Experimental investigation on using CO_(2)/H_(2)O emulsion with high water cut in enhanced oil recovery

CO_(2) emulsions used for EOR have received a lot of interest because of its good performance on CO_(2)mobility reduction.However,most of them have been focusing on the high quality CO_(2) emulsion(high CO_(2) fraction),while CO_(2) emulsion with high water cut has been rarely researched.In this paper,we carried out a comprehensive experimental study of using high water cut CO_(2)/H_(2)O emulsion for enhancing oil recovery.Firstly,a nonionic surfactant,alkyl glycosides(APG),was selected to stabilize CO_(2)/H_(2)O emulsion,and the corresponding morphology and stability were evaluated with a transparent PVT cell.Subsequently,plugging capacity and apparent viscosity of CO_(2)/H_(2)O emulsion were measured systematically by a sand pack displacement apparatus connected with a 1.95-m long capillary tube.Furthermore,a high water cut(40 vol%) CO_(2)/H_(2)O emulsion was selected for flooding experiments in a long sand pack and a core sample,and the oil recovery,the rate of oil recovery,and the pressure gradients were analyzed.The results indicated that APG had a good performance on emulsifying and stabilizing CO_(2) emulsion.An inversion from H_(2)O/CO_(2) emulsion to CO_(2)/H_(2)O emulsion with the increase in water cut was confirmed.CO_(2)/H_(2)O emulsions with lower water cuts presented higher apparent viscosity,while the optimal plugging capacity of CO_(2)/H_(2)O emulsion occurred at a certain water cut.Eventually,the displacement using CO_(2)/H_(2)O emulsion provided 18.98% and 13.36% additional oil recovery than that using pure CO_(2) in long sand pack and core tests,respectively.This work may provide guidelines for EOR using CO_(2) emulsions with high water cut. 展开更多

In this study,precise control over the thickness and termination of Ti3C2TX MXene flakes is achieved to enhance their electrical properties,environmental stability,and gas-sensing performance.Utilizing a hybrid method involving high-pressure processing,stirring,and immiscible solutions,sub-100 nm MXene flake thickness is achieved within the MXene film on the Si-wafer.Functionalization control is achieved by defunctionalizing MXene at 650℃ under vacuum and H2 gas in a CVD furnace,followed by refunctionalization with iodine and bromine vaporization from a bubbler attached to the CVD.Notably,the introduction of iodine,which has a larger atomic size,lower electronegativity,reduce shielding effect,and lower hydrophilicity(contact angle:99°),profoundly affecting MXene.It improves the surface area(36.2 cm^(2) g^(-1)),oxidation stability in aqueous/ambient environments(21 days/80 days),and film conductivity(749 S m^(-1)).Additionally,it significantly enhances the gas-sensing performance,including the sensitivity(0.1119Ωppm^(-1)),response(0.2% and 23%to 50 ppb and 200 ppm NO_(2)),and response/recovery times(90/100 s).The reduced shielding effect of the–I-terminals and the metallic characteristics of MXene enhance the selectivity of I-MXene toward NO2.This approach paves the way for the development of stable and high-performance gas-sensing two-dimensional materials with promising prospects for future studies. 展开更多

Superwetting materials have drawn unprecedented attention in the treatment of oily wastewater due to their preferable anti-fouling property and selective oil/water separation.However,it is still a challenge to fabricate multifunctional and environmentally friendly materials,which can be stably applied to purify the actual complicated wastewater.Here,a Ag/Ag/α-Fe_(2)O_(3) heterostructure anchored copper mesh was intentionally synthesized using a facile two-step hydrothermal method.The resultant mesh with superhydrophilicity and underwater superoleophobicity was capable of separating various oil/water mixtures with superior separation efficiency and high permeationflux driven by gravity.Benefiting from the joint effects of the smaller band gap of Ag/α-Fe_(2)O_(3) heterojunction,inherent antibacterial capacity of Ag/α-Fe_(2)O_(3) and Ag nanoparticles,favorable conductive substrate,as well as the hierarchical structure with superwettability,such mesh presented remarkably enhanced degradation capability toward organic dyes under visible light irradiation and antibacterial activity against both Escherichia coli(E.coli)and Staphylococcus aureus(S.aureus)compared with the pure Ag/α-Fe_(2)O_(3) coated mesh.Impressively,the mesh exhibited bifunctional water purification performance,in which organic dyes were eliminated simultaneously from water during oil/water separation in onefiltration process.More importantly,this mesh behaved exceptional chemical resistance,mechanical stability and long-term reusability.Therefore,this material with multifunctional integration may hold promising potential for steady water purification in practice. 展开更多

