Interaction of major facilitator superfamily domain containing 2A with the blood-brain barrier

The functional and structural integrity of the blood-brain barrier is crucial in maintaining homeostasis in the brain microenvironment;however,the molecular mechanisms underlying the formation and function of the blood-brain barrier remain poorly understood.The major facilitator superfamily domain containing 2A has been identified as a key regulator of blood-brain barrier function.It plays a critical role in promoting and maintaining the formation and functional stability of the blood-brain barrier,in addition to the transport of lipids,such as docosahexaenoic acid,across the blood-brain barrier.Furthermore,an increasing number of studies have suggested that major facilitator superfamily domain containing 2A is involved in the molecular mechanisms of blood-brain barrier dysfunction in a variety of neurological diseases;however,little is known regarding the mechanisms by which major facilitator superfamily domain containing 2A affects the blood-brain barrier.This paper provides a comprehensive and systematic review of the close relationship between major facilitator superfamily domain containing 2A proteins and the blood-brain barrier,including their basic structures and functions,cross-linking between major facilitator superfamily domain containing 2A and the blood-brain barrier,and the in-depth studies on lipid transport and the regulation of blood-brain barrier permeability.This comprehensive systematic review contributes to an in-depth understanding of the important role of major facilitator superfamily domain containing 2A proteins in maintaining the structure and function of the blood-brain barrier and the research progress to date.This will not only help to elucidate the pathogenesis of neurological diseases,improve the accuracy of laboratory diagnosis,and optimize clinical treatment strategies,but it may also play an important role in prognostic monitoring.In addition,the effects of major facilitator superfamily domain containing 2A on blood-brain barrier leakage in various diseases and the research progress on cross-blood-brain barrier drug delivery are summarized.This review may contribute to the development of new approaches for the treatment of neurological diseases. 展开更多

BACKGROUND Thrombocytopenia 2,an autosomal dominant inherited disease characterized by moderate thrombocytopenia,predisposition to myeloid malignancies and normal platelet size and function,can be caused by 5’-untranslated region(UTR)point mutations in ankyrin repeat domain containing 26(ANKRD26).Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1)have been identified as negative regulators of ANKRD26.However,the positive regulators of ANKRD26 are still unknown.AIM To prove the positive regulatory effect of GATA binding protein 2(GATA2)on ANKRD26 transcription.METHODS Human induced pluripotent stem cells derived from bone marrow(hiPSC-BM)INTRODUCTION Ankyrin repeat domain containing protein 26(ANKRD26)acts as a regulator of adipogenesis and is involved in the regulation of feeding behavior[1-3].The ANKRD26 gene is located on chromosome 10 and shares regions of homology with the primate-specific gene family POTE.According to the Human Protein Atlas database,the ANKRD26 protein is localized to the Golgi apparatus and vesicles,and its expression can be detected in nearly all human tissues[4].Moreover,UniProt annotation revealed that ANKRD26 is localized in the centrosome and contains coiled-coil domains formed by spectrin helices and ankyrin repeats[5,6].The most common disease related to ANKRD26 is thrombocytopenia 2(THC2),which is a rare autosomal dominant inherited disease characterized by lifelong mild-to-moderate thrombocytopenia and mild bleeding[7-9].Caused by the variants in the 5’-untranslated region(UTR)of ANKRD26,THC2 is defined by a decrease in the number of platelets in circulating blood and results in increased bleeding and decreased clotting ability[8,10].Due to the point mutations that occur in the 5’-UTR of ANKRD26,its negative transcription factors(TFs),Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1),lose their repression effect[11].The persistent expression of ANKRD26 increases the activity of the mitogen activated protein kinase and extracellular signal regulated kinase 1/2 signaling pathways,which are potentially involved in the regulation of thrombopoietin-dependent signaling and further impair proplatelet formation by megakaryocytes(MKs)[11].However,the positive regulators of ANKRD26,which might be associated with THC2 pathology,are still unknown. 展开更多

