To keep nostalgia in new urbanization is to preserve history, culture, memories, and humanistic spirits of the homeland, and also to maintain local cultural genes and carry forward rural civilization. The foundation of remembering the nostalgia is to intensify culture protection. And by means of protecting cultural heritage and style of the traditional settlement practically, preserving cultural gene, inheriting cultural memory, creating cultural brand and characteristic landscape etc., the nostalgia can meet the spiritual demands of the residents, such as homesickness, life emotion, historical emotion and cultural emotion in the process of new urbanization. Human primary epithelial cells of renal pelvis was established to investigate the adherence of uropathogenic Escherichia coli (UPEC) to this cell line, in which the primary cell culture was performed by using cultiv...Human primary epithelial cells of renal pelvis was established to investigate the adherence of uropathogenic Escherichia coli (UPEC) to this cell line, in which the primary cell culture was performed by using cultivation of the normal epithelium of renal pelvis in keratinocyte serum free medium (K-SFM) with epidermal growth factor (EGF) and bovine pituitary extract (BPE). Both UPEC132 obtained from urine specimen of patients with pyelonephritis and the pilus-free representative strain E. coli K-12p678- 54 were used to study the adherence of these strains on human primary epithelial cells of renal pelvis. The UPEC adherence was performed with observation on the morphological changes of the adhered cells, while the adhesion rates and indices were calculated in different times of experiment. In addition, the virulence genes hly and cnfl of UPEC132 were detected by multiplex PCR assay. In this study, the human primary epithelial cells of renal pelvis was found to exhibit the character of the transitional epithelial cells. Compared with the control group, the adhesion rates and indices began to increase from 15 min of the experiment time and reached its peak in 120 min. The adhesion rate and index of UPEC132 to human primary epithelial cells of renal pelvis were 74.4% and 34.0 respectively. Many microscopic changes in the primary cells adhered with UPEC132 could be detected, such as rounding or irregularity in shape, unevenness in staining and the cytoplasmic and nuclear changes. It suggests that human primary epithelial cells of renal pelvis can be used for the experiment on UPEC adhesion, thus providing a basis for the further study on the pathogenesis of UPEC.展开更多

Over past decades plant tissue culture has emerged as an alternative of whole plant cultivation in the production of valuable secondary metabolites.Adventitious roots culture of Panax ginseng and Echinacea purpure has...Over past decades plant tissue culture has emerged as an alternative of whole plant cultivation in the production of valuable secondary metabolites.Adventitious roots culture of Panax ginseng and Echinacea purpure has reached the scale of 1-10 kL.Some molecular biological techniques,such as transgenic technology and genetic stability are increasingly used in the studies on plant tissue cultures.The studies on elicitors have deepened into the induction mechanism,including signal molecules,functional genes,and so on.More and more biological elicitors,such as A.niger and yeast are used to increase the active compounds in plant tissue cultures.We also discussed the application of synthetic biology in the studies on biosynthesis of artemisinin,paclitaxel,and tanshinon.The studies on active ingredients biosynthesis of medicinal plants provide unprecedented possibilities to achieve mass production of active ingredients.Plant tissue cultures can not only produce active ingredients but also as experimental materials for biosynthesis.In order to improve the contents of active compounds in medicinal plants,following aspects could be carried out gene interference or gene silencing,gene overexpression,combination with chemical synthesis,application of elicitors,and site-directed mutagenesis of the key enzymes.展开更多
Abstract Objecitve To explore whether phosphoinositide specific phospholipase C (PLC) activation via G protein in vascular smooth muscle cells (VSMCs) is altered in spontaneously hypertensive rats (SHR). Met...Abstract Objecitve To explore whether phosphoinositide specific phospholipase C (PLC) activation via G protein in vascular smooth muscle cells (VSMCs) is altered in spontaneously hypertensive rats (SHR). Methods The VSMCs derived from aortae of SHR and Wistar Kyoto (WKY) rats were loaded for 48 hours with myo inositol. Inositol phosphate release was initiated by the addition of 10 5 mol/L norepinephrine in intact cells or by guanosine 5' 0 (3 thio tri sphosphate) (GTP gamma S) in permeabilized cells. In the meantime, growth arrested VSMCs were stimulated by 10% calf serum for 0, 30, 45, or 60 min, then gene expressions of Gq alpha subunit (G alph a q) were observed. Results There were no significant differences in inositol 1, 4,5 triphosphate (IP 3) level and expression of G alpha q mRNA between quiescent VSMCs from SHR and that from WKY. When stimulated by norepinephrine, IP 3 production increased transiently with a peak level at 10 s in VSMCs from WKY, and a rapid biphasic IP 3 response, which was significantly higher than that of WKY, in VSMCs from SHR had been observed. G proteins activated by GTP gamma S significantly raised IP 3 production in VSMCs from SHR compared to WKY (SHR vs WKY: 234.8%±29.2% vs 142.4%±12.0% of basal IP 3, P<0.05). In addition, the serum effect showed an significant increase in expression of G alpha q mRNA in VSMCs from SHR. Conclusions The hereditary factors are not the only variable regulating IP 3 metabolism and G alpha q gene expression. Influences of multi environmental factors such as vasoactive compounds, together with genetic predisposition, palys an important role in the highly sensitive response of IP 3 production and G alpha q gene over expression in SHR.展开更多
