BACKGROUND Di(2-ethylhexyl)phthalate(DEHP)is a common plasticizer known to cause liver injury.Green tea is reported to exert therapeutic effects on heavy metal exposureinduced organ damage.However,limited studies have examined the therapeutic effects of green tea polyphenols(GTPs)on DEHP-induced liver damage.AIM To evaluate the molecular mechanism underlying the therapeutic effects of GTPs on DEHP-induced liver damage.METHODS C57BL/6J mice were divided into the following five groups:Control,model[DEHP(1500 mg/kg bodyweight)],treatment[DEHP(1500 mg/kg bodyweight)+GTP(70 mg/kg bodyweight),oil,and GTP(70 mg/kg bodyweight)]groups.After 8 wk,the liver function,blood lipid profile,and liver histopathology were examined.Differentially expressed micro RNAs(miRNAs)and mRNAs in the liver tissues were examined using high-throughput sequencing.Additionally,functional enrichment analysis and immune infiltration prediction were performed.The miRNA-mRNA regulatory axis was elucidated using the starBase database.Protein expression was evaluated using immunohistochemistry.RESULTS GTPs alleviated DHEP-induced liver dysfunction,blood lipid dysregulation,fatty liver disease,liver fibrosis,and mitochondrial and endoplasmic reticulum lesions in mice.The infiltration of macrophages,mast cells,and natural killer cells varied between the model and treatment groups.mmu-miR-141-3p(a differentially expressed miRNA),Zcchc24(a differentially expressed mRNA),and Zcchc24(a differentially expressed protein)constituted the miRNA-mRNA-protein regulatory axis involved in mediating the therapeutic effects of GTPs on DEHP-induced liver damage in mice.CONCLUSION This study demonstrated that GTPs mitigate DEHP-induced liver dysfunction,blood lipid dysregulation,fatty liver disease,and partial liver fibrosis,and regulate immune cell infiltration.Additionally,an important miRNAmRNA-protein molecular regulatory axis involved in mediating the therapeutic effects of GTPs on DEHP-induced liver damage was elucidated. 展开更多

Objective Di-(2-ethylhexyl) phthalate(DEHP) is a ubiquitous environmental contaminant.As an endocrine disruptor,it seriously threatens human health and ecological environmental safety.This study examines the impact of intervention with soybean isoflavones(SIF) on DEHP-induced toxicity using a metabonomics approach.Methods Rats were randomly divided into control(H),SIF-treated(A,86 mg/kg body weight),DEHP-treated(B,68 mg/kg),and SIF plus DEHP-treated(D) groups.Rats were given SIF and DEHP daily through diet and gavage,respectively.After 30 d of treatment,rat urine was tested using UPLC/MS with multivariate analysis.Metabolic changes were also evaluated using biochemical assays.Results Metabolomics analyses revealed that p-cresol glucuronide,methyl hippuric acid,N1-methyl-2-pyridone-5-carboxamide,lysophosphatidycholine [18:2(9 Z,12 Z)] {lyso PC [18:2(9 Z,12 Z)]},lyso PC(16:0),xanthosine,undecanedioic acid,and N6-acetyl-l-lysine were present at significantly different levels in control and treatment groups.Conclusion SIF supplementation partially protects rats from DEHP-induced metabolic abnormalities by regulating fatty acid metabolism,antioxidant defense system,amino acid metabolism,and is also involved in the protection of mitochondria. 展开更多