C50混凝土的制备及其对Cs^(+)、Ni^(2+)的吸附性能研究

为研究处置库屏障材料混凝土对放射性核素的阻滞能力,制备了C50混凝土材料并研究其对放射性核素的吸附能力。发现屏障材料C50混凝土主要由含钙和铝的硅酸盐矿物组成,其在微观尺度下呈现不规则多面体与板层结构,比表面积为2.786 m^(2)... 为研究处置库屏障材料混凝土对放射性核素的阻滞能力,制备了C50混凝土材料并研究其对放射性核素的吸附能力。发现屏障材料C50混凝土主要由含钙和铝的硅酸盐矿物组成,其在微观尺度下呈现不规则多面体与板层结构,比表面积为2.786 m^(2)·g^(-1),且具有较强的非定形特征。通过吸附试验,发现实验条件下C50混凝土对Cs^(+)、Ni^(2+)具有较好的吸附能力,达到平衡后对Cs^(+)的K_(d)值达到19.400 L·mg^(-1),吸附容量达到0.316 mg·g^(-1);吸附Ni^(2+)的K_(d)值达到465.142 L·mg^(-1),吸附容量达到96.375 mg·g^(-1)。随着Cs^(+)、Ni^(2+)初始浓度的增大,C50混凝土对Cs^(+)的K_(d)值逐渐减小后趋于稳定,对Ni^(2+)的K_(d)值逐渐减小后稳定增大,吸附容量均逐渐增大。通过环境因素影响试验,发现实验条件下pH对C50混凝土吸附Cs^(+)的影响相对较小,C50混凝土对Ni^(2+)的能力随着pH上升而增大。不同离子对Cs^(+)抑制作用由大到小的顺序依次为:K^(+)、Ca^(2+)、Mg^(2+)、SO_(4)^(2-)、Cl^(-)、NO_(3)^(-),随着温度的升高,C50混凝土对Cs^(+)的吸附能力缓慢减小;对Ni^(2+)抑制作用从大到小的顺序依次为:Mg^(2+)、Ca^(2+)、SO_(4)^(2-)、CO_(3)^(2-)。Ni^(2+)的吸附能力随着温度上升缓慢增大。研究结果可为处置库的建设提供基础数据支持。 展开更多

Nitrobenzene-containing industrial wastewater was degraded in the presence of ozone coupled with H2O2 by high gravity technology. The effect of high gravity factor, H2O2 concentration, pH value, liquid flow-rate, and reaction time on the efficiency for removal of nitrobenzene was investigated. The experimental results show that the high gravity technology enhances the ozone utilization efficiency with O3/H202 showing synergistic effect. The degradation efficiency in terms of the COD removal rate and nitrobenzene removal rate reached 45.8% and 50.4%, respectively, under the following reaction conditions, viz.: a high gravity factor of 66.54, a pH value of 9, a H2O2/O3 molar ratio of 1:1, a liquid flow rate of 140 L/h, an ozone concentration of 40 rag/L, a H2O2 multiple dosing mode of 6 mL/h, and a reaction time of 4 h. Compared with the performance of conventional stirred aeration mixers, the high gravity technology could increase the COD and nitrobenzene removal rate related with the nitrobenzene-containing wastewater by 22.9% and 23.3%, respectively. 展开更多

