Background The development of a sustainable business model with social acceptance,makes necessary to develop new strategies to guarantee the growth,health,and well-being of farmed animals.Debaryomyces hansenii is a yeast species that can be used as a probiotic in aquaculture due to its capacity to i)promote cell proliferation and differen-tiation,ii)have immunostimulatory effects,iii)modulate gut microbiota,and/or iv)enhance the digestive function.To provide inside into the effects of D.hansenii on juveniles of gilthead seabream(Sparus aurata)condition,we inte-grated the evaluation of the main key performance indicators coupled with the integrative analysis of the intestine condition,through histological and microbiota state,and its transcriptomic profiling.Results After 70 days of a nutritional trial in which a diet with low levels of fishmeal(7%)was supplemented with 1.1%of D.hansenii(17.2×10^(5) CFU),an increase of ca.12%in somatic growth was observed together with an improve-ment in feed conversion in fish fed a yeast-supplemented diet.In terms of intestinal condition,this probiotic modu-lated gut microbiota without affecting the intestine cell organization,whereas an increase in the staining intensity of mucins rich in carboxylated and weakly sulphated glycoconjugates coupled with changes in the affinity for certain lectins were noted in goblet cells.Changes in microbiota were characterized by the reduction in abundance of several groups of Proteobacteria,especially those characterized as opportunistic groups.The microarrays-based transcrip-tomic analysis found 232 differential expressed genes in the anterior-mid intestine of S.aurata,that were mostly related to metabolic,antioxidant,immune,and symbiotic processes.Conclusions Dietary administration of D.hansenii enhanced somatic growth and improved feed efficiency param-eters,results that were coupled to an improvement of intestinal condition as histochemical and transcriptomic tools indicated.This probiotic yeast stimulated host-microbiota interactions without altering the intestinal cell organization nor generating dysbiosis,which demonstrated its safety as a feed additive.At the transcriptomic level,D.hansenii pro-moted metabolic pathways,mainly protein-related,sphingolipid,and thymidylate pathways,in addition to enhance antioxidant-related intestinal mechanisms,and to regulate sentinel immune processes,potentiating the defensive capacity meanwhile maintaining the homeostatic status of the intestine. 展开更多

Background Pink bollworm,Pectinophora gossypiella(Saunders)(Lepidoptera:Gelechiidae)has become a poten-tial pest of cotton by causing substantial yield losses around the world including Pakistan.Keeping in view the facts like limited research investigations,unavailability,and high cost of artificial diet’s constituents and their premixes,the present research investigations on the dietary aspect of P.gossypiella were conducted.The larvae of P.gossypiella were reared on different diets that were prepared using indigenous elements.The standard/laboratory diet com-prised of wheat germ meal 34.5 g,casein 30.0 g,agar–agar 20.0 g,sucrose 10.0 g,brewer’s yeast 5.0 g,α-cellulose 1.0 g,potassium-sorbate1.5 g,niplagin 0.5 g,decavitamin 0.01 g,choline-chloride 0.06 g,maize-oil 3.30 g,honey 2.0 g,and water 730.0 mL.Alternatives to cotton bolls and wheat germ meal were okra seed sprouts,okra fruit,cottonseed meal,and okra seed meals,which were included in the study to introduce an efficient and economic mass-rearing system.Results The larval development completed in 19.68d±0.05 d with a weight of 20.18mg±0.20 mg at the fourth instar fed on the cottonseed meal-based diet instead of wheat germ meal based diet.On the same diet,84.00%±4.00%,17.24 mg±0.03 mg,and 7.76d±0.06 d were recorded as pupae formation,pupal weight,and pupal duration,respectively.Adult emergence,76.00%±1.00%was recorded from pupae collected from larvae raised on cottonseed meal-based diet.These male and female moths lived for 40.25d±0.10 d,and 44.34d±0.11 d,respectively.Females deposited 21.28±0.04 eggs per day with the viability of 65.78%±0.14%.The larval mortal-ity at the fourth instar was 37.20%±1.36%and malformed pupation of 12.00%±1.41%was recorded.Replacement of wheat germ meal with that of local meals(cottonseed and okra seed)in the standard laboratory diet has saved 463.80 to 467.10 PKR with 1.62 to 1.63 cost economic returns,respectively.Conclusion This research is of novel nature as it provides a concise and workable system for the economic and suc-cessful rearing of P.gossypiella under laboratory conditions. 展开更多