The development of 3D structural composites with electromagnetic(EM)wave absorption could attenuate EM waves.Herein,magnetized flower-like Cu_(9)S_(5)/ZnFe_(2)O_(4)composites were fabricated through a multistep hydrothermal method.The crystallographic and surface phase chemical information,morphological structure,and magnetic and EM parameters of the composites were analyzed.The prepared Cu_(9)S_(5)/ZnFe_(2)O_(4)composites have multiple loss paths for EM waves and present an overall 3D flower-like structure.The Cu_(9)S_(5)/ZnFe_(2)O_(4)composites exhibit a minimum reflection loss of-54.38 dB and a broad effective absorption bandwidth of 5.92 GHz.Through magnetization,ZnFe_(2)O_(4)particles are self-assembled and grown on the surfaces of Cu_(9)S_(5).Such a modification is conducive to the generation of additional cross-linking contact sites and the effective introduction of a large number of phase interfaces,crystalline defects,special three-dimensional flower-like structures,and magneto-electrical coupling loss effects.Moreover,the synergistic effect of multiple loss strategies effectively improves EM wave absorption by the material.This work can provide a strategy for the use of magnetizationmodified sulfide composite functional materials in EM wave absorption. 展开更多

Undesirable gas channeling always occurs along the high-permeability layers in heterogeneous oil reservoirs during water-alternating-CO_(2)(WAG)flooding,and conventional polymer gels used for blocking the“channeling”paths usually suffer from either low injectivity or poor gelation control.Herein,we for the first time developed an in-situ high-pressure CO_(2)-triggered gel system based on a smart surfactant,N-erucamidopropyl-N,N-dimethylamine(UC22AMPM),which was introduced into the aqueous slugs to control gas channeling inWAG processes.The water-like,low-viscosity UC22AMPM brine solution can be thickened by high-pressure CO_(2) owing to the formation of wormlike micelles(WLMs),as well as their growth and shear-induced structure buildup under shear flow.The thickening power can be further potentiated by the generation of denser WLMs resulting from either surfactant concentration augmentation or a certain range of heating,and can be impaired via pressurization above the critical pressure of CO_(2) because of its soaring solvent power.Core flooding tests using heterogeneous cores demonstrated that gas channeling was alleviated by plugging of high-capacity channels due to the in-situ gelation of UC22AMPM slugs upon their reaction with the pre-or post-injected CO_(2) slugs under shear flow,thereupon driving chase fluids into unrecovered low-permeability areas and producing an 8.0% higher oil recovery factor than the conventional WAG mode.This smart surfactant enabled high injectivity and satisfactory gelation control,attributable to low initial viscosity and the combined properties of one component and CO_(2)-triggered gelation,respectively.This work could provide a guide towards designing gels for reducing CO_(2) spillover and reinforcing the CO_(2) sequestration effect during CO_(2) enhanced oil recovery processes. 展开更多

Electronic properties of two-dimensional(2D) materials can be strongly modulated by localized strain. The typical spatial resolution of conventional Kelvin probe force microscopy(KPFM) is usually limited in a few hundreds of nanometers, and it is difficult to characterize localized electronic properties of 2D materials at nanoscales. Herein, tip-enhanced Raman spectroscopy(TERS) is proposed to combine with KPFM to break this restriction. TERS scan is conducted on ReS2bubbles deposited on a rough Au thin film to obtain strain distribution by using the Raman peak shift. The localized contact potential difference(CPD) is inversely calculated with a higher spatial resolution by using strain measured by TERS and CPD-strain working curve obtained using conventional KPFM and atomic force microscopy. This method enhances the spatial resolution of CPD measurements and can be potentially used to characterize localized electronic properties of 2D materials. 展开更多

