Methods ICR mice and mouse lymphoma cell line (EL-4 cells) was used. The expressions of CD4, CD25, Nrpl, calcineurin and PKC-α were detected by flow cytometry. The expressions of TGF-131, IL-10, PKA and cAMP were estimated with ELISA. Results At 12 h after irradiation, the expression of Nrpl increased significantly in 4.0 Gy group, compared with sham-irradiation group (P〈0.05) in the spleen and thymus, respectively, when ICR mice received whole-body irradiation (WBI). Meanwhile the synthesis of Interleukin 10 (IL-20) and transforming growth factor-β1 (TGF-β1) increased significantly after high dose irradiation (HDR) (〉 or = 1.0 Gy). In addition, the expression of cAMP and PKA protein increased, while PKC-α, calcineurin decreased at 12h in thymus cells after 4.0 Gy X-irradiation. While TGF-β1 was clearly inhibited when the PLC-PIP2 signal pathway was stimulated or the cAMP-PKA signal pathway was blocked after 4.0 Gy X-irradiation, this did not limit the up-regulation of CD4+CD25+Nrpl+Treg cells after ionizing radiation. Conclusion These results indicated that HDR might induce CD4+CD25+Nrpl+Treg cells production and stimulate TGF-β1 secretion by regulating signal molecules in mice.展开更多

Signals during the test are detected in ultralow frequency band. The experimental results may be outlined as follows: original waveforms of the electro-magnetic radiation (EMR) and AE in the range of 0-20 kHz frequencies are obtained; the EMR's intensity in fracture process is related to some factors as rock type, content of water, fracture intensity of rocks, loading rates and fracture state etc.; the EMR'S rate is proportional to that of AE during fracture statistically. About 70% of maximum values of EMR rate and AE rate are corresponded with each other; furthermore, the EM signals (EMS) are generally more developed and longer duration than AE signals; the principal energy of EMS detected by copper coin antenna as sensor in power spectra are distributed in frequencies lower than 6 kHz and otherwise those detected by EMinduction coil are at about 10 kHz, and in contrast the major energies of AE are in frequencies less than3 kHz; another notable phenomenon is that distinct electro-magnetic signals are detected before AE occurrence. Since EMR and AE signals obtained in the process of rock fracture are better corresponded to eachother, therefore it'S reasonable to propose that the EMR anormalies observed prior to some earthquakes aremainly produced by fractures of rock in source region, as for the EMS occurred before AE may be related topiezoelectric and piezomagnetic effects.展开更多

Methods: Androgen sensitive prostate cancer cell line LNCaP was cultured and treated by radiation, androgen withdrawal and combination of the two. Apoptosis was determined using apoptotic cells staining and mononuclear cell direct cytotox-icity assay. The total RNA were extracted and harvested. cDNA probes were prepared and labeled with biotin-16-dUTP and then hybridized to commercially available cDNA arrays, including apoptosis pathway-specific genes. The expression of important gene was further determined using RT-PCR. Results: Radiation induced additive apoptosis of prostate cancer cells; androgen withdrawal exhibited synergetic action. TNFRSF8 variant 2, DFFA, LTbR, mdm2, Myd88, TNFRSF14 and TNFSF4 mRNA were up regulated by radiation, while Survivin and Bar mRNA were down regulated. Mcl-1, TNFRSF14, MyD88 and TNFSF4 mRNA were up regulated by androgen withdrawal, while Bar, Survivin and TRAIL-R3 mRNA were down regulated. Conclusion: Radiation and androgen withdrawal altered the expression of apoptosis pathway genes of prostate cancer cells in different patterns, which may contribute to the additive apoptotic effect induced by the combined treatment.展开更多