The inclusion of PepSoyGen (PSG), a commercially-available fermented soybean meal product, was evaluated with juvenile rainbow trout, Oncorhynchus mykiss in an initial 70-day feeding trial, with a supplemental trial involving a subset of the experimental diets continuing for an additional 40 d. Six diets containing 0%, 10%, 20%, 30%, 40%, or 50% PSG, with the PSG directly replacing fish meal, were used in the first trial. There were no significant differences in weight gain or feed conversion ratio between the fish meal-based control diet and diets containing up to 30% PSG. However, weight gain was significantly reduced and feed conversion ratio significantly increased with the 40% and 50% PSG diets. No health assessment differences were observed in fish receiving any of the diets, and no evidence of gross gut inflammation was evident. There were no significant differences in weight gain or feed conversion ratio among the four dietary treatments ranging from 0% to 30% PSG which were fed for an additional 40 d after the initial 70-d trial (110 days total). Based on these results, juvenile rainbow trout diets can contain up to 30% PSG without any loss of rearing performance, thereby replacing at least 60% of the fish meal. 展开更多

Substitution of silkworm pupae (Bombyx mori) for fish meal in broodstock diets for snakeskin gourami (Trichogaster pectoralis) was conducted in order to evaluate the effect of silkworm pupae on feed digestibility and broodstock performance. Snakeskin gourami broodfish were fed with isocaloric diets (DE 3,000 a: 50 Kcal/kg) contained 32% crude protein. The levels of substitution were 0%, 50%, 75% and 100% of protein from fish meal (equal to 0%, 14.57%, 21.75% and 29.03% silk worm by weight). The study was assigned in CRD with 4 treatments and 3 replications. Six hundred of snakeskin gourami with five-month old and average weight of 78.67 ± 2.15 g in female and average weight of 73.00 ± 2.49 g in male were stocked in 12 cages. Fifty fishes of 25 females and 25 males were reared in each cage and fed experimental diet for five months. The result indicated that protein digestibility of test diets were significantly decrease (P 〈 0.05) according to the level of silkworm pupae in diets where as the dry matter digestibility showed no significantly different (P 〉 0.05). The growth performance and egg fecundity were significantly decreased (P 〈 0.05) according to the levels of silkworm pupae. In contrast, egg quality in term of fry number on first spawned, fingering number on first spawned were not significantly different (P 〉 0.05). The survival rate of one month nursing fish on first spawned were significantly different (P 〈 0.05). Hence, silkworm pupae could substitution for 50% of protein from fish meal (14.57% by weight) in snakeskin gourami broodstock diets without any adverse effect on egg quality. 展开更多

This 121-day experiment evaluated the rearing performance of brown trout Salmo trutta fed one of two isonitrogenous and isocaloric diets and reared at velocities of either 2.8 or 16.1 cm/s. Fishmeal was the primary protein source for the reference diet, and bioprocessed soybean meal replaced approximately 67% of the fishmeal in the experimental diet. At the end of the experiment, there were no significant differences in gain, percent gain, feed conversion rates, or specific growth rates between the dietary treatments. There were also no significant differences in intestinal morphology, splenosomatic, hepatosomatic, and viscerosomatic indices related to diet composition. However, gain, percent gain, feed fed, and specific growth rate were all significantly greater in brown trout reared at the higher velocity. No significant differences in any of the other variables measured were observed between the velocity treatments. There were no significant interactions between diet and velocity in any of the variables. Based on the results of this study, bioprocessed soybean meal can replace at least 67% of the fishmeal in brown trout diets, regardless of the rearing velocities used in this study. However, higher rearing velocities are recommended to maximize juvenile brown trout growth rates. 展开更多