气藏注CO_(2)提高天然气采收率并实现碳封存有望成为大幅度提高天然气产量与碳减排协同的潜在关键技术。为了给底水气藏注CO_(2)高效开发提供指导,针对地层水盐度对CO_(2)-CH_(4)-H_(2)O-NaCl体系相平衡影响、气藏注气过程中压力变化对C... 气藏注CO_(2)提高天然气采收率并实现碳封存有望成为大幅度提高天然气产量与碳减排协同的潜在关键技术。为了给底水气藏注CO_(2)高效开发提供指导,针对地层水盐度对CO_(2)-CH_(4)-H_(2)O-NaCl体系相平衡影响、气藏注气过程中压力变化对CO_(2)-CH_(4)-H_(2)O-NaCl体系相平衡影响、注采方案对注CO_(2)提高气藏采收率影响、盐度对注CO_(2)提产及封存影响等目前认识不清的问题开展了CO_(2)-CH_(4)-H_(2)O-NaCl体系相平衡规律及注CO_(2)提采与封存数值模拟研究。研究结果表明:①随着盐度增加,CO_(2)和CH_(4)在盐水中的溶解度降低,液相的密度和黏度增加,盐度对气相性质几乎没有影响;②随着压力增加,CO_(2)和CH_(4)在液相中的溶解度均增加,气相、液相密度和黏度均增加,液相偏差因子随压力增加而增加,气相偏差因子先减小后增加;③同注同采方案CH_(4)产量更稳定且产出的CO_(2)少,而先注后采方案则会加速CO_(2)与CH_(4)的混合,CO_(2)封存量低,前者更适合注CO_(2)提采及封存;④在不考虑盐析效应的前提下,盐度对CH_(4)采收率和CO_(2)封存量的影响几乎可以忽略不计,不同盐度的衰竭底水气藏中CH_(4)采收率均超过80%、CO_(2)封存率均超过99%,短期注CO_(2)过程中,CO_(2)主要以气态或超临界态的形式被封存,少部分CO_(2)溶解在液相中,100年后CO_(2)在液相中的溶解质量分数约为5%。结论认为,衰竭底水气藏注CO_(2)能增压补能、驱替置换残余天然气,提高采收率并实现碳封存。 展开更多

目的通过超声造影(contrast-enhanced ultrasound,CEUS)评估小腿三头肌的微循环灌注情况,探讨CEUS在2型糖尿病(type 2 diabetes mellitus,T2DM)微血管病变评估中的应用价值。方法收集2020年8月至2023年1月在温州医科大学附属第二医院内... 目的通过超声造影(contrast-enhanced ultrasound,CEUS)评估小腿三头肌的微循环灌注情况,探讨CEUS在2型糖尿病(type 2 diabetes mellitus,T2DM)微血管病变评估中的应用价值。方法收集2020年8月至2023年1月在温州医科大学附属第二医院内分泌科住院治疗的T2DM患者51例,分为单纯T2DM组(n=15)、T2DM微循环病变组(n=36),记录每位患者的糖化血红蛋白(hemoglobin A1c,HbA1c)和胰岛素抵抗指数(homeostatic model assessment for insulin resistance,HOMA-IR)。CEUS观察小腿三头肌的造影情况,获取定量造影指标区域增强强度(enhanced intensity,PI-BI)和区域达峰时间(regional peak time,TTP-AT),并进行组间对比分析。结果T2DM微变组的HbA1c及HOMA-IR高于单纯T2DM组(P<0.05)。T2DM微变组腓肠肌内侧头(musculi gastrocnemii,MG)、腓肠肌外侧头(laterale musculi gastrocnemi,LG)、比目鱼肌(soleus,SOL)及交界区的TTP-AT均长于单纯T2DM组(P<0.05)。两组SOL的TTP-AT均为最长,LG次之,MG最短(P<0.05)。T2DM微变组MG的PI-BI高于SOL(P<0.05)。小腿三头肌交界区的TTP-AT与HbA1c、HOMA-IR之间均呈正相关(P<0.05)。结论CEUS小腿三头肌TTP-AT是评估T2DM患者微血管病变的新指标。 展开更多