Human beings are increasingly exposed to phthalates,which are a group of chemicals used to make plastics more flexible and harder to break,and simultaneously ingesting abundant food emulsifiers via daily diet.The purpose of this study was to investigate the effect of the food emulsifier glycerin monostearate(GMS)on male reproductive toxicity caused by di(2-ethylhexyl)phthalate(DEHP,one of the phthalates)and explore the underlying mechanism.Thirty male Sprague-Dawley rats were randomly divided into control group,DEHP group and DEHP+GMS group.Rats in the DEHP group and DEHP+GMS group were orally administered with 200 mg/kg/d DEHP with or without 20 mg/kg/d GMS.After 30 days of continuous intervention,it was found that the serum testosterone level was significantly lowered in DEHP group and DEHP+GMS group than that in control group(P<0.01).The serum testosterone level and the relative testis weight were significantly decreased in the DEHP+GMS group as compared with those in the DEHP group and control group(P<0.05).More spermatids were observed to be shed off in DEHP+GMS group than in DEHP group.The expression levels of cell cycle checkpoint kinase 1(Chkl),cell division cycle gene 2(Cdc2),and cyclin-dependent kinase 2(CDK2)were down-regulated in DEHP group,and this tendency was more significant in DEHP+GMS group(P<0.05 or P<0.01).There was no significant difference in the P-glycoprotein(P-gp)expression between DEHP group and control group.However,P-gp was markedly down-regulated in DEHP+GMS group(P<O.Ol).The results indicated that the food emulsifier GMS aggravated the toxicity of DEHP on male reproduction by inhibiting the cell cycle of testicular cells and the expression of P-gp in testis tissues. 展开更多

Immunity is crucial to the health of animals and it can determine their survival and fitness. Di(2-ethylhexyl) phthalate (DEHP) is widely used as a plasticizer and hence is the most abundant phthalate in the environment. Exposure to DEHP is of great concern for human health. In the present study, we tested the hypothesis that exposure to DEHP would suppress T cell-mediated immunity in mice. Twenty adult male Kunming mice were randomly assigned into the control (n = 10) and the DEHP treatment (n = 10) groups. Both groups have free access to food and water, while the mice in the latter group drank DEHP solution (2000 mg/L) for 42 days. T cell-mediated immunity assessed by phytohaemagglutinin (PHA) response was depressed in the DEHP treated mice compared with the controls, however, wet thymus and spleen mass, white blood cells were not influenced by DEHP treatment. Taken together, different immunological parameters responded differently to DEHP treatment in Kunming mice. 展开更多

The extraction of Nd^3+ and Sm^3+, including the extraction and stripping capability as well as the separation effect of Nd^3+ or Sm^3+, from a sulfuric acid medium, by mixtures of di-(2-ethylhexyl) phosphoric acid (HDEHP, H2A2(0)) and 2-ethylhexyl phosphoric acid mono-2-ethylhexyl ester (HEH/EHP, H2L2(0)) were studied. The distribution ratios and synergistic coefficients of Nd^3+ and Sm^3+ in different acidities were also determined. A synergistic extractive effect was found when HDEHP and HEH/EHP were used as mixed extractants for Sm^3+ or Nd^3+. The chemical compositions of the extracted complex were determined as Nd.(HA2)2-HL2 and Sm.(HA2)2-HL2. The extraction equilibrium constants, enthalpy change, and entropy change of the extraction reaction were also determined. 展开更多

Objective To estimate the daily intake of DEHP among workers in flavoring factories. Methods 71 workers in two flavoring manufacturers, 27 administrators in those factories and 31 laboratory technicians in a research institute were recruited and assigned to exposure group, control group 1 and control group 2 respectively. Their urinary DEHP metabolites, mono(2-ethylhexyl) phthalate (MEHP), mono-(2-ethyl-5-hydroxyhexyl) phthalate (MEHHP), mono-(2-ethyl-5-oxohexyl) phthalate (MEOHP), were detected by isotope dilution-ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS). The urinary metabolites concentrations were converted into DEHP intake levels using two pharmacokinetic models: the urine creatinine-excretion (UCE) one and the urine volume (UV) one. Results No significant differences were found among the three groups. Based on the urinary concentrations of Z3MEHP, we got a median daily DEHP intake of 3.22 or 1.85 μg/kg body-weight/day applying the UV or UCE models respectively. Depending on the UV model, three subjects (2.34%) exceeded the RfD value given by US EPA and the P50 of estimate daily DEHP intakes accounted for 16.10% of the RfD value. No subjects exceeded the limitation depending on the UCE model. Conclusion The workers in flavoring factories were not supposed to be the high DEHP exposure ones and their exposure level remained at a low risk. 展开更多