Effect of Fish Meal Replacement by Unconventional Meal in the Post-Larval Diet of Clarias Gariepinus in Benin (West Africa)

To reduce the pressure on aquatic resources due to the use of fishmeal in fish feed, the mastery and use of alternative sources of fishmeal in all stages of fish development are essential. Five diets including 4 experimental and one control diet were considered. Experimental diets of post-larvae were developed from unconventional ingredients in which fishmeal was completely replaced by a mixture of brewer’s yeast, chicken viscera and maggots. These foods were tested on Clarias gariepinus post-larvae with an initial average weight of 80 ± 1 mg. 1500 fish individuals were equally distributed in 15 concrete basins with a volume of 500 L forming five treatments in triplicate. Fish individuals were fed, manually at a ration rate of 20%, four times per day. The specific growth rates were 8.38%/day in imported food and 6.35%/day in the experimental food receiving 45.5% of protein from unconventional meal. Similar results were obtained with the feed consumption index. These encouraging results show that it is possible to partially or completely replace fishmeal in catfish feed and obtain an economically profitable production. The recovery of waste such as chicken viscera, brewer’s yeast and soybean meal in fish food formulation as tested in this study is a good contribution to environmental sanitation. 展开更多

The main animal protein ingredient in fish diets is most often fishmeal because of its nutritional quality.However,the limited availability and increasingly cost of fishmeal has lead to investigations of either lowering or replacing the fishmeal content with more economic protein sources of animal and/or plant origin.The research for appropriate and cheap cost alternative sources of protein to use in commercial fish diets will be the most important factor in intensive fish culture development.Insect meals are healthy and nutritious alternatives to fish meal due to their rich nutritional values,particularly protein,fat and minerals.Housefly maggots(Musca domestica)meal is also rich in B complex vitamins,trace elements and phosphorus.From the results of previous studies,Housefly maggots meal can be used successfully to replace the fish meal portion partially or completely in the fish diets.Also,the results observed that not physiological stressful was introduced in the fish by feeding Housefly maggots meal diets.This indicates that Housefly maggots meal were well utilized by the fish thus resulting in good growth of fish.In other study,observed a best growth performance with fish feeding on diets containing maggot’s meal compared with fish feeding on fishmeal diet.This indicates the high nutritional quality and fish acceptance of maggot’s meal. 展开更多

本试验旨在研究液态饲喂杂粕型饲粮对生长猪生长性能、养分消化率、血清炎性因子及粪便短链脂肪酸(SCFAs)含量的影响。选取平均体重为(30.00±4.26)kg的“杜×长×大”三元杂交猪126头,随机分为3组,每组7个重复,每个重复6... 本试验旨在研究液态饲喂杂粕型饲粮对生长猪生长性能、养分消化率、血清炎性因子及粪便短链脂肪酸(SCFAs)含量的影响。选取平均体重为(30.00±4.26)kg的“杜×长×大”三元杂交猪126头,随机分为3组,每组7个重复,每个重复6头猪。试验Ⅰ组饲喂豆粕型颗粒饲粮;试验Ⅱ组饲喂杂粕型颗粒饲粮(由双低菜籽粕、玉米胚芽粕、喷浆玉米皮及米糠替代部分豆粕和玉米组成);试验Ⅲ组饲喂杂粕型液态饲粮,即在杂粕型饲粮的基础上加入2.7倍的水,混合均匀后饲喂。各组饲粮营养水平基本一致。试验期56 d。结果表明:1)试验全期,与试验Ⅰ组相比,试验Ⅱ组平均日采食量和平均日增重分别降低了2.29%和5.33%(P>0.05),料重比提高了3.40%(P>0.05);与试验Ⅱ组相比,试验Ⅲ组平均日采食量和平均日增重分别提高了7.04%(P<0.05)和6.32%(P>0.05)。2)试验Ⅱ组和试验Ⅲ组干物质、总能和粗蛋白质消化率极显著低于试验Ⅰ组(P<0.01),粗脂肪、磷和粗灰分消化率显著低于试验Ⅰ组(P<0.05)。试验Ⅱ组和试验Ⅲ组间各养分消化率均无显著差异(P>0.05)。3)试验Ⅲ组粪便丁酸含量显著高于试验Ⅱ组(P<0.05),与试验Ⅰ组差异不显著(P>0.05)。4)与试验Ⅰ组相比,试验Ⅱ组和试验Ⅲ组血清抗炎因子白细胞介素-10(IL-10)含量极显著提高(P<0.01),试验Ⅱ组血清促炎因子白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)含量显著提高(P<0.05);与试验Ⅱ组相比,试验Ⅲ组血清IL-1β含量显著降低(P<0.05),血清白细胞介素-6(IL-6)、白细胞介素-2(IL-2)和TNF-α含量有一定程度降低但未达到显著水平(P>0.05)。综上可知,与豆粕型颗粒饲粮相比,杂粕型颗粒饲粮会降低生长猪采食量和养分消化率,提高血清炎性因子含量,而通过液态方式饲喂杂粕型饲粮可改善饲粮适口性,提高生长猪采食量,降低血清促炎因子IL-1β含量,对养分消化率无改善作用。 展开更多