南川常压海相页岩气注CO_(2)吞吐提高采收率工程实践

当前研究实验和模拟论证了注CO_(2)吞吐提高页岩气采收率的理论可行性,然而其实际工程效果尚未得到现场试验的验证。为此,基于四川盆地南川常压海相页岩气藏地质赋存及生产特征,开展了页岩等温吸附和CO_(2)—CH_(4)竞争吸附机理研究,实... 当前研究实验和模拟论证了注CO_(2)吞吐提高页岩气采收率的理论可行性,然而其实际工程效果尚未得到现场试验的验证。为此,基于四川盆地南川常压海相页岩气藏地质赋存及生产特征,开展了页岩等温吸附和CO_(2)—CH_(4)竞争吸附机理研究,实施了国内首次常压海相页岩气衰减井CO_(2)吞吐现场试验,并提出了构建成套技术体系的攻关方向。研究结果表明:①气井生产到后期进入低压低产阶段,表现出地层能量不足的特征,是提高气井采出程度的重要阶段;②常压页岩的CH_(4)和CO_(2)吸附能力明显高于低压或高压页岩,采用CO_(2)提采更具可行性和必要性;③相同条件下的常压、高压和低压3组页岩样品均表现出对CO_(2)的吸附量大于对CH_(4)的吸附量,且对气体的吸附能力依次为南川(常压海相)页岩>长宁(高压海相)页岩>延长(低压陆相)页岩;④试验井注CO_(2)吞吐增产效果明显,页岩气单井预测最终可采储量(EUR)从0.75×10^(8) m^(3)上升到0.90×10^(8) m^(3),单井控制储量5.186×10^(8) m^(3),预计采收率提高了2.9%。结论认为:①实施的国内首次常压海相页岩气衰减井注CO_(2)吞吐试验取得突破性进展,为我国页岩气井提高采收率技术研发与应用提供了重要借鉴与参考;②亟需开展页岩气衰减井CO_(2)提采资源潜力与时机评估、提采效果定量评价与工艺优化等研究,并形成页岩气衰减井注CO_(2)吞吐提高采收率成套关键技术及标准体系。 展开更多

急性缺血性脑卒中患者血清长链非编码RNA母系表达基因3和Zeste同源物增强子-2表达与神经功能损伤的相关性分析

目的探讨长链非编码RNA(lncRNA)母系表达基因3(MEG3)与Zeste同源物增强子-2(EZH2)在急性缺血性脑卒中(AIS)患者血清中的表达水平及其与神经功能损伤之间的关系。方法选取延安市人民医院2021年1月至2022年11月收治的92例AIS患者为脑卒中... 目的探讨长链非编码RNA(lncRNA)母系表达基因3(MEG3)与Zeste同源物增强子-2(EZH2)在急性缺血性脑卒中(AIS)患者血清中的表达水平及其与神经功能损伤之间的关系。方法选取延安市人民医院2021年1月至2022年11月收治的92例AIS患者为脑卒中组,92例健康体检者为对照组,根据NIHSS评分将脑卒中组患者分为致残性损伤组19例,非致残性损伤组73例。采用实时荧光定量聚合酶链式反应、酶联免疫吸附法分别检测血清lncRNAMEG3、EZH2水平。采用Spearman相关分析法进行AIS患者血清lncRNAMEG3、EZH2表达水平与美国国立卫生研究院卒中量表(NIHSS)评分之间的相关性分析;采用多因素Logistic回归分析AIS患者合并神经功能致残性损伤的影响因素,并绘制受试者工作特征(ROC)曲线分析血清lncRNA MEG3、EZH2水平对AIS患者合并神经功能致残性损伤的诊断价值。结果脑卒中组患者血清lncRNA MEG3、EZH2表达水平均高于对照组(t=11.817、11.542,P<0.05)。Spearman相关分析显示,AIS患者血清lncRNA MEG3、EZH2水平与NIHSS评分均呈正相关(r=0.540、0.603,P<0.01)。致残性损伤组体质量指数、吸烟史占比、甘油三酯、总胆固醇、同型半胱氨酸、发病-到院时间(ODT)、lncRNAMEG3、EZH2水平均高于非致残性损伤组(t/χ2=2.103、5.050、9.121、5.585、2.276、5.448、4.638、4.682,P<0.05)。多因素Logistic回归分析显示,甘油三酯、总胆固醇、lncRNA MEG3、EZH2以及ODT均是AIS患者合并神经功能致残性损伤的影响因素(OR=4.853、4.277、2.674、3.052、3.901,P<0.05)。lncRNA MEG3、EZH2联合诊断AIS患者合并神经功能致残性损伤的曲线下面积(AUC)大于lncRNAMEG3以及EZH2单独诊断的AUC(Z=2.626、2.954,P<0.01),敏感度、特异度分别为94.74%、82.15%。结论AIS患者血清lncRNA MEG3、EZH2水平均上升,与AIS患者神经功能损伤程度均正相关,可用作AIS患者合并神经功能致残性损伤的诊断指标,且联合诊断效果更好。 展开更多