Exposure to di(2-ethylhexyl)phthalate(DEHP)in the indoor environment has been linked with significant health risks for Chinese children.Multi-phase DEHP concentrations in Chinese residences and kindergartens were estimated using a mass balance model based on the current baseline condition and control strategies(i.e.,increasing ventilation rate,reducing area of sources,using mechanical ventilation systems,and using portable air cleaners).The health benefits of each control strategy were quantified as the reduction in lifetime cancer risks(LCR)and burden of disease(BoD).In the current situation,the mean LCR and disability-adjusted life years(DALY)number attributable to indoor DEHP exposure for Chinese children were around 6.0×10^(−6) and 155 thousand,respectively.The mean LCR and DALY might be reduced by 25%-54%and 16%-40%,respectively,by increasing air exchange rates by 100%,reducing the use of source materials by two-thirds or deploying commercial air cleaners in naturally ventilated buildings.Meanwhile,avoidable DALYs could result in a reduction of mean economic losses of 2.2-5.3 billion RMB.Mechanical ventilation systems with filtration units may not be helpful for reducing children’s health risks.House-specific and tailor-made control measures are critical in lowering indoor exposure to DEHP to promote sustainable buildings and children’s health in China. 展开更多

Chinese forest musk deer(FMD),an endangered species,have exhibited low reproductive rates even in captivity due to stress conditions.Investigation revealed the presence of di(2-ethylhexyl)phthalate(DEHP),an environmental endocrine disruptor,in the serum and skin of captive FMDs.Feeding FMDs with maslinic acid(MA)has been observed to alleviate the stress response and improve reproductive rates,although the precise molecular mechanisms remain unclear.Therefore,this study aims to investigate the molecular mechanisms underlying the alleviation of DEHP-induced oxidative stress and cell apoptosis in primary peritubular myoid cells(PMCs)through MA intake.Primary PMCs were isolated and exposed to DEHP in vitro.The results demonstrated that DEHP significantly suppressed antioxidant levels and promoted cell apoptosis in primary PMCs.Moreover,interfering with the expression of PRDX6 was found to induce excessive reactive oxygen species(ROS)production and cell apoptosis in primary PMCs.Supplementation with MA significantly upregulated the expression of PRDX6,thereby attenuating DEHP-induced oxidative stress and cell apoptosis in primary PMCs.These findings provide a theoretical foundation for mitigating stress levels and enhancing reproductive capacity of in captive FMDs. 展开更多

This study is concerned with the effects of di (2-ethylhexyl) phthalate (DEHP) on two kinds of duckweeds (Spirodela polyrhiza and Lemna minor).The results indicate that DEHP has aquatic toxicity to Spirodela polyrhiza at 0.4 mg/L and to Lemna minor at over 0.1 mg/L by changing their physiologic-biochemical characteristics.The contents of duckweed chlorophyll and soluble protein decrease with increasing DEHP concentration after 7 d of exposure.DEHP shows the stimulating role in catalase (CAT) and superoxide dismutase (SOD) systems at relative low levels.At 0.01 mg/L and 0.005 mg/L,SOD activities of Spirodela polyrhiza and Lemna minor reach their peak values respectively,while CAT activity reaches its maximum value at 0.05 mg/L and 0.01 mg/L.When DEHP levels are too high,the protection enzyme system would be destroyed and plant growth is inhibited.The analysis of malondialdehyde (MDA) and Fourier transform infrared spectroscopy manifest that DEHP could affect the tested duckweeds by destroying its cell membranes,and Spirodela polyrhiza is more resistant to DEHP exposure than Lemna minor. 展开更多

In this work, a novel room temperature ionic liquid (2-butyl-imidazolium di-(2-ethylhexyl) phosphate) ([C4mim] [D2EHPA]) was synthesized and tested as extractant in the mercury(II) liquid-liquid extraction. The effects of parameters such as aqueous to organic phase’s volume ratio, metal concentration IL concentration, pH levels, ionic strength, and temperature were reported. For the extraction of metal, [C4mim]3[R.HR]3[HgCl2]org and [C4mim]3[R.HR]3 [HgClOH]org species were formed where (H2R2) was D2EHPA. In the case of ionic strength, the results showed that the addition of sodium acetate at 0.302 mmol·L?1 to the aqueous phase strongly increased the mercury extraction yield (R = 100%). The extracted species were investigated by a calculation program using CHEAQS V. L20.1 inorder to determine the relation between the percentages of the extracted species and the extraction yield. The results showed that the extracted species in the best conditions were HgCl2 and HgClOH with respective percentages 80.66% and 18.29%. 展开更多

