Mitochondria undergo morphological changes during spermatogenesis in some animals.The mechanism and role of mitochondrial morphology regulation,however,remain somewhat unclear.In this study,we analyzed the molecular characteristics,expression dynamics and subcellular localization of optic atrophy protein 1(OPA1),a mitochondrial fusion and cristae maintenance-related protein,to reveal the possible regulatory mechanisms underlying mitochondrial morphology in Phascolosoma esculenta spermiogenesis.The full-length cDNA of the P.esculenta opa1 gene(Pe-opa1)is 3743 bp in length and encodes 975 amino acids.The Pe-OPA1 protein is highly conservative and includes a transmembrane domain,a GTPase domain,two helical bundle domains,and a lipid-interacting stalk.Gene and protein expression was higher in the coelomic fluid(a site of spermatid development)of male P.esculenta and increased first and then decreased from March to December.Moreover,their expression during the breeding stage was significantly higher than during the non-breeding stage,suggesting that Pe-OPA1 is involved in P.esculenta reproduction.The Pe-OPA1 protein was more abundant in components consisting of many spermatids than in components without,indicating that Pe-OPA1 mainly plays a role in the spermatid in coelomic fluid.Moreover,Pe-OPA1 was mainly detected in the spermatid mitochondria.Immunofluorescence experiments showed that the Pe-OPA1 are constitutively expressed and co-localized with mitochondria during spermiogenesis,suggesting its involvement in P.esculenta spermiogenesis.These results provide evidence for Pe-OPA1's involvement in the regulation of mitochondrial morphology during spermiogenesis. 展开更多

Hepatitis E virus(HEV)is hyperendemic in South Asia and Africa accounting for half of total Global HEV burden.There are eight genotypes of HEV.Among them,the four common ones known to infect humans,genotypes 1 and 2 are prevalent in the developing world and genotypes 3 and 4 are causing challenge in the industrialized world.Asymptomatic HEV viremia in the general population,especially among blood donors,has been reported in the literature worldwide.The clinical implications related to this asymptomatic viremia are unclear and need further exploration.Detection of viremia due to HEV genotype 1 infection,apparently among healthy blood donors is also reported without much knowledge about its infection rate.Similarly,while HEV genotype 3 is known to be transmitted via blood transfusion in humans and has been subjected to screening in many European nations,instances of transmission have also been documented albeit without significant clinical consequences.Epidemiology of HEV genotype 1 in endemic areas often show waxing and waning pattern.Occasional sporadic occurrence of HEV infection interrupted by outbreaks have been frequently seen.In absence of known animal reservoir,where HEV exists in between outbreak is a mystery that needs further exploration.However,occurrence of asymptomatic HEV viremia due to HEV genotype 1 during epidemiologically quiescent period may explain that this phenomenon may act as a dynamic reservoir.Since HEV genotype 1 infection cannot cause chronicity,subclinical transient infection and transmission of virus might be the reason it sustains in interepidemic period.This might be the similar phenomenon with SARS COVID-19 corona virus infection which is circulating worldwide in distinct phases with peaks and plateaus despite vaccination against it.In view of existing evidence,we propose the concept of“Dynamic Human Reservoir.”Quiescent subclinical infection of HEV without any clinical consequences and subsequent transmission may contribute to the existence of the virus in a community.The potential for transmitting HEV infection by asymptomatic HEV infected individuals by fecal shedding of virus has not been reported in literature.This missing link may be a key to Pandora's box in understanding epidemiology of HEV infection in genotype 1 predominant region. 展开更多

Since the Dongfeng-2 missile, full-vehicle modal testing has been established as an indispensable part of the development and testing of rocket and missile models. However, as rockets have been developed larger, the cost and duration of such tests have significantly increased, magnifying their impact on model development. This article follows the process of the modal testing practice of the Gravity-1 rocket, reviewing and summarizing the design process of the rocket's dynamic characteristics. Initially, the article introduces common modeling techniques for launch rockets, including the mass-beam model and the hybrid element model. It then discusses the relationship between the structural dynamics model of the launch rocket and modal testing, aiming to reduce testing costs through refined structural dynamics modeling methods. Subsequently, the article describes the dynamic characteristics design process of the Gravity-1 carrier rocket, categorizes structural parameters, and studies how the selection of structural parameters affects the predicted dynamic characteristics of the rocket. Finally, it elaborates on the design of the modal testing scheme and the dynamic characteristics design based on the test data. 展开更多

