The lyophilized cell extract injected intraperitoneally at 1 and 2 LD50 (15.8 and 31.6 mg/kg, respectively) produced significant increase in liver-specific enzymes viz. plasma alkaline phosphatase,γ-glutamyl transferase, lactate dehydrogenase with a concomitant decrease in hepatic glutamic pyruvic transaminase. A corresponding increase in liver body weight index and histopathological changes in liver (degeneration of hepatocytes, congestion and hemorrhage etc.) are indicative of a dose and time dependent hepatotoxic nature of the algal extract展开更多

Theoretically, nimodipine can lead to L-type channel inactivation and prevent calcium ion inflow, thereby exhibiting protective effects on nerve cells. OBJECTIVE: To observe the protective effects of nimodipine on glutamate-induced injury to embryonic rat neural stem cells, and to make a comparison with MK-801, a nonselective glutamate receptor antagonist. DESIGN, TIME AND SETTING: The present in vitro experiment pertaining to neural stem cells was performed at the Department of Neurology, First Hospital, Jilin University between January 2005 and December 2006. MATERIALS: Glutamate was sourced from the Shanghai Biological Research Institute of the Chinese Academy of Sciences. Nimodipine was provided by Bayer Company, Germany. Brain tissue was taken from Wistar rats on day 15 of gestation for isolation and culture of neural stem cells. METHODS: Passage 2 neural stem cell spheres were taken for preparation of single cell suspension. The prepared single cell suspension was divided into 4 groups: (1) Normal control, normally cultured. (2) Glutamate, cultured with 50, 100, 200, 500, and 1 000 μmol/L glutamate. (3) Nimodipine, received a 30- minute nimodipine [1×(10^-8-10^-2) g/L] culture followed by a glutamate (200 μmol/L) treatment step. (4) MK-801, given as 30- minute MK-801 (100 μmol/L) culture, followed by a glutamate (200 μmol/L) treatment step. MAIN OUTCOME MEASURES: Determination of glutamate-induced cell death by methyl thiazolyl tetrazolium (MTT) assay; calculation of neural stem cell survival rate following addition of nimodipine. RESULTS: The survival rate of neural stem cells was approximately 25.26% following 24 hour 50 μmol/L glutamate culture and gradually decreased as the glutamate dose increased (P 〈 0.05 0.01). Only 9.27% of neural stem cells survived when the glutamate dose was 1000 μmol/L. The survival rate of neural stern cells was significantly higher, in a dose-dependent manner, in the nimodipine [1×(10^-7-10^-2) g/L] group than in the glutamate group. In addition, the MK-801 group exhibited a higher survival rate after 24 hour treatment than the glutamate group (P 〈 0.01). CONCLUSION: Glutamate (50-1 000 μmol/L) induces injury to neural stem cells dose-dependently. Nimodipine exhibits protective effects on injury to neural stem cells and presents the effects in a dose-dependent manner at 1 ×(10^-7-10^-2) g/L. Nimodipine displays neuroprotective effects equivalent to MK-801.展开更多

It is unclear how excessive Glu plays roles in cochlear dysfunction in cochlear organotypic cultures. In this study neonatal rat cochlear organotypic cultures were prepared, and then the cochlear tissues were incubated with a new medium containing specific concentrations of Glu(0.1, 0.5, 1, 10 or 20 mmol/L) for 24 h, or incubated with the medium containing a concentration of 20 mmol/L Glu for 6, 12, 24 or 72 h, respectively. It was found that when the cochlear tissues were cultured for 24 h, the inner hair cells(IHCs) were damaged at the concentration of 0.5 mmol/L Glu, and with the increases of the concentrations, the injury was gradually aggravated, and 20 mmol/L Glu resulted in the significant loss of IHCs. In the 20 mmol/L Glu groups, the stereocilia bundles were missing or disarrayed on a few IHCs after culture for 6 h and the damage effect was time-dependent. The missing of IHCs was more significant in the basal turn of the cochlea than in the middle turn of the cochlea under the same concentration of Glu exposure. These results suggest that excessive exogenous Glu affects the morphology of IHCs, but not affects the outer hair cells(OHCs) in cochlear organotypic cultures, and the excitotoxic effects are different on IHCs of different parts of the cochlea under the same concentration of Glu exposure.展开更多

