Using selective extraction together with embeddment ...Dinoflagellate is one of the primitive eukaryotes, whose nucleus may represent one of the transition stages from prokaryotic nucleoid to typical eukaryotic nucleus. Using selective extraction together with embeddment - free section and whole mount electron microscopy, a delicate nuclear matrix filament network was shown, for the first time, in dinoflagellate Crypthecodinium cohnii nucleus. Chromosome residues are connected with nuclear matrix filaments to form a complete network spreading over the nucleus. Moreover, we demonstrated that the dinoflagellate chromosome retains a protein scaffold after the depletion of DNA and soluble proteins. This scaffold preserves the characteristic morphology of the chromosome. Two dimensional elec-trophoreses indicated that the nuclear matrix and chromosome scaffold are mainly composed of acidic proteins. Our results demonstrated that a framework similar to the nuclear matrix and chromosome scaffold in mammalian cells appears in this primitive eukaryote,suggesting that these structures may have been originated from the early stages of eukaryote evolution.展开更多
An important filamentous industrial fungus,Rhizopus cohnii(R.cohnii),was used as an efficient biosorbent for removing cadmium from wastewater.The sorption conditions,such as pH,the dose of biomass and the initial conc...An important filamentous industrial fungus,Rhizopus cohnii(R.cohnii),was used as an efficient biosorbent for removing cadmium from wastewater.The sorption conditions,such as pH,the dose of biomass and the initial concentration of cadmium were examined.Two kinds of adsorption models were applied to simulate the biosorption data.The uptake of cadmium was higher in weak acid condition than in strong acid condition.Nearly no sorption of cadmium occurred when the pH value was lower than 2.0. Biosorption isothermal data could be well simulated by both Langmuir and Freundlich models.Langmuir simulation of the biosorption showed that the maximum uptake of cadmium was 40.5 mg/g(0.36 mmol/g)in the optimal conditions,which was higher than many other adsorbents,including biosorbents and activated carbon.In addition,the reusability results showed that after five times of sorption and desorption process,the sorption capacity of R.cohnii could still maintain nearly 80%,confirming its practical application in cadmium treatment.Fourier transform infrared spectrum revealed that carboxyl,amino and hydroxyl groups on biosorbent R.cohnii surface were responsible for the biosorption of cadmium.展开更多

Single nitrogen tryptone and peptone are suitable to growth, the dry weight biomass is up to 2.78 g/L and 2.70g/L respectively on medium containing 0.34 g/L nitrogen. Peptone is a favorable nitrogen source for DHA accumulation, DHA yield increases up to 338.56 mg/L. Using peptone and KNO3 as a multiple nitrogen source, the highest biomass and DHA yield are obtained from media containing 0.34 g/L nitrogen in which the ratio of peptone-N:NO3--N is 1:2,and the biomass and DHA yield are 2.98 g/L(DW) and 527.97mg/L respectively.展开更多

The assembled nuclei, being surrounded by a continuous double membrane containing nuclear pores and the uniformly dispersed chromatin fibers are morphologically distinguishable from that of Dinoflagellate Crythecodinium cohnii. However, incubation of dinoflagellate Crythecodinium cohnii chromosomes in the extracts from dinoflagellate Crythecodinillm cohnii cells does not induce nuclear reconstitution.展开更多

We are highlighting this case not only for its rarity but rapidity of worsening also. This report indicates that Staphylococcus cohnii are not as innocuous as once thought to be and these organisms should not be disregarded as possible skin contaminants.
