Therefore, a field experiment was conducted to test the performance of alternate wetting and d...Sustainable irrigation method is now essential for adaptation and adoption in the areas where water resources are limited. Therefore, a field experiment was conducted to test the performance of alternate wetting and drying furrow irrigation(AWDFI) on crop growth, yield, water use efficiency(WUE), fruit quality and profitability analysis of tomato. The experiment was laid out in randomized complete block design with six treatments replicated thrice during the dry seasons of 2013-2014 and 2014-2015. Irrigation water was applied through three ways of furrow: AWDFI, fixed wetting and drying furrow irrigation(FWDFI) and traditional(every) furrow irrigation(TFI). Each irrigation method was divided into two levels: irrigation up to 100 and 80% field capacity(FC). Results showed that plant biomass(dry matter) and marketable fruit yield of tomato did not differ significantly between the treatments of AWDFI and TFI, but significant difference was observed in AWDFI and in TFI compared to FWDFI at same irrigation level. AWDFI saved irrigation water by 35 to 38% for the irrigation levels up to 80 and 100% FC, compared to the TFI, respectively. AWDFI improved WUE by around 37 to 40% compared to TFI when irrigated with 100 and 80% FC, respectively. Fruit quality(total soluble solids and pulp) was found greater in AWDFI than in TFI. Net return from AWDFI technique was found nearly similar compared to TFI and more than FWDFI. The benefit cost ratio was viewed higher in AWDFI than in TFI and FWDFI by 2.8, 8.7 and 11, 10.4% when irrigation water was applied up to 100 and 80% FC, respectively. Unit production cost was obtained lower in AWDFI compared to TFI and FWDFI. However, AWDFI is a useful water-saving furrow irrigation technique which may resolve as an alternative choice compared with TFI in the areas where available water and supply methods are limited to irrigation.展开更多

The experiment was designed as a 2 × 2 factorial arrangement with two sources of dietary proteins(cottonseed meal, CSM vs. soybean meal, SBM) and two levels of CP(12 % vs. 14 %, as-fed basis). Seventy-two crossbred gilts(89.5 ± 0.9 kg) were allotted to one of four dietary treatments in a randomized complete block design for a period of 28 d. All diets were formulated to be isoenergetic and similar concentrations of standardized ileal digestible essential AA covering the nutrient requirements of pigs.Results: Growth, carcass characteristics and meat quality were not affected by dietary protein source nor crude protein level(P > 0.10) except that average daily feed intake was increased by CSM diets(P = 0.03). Gilts offered reduced protein diets had lower muscle p H45min(P 〈 0.05). Neither dietary protein source nor crude protein level influenced N deposition. However, reduced protein diets decreased N intake, N excretion, and serum urea nitrogen content, whilst improved N efficiency(P 〈 0.01). CSM diets increased N intake(P = 0.04),but did not depress N efficiency. The concentrations of phenylalanine, tryptophan, cysteine and tyrosine(P 〈 0.05) of the longissimus muscle were decreased when gilts offered CSM diets, while muscle intracellular free valine concentration was increased(P = 0.03). The gilts offered reduced protein diets had greater intracellular concentrations of free methionine, lysine, and total AA in muscle(P 〈 0.05).Conclusion: These results suggest that CSM could replace SBM as a primary protein source in finishing pig diets in terms of performance, N efficiency, carcass characteristics, and meat quality, but decrease the concentrations of muscle specific AA. Furthermore, the reduced protein diet played an important role in increasing muscle intracellular concentrations of specific free amino acids(FAA), and in reducing the relative ratios of specific FAA to lysine in longissimus dorsi muscle of pig, whose biological meaning needs further studies.展开更多

Differentiated cells can be reprogrammed into pluripotent stem cells,known as“induced pluripotent stem cells”(iPSCs),through the overexpression of defined transcription factors.The creation of iPSC lines has opened ...Differentiated cells can be reprogrammed into pluripotent stem cells,known as“induced pluripotent stem cells”(iPSCs),through the overexpression of defined transcription factors.The creation of iPSC lines has opened new avenues for patient-specific cell replacement therapies for regenerative medicine.However,the clinical utilization of iPSCs is largely impeded by two limitations.The first limitation is the low efficiency of iPSCs generation from differentiated cells.The second limitation is that many iPSC lines are not authentically pluripotent,as many cell lines inefficiently differentiate into differentiated cell types when they are tested for their ability to complement embryonic development.Thus,the“quality”of iPSCs must be increased if they are to be differentiated into specialized cell types for cell replacement therapies.Overcoming these two limitations is paramount to facilitate the widespread employment of iPSCs for therapeutic purposes.Here,we summarize recent progress made in strategies enabling the efficient production of high-quality iPSCs,including choice of reprogramming factors,choice of target cell type,and strategies to improve iPSC quality.展开更多