Plastics such as polyvinyl chlorides (PVC) are widely used in many indoor constructed environments; however, their unbound chemicals, such as di-(2-ethylhexyl) phthalates (DEHP), can leach into the surrounding environment. This study focused on DEHP's effect on the central nervous system by determining the precise DEHP content in mice brain tissue after exposure to the chemical, to evaluate the specific exposure range. Primary neuronal-astrocyte co-culture systems were used as in vitro models for chemical hazard identification of DEHP. Oxidative stress was hypothesized as a probable mechanism involved, and therefore the total reactive oxygen species (ROS) concentration was determined as a biomarker of oxidative stress. In addition, NeuriteTracer, a neurite tracing plugin with ImageJ, was used to develop an assay for neurotoxicity to provide quantitative measurements of neurological parameters, such as neuronal number, neuron count and neurite length, all of which could indicate neurotoxic effects. The results showed that with 1 nmol/L DEHP exposure, there was a significant increase in ROS concentrations, indicating that the neuronal-astrocyte cultures were injured due to exposure to DEHP. In response, astrocyte proliferation (gliosis) was initiated, serving as a mechanism to maintain a homeostatic environment for neurons and protect neurons from toxic chemicals. There is a need to assess the cumulative effects of DEHP in animals to evaluate the possible uotake and effects on the human neuronal system from exoosure to DEHP in the indoor environment. 展开更多

Reference gene is essential for correct interpretation of q PCR data. However, an ideal universal reference gene that is stable under all experimental circumstances has not been described. Researchers should validate their reference genes while performing q PCR analysis. The expression of 6 candidate reference genes: 18 s r RNA,28 s r RNA, Cathepsin Z, Elongation factor 1-α, Glyceraldehyde-3-phosphate dehydrogenase andβ-actin were investigated during final oocyte maturation induced by different compounds(DES and DEHP) in common carp(Cyprinus carpio). Four softwares(Bestkeeper, ge Norm,Norm Finder and Ref Finder) were used to screen the most stable gene in order to evaluate their expression stability. The results revealed that EF1α was highly stable expressed when final oocyte maturation was induced by DES, while gapdh was the most stable gene when final oocyte maturation was induced by DEHP. Stable expressed reference gene selection is critical for all q PCR analysis to get accurate target gene m RNA expression information. 展开更多

Compared with the traditional liquid–liquid extraction method,solid-phase extraction agents are of great significance for the recovery of indium metal due to their convenience,free of organic solvents,and fully exposed activity.In this study,P_(2)O_(4)(di-2-ethylhexyl phosphoric acid)was chemically modified by using UiO-66 to form the solid-phase extraction agent P_(2)O_(4)-UiO-66-MOFs(di-2-ethylhexyl phosphoric acid-UiO-66-metal-organic frameworks)to adsorb In(Ⅲ).The results show that the Zr of UiO-66 bonds with the P-OH of P_(2)O_(4) to form a composite P_(2)O_(4)-UiO-66-MOF,which was confirmed by X-ray photoelectron spectroscopy(XPS)and Fourier transform infrared spectroscopy(FT-IR).The adsorption process of indium on P_(2)O_(4)-UiO-66-MOFs followed pseudo first-order kinetics,and the adsorption isotherms fit the Langmuir adsorption isotherm model.The adsorption capabilities can reach 192.8 mg/g.After five consecutive cycles of adsorption-desorption-regeneration,the indium adsorption capacity by P_(2)O_(4)-UiO-66-MOFs remained above 99%.The adsorption mechanism analysis showed that the P=O and P-OH of P_(2)O_(4) molecules coated on the surface of P_(2)O_(4)-UiO-66-MOFs participated in the adsorption reaction of indium.In this paper,the extractant P_(2)O_(4) was modified into solid P_(2)O_(4)-UiO-66-MOFs for the first time.This work provides a new idea for the development of solid-phase extractants for the recovery of indium. 展开更多