Fusarium head blight(FHB) caused by Fusarium graminearum is a devastating fungal disease on small grain cereal crops,because it reduces yield and quality and causes the mycotoxin contamination to the grain.Dynamins and dynamin-related proteins(DRPs) are large GTPase superfamily members,which are typically involved in the budding and division of vesicles in eukaryotic cells,but their roles in Fusarium spp.remain unexplored.Here,we found that FgDnm1,a DRP and homolog to Dnm1 in Saccharomyces cerevisiae,contributes to the normal fungal growth,sexual reproduction and sensitivity to fungicides.In addition,we found FgDnm1 co-localizes with mitochondria and is involved in toxisome formation and deoxynivalenol(DON) production.Several quinone outside inhibitors(QoIs) and succinate dehydrogenase inhibitors(SDHIs) cause fragmentated morphology of mitochondria.Importantly,the deletion of FgDnm1displays filamentous mitochondria and blocks the mitochondrial fragmentation induced by QoIs and SDHIs.Taken together,our studies uncover the effect of mitochondrial dynamics in fungal normal growth and how such events link to fungicides sensitivity and toxisome formation.Thus,we concluded that altered mitochondrial morphology induced by QoIs and SDHIs depends on FgDnm1. 展开更多

The solubility of 2,3,4-trichloro-1,5-dinitrobenzene(TCDNB) was measured by a laser dynamic method over the temperature range from 278.15 K to 323.15 K under 0.1 MPa in fifteen mono-solvents(methanol,ethanol,isopropanol,n-butanol,toluene,dichloromethane,chloroform,tetrachloromethane,1,2-dichloroethane,acetone,ethyl acetate,acetonitrile,N-methylpyrrolidone(NMP),N,Ndimethylformamide dimethyl sulfoxide(DMF),dimethyl sulfoxide(DMSO).The solubility of TCDNB could be increased with increasing temperature in fifteen mono-solvents.TCDNB solubility is in the following order at 298.15 K:NMP>DMF>DMSO>toluene>acetone>ethyl acetate>dichloromethane>1,2-dich loroethane>chloroform>acetonitrile>tetrachloromethane>methanol>ethanol>n-butanol>isopropanol.The KAT-LSER model was used to investigate the solvent effect,which revealed that the hydrogen bond acidity of solvents has a greater effect on TCDNB solubility.The van't Hoff model,the modified Apelblat model,theλh model,and the non-random two liquid(NRTL)model were used to correlate the solubility of TCDNB.The calculated solubility data agreed well with the experimental data,and the modified Apelblat model fit best.Furthermore,the van't Hoff and Gibbs equations were also used to calculate the dissolution thermodynamic properties of TCDNB in various solvents.TCDNB dissolution could be an enthalpy-driven,non-spontaneous,and endothermic process in fifteen mono-solvents.The determination and fitting solubility of TCDNB,as well as the calculation of its thermodynamic properties,would be critical in the purification and crystallization of its preparation process research. 展开更多

[目的]探讨血清N-乙酰神经氨酸(Neu5Ac)及S100钙结合蛋白A1(S100A1)水平预测急性脑梗死(ACI)患者预后的作用.[方法]选择2021年8月—2022年8月间长葛市人民医院收治的80例ACI患者纳入研究组,根据病情分为轻度组(n=35)、中度型(n=30)和重度组(n=15),根据治疗后3个月的随访结果分为预后良好组(n=54)和预后不良组(n=26);另选择同时期行体格检查的70例健康者纳入对照组.所有研究对象均在入院后24 h内检测血清Neu5Ac、S100A1水平,并进行组间比较;采用受试者工作特征曲线(ROC)评估ACI患者预后临床效能.[结果]研究组血清Neu5Ac、S100A1水平均明显高于对照组(P<0.05);重度组血清Neu5Ac、S100A1水平明显高于中度组和轻度组(重度组>中度组>轻度组,P<0.05);预后不良组血清Neu5Ac、S100A1水平均明显高于预后良好组(P<0.05);血清Neu5Ac、S100A1预测ACI预后的AUC分别为0.830和0.882,二者联合预测的AUC为0.927.[结论]ACI患者血清Neu5Ac、S100A1水平均明显升高,其水平变化与病情及预后密切相关,两者联合应用预测ACI预后的效果更佳. 展开更多