本试验旨在研究无鱼粉无豆粕饲料中补充蛋氨酸对鲤生长性能、氮循环和氨基酸沉积的影响,以及投喂频率和补充蛋氨酸对鲤餐后血浆氨基酸含量的影响。选取初始体重为27 g的鲤幼鱼540尾,随机分为2组(每组9个重复,每个重复30尾),分别投喂添加... 本试验旨在研究无鱼粉无豆粕饲料中补充蛋氨酸对鲤生长性能、氮循环和氨基酸沉积的影响,以及投喂频率和补充蛋氨酸对鲤餐后血浆氨基酸含量的影响。选取初始体重为27 g的鲤幼鱼540尾,随机分为2组(每组9个重复,每个重复30尾),分别投喂添加0(C组)和3 g/kg蛋氨酸(DL-蛋氨酸形式,纯度为98%)(M组)的无鱼粉无豆粕饲料,2种试验饲料中蛋氨酸含量分别为3.85和6.83 g/kg饲料(1.14和1.99 g/16 g N),进行为期8周的生长试验。生长试验结束后称重,采样后继续进行1周的投喂频率试验。投喂频率试验中,将C组和M组试验鱼分别随机分为3组,投喂频率分别为1、2和3次,共6组,每组3个重复,每个重复25尾。采集餐后不同时间点血浆样品,测定血浆游离氨基酸含量。结果显示:1)在无鱼粉无豆粕饲料中补充蛋氨酸显著提高了鲤的总摄食量、特定生长率和蛋白质效率(P<0.05),显著降低了摄食率和饲料系数(P<0.05);2)与C组相比,M组鲤氮摄入量从54.33 g/桶显著提高到60.73 g/桶(P<0.05),但氮排放量从41.08 g/桶显著降低到37.93 g/桶(P<0.05),各氨基酸以及总必需氨基酸和总非必需氨基酸沉积率均显著提高(P<0.05),血浆尿素氮和总胆固醇含量显著下降(P<0.05);3)投喂频率达到3次时,饲料中补充蛋氨酸可显著提高鲤血浆蛋氨酸、赖氨酸曲线下面积(AUC)(P<0.05)。综上所述,在无鱼粉无豆粕饲料中补充蛋氨酸至鲤需求量,在日投喂3次的养殖模式下,鲤的生长性能得到显著改善,饲料效率和氮沉积率显著提高,说明提高投喂频率是提高晶体氨基酸利用率的有效方法。 展开更多