METHODS: Rat hepatocytes were incubated with AG, L-NAME, L-NNA, Actinomycin D (ActD) and dexamethasone in a medium containing CM (LPS plus TNF-alpha, IL-1 beta, and IFN-gamma) for 24h. NO production in the cultured supernatant was measured with the Griess reaction. Intracellular cGMP level was detected with radioimmunoassy. RESULTS: NO production was markedly blocked by AG and L-NAME in a dose-dependent manner under inflammatory stimuli condition triggered by CM in vitro. The rate of the maximum inhibitory effects of L-NAME (38.9%) was less potent than that obtained with AG(53.7%, P 【 0.05). There was no significant difference between the inhibitory effects of AG and two L-arginine analogues on intracellular cGMP accumulation in rat cultured hepatocytes. Non-specific NOS expression inhibitor dexamethasone (DEX)and iNOS mRNA transcriptional inhibitor ActD also significantly inhibited CM-induced NO production. AG(0.1 mmol x L(-1)) and ActD (0.2 ng x L(-1)) were equipotent in decreasing NO production induced by inflammatory stimuli in vitro, and both effects were more potent than that induced by non-selectivity NOS activity inhibitor L-NAME (0.1 mmol x L(-1)) under similar stimuli conditions (P【0.01). CONCLUSION: AG is a potent selective inhibitor of inducible isoform of NOS,and the mechanism of action may be not only competitive inhibition in the substrate level, but also the gene expression level in rat hepatocytes.展开更多

Different combination of physical geographical elements induced the difference of spatio-temporal pattern of land use change. There are four physical geographical regions in Mongolian Autonomou County of Qian Gorlos of Jilin Province. Based on spatial analysis and statistical analysis, we conclude that the primary pattern of land use and the tendency of land use changes are all different in four physical geographical regions. During 1987 – 1996, the dominant land use change processes were from grassland or forest to arableland, from unused land to paddy and grassland to unused land. Though land use change is mainly affected by social and economic condition in short period, the composite characters of physical geographical elements controls land use dynamic process. The relationship between land use dynamic process and the character of physical geographical units differ in different regions. Possible human impacts on land use change are explored with application of buffer areas of series distance along main roads and radius around main settlements. A few models are built to describe the relationship between land use spatial change rates and distance to road and settlements. According to our result, the relationship with the proximity to roads was a negative liner function, with the change rate decreasing rapidly when moving away from roads. Within a distance of less 7.5km from main traffic lines, land use changes occur red more. The bulk of grassland was apt to be opened up for cultivation around the settlements and the transformation from dryland to paddy occurred within the distance of 1km away from settlements.展开更多

Two kinds of inbred-line, R18-599 (red) with very excellent embryo culturing capacity and R15 with very poor embryo culturing capacity, were used as P1 and P2 for obtaining six generations. By culturing immature embryos of the six generations, four culturing characters, namely embryonic callus induction efficiency, nonembryonic callus induction efficiency, cloning ability of the embryonic callus, and number of regenerating plants, were analyzed using the general mean analysis and generation joint analysis. Results showed that the embryonic callus induction efficiency accorded with two major additive-dominance-epistatic genes and polygene-mixed additive-dominance-epistatic inheritance model. The induction efficiency of the nonembryonic callus accorded with two major additive-dominance-epistatic genes. The number of regenerating plants accorded with one major gene and polygene-mixed additive-dominance inheritance model. The cloning ability of the embryo callus accorded with two major genes and polygene-mixed inheritance model, whereas the effect of epistatic gene on this character was identified results of the two methods, generation joint analysis may genetic information. to be different using the two methods. METHODS: A monoclonal antibody against AM developed by our laboratory was used to detect the localization of AM protein in rat kidney tissue by avidin-biotin complex immunohistochemistry. The expressions of AM and its receptor CRLR mRNA on cultured glomerular epithelial cells (GEC) and MsC were investigated by Northern blot assay, and the possible effect of AM secreted by GEC on MsC proliferation was observed using [3H]thymidine incorporation as an index. RESULTS: A specific monoclonal antibody against AM was succesfully developed. AM was immunohistochemically localized mainly in glomeruli (GEC and endothelial cells), some cortical proximal tubules, medullary collecting duct cells, interstitial cells, vascular smooth muscle cells and endothelial cells. Northern blot assay showed that AM mRNA was expressed only on cultured GEC, but not on MsC, however, AM receptor CRLR mRNA was only expressed on MsC. GEC conditioned medium containing AM can inhibit MsC growth and AM receptor blocker CGRP8-37 may partially decreased this inhibitory effect. CONCLUSION: AM produced by GEC inhibits the proliferation of MsC, which suggests that AM as an important regulator is involved in glomerular normal physiological functions and pathologic processes.展开更多