High mobility group protein 1(HMGB1) is a multifunctional protein that interacts with DNA and chromatin to influence the regulation of transcription, DNA replication and repair and recombination. We show that HMGB1 alters the structure and stability of the canonical nucleosome(N) in a nonenzymatic,adenosine triphosphate-independent manner. As a result, the canonical nucleosome is converted to two stable, physically distinct nucleosome conformers. Although estrogen receptor(ER) does not bind to its consensus estrogen response element within a nucleosome, HMGB1 restructures the nucleosome to facilitate strong ER binding. The isolated HMGB1-restructured nucleosomes(N' and N'') remain stable and exhibit a number of characteristics that are distinctly different from the canonical nucleosome. These findings complement previous studies that showed(1) HMGB1 stimulates in vivo transcriptional activation at estrogen response elements and(2) knock down of HMGB1 expression by siR NA precipitously reduced transcriptional activation. The findings indicate that a major facet of the mechanism of HMGB1 action involves a restructuring of aspects of the nucleosome that appear to relax structural constraints within the nucleosome. The findings are extended to reveal the differences between ER and the other steroid hormone receptors. A working proposal outlines mechanisms that highlight the multiple facets that HMGB1 may utilize in restructuring the nucleosome. 展开更多

Texture evolution of an extruded dilute Mg-1Sn-1Zn-1Al alloy was thoroughly investigated based on the twinning and dynamic recrystallization(DRX)behavior via hot compression at a strain rate of 10 s^(-1)and temperature of 225℃.It was found that the types and intensities of the texture are strongly dependent on the fraction of twins and DRX modes as well as regions where sub-grain boundaries(sub-GBs)are intensively accumulated.At the initial stage of deformation,the formation of compression direction(CD)-tilted basal texture was mainly determined by the occurrence of{101^(-)2}extension twins.As the strain increases,the variation in the texture intensity was greatly dominated by the DRX modes but the type of main texture remained unchanged.These findings are of great importance for texture modification of wrought Mg-Sn-based alloys during post-deformation. 展开更多

The Pan-1 coal mine located in Huainan municipality, Anhui province, is abundant in coal resources. In order to discover the natural conditions of gas in its coal seams, we inverted the burial history of these coal seams using the software of Easy%Ro method and simulated the development process of gas volumes and pressure of the major coal seams using CBM History Simulation Software. Our analysis shows that the devolution of gas volumes and levels of pressure can be divided into four stages: i.e., a biogas stage (P1-P3), a pyrolysis gas stage (T1-T2), an active gas enrichment stage (T3-K1) and a gas dissipating stage (K2-present). Currently, the average amounts of gas and gas pressure in coal seams Nos. 13-1, 11-2 and 8 of the Pan-1 coal mine are 8.18 m3/t and 2.20 MPa; 3.89 m3/t and 2.47 MPa and 6.35 m3/t and 2.89 MPa, respectively. N-1最大供电能力(total supply capability,TSC)是评估配电网的重要指标。该文提出TSC判定定理并证明。定理1:每回馈线均能找到与其具有转带关系的一回馈线/馈线段,使两者负荷互补;且N供一备和不对称分段联络接线的备用馈线负荷最小,对称分段联络接线由同一馈线提供备用的馈线段负荷相等。定理2:任意馈线N-1后,所有与故障馈线相联络馈线都满载。定理3:冗余量最小。冗余量是由严格边界等式约束得到的冗余式的取值。这3个定理分别从N-1前负载特征、N-1后负载特征和数学模型的不同角度进行判定。该文证明以上3个定理,并采用我国常见接线模式和配电网算例验证;进一步给出定理的应用举例。判定定理是TSC理论体系的一个基础性工作。与现有研究相比,判定定理提炼出了TSC点的负荷分布特征,能方便地确定所有达到TSC的系统状态,为规划运行中优化负荷分布提供方向,便于发挥配电网的供电能力。 展开更多

