[ Method] The "Quxie" ( i. e. eliminating pathogenic factors) drugs including Hedyotis diffusa and Loni- cera japonica and "Fuzheng" ( i. e. strengthening body resistance) drugs including Radix astragali and Glycyrrhiza uralensis were taken to form an antiviral herbal compound preparation. Taking the astragalus polysaccharide monomer as control, the experiment was constructed by using the agent to treat some naturally occurring viral diseases, including Newcastle disease (ND), infectious bursal disease (IBD), duck viral hepatitis (DVH), avian influenza (Al) H9 subtype and avian swollen head syndrome. [Ressult] The effective rates of the agent for treatment of the above- mentioned diseases were 87%, 87%, 69%, 100% and 100%, respectively, which were higher than that of astragalus polysaccharide monomer. The cure rates for these diseases were 33%, 37%, 13%, 69% and 75%, respectively, which were significantly higher than that of astragalus polysacchadde monomer ( P 〈 0.01 ). The rates of poor therapeutic efficacy were 15%, 21%, 24%, 6% and 7%, respectively, which were significantly lower than that of astragalus polysaccharide monomer (P 〈 0.01 ). [ Condusion] The Chinese herbal compound preparation shows good therapeutic effects on some avian viral diseases, which can decrease mortality rate and improve production performance of poultry more greatly than astragalus polysaccharide monomer.展开更多

The preliminary results indicated that combination treatment seemed to possess better antitumor activity than chemotherapy alone. The treatment with CHC alone however had neither an obvious antitumor effect in tumor bearing mice nor toxicity in normal mice. These results show that CHC may stimulate organs of the immune system such as the spleen to be im-munomodulators and enhance the antitumor activity of some chemotherapeutic agents.展开更多

[ Metho] Myelasuppressive mice model was established by intraperitoneal injection of 80 mg/kg cyclophosphamide. Chinese herbal compound was composed of Houttuynia cordata, Taraxacum mongolicum, Citrus reticulata peel, Atractylodes chinensis, Paeonia sterniana, Atractylodes macrocephala and Angelica sinensis. The effects of Chinese herbal compound and Astragalus polysaccharide at different doses on the blood physiological indices and hematopoietic growth factors of myelosuppressive mice were discussed. [ Result] Myelosuppressive mice medel was successfully established. The total white blood cell count, total nentrephile granulocyte count, total lymphocyte count, total platelet count and the contents of serum interleukin-6 and erythropoietin in myelosuppressive mice were significantly decreased. The total erythrocyte count, the contents of hemoglobin and imerleukin-3 were decreased, without significant difference. The blood physiological indices and the contents of interleukin-3 and erythropoietin in myelosuppressive mice could be improved by intragastric administration of Astragalus polysaccharide and Chinese herbal compound at different doses for 3 days or 7 days. The effect of 20 g/kg Chinese herbal compound was the best after administration for 3 days, and the effect of 10 g/kg Chinese herbal compound was the best after administration for 7 days. The total white blcod cell count, total neutrophile granulocyte count, percentage of neutrephile granulocyte, total lymphocyte count, total count of middle cells, percentage of middle cells, total platelet count, contents of interleukin-3 and erythropoietin in myelosuppressive mice could be extremely significantly improved by intragastric administration of 10 g/kg Chinese herbal compound for 7 days( P 〈 0. 01 ) , and the reduction of red blood cell count, hematecfit, the contents of hemoglobin and interleukin-6 induced by cyclophesphamide could be inhibited (P 〈 0.01 ). [ Conclusion ] Chinese herbal compound could improve the hematopoietic function of myclosuppressivc mice induced by cyclophosphamide and its effect was better than Astragalus polysaccharide. 20 g/kg Chinese herbal compound reacted fast and the reaction of 10 g/kg Chinese herbal compound was slow ,but its efficacy was lasting.展开更多

Methods: The microglia BV-2 cells incubated in vitrowere divided into different groups: the control group(25 mmol/L glucose), the model group(75 mmol/L glucose), high glucose media containing different dose medicated serum of NFC. After being cultured for 24 h, changes in IL-6 and TNF-α were measured by quantitative real-time polymerase chain reaction and enzyme-linked immunosorbent assay. The expression of surface marker CD11 b of activated microglia was measured by confocal laser scanning microscope and Western blot. Nuclear factor-κB(NF-κB) p-p65 expression was analyzed by Western blot. Results: The model group obviously increased the expression of microglial surface marker CD11 b and NF-κB p-p65(all P<0.01), induced a significant up-regulation of release and the m RNA expression of IL-6 and TNF-α(P<0.01 or P<0.05). The medicated serum of NFC could obviously down-regulate the transcription and expression of surface marker CD11 b and NF-κB p-p65(all P<0.01), and inhibit the m RNA and protein expression(P<0.01 or P<0.05) of inflammatory cytokines, such as IL-6 and TNF-α, in microglia BV-2 cells cultured with high glucose for 24 h. Conclusions: The inhibition of microglial activation and IL-6 and TNF-α expression induced by high glucose may at least partly explain NFC therapeutic effects on diabetes-associated cognitive decline diseases. Its underlying mechanism could probably be related to the inhibition of NFC on NF-κB phosphorylation.展开更多

It has been used in China for many years for treating amnesia. Kai Xin San ameliorates amyloid-β (Aβ) induced cognitive dysfunction and is neuroprotective in vivo, but its precise mechanism remains unclear. Expression of insulin-degrading enzyme (IDE), which degrades Aβ, is strongly correlated with cognitive function. Here, we injected rats with exogenous Aβ42 (200 μM, 5 μL) into the hippocampus and subsequently administered Kai Xin San (0.54 or 1.08 g/kg/d) intragastrically for 21 consecutive days. Hematoxylin eosin and Nissl staining revealed that Kai Xin San protected neurons against Aβ-induced damage. Furthermore, enzyme linked immunosorbent assay, western blot and polymerase chain reaction results showed that Kai Xin San decreased Aβ42 protein levels and increased expression of IDE protein, but not mRNA, in the hippocampus. Our findings reveal that Kai Xin San facilitates hippocampal Aβ degradation and increases IDE expression, which leads, at least in part, to the alleviation of hippocampal neuron injury in rats.展开更多

