尿病的疗效及对血清C反应蛋白(CRP)、白细胞介素-6(IL-6)、血清尿酸的影响。方法选择2014年8月-2016年8月收治的120例2型糖尿病患者,随机分为两组,每组60例。对照组采用二甲双胍治疗,治疗组在对照组治疗的基础上联合补虚活血方治疗,1个月后对比两组疗效及对血清CRP、IL-6、血清尿酸的影响。结果与治疗前比较,两组空腹血糖(FPG)、糖化血红蛋白(Hb Alc)及餐后2 h血糖(2 h PG)均降低(P<0.05);与对照组比较,治疗组治疗后FPG及Hb Alc水平降低更显著(P<0.05)。与治疗前比较,两组血清CRP、IL-6及血清尿酸水平均降低(P<0.05);与对照组比较,治疗组治疗后血清CRP、IL-6及血清尿酸水平降低更显著(P<0.05)。与对照组比较,治疗组不良反应发生率并未增加,差异无统计学意义(P>0.05)。结论二甲双胍联合补虚活血方治疗2型糖尿病患者能很好地控制血糖水平,改善相关代谢指标及炎症反应,且不良反应少,临床应用安全、有效。 展开更多

Objective: To evaluate effects of docosahexaenoic acid-enriched phosphatidylcholine(DHAPC) on cytokine production induced by lipopolysaccharide(LPS). Methods: The culture supernatants of splenocytes exposed to DHA-PC along with LPS were harvested to determine the production of Th 1(IFN-kines. Cytokines were m毭 and IL-2) and Th2 [IL-4, IL-5, IL-6 and IL-12/IL-23(p40)] cytoeasured using ELISA. Results: Co-administration of DHAPC with LPS resulted in significantly lower IL-2 expression compared to that observed with administration of only LPS(P<0.01). Treatment with DHA-PC and LPS significantly increased IL-5 expression(P<0.01). Moreover, co-administration of DHA-PC with LPS significantly decreased IL-6 and IL-12/IL-23(p40) expressions compared to that observed with administration of only LPS(P<0.01). Conclusions: Our results suggest that DHA-PC inhibits pro-inflammatory cytokines [IL-2, IL-6 and IL-12/IL-23(p40)] expression on induction of inflammation. 展开更多

Recombinant interleukin-33(IL-33)inhibits tumor growth,but the detailed immunological mechanism is still unknown.IL-33-mediated tumor suppression did not occur in Batf3^(−/−)mice,indicating that conventional type 1 dendritic cells(cDC1s)play a key role in IL-33-mediated antitumor immunity.A population of CD103^(+)cDC1s,which were barely detectable in the spleens of normal mice,increased significantly in the spleens of IL-33-treated mice.The newly emerged splenic CD103^(+)cDC1s were distinct from conventional splenic cDC1s based on their spleen residency,robust effector T-cell priming ability,and surface expression of FCGR3.DCs and DC precursors did not express Suppressor of Tumorigenicity 2(ST2).However,recombinant IL-33 induced spleen-resident FCGR3^(+)CD103^(+)cDC1s,which were found to be differentiated from DC precursors by bystander ST2+immune cells.Through immune cell fractionation and depletion assays,we found that IL-33-primed ST2^(+)basophils play a crucial role in the development of FCGR3^(+)CD103^(+)cDC1s by secreting IL-33-driven extrinsic factors.Recombinant GM-CSF also induced the population of CD103^(+)cDC1s,but the population neither expressed FCGR3 nor induced any discernable antitumor immunity.The population of FCGR3^(+)CD103^(+)cDC1s was also generated in vitro culture of Flt3L-mediated bone marrow-derived DCs(FL-BMDCs)when IL-33 was added in a pre-DC stage of culture.FL-BMDCs generated in the presence of IL-33(FL-33-DCs)offered more potent tumor immunotherapy than control Flt3L-BMDCs(FL-DCs).Human monocyte-derived DCs were also more immunogenic when exposed to IL-33-induced factors.Our findings suggest that recombinant IL-33 or an IL-33-mediated DC vaccine could be an attractive protocol for better tumor immunotherapy. 展开更多