血清Neu5Ac、S100A1水平与急性心源性脑梗死患者病情、预后的关系

目的 探讨血清N-乙酰神经氨酸(Neu5Ac)、S100钙结合蛋白质A1(S100A1)水平与急性心源性脑梗死患者病情、预后的关系。方法 选取2021年9月至2023年9月承德市中心医院收治的165例急性心源性脑梗死患者,根据梗死面积将患者分为小/中面积组(... 目的 探讨血清N-乙酰神经氨酸(Neu5Ac)、S100钙结合蛋白质A1(S100A1)水平与急性心源性脑梗死患者病情、预后的关系。方法 选取2021年9月至2023年9月承德市中心医院收治的165例急性心源性脑梗死患者,根据梗死面积将患者分为小/中面积组(梗死直径≤4 cm,97例)和大面积组(梗死直径>4 cm,68例);根据美国国立卫生研究院卒中量表(NIHSS)评分将患者分为低NIHSS组(NIHSS<16分,108例)和高NIHSS组(NIHSS≥16分,57例)。检测患者Neu5Ac、S100A1水平,根据患者出院后3个月mRS评分将其分为预后不良组(n=41)和预后良好组(n=124)。采用Pearson相关法探讨血清Neu5Ac、S100A1与脑梗死面积、NIHSS评分的关系,采用多因素logistic回归分析影响因素。结果小/中面积组Neu5Ac、S100A1水平低于大面积组,差异有统计学意义(P<0.05)。低NIHSS组Neu5Ac、S100A1水平低于高NIHSS组,差异有统计学意义(P<0.05)。血清Neu5Ac、S100A1与脑梗死面积、NIHSS评分均呈正相关(均P<0.001)。预后良好组血清Neu5Ac、S100A1低于预后不良组,差异有统计学意义(P<0.05)。预后良好组梗死面积大面积比例、入院NIHSS评分低于预后不良组,差异有统计学意义(P<0.05)。梗死面积大面积、入院NIHSS评分≥15.33分、Neu5Ac≥274.58 ng/mL、S100A1≥5.08 ng/mL是急性心源性脑梗死患者预后危险因素(P<0.05)。结论 血清Neu5Ac、S100A1与急性心源性脑梗死患者脑梗死面积、NIHSS评分有关,有望作为预测患者病情及预后的潜在标记物。 展开更多

目的探讨真核翻译起始因子4γ2(EIF4G2)通过调控N-乙酰氨基半乳糖转移酶1(GALNT1)mRNA对胃癌进展的影响。方法通过生物信息数据库分析GALNT1、EIF4G2在胃癌细胞和胃癌组织中的表达及与胃癌患者预后的关系,并分析胃癌组织中EIF4G2、GALNT... 目的探讨真核翻译起始因子4γ2(EIF4G2)通过调控N-乙酰氨基半乳糖转移酶1(GALNT1)mRNA对胃癌进展的影响。方法通过生物信息数据库分析GALNT1、EIF4G2在胃癌细胞和胃癌组织中的表达及与胃癌患者预后的关系,并分析胃癌组织中EIF4G2、GALNT1表达的相关性。采用实时荧光定量聚合酶链反应(RT-qPCR)检测GALNT1 mRNA、EIF4G2 mRNA在人胃黏膜上皮细胞GES-1和人胃癌细胞(AGS细胞等)中的表达水平,并采用蛋白质印迹法(Western blot)检测GALNT1、EIF4G2蛋白表达水平。建立干扰GALNT1和过表达EIF4G2的AGS细胞株,采用CCK-8法和克隆形成实验检测细胞增殖能力,采用TUNEL实验检测细胞凋亡能力,采用划痕实验和Transwell实验分别检测细胞迁移和侵袭能力,采用Western blot检测凋亡和转移相关蛋白表达水平,采用RIP实验和RNA下拉实验检测GALNT1 mRNA与EIF4G2的结合关系。结果胃癌组织和胃癌细胞中的GALNT1、EIF4G2水平分别高于正常组织和人胃黏膜上皮细胞,差异有统计学意义(P<0.001),且与胃癌患者预后差相关;胃癌组织中,EIF4G2表达与GALNT1表达呈正相关。敲低GALNT1后,AGS细胞活力降低,克隆形成数、迁移和侵袭细胞数减少,凋亡细胞数增加,Bcl-2、MMP2、MMP9蛋白表达降低,Bax、cleaved-caspase3、cleaved-caspase9蛋白表达增加,差异有统计学意义(P<0.001)。EIF4G2可与GALNT1 mRNA结合,促进GALNT1 mRNA、GALNT1蛋白表达及GALNT1 mRNA稳定性,差异有统计学意义(P<0.001)。共转染sh-GALNT1-1与oe-EIF4G2可逆转sh-GALNT1-1对AGS细胞增殖、凋亡、迁移和侵袭的作用,差异有统计学意义(P<0.05或P<0.01或P<0.001)。结论EIF4G2可通过稳定GALNT1 mRNA促进胃癌细胞增殖、迁移、侵袭并抑制细胞凋亡,其或可成为胃癌临床诊疗的新靶点。 展开更多