近红外荧光转换发光二极管(pc-LED)在成像和检测领域的快速发展突显了对近红外二区(NIR-Ⅱ区)发光材料的迫切需求。然而,商用紫外LED芯片激发的高效NIR-Ⅱ荧光材料匮乏,制约了近红外光谱技术的应用。本文通过高温固相法成功制备了MgAl_(... 近红外荧光转换发光二极管(pc-LED)在成像和检测领域的快速发展突显了对近红外二区(NIR-Ⅱ区)发光材料的迫切需求。然而,商用紫外LED芯片激发的高效NIR-Ⅱ荧光材料匮乏,制约了近红外光谱技术的应用。本文通过高温固相法成功制备了MgAl_(x)Ga_(2-x)O_(4)∶Ni^(2+)荧光材料,该材料的激发波长为390 nm,发射峰位于1300 nm,覆盖了NIR-Ⅱ区域。半高宽约为220 nm的超宽带NIR发射是由于Ni^(2+)处于一个弱晶体场环境中,这种环境是由于MgAl_(x)Ga_(2-x)O_(4)基体中[(Al/Ga)O_(6)]八面体中心周围的高电荷极化导致的空间不对称畸变造成的。通过改变体系中Al^(3+)与Ga^(3+)的离子比例,将NIR-Ⅱ区的发射强度大大提高,发光强度提升为原来的9倍左右。着重研究了Ni^(2+)掺杂最优比例MgAl_(1.5)Ga_(0.5)O_(4)的光谱特性。探讨了体系的温度特性,使用掺Ni^(2+)的近红外荧光粉和商业高效紫外LED芯片(@395 nm)构建了NIR pc-LED,研究表明该材料体系可能在pc-LED光谱技术中展现出较大潜力。这种阳离子调制策略将给近红外发光材料的性能提升提供有效的方法。 展开更多

A kind of active composite antibacterial material was prepared with CaHPO4 as the container, Ag^+ and Zn^2+ were adsorbed through ion-exchange, then it was doped with small scale of rare earth and photocatalyst, and was finally calcined at a certain temperature. The properties and application of the composite material antibacterial were investigated. Some tests show that the as-prepared antibacterial powders modified by opaque agents such os SnO2 and ZrO2, possess beautiful white and excellent climate resistance at normal temperatures and are promising candidate materials for antibacterial plastics and dope. The result of the application in glaze indicates that Ag^+ can still exist stably, with no color change for the glaze, even being sintered at 1200℃ . SEM , EDS , antibacterial activity analyses and contrast tests reveal that the as-prepared antibacterial powders and the antibacterial glaze both have excellent antibacterial activities, without color change, in the case of dark or brightness. 展开更多

近红外光谱技术在食品科学、信息安全、生物医疗等重大国计民生领域的应用对近红外光源提出了越来越高的要求。研发具有高效超宽带发射的近红外光源因此成为一项重要且迫切的研究课题。本文采用熔融淬火法在氟硅酸盐玻璃体系SiO_(2)-K_(... 近红外光谱技术在食品科学、信息安全、生物医疗等重大国计民生领域的应用对近红外光源提出了越来越高的要求。研发具有高效超宽带发射的近红外光源因此成为一项重要且迫切的研究课题。本文采用熔融淬火法在氟硅酸盐玻璃体系SiO_(2)-K_(2)CO_(3)-KF·2H_(2)O-MgF_(2)中成功析出了钙钛矿型KMgF_(3)纳米晶体。通过改变玻璃组分和热处理温度可以调控氟化物纳米晶相的析出,得到析晶和透明度最佳的微晶玻璃样品。玻璃中KMgF_(3)纳米晶体为Cr^(3+)和Ni^(2+)提供了稳定的八面体配位和低声子能量发光环境,在450 nm蓝光激发下,基于Cr^(3+)到Ni^(2+)的能量传递实现了Cr^(3+)(700~1200 nm)和Ni^(2+)(1400~1700 nm)双宽带近红外发射,并且双宽带近红外发光强度随离子掺杂浓度而可调变化。荧光光谱和荧光衰减曲线表征证明了Cr^(3+)到Ni^(2+)的能量传递过程,对应的能量传递效率为52.2%,能量传递机制为电子偶极-四极相互作用。研究结果不仅可以为系统掌握透明光学材料的超宽带发光规律提供基础数据,同时有助于设计开发低成本、高效率的近红外宽带光源。 展开更多