为了探讨中草药复方对断奶仔兔肠道发育及肠道免疫水平的影响,试验选取30日龄、体重相近且健康状况良好的断奶伊拉兔仔兔60只,公母各半,随机分为2组,每组3个重复,每个重复10只,对照组饲喂基础日粮,试验组饲喂基础日粮+1%中草药复方,预试... 为了探讨中草药复方对断奶仔兔肠道发育及肠道免疫水平的影响,试验选取30日龄、体重相近且健康状况良好的断奶伊拉兔仔兔60只,公母各半,随机分为2组,每组3个重复,每个重复10只,对照组饲喂基础日粮,试验组饲喂基础日粮+1%中草药复方,预试期5 d,试验期30 d。试验结束后,取小肠制作石蜡切片,观察小肠组织结构;测定空肠消化酶活性及小肠白细胞介素2(interleukin-2,IL-2)mRNA表达量。结果表明:(使用中草药复方后,试验组十二指肠、空肠和回肠的绒毛高度均较对照组显著增加(P<0.05),空肠的隐窝深度显著降低(P<0.05),三段肠段的绒毛高度与隐窝深度比值均极显著升高(P<0.01)。)试验组淀粉酶、胰蛋白酶活性较对照组显著或极显著提高(P<0.05或P<0.01);糜蛋白酶活性也有所提高,但差异不显著(P>0.05)。试验组空肠和回肠IL-2 mRNA表达量较对照组显著升高(P<0.05);十二指肠IL-2 mRNA表达量有升高趋势,但差异不显著(P>0.05)。说明中草药复方能够有效促进仔兔小肠发育,并增强其肠道免疫力。 展开更多

Interleukin-2(IL-2) is an important immunoregulatory factor. After the interaction between IL-2 and its receptor, intracellular signal molecules were activated and pro-oncogenes, e.g. c-Myc, c-Fos, c-Jun and Bcl-2 were induced, then cells stimulated with IL-2 would proliferate or differentiate. During these processes, IL-2 were internalized. The internalization of IL-2 mediated by IL-2R decreases the number of IL-2R on cell membranes, and makes extracellular IL-2 enter cells and degrade. It will down-regulate IL-2 biological activities. In order to identify the IL-2R subunit responsible for IL-2 internalization, chimeric IL-2Rs which consisted of different endocellular domains and extracellular domains from various IL-2R subunits were constructed. After they were transfected into L929 cell line, the IL-2 internalization was detected. Results showed that intracellular domains from IL-2R α and β subunits could not mediate IL-2 internalization alone; the signal for IL-2 internalization was located only in the endocellular domain of IL-2R γ subunit. Furthermore, IL-2 internalization extent would be affected by affinity between IL-2 and IL-2R. 展开更多

Seaweed has been used in traditional cosmetics and as a herbal medicine in treatments for cough,boils,goiters,stomach ailments,and urinary diseases,and for reducing the incidence of tumors,ulcers,and headaches.Despite the fact that seaweeds are frequently used in the practice of human health,little is known about the role of seaweed in the context of inflammation.This study aimed to investigate the influence of Jeju endemic seaweed on a mouse macrophage cell line(RAW 264.7) under the stimulation of lipopolysaccharide(LPS).Ethyl acetate extracts obtained from 14 different kinds of Jeju seaweeds were screened for inhibitory effects on pro-inflammatory mediators.Our results revealed that extracts from five seaweeds,Laurencia okamurae,Grateloupia elliptica,Sargassum thun-bergii,Gloiopeltis furcata,and Hizikia fusiformis,were potent inhibitors of the production of pro-inflammatory mediators such as nitric oxide(NO),prostaglandin E2(PGE2),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α).Based on these results,the anti-inflammatory effects and low cell toxicity of these seaweed extracts suggest potential thera-peutic applications in the regulation of the inflammatory response. 展开更多