目的:基于线粒体动力学途径变化下探究小分子Sigma-1受体对顺铂(CDDP)诱导的急性肾损伤(AKI)的影响。方法:实验分为对照组、PRE-084组、CDDP组、CDDP+PRE-084组,共4组,每组10只小鼠,CDDP组和CDDP+PRE-084组经腹腔注射CDDP(15 mg/kg)诱导... 目的:基于线粒体动力学途径变化下探究小分子Sigma-1受体对顺铂(CDDP)诱导的急性肾损伤(AKI)的影响。方法:实验分为对照组、PRE-084组、CDDP组、CDDP+PRE-084组,共4组,每组10只小鼠,CDDP组和CDDP+PRE-084组经腹腔注射CDDP(15 mg/kg)诱导AKI模型,随后PRE-084组和CDDP+PRE-084组经腹腔注射PRE-084(0.6 mg/kg),每日1次,连续7 d;结束后,全自动生化仪进行血清尿素氮(BUN)与血清肌酐(Scr)水平检测,酶联免疫吸附法(ELISA)测定尿液中肾损伤分子1(KIM-1)含量,苏木精-伊红(HE)染色观察肾组织病理形态学变化,TdT介导的dUTP缺口末端标记(TUNEL)法检测肾脏细胞凋亡情况,透射电子显微镜观察肾脏组织线粒体超微结构,生物发光技术检测肾脏组织三磷酸腺苷(ATP)产生水平,免疫荧光双染测定肾脏组织中Sigma-1受体与Mfn2定位表达,蛋白质免疫印迹法(Western blot)测定肾脏组织内线粒体分裂蛋白Drp1、Fis1与线粒体融合蛋白Opa1、Mfn1的表达。结果:与CDDP组比较,CDDP+PRE-084组小鼠血清BUN和Scr水平、尿液KIM-1水平均降低(P<0.05),肾脏组织病理损伤情况得到明显改善,TUNEL阳性细胞率减少(P<0.05),线粒体肿胀、碎裂及空泡变性程度减小,形态恢复,肾脏组织ATP含量升高(P<0.05),Sigma-1受体与Mfn2荧光染色增强,Drp1、Fis1蛋白相对表达量下调且Opa1、Mfn1蛋白相对表达量上调(P<0.05)。结论:线粒体分裂与融合途径介导了CDDP所致小鼠AKI的发生,Sigma-1受体通过调控线粒体动力学途径对CDDP诱导的AKI小鼠起到保护作用。 展开更多

本研究旨在探讨N-乙酰-L-半胱氨酸(NAC)对1型糖尿病模型比格犬晶状体上皮氧化损伤的影响。将40只2~3岁比格犬随机分为对照组(CON组)、糖尿病模型组(DM组)、胰岛素治疗组(INS组)、NAC联合胰岛素治疗组(NAC+INS组)和NAC治疗组(NAC组),每组... 本研究旨在探讨N-乙酰-L-半胱氨酸(NAC)对1型糖尿病模型比格犬晶状体上皮氧化损伤的影响。将40只2~3岁比格犬随机分为对照组(CON组)、糖尿病模型组(DM组)、胰岛素治疗组(INS组)、NAC联合胰岛素治疗组(NAC+INS组)和NAC治疗组(NAC组),每组8只,各治疗组试验犬模型建立持续120 d。在造模前及造模后按试验设定时间对犬进行空腹血糖监测、胰岛素释放试验、房水葡萄糖测定、用裂隙灯进行白内障等级评定。对眼晶状体上皮进行病理组织学观察的同时,利用ELISA、RT-PCR等方法对晶状体内氧化应激指标进行检测。试验结果显示:空腹血糖监测和胰岛素释放试验显示,成功建立了1型糖尿病比格犬模型;与CON组相比,DM组犬在试验结束时,血清及房水葡萄糖显著升高(P<0.05),晶状体功能明显下降;裂隙灯结果显示,DM组犬的晶状体混浊等级在试验结束时达Ⅳ级,其余4组并未出现明显皮质混浊迹象,且DM组的晶状体上皮出现细胞核固缩和坏死小体等较为明显的病理变化;NAC、INS和NAC+INS组血糖虽呈下降趋势,但仍高于CON组,其中,NAC+INS组血糖较DM组显著下降(P<0.05),晶状体混浊也明显改善;试验结束时,与CON组相比,DM组犬晶体上皮和房水MDA含量升高(P<0.05或P<0.01),GSH-Px活性和GSH/GSSG均显著下降(P<0.05或P<0.01),GR活性下降(晶状体上皮,P>0.05;房水,P<0.01),给予NAC联合胰岛素治疗后,晶体上皮内的氧化应激标志物均出现相反趋势;DM组及各治疗组中抗氧化相关基因SOD2和GR表达量均显著升高(P<0.05)。综上:NAC联合胰岛素治疗能有效改善糖尿病犬的血糖和晶状体功能,NAC联合胰岛素治疗后对糖尿病犬晶状体上皮损伤的保护作用可能和抑制晶状体的氧化应激有关。 展开更多

