目的:探讨exosomes(Exo)联合卡介苗(bacillus Calmette-Gurin vaccine,BCG)的体内抗肿瘤效应。方法:通过密度梯度离心法分离和纯化E.G7-OVA肿瘤细胞来源的Exo,Western blotting检测其蛋白成分。分别以Exo、BCG、Exo+BCG、PBS免疫小鼠,... 目的:探讨exosomes(Exo)联合卡介苗(bacillus Calmette-Gurin vaccine,BCG)的体内抗肿瘤效应。方法:通过密度梯度离心法分离和纯化E.G7-OVA肿瘤细胞来源的Exo,Western blotting检测其蛋白成分。分别以Exo、BCG、Exo+BCG、PBS免疫小鼠,以E.G7-OVA细胞攻击,观察各组免疫保护效应;建立E.G7-OVA荷瘤小鼠模型,观察各组免疫治疗效应。LDH法检测4组免疫小鼠脾细胞CTL细胞毒性。结果:Western blotting检测显示,Exo含有HSP60、OVA、HSC70和CD63分子;免疫保护实验结果显示,Exo+BCG组免疫小鼠90d无瘤率显著高于Exo组和BCG组(60%vs20%、0%,P<0.01);免疫治疗实验结果显示,Exo+BCG对小鼠移植瘤的抑制显著强于Exo组和BCG组(P<0.01)。CTL检测结果显示,Exo+BCG免疫小鼠的CTL细胞特异性杀伤E.G7-OVA细胞的活性显著高于其他各组(P<0.01)。结论:BCG作为免疫佐剂能显著增强exosomes的体内抗肿瘤效应。 展开更多

Edge Computing is one of the radically evolving systems through generations as it is able to effectively meet the data saving standards of consumers,providers and the workers. Requisition for Edge Computing based items havebeen increasing tremendously. Apart from the advantages it holds, there remainlots of objections and restrictions, which hinders it from accomplishing the needof consumers all around the world. Some of the limitations are constraints oncomputing and hardware, functions and accessibility, remote administration andconnectivity. There is also a backlog in security due to its inability to create a trustbetween devices involved in encryption and decryption. This is because securityof data greatly depends upon faster encryption and decryption in order to transferit. In addition, its devices are considerably exposed to side channel attacks,including Power Analysis attacks that are capable of overturning the process.Constrained space and the ability of it is one of the most challenging tasks. Toprevail over from this issue we are proposing a Cryptographic LightweightEncryption Algorithm with Dimensionality Reduction in Edge Computing. Thet-Distributed Stochastic Neighbor Embedding is one of the efficient dimensionality reduction technique that greatly decreases the size of the non-linear data. Thethree dimensional image data obtained from the system, which are connected withit, are dimensionally reduced, and then lightweight encryption algorithm isemployed. Objective To study the effects of TYRO protein kinase-binding protein(TYROBP)deficiency on learning behavior,glia activation and pro-inflammatory cycokines,and Tau phosphorylation of a new Alzheimer’s disease(AD)mouse model carrying a PSEN1 p.G378E mutation.Methods A new AD mouse model carrying PSEN1 p.G378E mutation was built based on our previously found AD family which might be ascribed to the PSEN1 mutation,and then crossed with TYROBP deficient mice to produce the heterozygous hybrid mice(PSEN1^(G378E)/WT;Tyrobp^(+/-))and the homozygous hybrid mice(PSEN1^(G378E/G378E);Tyrobp^(-/-)).Water maze test was used to detect spatial learning and memory ability of mice.After the mice were sacrificed,the hippocampus was excised for further analysis.Immunofluorescence was used to identify the cell that expresses TYROBP and the number of microglia and astrocyte.Western blot was used to detect the expression levels of Tau and phosphorylated Tau(p-Tau),and ELISA to measure the levels of pro-inflammatory cytokines.Results Our results showed that TYROBP specifically expressed in the microglia of mouse hippocampus.Absence of TYROBP in PSEN1^(G378E) mutation mouse model prevented the deterioration of learning behavior,decreased the numbers of microglia and astrocytes,and the levels of interleukin-6,interleukin-1βand tumor necrosis factor-αin the hippocampus(all P<0.05).The ratios of AT8/Tau5,PHF1/Tau5,pT181/Tau5,pT231/Tau5 and p-ERK/ERK were all higher in homozygous hybrid mice(PSEN1^(G378E/G378E);Tyrobp^(-/-) mice)compared with PSEN1^(G378E/G378E) mice(all P<0.05).Conclusions TYROBP deficiency might play a protective role in the modulation of neuroinflammation of AD.However,the relationship between neuroinflammation processes involving microglia and astrocyte activation,and release of pro-inflammatory cytokines,and p-Tau pathology needs further study. 展开更多

The production-oriented approach (POA) has been developed over a decade. It is driven by the need to improve English classroom instruction for university students in China (Wen, 2016). It is also motivated by the ... The production-oriented approach (POA) has been developed over a decade. It is driven by the need to improve English classroom instruction for university students in China (Wen, 2016). It is also motivated by the aspiration to enhance the quality of foreign language education in other similar pedagogical contexts outside China. A volume of research has been done by Wen Qiufang and her research team, to formulate the theory of POA and to test its effectiveness in classroom pedagogy (e.g. Wen, 2016, 2015; Yang, 2015; Zhang, 2015). At the moment, the POA is still at an early stage of theory building and almost all empirical research is done in the Chinese context. In order to improve the quality of this theory and to make it intelligible to the international academic community, a one-day symposium was held in Beijing Foreign Studies University on May 15, 2017. The symposium was entitled 'The first international forum on innovative foreign language education in China: Appraisal of the POA'. In the forum, leading experts in applied linguistics were invited to discuss the strengths and weaknesses of the POA and the directions for its future development. The symposium was the first attempt for the POA research team to discuss its latest work with international scholars. This Viewpoint section collects the responses of four experts who participated in the symposium, listed in alphabetical order. The collection of articles covers three topics related to the POA: its pedagogical application, its use for teacher training, and its research. Alister Cumming is Professor Emeritus and the former Head of the Centre for Educational Research on Languages and Literacies, University of Toronto, Canada. His article focuses primarily on POA research as an exemplary case of design-based research. Rod Ellis is Research Professor in the School of Education at Curtin University, Australia. He discusses POA in terms of pedagogy, teacher training and research, with both critiques and constructive suggestions. Paul Kei Matsuda is Professor of English and Director of Second Language Writing at Arizona State University, the writed states. He responds to POA from the perspective of an expert researcher and teacher of L2 writing. Charlene Polio is Professor and Associate Chair in the Department of Linguistics & Germanic, Slavic, Asian & African Languages atMichigan State University, the writed states. She conceptualises POA as a useful method to address some issues in pre-service teacher development. Overall, the articles in this section are insightful and reader-friendly. They are not only useful for the development of POA in particular, but may also be valuable to a broad range of researchers as they touch upon pertaining issues, as well as emerging topics, in the field of applied linguistics.
