Conserved dopamine neurotrophic factor protects and rescues dopaminergic neurodegeneration induced by 6-hydroxydopamine in vivo,but its potential value in treating Parkinson's disease remains controversial.Here,we us...Conserved dopamine neurotrophic factor protects and rescues dopaminergic neurodegeneration induced by 6-hydroxydopamine in vivo,but its potential value in treating Parkinson's disease remains controversial.Here,we used the proteasome inhibitors lactacystin and MG132 to induce neurodegeneration of PC12 cells.Afterwards,conserved dopamine neurotrophic factor was administrated as a therapeutic factor,both pretreatment and posttreatment.Our results showed that(1)conserved dopamine neurotrophic factor enhanced lactacystin/MG132-induced cell viability and morphology,and attenuated alpha-synuclein accumulation in differentiated PC12 cells.(2)Enzyme linked immunosorbent assay showed up-regulated 26S proteasomal activity in MG132-induced PC12 cells after pre-and posttreatment with conserved dopamine neurotrophic factor.Similarly,26S proteasome activity was upregulated in lactacystin-induced PC12 cells pretreated with conserved dopamine neurotrophic factor.(3)With regard proteolytic enzymes(specifically,glutamyl peptide hydrolase,chymotrypsin,and trypsin),glutamyl peptide hydrolase activity was up-regulated in lactacystin/MG132-administered PC12 cells after pre-and posttreatment with conserved dopamine neurotrophic factor.However,upregulation of chymotrypsin activity was only observed in MG132-administered PC12 cells pretreated with conserved dopamine neurotrophic factor.There was no change in trypsin expression.We conclude that conserved dopamine neurotrophic factor develops its neurotrophic effects by modulating proteasomal activities,and thereby protects and rescues PC12 cells against neurodegeneration.展开更多
Nest-site selection patterns of Red-crowned cranes(Grus japonensis) and the effects of environmental variables were studied during the years of 2002-2008 in Zhalong Nature Reserve,Qiqihar city,northeast China.The ne...Nest-site selection patterns of Red-crowned cranes(Grus japonensis) and the effects of environmental variables were studied during the years of 2002-2008 in Zhalong Nature Reserve,Qiqihar city,northeast China.The nest-site selection pattern of Red-crowned cranes included two orders and three choices:the choice of nest-site habitat type within the macro-habitat order,nest zone selection and nest-site micro-habitat selection within the micro-habitat order.Various habitats(such as Carex swamps and reed fire districts) can be selected as the nest sites for Red-crowned cranes,of which reed swamps(93.15%) are given a preference.Factor Analysis reveals that the micro-habitat selection are affected by four main factors:fire,security(concealment /disturbance),incubation(conditions,nest-material),and food.Further analysis reveals that Red-crowned cranes have certain adaptability to the changes of nesting habitat quality in the Zhalong wetlands.In conclusion,fire,reeds,and water were the most important variables for nest-site habitat selection of Red-crowned Cranes in Zhalong Nature Reserve.展开更多
The Revised Universal Soil Loss Equation(RUSLE)is widely used to estimate regional soil erosion.However,quantitative impacts of soil and water conservation(SWC)measures on conservation practice factor(P)of the RUSLE r...The Revised Universal Soil Loss Equation(RUSLE)is widely used to estimate regional soil erosion.However,quantitative impacts of soil and water conservation(SWC)measures on conservation practice factor(P)of the RUSLE remain largely unclear,especially for the mountainous and hilly areas.In this study,we improved the RUSLE by considering quantitative impacts of different SWC measures on the P factor value.The improved RUSLE was validated against the long-term(2000-2015)soil erosion monitoring data obtained from 96 runoff plots(15—35°)in mountainous and hilly areas of Hubei Province,China;the result presented a high accuracy with the determination coefficient of 0.89.Based on the erosion monitoring data of 2018 and 2019,the Root Mean Square Error of the result by the improved RUSLE was 28.0%smaller than that by the original RUSLE with decrement of 19.6%—24.0%in the average P factor values,indicating that the soil erosion modelling accuracy was significantly enhanced by the improved RUSLE.Relatively low P factor values appeared for farmlands with tillage measures(P<0.53),grasslands with engineering measures(P<0.23),woodlands with biological measures(P<0.28),and other land use types with biological measures(P<0.51).The soil erosion modulus showed a downward trend with the corresponding values of 1681.21,1673.14,1594.70,1482.40 and 1437.50 t km^(-2)a-1 in 2000,2005,2010,2015 and 2019,respectively.The applicability of the improved RUSLE was verified by the measurements in typical mountainous and hilly areas of Hubei Province,China,and arrangements of SWC measures of this area were proposed.展开更多