Background: The aim of this study was to evaluate the clinical potential of new biomarkers such as IL-2, TNF-α, MCP-1, MIP-1β including IFN-γ for the diagnosis of active tuberculosis (TB) disease. Materials and Methods: The subjects consisted of 31 patients with active TB disease and 62 patients with non-TB disease. We measured IFN-γ using QuantiFERON-TB Gold In Tube, and IL-2, TNF-α, MCP-1, and MIP-1β using the supernatant from whole blood stimulated with MTB (Mycobacterium tuberculosis)-specific antigens. Results: In the patient group with active TB disease, while the positive response rate of IFN-γ was 74%, that of IL-2 using the supernatant was 61%, TNF-α was 71%, MCP-1 was 81% and MIP-1β was 81%. In the patient group with non-TB disease, while the positive response rate of IFN-γ was 11%, that of IL-2using the supernatant was 32%, TNF-α was 26%, MCP-1 was 23% and MIP-1β was 19%. All biomarker levels of the patients with active TB disease using the supernatant were significantly higher than those of the patients with non-TB disease. While MIP-1β was the most sensitive of all the biomarkers (sensitivity: 80.6%), IFN-γ was the best in terms of specificity (specificity: 87.0%). Conclusions: Several biomarkers, apart from IL-2, showed similar results compared to IFN-γ. The combination of IFN-γ and other new biomarkers may increase the diagnostic accuracy of active TB. 展开更多

目的:研究复方柴芩退热颗粒对2,4二硝基苯酚及细菌内毒素所致大鼠发热模型的退热作用,进一步明确该颗粒的退热效果,初步探讨其退热机制。方法:将SPF级SD大鼠随机分为正常对照组、模型组、酚麻美敏片组[给药剂量0.148 g/(kg·d)]、... 目的:研究复方柴芩退热颗粒对2,4二硝基苯酚及细菌内毒素所致大鼠发热模型的退热作用,进一步明确该颗粒的退热效果,初步探讨其退热机制。方法:将SPF级SD大鼠随机分为正常对照组、模型组、酚麻美敏片组[给药剂量0.148 g/(kg·d)]、小柴胡冲剂组[给药剂量2.160 g/(kg·d)]、复方柴芩退热颗粒高、中、低剂量组[给药剂量分别为6.480、3.240、1.620 g/(kg·d)]。各给药组按相应剂量灌胃给药,正常对照组、模型组同法给予等体积的蒸馏水,每天1次,连续给药5天,第4天各组动物均开始禁食不禁水24 h,末次给药前开始造模。造模实验一:除正常对照组外,余各组大鼠均皮下注射2,4-二硝基苯酚17 mg/kg,正常对照组皮下注射同体积生理盐水;造模实验二:除正常对照组外,余各组大鼠均腹腔注射细菌内毒素80μg/kg。测大鼠各时间点的体温和血清中白细胞介素-1β(IL-1β)、前列腺素E2(PGE2)、肿瘤坏死因子-α(TNF-α)的含量。结果:实验一:与正常对照组比较,模型组大鼠造模后1 h、2 h、3 h体温上升值均显著升高,大鼠血清IL-1β、PGE2含量均显著升高(P<0.01)。与模型组比较,酚麻美敏片组、小柴胡冲剂组、复方柴芩退热颗粒高、中、低剂量组大鼠血清IL-1β、PGE2含量均明显降低(P<0.05,P<0.01);酚麻美敏片组大鼠在造模后1 h、2 h、3 h体温上升值均显著降低(P<0.05,P<0.01);小柴胡冲剂组大鼠在造模后2 h、3 h体温上升值均显著降低(P<0.05,P<0.01);复方柴芩退热颗粒高、低剂量组在造模后1 h、2 h、3 h体温上升值均明显降低(P<0.05,P<0.01);复方柴芩退热颗粒中剂量组在造模后2 h、3 h体温上升值均明显降低(P<0.05)。实验二:与正常对照组比较,模型组大鼠造模后2 h、4 h、6 h、8 h体温上升值均显著升高,大鼠血清PGE2、TNF-α含量均显著升高(P<0.01)。与模型组比较,酚麻美敏片组、小柴胡冲剂组、复方柴芩退热颗粒高、中、低剂量组大鼠血清PGE2、TNF-α含量均明显降低(P<0.05,P<0.01);酚麻美敏片组大鼠在造模后2 h、4 h、6 h体温上升值均显著降低(P<0.05,P<0.01);小柴胡冲剂组、复方柴芩退热颗粒高、中、低剂量组在造模后2 h、4 h体温上升值均明显降低(P<0.05,P<0.01)。结论:复方柴芩退热颗粒对2,4二硝基苯酚及细菌内毒素所致大鼠发热均有降温作用,能有效降低大鼠血清PGE2、IL-1β和血清TNF-α含量,对大鼠实验性高热模型有较好的解热作用。 展开更多