Background Mono-(2-ethylhexyl) phthalate (MEHP), the metabolite of di-(2-ethylhexyl) phthalate (DEHP), was suspected to be toxic to human embryos. This study contributes to investigating its toxic effects by an embryonic stem cell test (EST) based on two human embryonic stem cell (hESCs) lines. Methods CH1 established in our own lab and H1, a federally registered cell line were two human embryonic stem cell lines used in this test. Four endpoint measurements were performed consisting of cell viability, proliferation ability, apoptosis as well as changes of gene expression patterns after spontaneous differentiation were determined. For measuring effects on the first three endpoints, the cells were treated with various concentrations of MEHP dissolved in dimethyl sulfoxide (DMSO) and only with DMSO which served as control and harvested after 5 days. For measuring effects during spontaneous differentiation, the RNA of embryoid bodies (EBs) formed after 8 days' MEHP exposure was collected and changes in differentiation specific gene expression patterns were analyzed by quantitative real time RT-PCR. Results As a result the viability and proliferation ability of both cell lines decreased significantly at 1000 μmol/L MEHP, while there was no effect on apoptosis or cell morphology. In addition MEHP also changed the gene expression pattern in the EBs of both cell lines. Conclusion MEHP in a high dose was cytotoxic and affected the development of hESCs, which indicates its embryo toxicitv in human embrvos. 展开更多

Common additives in plastics such as bisphenol A (BPA) or phthalates like di-(2-ethylhexyl) phthalate (DEHP) are environmental estrogens that have been shown to be endocrine disruptors in some experimental animal models. This project used the C2C12 cell culture model to examine how exposure to BPA or DEHP affects two aspects of skeletal muscle development, the fusion of myoblasts into myotubes and agrin-induced clustering of acetylcholine receptors (AChRs). During myotube formation AChRs cluster spontaneously. Treatment with motor neuron derived agrin increases the frequency of AChR clusters through an agrin signaling pathway that also clusters other postsynaptic components of the neuromuscular synapse. For this project C2C12 cells were exposed to BPA or DEHP while myoblasts fused into myotubes. After exposure to 10 μM BPA or 100 μM DEHP the frequency of agrin-induced AChR clusters decreased. In addition, myotube formation decreased as a higher percentage of nuclei remained in myoblasts. Furthermore, BPA or DEHP reduced the amount of the myogenic regulatory factor myogenin. This suggests that BPA and DEHP decrease AChR clustering by reducing myogenin. Moreover, plastic additives like BPA and DEHP may pose a risk for skeletal muscle development in humans. 展开更多

Solvent extraction is generally accepted as the most appropriate commercial technology for separating REEs due to the need to be able to handle larger volumes of diluted pregnant solutions. This study focused on the development of selective separation of light and heavy REEs from the pregnant leach solution obtained from leaching of apatite ore in 1 M sulfuric acid (H2SO4) using solvent extraction. Three different commercial organophosphorus extractants (di-(2-ethylhexyl) phosphoric acid (D2EHPA), 2-ethylhexyl phosphonic acid mono-2-ethylhexyl ester (PC88A) and tributyl phosphate (TBP)), and the influences of experimental parameters such as extractant concentration, organic/aqueous phase ratio, diluent type, pH, extraction time and stripping agent concentration were examined. Results showed that light REEs (LREEs) and heavy REEs (HREEs) in the pregnant leach solution were selectively separated with D2EHPA via a two-stage extraction process. In the first-stage of solvent extraction, >90% of (0.05 g/L) HREEs was extracted with 1.8 M D2EHPA in kerosene while the vast majority (>95%) of LREEs was remained in raffinate. In the second-stage, >93% (1.01 g/L) of LREEs was extracted from the raffinate with 1.8 M D2EHPA dissolved in kerosene at pH 1.6. HREEs (>95%) and LREEs (>90%) loaded with D2EHPA after the first and second-stage of extraction were stripped by 4 M H2SO4 and 1 M H2SO4 solutions, respectively. Distribution of middle rare earth elements (MREEs) was discussed through the extraction processes in this study. 展开更多

