酵母双杂交法筛选与受翻译调节的肿瘤蛋白相互作用的蛋白

目的:采用酵母双杂技术筛选与受翻译调节的肿瘤蛋白(translationally controlled tumor protein,TCTP)相互作用的蛋白,以探讨TCTP的生物学功能。方法:采用PCR法扩增获得TCTP基因,并将其插入酵母双杂交诱饵载体中构建获得pDEST32-TCTP;检... 目的:采用酵母双杂技术筛选与受翻译调节的肿瘤蛋白(translationally controlled tumor protein,TCTP)相互作用的蛋白,以探讨TCTP的生物学功能。方法:采用PCR法扩增获得TCTP基因,并将其插入酵母双杂交诱饵载体中构建获得pDEST32-TCTP;检测pDEST32-TCTP在酵母菌中的表达和自激活情况,利用酵母双杂交系统筛选出带有靶蛋白基因的阳性克隆并测序鉴定;免疫共沉淀(co-immunoprecipitation,Co-IP)法验证人肝癌细胞Huh7中TCTP与细胞S期激酶相关蛋白-2(S-phase kinase-associated protein2,Skp2)的相互作用。结果:成功构建获得酵母双杂交诱饵载体pDEST32-TCTP,pDEST32-TCTP在酵母菌中可以表达,且在3-氨基-1,2,4-三唑(3-amino-1,2,4-triazole,3AT)浓度为20mmol/L时无自激活作用。采用酵母双杂交系统共筛选获得6个可与TCTP相互作用蛋白的基因,包括Smad4、人热休克蛋白60(heat shock protein60,HSP60)、TAKl、Bcl-xl、卵泡抑素(follistatin,FS)和Skp2。其中Skp2与TCTP蛋白的相互作用在Co-IP中得到验证。结论:应用酵母双杂交系统可筛选获得一组与TCTP相互作用的蛋白,为进一步研究TCTP的生物学功能提供了可靠的实验依据。 展开更多
