Assay for Hydroxyproline of Collagen Sponge

The content of hydroxyproline of collagen sponge is tested by Woessner Method I.The hydroxyproline standard curve and regression equation are described.The content of hydroxyproline of collagen sponge is tested subsequently.The result show that the hydroxyproline content is consistent with that of collagen protein,which could be concluded that the purity of collagen sponge,either prepared in our institute or that from abroad is high.

胶原海绵的羟脯氨酸含量测定

选用普通实验室均能实现的 Woessner第 法对自制胶原海绵和 Gelfix(国外样品 )进行了羟脯氨酸含量测定 ,结果表明 ,该方法操作简单 ,重复性好 :自制胶原海绵的羟脯氨酸含量稳定 ,与国外样品的羟脯氨酸含量接近 ,且与胶原蛋白的羟脯氨酸含量接近 。

三重四级杆液质联用测定胶原蛋白海绵中L-羟脯氨酸含量

目的 建立液质联用法测定胶原蛋白海绵中L-羟脯氨酸的方法。方法 采用6 mol·L^(-1)盐酸-0.5%苯酚酸解胶原蛋白海绵,以乙腈-0.1%甲酸(5∶95)为流动相,质谱测定特征氨基酸L-羟脯氨酸,以茶氨酸为内标定量分析。结果 方法检出限为0.02 ng,线性范围为2.04~30.6μg·m L^(-1),方法的重复性RSD为2.4%,准确度为98.3%(RSD=2.3%)。结论 本方法准确可靠,可作为胶原蛋白类产品的含量测定方法。