Reverse transcription PCR results showed that tyrosine hydroxylase mRNA expression in the rat substantia nigra was significantly increased, while monoamine oxidase B mRNA expression was significantly decreased in the Zhichan powder group, compared with the model group. In addition, the levels of striatal dopamine and homovanillic acid, the ratio of dopamine to homovanillic acid, and the activity of blood superoxide dismutase were all higher in the Zhichan powder group than in the model group but the content of malondialdehyde in blood was lower. Our experimental findings indicate that Zhichan powder has an antioxidant effect, it can regulate the expression of monoamine oxidase B and tyrosine hydroxylase in the substantia nigra of Parkinson's disease rats, and it can facilitate the secretion of striatal dopamine and its metabolite homovanillic acid.展开更多

Methods: Based on the previous 3-(4,5)-dimethylthiahiazo (z-yl)-3-5-diphenytetrazoliumromide (MI-I-) experiment, under different medium concentrations of glucose and Dangua liquor, the endothelial cells of vein-304 (ECV-304) were divided into 6 groups as follows: standard culture group (Group A, 5.56 mmol/L glucose); 11300 herb-standard group (Group B); high glucose culture group (Group C, 16.67 mmol/L glucose); 1/150 herb-high glucose group (Group D); 1/300 herb-high glucose group (Group E); and 1/600 herb-high glucose group (Group F). The cell cycle was assayed using flow cytometry after cells were cultivated for 36, 72 and 108 h, respectively. Results: (1) The percentage of cells in the Go/G1 phase was significantly increased in Group C compared with that in Group A (P〈0.05), while the percentage of S-phase (S%) cells in Group C was significantly reduced compared with Group A (P〈0.05); the latter difference was dynamically related to the length of growing time of the endothelial cells in a high glucose environment. (2) The S% cells in Group A was decreased by 30.25% (from 40.23% to 28.06%) from 36 h to 72 h, and 12.33% (from 28.06% to 24.60%) from 72 h to 108 h; while in Group C, the corresponding decreases were 23.05% and 21.87%, respectively. The difference of S% cells between the two groups reached statistical significance at 108 h (P〈0.05), (3) The percentage difference of cells in the Gz/M phase between Group C and Group A was statistically significant at 72 h (P〈0.01). (4) 1/300 Dan Gua-Fang completely reversed the harmful effect caused by 16.67 mmol/L high glucose on the cell cycle; moreover it did not disturb the cell cycle when the cell was cultivated in a glucose concentration of 5.56 mmol/L. Conclusions: High glucose produces an independent impact on the cell cycle. Persistent blocking of the cell cycle and its arrest at the Go/G1 phase are toxic effects of high glucose on the endothelial cell cycle. The corresponding variation of the arrest appears in the S phase. 1/300 Dan Gua-Fang completely eliminates the blockage of high glucose on the endothelial cell cycle.展开更多

文摘The effect of the Chinese herbal compound (CHC) on solid sarcoma 180 (S180) in Swiss mice was studied either alone or in combination with 5-fluorouracil (5FU), cyclophosphamide (CYT) or mitomycin C (MMC). The preliminary results indicated that combination treatment seemed to possess better antitumor activity than chemotherapy alone. The treatment with CHC alone however had neither an obvious antitumor effect in tumor bearing mice nor toxicity in normal mice. These results show that CHC may stimulate organs of the immune system such as the spleen to be im-munomodulators and enhance the antitumor activity of some chemotherapeutic agents.

文摘Objective: To determine the effect of medicated serum of Chinese herbal compound Naofucong(脑复聪, NFC) on the microglia BV-2 cells viability and the transcription and expression of interleukin-6(IL-6) and tumor necrosis factor α(TNF-α) in microglia BV-2 cells to further explore the mechanisms underlying the protective effect of NFC on inflammatory process induced by high glucose. Its underlying mechanism could probably be related to the inhibition of NFC on NF-κB phosphorylation.

文摘Kai Xin San is a Chinese herbal formula composed of Radix Ginseng, Poria, Radix Polygalae and Acorus Tatarinowii Rhizome. Our findings reveal that Kai Xin San facilitates hippocampal Aβ degradation and increases IDE expression, which leads, at least in part, to the alleviation of hippocampal neuron injury in rats.
文摘In this study, rat models of Parkinson's disease induced by substantia nigra injection of 6-hydroxy-dopamine were intragastrically administered Zhichan powder daily for 50 days. Our experimental findings indicate that Zhichan powder has an antioxidant effect, it can regulate the expression of monoamine oxidase B and tyrosine hydroxylase in the substantia nigra of Parkinson's disease rats, and it can facilitate the secretion of striatal dopamine and its metabolite homovanillic acid.

文摘Objective: To study the toxicity features of high glucose on the endothelial cell cycle and the influence of Dan Gua-Fang , a Chinese herbal compound prescription, on the reproductive cycle of vascular endothelial cells cultivated under a high glucose condition; to reveal the partial mechanisms of Dan Gua- Fang in the prevention and treatment of endothelial injury caused by hyperglycemia in diabetes mellitus (DM); and offer a reference for dealing with the vascular complications of DM patients with long-term high blood glucose. 1/300 Dan Gua-Fang completely eliminates the blockage of high glucose on the endothelial cell cycle.