An analysis demonstrates that the movie and television industry of South Korea is one of the most essential factors to attract the potential tourist from China. South Korean Pop culture is directly affecting rather the image of South Korean as a tourist destination than tourists' purchasing. And the paper suggests a set of "4S" strategy for South Korean to combine the elements of South Korean movies with those of tourism, so as to create an industrial development pattern characterized as "movies + tourism".展开更多

CONCLUSION: AG is a potent selective inhibitor of inducible isoform of NOS,and the mechanism of action may be not only competitive inhibition in the substrate level, but also the gene expression level in rat hepatocytes.

文摘The diversified development of British novels in the Renaissance mainly displays in several aspects, that is,lateral reflection of the social development background, specific expression of forming complex psychology of developing social mission, and macro manifestation of the basic needs of the public. This is the important factor of contemporary cultural school to find the innovative development road, and also the crucial effect to explore and excavate the development characteristics of contemporary cultural school, expressing the time promoting effect of the development of the diversified development of British novels in the Renaissance.Seen from the development characteristics of the C ultural School, the development direction between the social characteristics of times and the basic demand of social groups on social culture agrees with the diversified development of British novels in the Renaissance, and fully demonstrates the effect and value of the latter on the overall development of social cultural school.
文摘This study introduces the individualism-collectivism dimension of the cultural dimension of cross-cultural communication initiated by Geert Hofstede.Different cultures must develop a way of correlating that strikes a balance between caring for themselves and showing concern for others.Individualist culture encourages uniqueness and independence while collectivist culture emphasizes conformity and mutual assistance. This article introduces how to use case analysis method to effectively carry out classroom teaching in this cultural dimension.
文摘The names in English-speaking countries are full of historical origin and cultural connotation. For the significance of family name and given name, this paper focus on the origin and culture connotation of family name and given name, and also the hidden influences of name played on human beings. Moreover, the relationship between development of society and English name will also be concerned because we can not ignore that the development of society has made great effects to English name.According to the whole essay, we can easily realize the origin and connotation of English name through all my researches and analyses.

文摘Land use changes are regarded as landscape pattern change driven by many interactive natural and social-economic factors. The bulk of grassland was apt to be opened up for cultivation around the settlements and the transformation from dryland to paddy occurred within the distance of 1km away from settlements.
文摘This study was to find the regularity in the hereditary variation for the main culturing characters of the immature embryo culture in maize. By comparison of the not only raise experimental precision but also provide more
文摘OBJECTIVE: To observe the localization of adrenomedullin (AM) in rat kidney tissue and its inhibitory effect on the growth of cultured rat mesangial cells (MsC). CONCLUSION: AM produced by GEC inhibits the proliferation of MsC, which suggests that AM as an important regulator is involved in glomerular normal physiological functions and pathologic processes.
文摘Following along with South Korean Pop culture sweeping across Chinese Mainland, Chinese market of South Korea tour is developing rapidly. An analysis demonstrates that the movie and television industry of South Korea is one of the most essential factors to attract the potential tourist from China. South Korean Pop culture is directly affecting rather the image of South Korean as a tourist destination than tourists' purchasing. And the paper suggests a set of "4S" strategy for South Korean to combine the elements of South Korean movies with those of tourism, so as to create an industrial development pattern characterized as "movies + tourism".