The thermodynamical properties of MgCl_2 in KCI-MgCl_2-LiCl molten electrolytes containing MgCl_2 below 0.5 (mole fraction, the same below) have been determined from the interchange energies of two binary systems KCI-MgCl_2 and LiCI-MgCl_2, by means of a model on the assumptions that the electrolytes in the solution are treated as independent particles instead of their ion forms and the interchange energy between the component pair KCI-LiCl is ignored when compared with those of component pairs KCl-MgCl_2 and MgCl_2-LiCl. The interchange energies, wKCl-MgCl_2 and wMgcCl_2-Licl, are obtained as-70000 and -13800 J.mol-1, from the corresponding binary solutions, respectively. 展开更多

The apparent 1^st order rate constant of photodegradation of formaldehyde by carbon containing TiO2 nanoparticles has been investigated by numerical integration of mass transfer equation with measured degradation degree using a tubular photoreactor. The carbon containing TiO2 nanoparticles are synthesized by the oxidation of TiCl4 in propane/air flame CVD process with futile fraction up to 0.3 and carbon mass fractions up to 0.22, respectively. Thin TiO2 film is coated on the wall of the tubular reactor by sedimentation method. Effects of rutile mass fraction and carbon content have been examined on the apparent 1 ^st order rate constant and results show that, at 570ppm of formaldehyde loaded air stream, 80% relative humidity and about 100nm thin TiOa film, the 1^st order rate constant increases with increasing rutile mass fraction up to 0.3, occurs a maximum at the carbon content of about 5% by weight and is about 2.5 times of that at carbon content about zero or above 10%. 展开更多

背景:卷曲螺旋结构域蛋白2(coiled-coil-helix-coiled-coil-helix domain-containing 2,CHCHD2)能否调控PINK1/Parkin介导的线粒体自噬在帕金森病中发挥神经保护作用尚未可知。目的:探讨CHCHD2在6-羟基多巴胺诱导的帕金森病细胞模型中对... 背景:卷曲螺旋结构域蛋白2(coiled-coil-helix-coiled-coil-helix domain-containing 2,CHCHD2)能否调控PINK1/Parkin介导的线粒体自噬在帕金森病中发挥神经保护作用尚未可知。目的:探讨CHCHD2在6-羟基多巴胺诱导的帕金森病细胞模型中对PINK1/Parkin信号通路介导的线粒体自噬发挥的作用及机制。方法:利用重组质粒转染技术过表达或敲低CHCHD2,用6-羟基多巴胺构建SH-SY5Y细胞帕金森病模型后分对照组、模型组、过表达阴性对照+6-羟基多巴胺组、敲低阴性对照+6-羟基多巴胺组、过表达CHCHD2+6-羟基多巴胺组和敲低CHCHD2+6-羟基多巴胺组。Western blot及RT-qPCR检测CHCHD2的表达;Western blot检测LC3Ⅰ/Ⅱ、p62、MFN1、COXⅣ、DRP1、PINK1、Parkin、TIM23、Bax、Bcl-2及cleavedcaspase3蛋白表达;CCK-8、JC-1和活性氧试剂盒检测细胞活性、线粒体膜电位和活性氧水平,单丹磺酰尸胺染色观察细胞自噬情况,透射电镜观察自噬溶酶体。结果与结论:①与对照组相比,模型组细胞活性、线粒体膜电位及CHCHD2、PINK1、Parkin蛋白表达降低,活性氧水平、凋亡水平及LC3Ⅰ/Ⅱ、p62蛋白表达升高(P<0.05),并观察到自噬溶酶体的存在;②与模型组相比,过表达CHCHD2能降低细胞活性氧水平,升高线粒体膜电位及PINK1、Parkin和MFN1蛋白表达水平,并观察到线粒体自噬溶酶体增多,而敲低CHCHD2后有与上述相反的作用,并伴有COXⅣ、TIM23和p-DRP1蛋白表达的升高(P<0.05);③与模型组相比,过表达CHCHD2能减少细胞凋亡,上调Bcl-2并下调Bax及cleavedcaspase3蛋白的表达,而敲低CHCHD2后有与上述相反的作用(P<0.05);④结果提示,CHCHD2在6-羟基多巴胺诱导的帕金森病细胞模型中可以通过促进PINK1/Parkin介导的线粒体自噬改善线粒体功能从而减轻细胞凋亡发挥神经保护作用。 展开更多