动态心电图及sST2、PAI-1在川崎病伴发冠脉损伤诊断中的研究

目的探讨动态心电图及可溶性人基质裂解素-2(sST2)、纤溶酶原激活物抑制物-1(PAI-1)在川崎病伴发冠脉损伤诊断中的意义。方法研究对象选取2018年1月至2024年1月郑州大学第一附属医院收治的102例川崎病患儿,根据患儿是否伴发冠脉损伤分... 目的探讨动态心电图及可溶性人基质裂解素-2(sST2)、纤溶酶原激活物抑制物-1(PAI-1)在川崎病伴发冠脉损伤诊断中的意义。方法研究对象选取2018年1月至2024年1月郑州大学第一附属医院收治的102例川崎病患儿,根据患儿是否伴发冠脉损伤分为两组,其中损伤组40例,未损伤组62例。比较两组患儿的临床病例资料[性别、年龄、身体质量指数(BMI)、发热持续时间、白细胞计数、25-羟维生素D3(25-(OH)D_(3))、血红蛋白、血小板计数、肌酸激酶同工酶(CK-MB)、氨基末端脑利钠肽前体(NT-proBNP)、C反应蛋白(CRP)、sST2、PAI-1]及动态心电图异常率,通过多因素分析川崎病伴发冠脉损伤的影响因素,绘制ROC曲线评价不同指标对川崎病患儿伴发冠脉损伤的诊断价值。结果损伤组患儿的发热持续时间、血小板计数、CRP、CK-MB、NT-proBNP、sST2、PAI-1水平高于未损伤组,25-(OH)D_(3)水平低于未损伤组,差异具有统计学意义(P<0.05)。损伤组患儿的动态心电图异常率为60.00%,未损伤组患儿的动态心电图异常率为16.13%,损伤组高于未损伤组,差异具有统计学意义(P<0.05)。二元Logistic分析结果显示,发热持续时间、25-(OH)D_(3)、血小板计数、CRP、CK-MB、NT-proBNP、sST2、PAI-1、动态心电图异常是川崎病患儿伴发冠脉损伤的影响因素(P<0.05)。以sST2、PAI-1、动态心电图异常及3项指标联合应用作为变量绘制诊断川崎病伴发冠脉损伤的ROC曲线,sST2的AUC为0.789,PAI-1的AUC为0.729,动态心电图异常的AUC为0.719,联合检测的AUC为0.978。结论动态心电图及sST2、PAI-1水平诊断川崎病是否伴发CAL具有较高的诊断价值。 展开更多

