In this study, Shotgun lipidomic methodology based on tandem mass spectrometry but without liquid chromatography was established for simple lipids analysis such as mono-, di-and triacylglycerols(MAGs, DAGs and TAGs) and the derivatives such as free fatty acids(FFAs). Six varieties of rapeseeds belonging to 3 types(B. juncea, B. campestris and B. oleracea) from different areas of China were investigated, with 59 TAG species, 10 DAG species, 5 MAG species and 6 FFA species identified. It was found that erucic acid was mainly present in the form of TAG 62:4(22:1/18:2/22:1), TAG 62:3(22:1/18:1/22:1), and TAG 62:5(22:1/18:3/22:1) in high erucic acid rapeseed oils, while oleic acid was mainly present in the form of TAG 54:2(18:0/18:1/18:1), and TAG 54:4(18:1/18:2/18:1) in high oleic acid rapeseed oils. It was difficult for DAG, MAG and FFA quantification due to their low content with high background content of TAGs. Results showed that the main differences in lipids were not derived from the types of rapeseed, but directly related to varieties.展开更多

Samples from a freshwater receiving pond (ABI-1002) and two upstream storm water ditches (ABI-1003) and (ABI-1004) yielded alarmingly high <em>Fecal coliform</em> MF densities of 220, >2000 and >2000 CFU/100ml respectively. The indicator, <em>Enterococcus</em> bacteria exceeded allowable limits in all but the equipment control (ABI-1001). Using MSS, the relative numerical abundance of pathogenic bacteria, virulence and antibiotic resistance genes revealed the status and potential pollution sources of each ditch. High levels of <em>Shigella</em><em> sp</em>. (0 (ABI-1001), 4945 (ABI-1002), 55,008 (ABI-1003), and 2221 (ABI-1004) genomic reads/100ml) correlated with virulence genes and antibiotic resistance genes found in fecal samples for ABI1003 and not ABI1004. Traditional culture methods (TCM) showed possible fecal contamination in two of the four samples, and no contamination in the others. MSS clearly distinguished between fecal and environmental bacteria contamination sources, and pinpointed actual risks from pathogens. Our data underscore the potential utility of MSS in precision risk assessment for public and biodiversity health and tracking of environmental microbiomes shifts by field managers and policy makers.展开更多

We used an optimized semi-s...Secretome, the totality of secreted proteins, is viewed as a promising pool of candidate cancer biomarkers. Simple and reliable methods for identifying secreted proteins are highly desired. We used an optimized semi-shotgun liquid chromatography followed by tandem mass spectrometry (LC-MS/MS) method to analyze the secretome of breast cancer cell line MDA-MB-231. A total of 464 proteins were identified. About 63% of the proteins were classified as secreted proteins, including many promising breast cancer biomarkers, which were thought to be correlated with tumorigenesis, tumor development and metastasis. These results suggest that the optimized method may be a powerful strategy for cell line secretome profiling, and can be used to find potential cancer biomarkers with great clinical significance.展开更多

A 49-year-old man presented to the emergency department due ...In this report, we present the case of a patient who presented with a pellet in his myocardium and was managed with cardiac tamponade due to shotgun injury. A 49-year-old man presented to the emergency department due to a close-range shotgun pellet injury. The patient was operatedemergently because arterial pressure was progressively decreasing and cardiac tamponade was detected echocardiographically. Three pellet entrance hole were detected on left ventricule posterior wall in surgical exploration after the cardiac tamponade relieved. No further intervention was considered because bleeding discontinued. We conclude that in shotgun pellet cardiac injuries, if the pellets are located in the myocardium, the operational decision and surgical procedure to be undertaken depends on the clinical situation of the patient and the echocardiographic signs.展开更多

However, des...The discipline of lipidomics, as coined in 2003, has made profound advances and been rapidly expanded. Direct infusion-based shotgun lipidomics contributes a major part of the research in this discipline. However, despite of its numerous advantages over other lipidomics platforms, the classical shotgun lipidomics approach suffers ion suppression and has the difficulties to assess low abundance lipid classes, particularly those less ionizable or that do not yield sensitive and specific fragments. To overcome this obstacle and broaden coverage, strategies have been developed based on unique chemistry and/or physics of lipid classes of interest to enhance shotgun lipidomics. This article overviews the recent enhancements of shotgun lipidomics, including prefractionation, acid/base hydrolysis, derivatization, charge feature utilization. We believe that, by this versatile platform, comprehensive coverage of cellular lipidomes would be achieved in more accurate and effective manner. Therefore, application of the enhanced shotgun lipidomics would greatly accelerate lipidomics research to substantiate the aberrant lipid metabolism, signaling, trafficking, and homeostasis under pathological conditions and their underpinning biochemical mechanisms. It would also facilitate precision medicine studies.展开更多