The distribution of Fe3+ ions on different structural aggregates was revealed by means of EPR, as depending on Fe2O3 concentration. Strongly distorted octahedral sites were detected for the isolated paramagnetic ions, and also clusters of Fe ions especially at high Fe2O3 content of samples. Reaction of 2-pyridone, copper acetate and terbium(or yttrium) perchlorate in acetone with the mole ratio 6: 2: 1 results in the formation of heteronuclear complex Ln_2Cu_4L_8 (HL)_4 (OH)_2 (ClO_4)_4 (H_20)_(10) 2CH_3... Reaction of 2-pyridone, copper acetate and terbium(or yttrium) perchlorate in acetone with the mole ratio 6: 2: 1 results in the formation of heteronuclear complex Ln_2Cu_4L_8 (HL)_4 (OH)_2 (ClO_4)_4 (H_20)_(10) 2CH_3COCH_3(Ln = Tb (1), Y (2)). By recrystallizing (1) in CHCl_3 single crystals were obtained and the structure was determined by four-circle diffractometer. Data showed that the crystal is in space group C2/m with a=27. 454(9)A, b=13, 608A, c=30. 556(11)A, β=99. 89(3)°, v=11245. 7(7. 5)A^3. The structure was solved by a combination of Patterson method and Fourier technique. The final R value is 0. 103. In the structure, four copper and two terbium ions are bridged by 2-pyridone anions to form an essentially octahedral Cu_4Tb_2 core. The terbium atoms are each eight-coordinate and the copper atoms are five-coordinate. 展开更多

Eight novel Schiff Bases, from 6,6′-diformyl-2,2′-bipyridyl (1a, 43%)with O, N, S and F containing amines: Thiosemicarbazide (2a, 70%), 4-Ethyl-3-thiosemicabazide (2b, 75%), 4,4-Dimethyl-3-thio-semicarbazide (2c, 75%), S-benzyldithiocarbazide, SBDTC (2d, 80%), (Trifluromethyl) phenylhy-drazine (2e, 80%), 4-Phenyl-3-thiosemcarbazide (2f, 80%), Thiocarbazide (2g, 70%), 2-Amino-thiophenol (2h, 65%), have been synthesized. The conventional method of synthesis of the Schiff bases involves refluxing the reaction mixture containing the diformyls and amines for 1 hour. The solid products that had formed were filtered off using suction filtration. In few reactions, 2 - 3 drops of conc. sulfuric acid were used to obtain high yield. The structures of all eight novel synthesized compounds have fully been characterized by spectroscopic (IR, NMR, MS) methods. 展开更多

The Ni 2+/poly(o-aminophenol)electropolymerization was carried out by a continuous cyclic voltammetric scanning at a carbon paste electrode in a mixture of 0.02 mol/L o-aminophenol,0.3 mol/L NiSO 4 and 0.6 mol/L HClO 4.dsDNA was linked to the conducting polymer via three-dimensional structure of polymer film and electrostatic attraction between the negatively charged phosphate backbone of DNA and Ni 2+ ion.The results were testified by the electrochemical methods by using Co(phen)2+3 as an electrochemical indicator.The currents of redox peaks of Co(phen)2+3 increased after immobilizing dsDNA.The conditions for the preparation of Ni 2+/P-OAP/CPE and DNA immobilization were optimized.The voltammetric behavior of dopamine was investigated at the modified electrode.It was found that the modified electrode exhibited a highly electrocatalytic activity toward dopamine oxidation.Differential pulse voltammetry was used for determination of dopamine in pH 5.1 phosphate buffer solution.A linear response of the peak current versus the concentration was found in the range from 5.0×10-6 to 2.0×10-4mol/L with 1.0×10-2 mol/L AA(ascorbic acid)coexisting.The presence of a high concentration of AA did not interfere with the determination.The proposed method exhibits a good recovery and reproducibility. 展开更多