伴随着可再生能源装机规模逐步扩大,电力系统的随机性和波动性进一步增强。为了制定更准确的机组启停和发电计划,保证系统运行安全,建立计及N-1线路故障的连续时间随机安全约束机组组合模型。首先采用Bernstein多项式将该无限维的连续... 伴随着可再生能源装机规模逐步扩大,电力系统的随机性和波动性进一步增强。为了制定更准确的机组启停和发电计划,保证系统运行安全,建立计及N-1线路故障的连续时间随机安全约束机组组合模型。首先采用Bernstein多项式将该无限维的连续时间优化模型转化为有限维的混合整数线性规划(mixed integer linear programming,MILP)模型。由于转化后的模型维度极高,为高效求解该模型,提出逐步对冲-Benders分解(progressive hedging-Bendersdecom-position, PH-BD)双层分解算法:外层采用逐步对冲(progressive hedging,PH)算法对场景打捆后的“场景束”进行分解,实现每个场景束对应问题的并行计算;内层采用Benders分解(Benders decomposition,BD)算法将每个场景束对应问题分解为无线路约束主问题和可并行求解的线路约束子问题。最后在IEEE118节点系统中进行算例验证。结果表明,与离散时间模型相比,连续时间随机安全约束机组组合模型可以更准确地制定机组启停和发电计划以及设置备用容量;与传统PH算法相比,所提出的PH-BD算法显著降低了求解时间。 展开更多

[目的]在保证变电站平均负载率相对合理、提高变电站利用小时数的前提下,为了解决目前电网存在的高峰负荷过高造成的变电站主变压器短时间重载问题,延缓电网的建设投资,保证电网经济运行,文章提出了基于满足主变N-1供电要求的变电站储能定容方法。[方法]首先,建立储能定容数学模型实现对负荷运行曲线削峰填谷、降低变压器最高运行负载率;然后,构建锂离子电化学储能定容经济目标函数,并开展建设锂离子电化学储能和扩建主变的建设投资对比,分析储能定容经济性;最后,结合实际变电站,提出储能配建规模的建议及经济可行的储能建设成本。[结果]通过分析,随着未来锂离子电化学储能单位造价降低,储能功率可按照变电站容量15%、充放电时长2 h考虑,储能系统单位造价降低至1 200元/kWh左右时,建设储能的经济优势明显。[结论]所提出的储能定容方法可以有效解决变电站短时间重载问题,并具备较常规输变电工程更优的经济性。 展开更多

目的研究谷胱甘肽S-转移酶Omega-1(GSTO1)在宫颈癌组织中的表达及其与患者生存时间的相关性,并探究GSTO1 N-糖基化对宫颈癌细胞增殖、迁移、侵袭和上皮间质转化的影响。方法用免疫组化检测82例宫颈癌患者癌组织中GSTO1的表达水平,分析GS... 目的研究谷胱甘肽S-转移酶Omega-1(GSTO1)在宫颈癌组织中的表达及其与患者生存时间的相关性,并探究GSTO1 N-糖基化对宫颈癌细胞增殖、迁移、侵袭和上皮间质转化的影响。方法用免疫组化检测82例宫颈癌患者癌组织中GSTO1的表达水平,分析GSTO1表达与临床病理特征的相关性。Kaplan-Meier法绘制生存曲线,评估宫颈癌组织中GSTO1的表达对患者生存时间的影响;单因素和多因素Cox回归分析评估影响宫颈癌预后的独立危险因素;NetNGlyc 1.0 Server数据库预测GSTO1潜在的N-糖基化修饰位点(Asn55、Asn135、Asn190);在宫颈癌细胞(HeLa)中转染GSTO155位、135位和190位N-糖基化突变载体以及GSTO1野生型载体和空载体,嘌呤霉素筛选稳定转染细胞;Western blot实验检测慢病毒干扰效果;EdU增殖实验、划痕愈合实验和Transwell实验检测GSTO1 N-糖基化定点突变对HeLa细胞增殖、迁移和侵袭的影响;Western blot实验检测GSTO1 N-糖基化定点突变对HeLa细胞上皮间质转化的影响。结果免疫组化结果显示GSTO1在宫颈癌组织中高表达;GSTO1在间质浸润深度≥1/2、有淋巴脉管间隙浸润、有淋巴结转移的宫颈癌组织中的表达率更高(P<0.05),且GSTO1高表达与较差的总生存时间相关;GSTO1 N-糖基化定点突变后,HeLa细胞的增殖、迁移和侵袭的细胞个数均减少(P<0.01);Western blot结果显示GSTO1 N-糖基化定点突变抑制HeLa细胞上皮间质转化。结论宫颈癌组织中GSTO1表达与患者间质浸润深度、淋巴脉管间隙浸润和淋巴结转移有关,且与患者较短的生存时间相关。GSTO1 N-糖基化定点突变能够抑制HeLa细胞的增殖、转移和上皮间质转化。 展开更多