目的研究在转化生长因子β1(TGF-β1)诱导肺泡上皮细胞间质转化(EMT)进程中lncSIL的作用及其相关信号通路。方法采用Western blot法研究沉默lncSIL后对TGF-β1诱导EMT进程中细胞标志蛋白E-钙黏蛋白(E-cad)、α-平滑肌肌动蛋白(α-SMA)... 目的研究在转化生长因子β1(TGF-β1)诱导肺泡上皮细胞间质转化(EMT)进程中lncSIL的作用及其相关信号通路。方法采用Western blot法研究沉默lncSIL后对TGF-β1诱导EMT进程中细胞标志蛋白E-钙黏蛋白(E-cad)、α-平滑肌肌动蛋白(α-SMA)和Ⅰ型胶原蛋白(ColⅠ)表达的影响;通过RNA pulldown分析lncSIL相互作用蛋白,并检测过表达或沉默lncSIL后对其靶基因组蛋白赖氨酸N-甲基转移酶(EZH2)以及下游因子P21蛋白(P21)和细胞周期蛋白依赖性激酶6(CDK6)表达的影响,并结合流式细胞术分析lncSIL对细胞周期进程的作用。结果沉默lncSIL后,间质细胞标志蛋白α-SMA和Col I表达升高,肺泡上皮细胞标志蛋白E-cad表达下降;RNA pulldown实验结果显示EZH2是与lncSIL相互作用的靶蛋白,并且沉默lncSIL后EZH2表达升高,其下游基因P21表达下调,CDK6表达上调,同时S期细胞的数量显著升高;过表达lncSIL时,EZH2与CDK6表达下调,P21表达上调,同时S期细胞的数量明显降低。结论lncSIL通过负向调控EZH2/P21/CDK6信号通路抑制细胞周期进程进而抑制TGF-β1诱导的肺泡上皮细胞向间质转化。 展开更多

Methane coupling reaction was studied in the system of continuous flow reactor on Ni, MoO3, MnO2 catalysts and different ZSM-5 carriers. The per pass conversion of methane can be as high as 22%, the selectivity of ethylene can be as high as 23.8%, of acetylene 60.8%, of ethane 5.4% and of total C2 hydrocarbons was more than 90%. ZSM-5-25 was the better carrier and MnO2 was the better active component. The efficiency of energy was as high as 7.81%. 展开更多

Opioid-sparing effect of selective cyclooxygenase-2 inhibitors on surgical outcomes after open colorectal surgery within an enhanced recovery after surgery protocol 被引量:7