Objectives:This study aimed to investigate the effect of the widely used food emulsifier glycerin monostearate(GM)on testicular toxicity caused by the mixture of three commonly used phthalate esters(MPEs)in rats,and further to explore the underlying mechanism.Materials and Methods:Thirty male Sprague-Dawley rats were randomly divided into three groups.Rats were orally treated with 160 mg/kg/d MPEs in the MPEs group;coinstantaneously treated with 160 mg/kg/d MPEs and 200 mg/kg/d GM in the MPEs+GM group;and treated with the excipient in the control group.The intervention lasted for 5 weeks.Testis weight,epididymis weight,testicular histopathology,and serum testosterone were detected for testicular toxicity evaluation.The testicular ultrastructure,the tight junction proteins zonula occluden(ZO)-1,and claudin were measured for the mechanism exploration.Results:The body weight,epididymis,serum testosterone level,and anogenital distance in the MPEs+GM group were significantly decreased compared with control group(P<0.05);Testicular histopathological observation showed that shed spermatids were observed in the MPEs+GM group.Ultrastructural observation of testicular cells showed that the cristae number was decreased in some mitochondria in the MPEs group,whereas the cristae were fused and disappeared in most mitochondria in the MPEs+GM group.The tight junctions were broken in the MPEs+GM group;meanwhile,the expression of ZO-1 and claudin were altered in the MPEs+GM group(P<0.01).Conclusions:The results from this study indicated that GM aggravated MPEs'testicular toxicity,which might relate to the injured mitochondria and damaged tight junctions in testicular tissue. 展开更多

Lock from Nigeria is being reported. The hydrodistilled oil was analyzed by gas chromatography-flame ionization detector (GC-FID) and gas chromatography-mass spectrometry (GC-MS) techniques. The main constituents of the oil were 1,2-benzenedicarboxylic acid, mono (2-ethylhexyl) ester phthalate (26.6%) and β-elemene (27.9%). There were significant amounts of caryophyllene oxide (7.3%) and urs-12-en-24-oic acid, 3-oxo-methyl ester (5.5%). The antimicrobial activity of the essential oil was evaluated against a panel of seven bacteria and two fungal strain using agar diffusion and broth microdilution methods. Results had shown that the oil exhibited moderate to strong antimicrobial activity against the tested microorganisms. The activity zones of inhibition (ZI) ranged between 10.0 ± 0.2 mm and 28.3 ± 2.9 mm while the minimum inhibitory concentrations (MIC) ranged between 0.3 mg/mL 5.0 mg/mL, respectively. The chemical constituents and antimicrobial activity of the essential oil of Senna podocarpa were being reported for the first time. 展开更多

In this paper,we proposed a microextraction approach for the extraction and separation of Mn(Ⅱ)and Co(Ⅱ)from sulfate solution simulating leachate of spent lithium-ion battery cathode materials using saponified di-(2-ethylhexyl)phosphoric acid system.The effects of the following operational variables were investigated:equilibrium pH,tri-«-butyl phosphate concentration,saponification rate,two-phase ratio and residence time.The results showcased that the microextractor can reach the extraction equilibrium within 20 s,thereby greatly reducing necessary extraction time comparing to that of conventional processes.The volumetric mass transfer coefficient showed 8-21 times larger than that of batch device.With the help of microextractor,95%of Mn(Ⅱ)was extracted with a single theoretical stage at a chosen two-phase ratio of 3:1,and the separation factor β_(Mn/Co) was as large as 65.5.In the subsequent stripping step,more than 99%of manganese from loaded phase was easily stripped under optimal conditions.The microextraction approach greatly enhances the mass transfer while enabling a continuous and controllable extraction process within a simple structure design.When extracting spent electrode material with microextractors,the comprehensive recovery of mangenese can reach 96%.The microextraction approach has a good applicability in the spent lithium-ion battery cathode materials recycling at both bench and industrial scales. 展开更多