本试验旨在研究玉米-杂粕型饲粮中添加不同水平的复合纤维素酶对蛋鸡生产性能、蛋品质、肝脏中糖代谢相关酶活性、内源消化酶活性以及肠道中葡萄糖转运载体基因表达的影响,并筛选出复合纤维素酶的适宜添加量。选取288只健康状况良好的5... 本试验旨在研究玉米-杂粕型饲粮中添加不同水平的复合纤维素酶对蛋鸡生产性能、蛋品质、肝脏中糖代谢相关酶活性、内源消化酶活性以及肠道中葡萄糖转运载体基因表达的影响,并筛选出复合纤维素酶的适宜添加量。选取288只健康状况良好的53周龄海兰褐蛋鸡,随机分为4组,每组6个重复,每个重复12只鸡。对照组饲喂玉米-杂粕(豆粕-棉籽粕-菜籽粕)型饲粮,3个试验组在玉米-杂粕型饲粮中分别添加200、400、600 mg/kg复合纤维素酶(主要成分为纤维素酶、木聚糖酶、淀粉酶和蛋白酶)。预试期7 d,正试期84 d。结果显示:1)与对照组相比,400 mg/kg组产蛋率显著提高(P<0.05),料蛋比和平均日采食量显著降低(P<0.05)。2)玉米-杂粕型饲粮中添加不同水平复合纤维素酶对蛋鸡蛋品质各项指标均无显著影响(P>0.05)。3)与对照组相比,玉米-杂粕型饲粮中添加400或600 mg/kg复合纤维素酶可以显著提高蛋鸡肝脏中丙酮酸激酶活性(P<0.05)。4)与对照组相比,400 mg/kg组蛋鸡空肠中淀粉酶与胰蛋白酶活性显著提高(P<0.05)。5)与对照组相比,玉米-杂粕型饲粮中添加400或600 mg/kg复合纤维素酶均可以显著提高蛋鸡空肠中钠-葡萄糖协同转运蛋白-1(SGLT-1)mRNA相对表达量(P<0.05)。综上可知,在玉米-杂粕型饲粮中添加复合纤维素酶可以通过提高饲粮中营养物质的释放量而增强蛋鸡小肠消化酶活性及肝脏中糖代谢相关酶活性,还能提高空肠中SGLT-1 mRNA相对表达量,进而改善生产性能。在本试验条件下,蛋鸡玉米-杂粕型饲粮中复合纤维素酶的最适添加量为400 mg/kg。 展开更多

核桃粕酶解物抑制脂质积累的机制

利用分化的3T3-L1脂肪细胞和高脂饮食(HFD)诱导的肥胖C57BL/6小鼠模型,研究核桃粕酶解物的抗肥胖作用。采用油红O脂肪染色法检测脂肪细胞脂质积累情况,采用试剂盒检测细胞甘油三脂(TG)、甘油(Gly)和游离脂肪酸(FFA)含量,采用实时荧光定... 利用分化的3T3-L1脂肪细胞和高脂饮食(HFD)诱导的肥胖C57BL/6小鼠模型,研究核桃粕酶解物的抗肥胖作用。采用油红O脂肪染色法检测脂肪细胞脂质积累情况,采用试剂盒检测细胞甘油三脂(TG)、甘油(Gly)和游离脂肪酸(FFA)含量,采用实时荧光定量PCR(PCR RT-qPCR)检测脂肪合成相关基因(mRNA)的表达水平。结果表明,核桃粕酶解物显著降低脂肪细胞内脂滴聚积,显著降低脂肪细胞内TG和FFA的含量,增加细胞上清液中Gly含量,同时核桃粕酶解物显著降低脂肪细胞中脂肪合成相关基因PPAR-γ和C/EBPαmRNA表达水平;当核桃粕酶解物的质量浓度达到50μg/mL时,TG和FFA分别降低了40.14%和39.50%,Gly水平增加了35.12%;PPAR-γ和C/EBPα的mRNA表达水平分别降低了31.42%和55.25%。此外,核桃粕酶解物显著降低HFD小鼠的体质量、脂肪组织质量和食物摄入量。研究表明,核桃粕酶解物可以抑制3T3-L1脂肪细胞脂质积累,其作用机制与抑制脂质合成相关基因表达相关。此外,其还具有抑制HFD脂质积累的作用。 展开更多

本试验旨在探讨芝麻粕型低蛋白质饲粮对育肥猪生长性能、养分表观消化率及血清生化和抗氧化指标的影响。试验选取平均体重(61.58±5.62)kg的“杜×长×大”三元育肥猪288头,随机分为4组,每组6个重复,每重复12头猪。各组分... 本试验旨在探讨芝麻粕型低蛋白质饲粮对育肥猪生长性能、养分表观消化率及血清生化和抗氧化指标的影响。试验选取平均体重(61.58±5.62)kg的“杜×长×大”三元育肥猪288头,随机分为4组,每组6个重复,每重复12头猪。各组分别饲喂4种粗蛋白质水平(15%、14%、13%、12%)的芝麻粕型低蛋白质饲粮。预试期7 d,正试期60 d,其中前30 d为育肥前期,后30 d为育肥后期。结果表明:1)不同粗蛋白质水平的芝麻粕型低蛋白质饲粮对育肥猪生长性能无显著影响(P>0.05)。2)不同粗蛋白质水平的芝麻粕型低蛋白质饲粮对育肥猪干物质、粗纤维、钙和磷表观消化率无显著影响(P>0.05)。随着粗蛋白质水平的下降,粗蛋白质表观消化率呈线性下降(P<0.05)。3)在育肥前期,12%粗蛋白质水平组的血清总蛋白(TP)、白蛋白(ALB)、肌酐(CREA)、尿素氮(UN)、葡萄糖(GLU)含量和谷草转氨酶(AST)、谷丙转氨酶(ALT)活性显著低于15%粗蛋白质水平组(P<0.05)。在育肥后期,12%粗蛋白质水平组的血清TP、ALB、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、CREA、GLU含量和碱性磷酸酶(ALP)、AST活性显著高于15%粗蛋白质水平组(P<0.05)。4)在育肥前期,14%、13%和12%粗蛋白质水平组的血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性显著高于15%粗蛋白质水平组(P<0.05),13%和12%粗蛋白质水平组的血清丙二醛(MDA)含量显著低于15%粗蛋白质水平组(P<0.05)。在育肥后期,14%和13%粗蛋白质水平组的血清SOD、GSH-Px活性显著高于15%粗蛋白质水平组(P<0.05),13%粗蛋白质水平组的血清MDA含量显著低于15%粗蛋白质水平组(P<0.05)。由此可见,芝麻粕型低蛋白质饲粮对育肥猪生长性能及干物质、粗纤维、钙和磷表观消化率无显著影响,且能提高育肥猪的抗氧化能力。 展开更多

研究旨在对复合益生菌发酵玉米-豆粕型日粮产生酸溶蛋白的工艺进行优化,为复合益生菌发酵饲料的应用提供参考。在单因素试验的基础上以酸溶蛋白含量为响应值,以接种量、料水比、发酵时间、发酵温度为自变量,通过Box-Behnken设计试验,优... 研究旨在对复合益生菌发酵玉米-豆粕型日粮产生酸溶蛋白的工艺进行优化,为复合益生菌发酵饲料的应用提供参考。在单因素试验的基础上以酸溶蛋白含量为响应值,以接种量、料水比、发酵时间、发酵温度为自变量,通过Box-Behnken设计试验,优化复合益生菌发酵玉米-豆粕型日粮的发酵工艺。结果表明:乳酸杆菌RSG-1∶枯草芽孢杆菌B-1∶地衣芽孢杆菌Y5-39为1∶1∶4时,酸溶蛋白含量最高为5.30%;响应面优化后复合益生菌最佳发酵工艺为:含水量48%,接种量10%,发酵温度48℃,发酵时间10 d,酸溶蛋白含量为13.31%,在考虑生产实际的情况下,将发酵温度调整为37℃。综上,研究确定了多菌种组合固态发酵玉米-豆粕型日粮的最佳工艺条件:含水量为48%、接种量(乳酸杆菌∶枯草芽孢杆菌∶地衣芽孢杆菌=1∶1∶4)10%、发酵温度37℃、发酵时间10 d,酸溶蛋白含量达到13.06%。 展开更多