AIM: To evaluate the opioid-sparing effect of selective cyclooxygenase-2(COX-2) inhibitors on short-term surgical outcomes after open colorectal surgery.METHODS: Patients undergoing open colorectal resection within an enhanced recovery after surgery protocol from 2011 to 2015 were reviewed. Patients with combined general anesthesia and epidural anesthesia, and those with acute colonic obstruction or perforation were excluded. Patients receiving selective COX-2 inhibitor were compared with well-matched individuals without such a drug. Outcome measures included numeric pain score and morphine milligram equivalent(MME) consumption on postoperative day(POD) 1-3, gastrointestinal recovery(time to tolerate solid diet and time to defecate), complications and length of postoperative stay.RESULTS: There were 75 patients in each group. Pain score on POD 1-3 was not significantly different between two groups. However, MME consumption and MME consumption per kilogram body weight on POD 1-3 was significantly less in patients receiving a selective COX-2 inhibitor(P < 0.001). Median MME consumption per kilogram body weight on POD 1-3 was 0.09, 0.06 and nil, respectively in patients receiving a selective COX-2 inhibitor and 0.22, 0.25 and 0.07, respectively in the comparative group(P < 0.001), representing at least 59% opioidreduction. Patients prescribing a selective COX-2 inhibitor had a shorter median time to resumption of solid diet [1(IQR 1-2) d vs 2(IQR 2-3) d; P < 0.001] and time to first defecation [2(IQR 2-3) d vs 3(IQR 3-4) d; P < 0.001]. There was no significant difference in overall postoperative complications between two groups. However, median postoperative stay was significantly 1-d shorter in patients prescribing a selective COX-2 inhibitor [4(IQR 3-5) d vs 5(IQR 4-6) d; P < 0.001]. CONCLUSION: Perioperative administration of oral selective COX-2 inhibitors significantly decreased intravenous opioid consumption, shortened time to gastrointestinal recovery and reduced hospital stay after open colorectal surgery. 展开更多

Sequestration of CO2 in deep and unmineable coal seams is one of the attractive alternatives to reduce its atmospheric concentration. Injection of CO2 in coal seams may help in enhancing the recovery of coalbed methane. An experimental study has been carried out using coal samples from three different coal seams, to evaluate the enhanced gas recovery and sequestration potential of these coals. The coals were first saturated with methane and then by depressurization some of the adsorbed methane was desorbed. After partial desorption, CO2 was injected into the coals and subsequently they were depressurized again. Desorption of methane after the injections was studied, to investigate the ability of CO2 to displace and enhance the recovery of methane from the coals. The coals exhibited varying behavior of adsorption of CO2 and release of methane. For one coal, the release of methane was enhanced by injection of CO2, suggesting preferential adsorption of CO2 and desorption of methane. For the other two coals, CO2 injection did not produce incremental methane initially, as there was initial resistance to methane release. However with continued CO2 injection, most of the remaining methane was produced. The study suggested that preferential sorption behavior of coal and enhanced gas recovery pattern could not be generalized for all coals. 展开更多

四环素类抗生素(TCs)是一类广谱抗菌药物,被广泛应用于治疗人畜疾病且效果显著,但过度使用TCs造成其在动物性食品中的残留问题会对自然环境和人体健康造成严重的危害。因此,对食品环境中TCs的残留进行快速检测是十分必要的。迄今为止,对... 四环素类抗生素(TCs)是一类广谱抗菌药物,被广泛应用于治疗人畜疾病且效果显著,但过度使用TCs造成其在动物性食品中的残留问题会对自然环境和人体健康造成严重的危害。因此,对食品环境中TCs的残留进行快速检测是十分必要的。迄今为止,对TCs在酸性溶液中荧光检测相关的研究报道较少。在弱酸性条件下,TCs属于弱荧光物质,但与富有氨基的阳离子表面活性剂聚乙烯亚胺(PEI)结合时,在370 nm的激发下,在525 nm处产生较强的绿色荧光;当TCs与PEI和二价金属镁离子(Mg^(2+))共存时,TCs的荧光强度被进一步增强。本研究利用PEI和Mg^(2+)在弱酸性条件下对TCs的协同荧光增敏效应,构建了一种快速、简单、高灵敏检测残留TCs的荧光检测法。实验结果表明:在最优检测条件下,该体系对TCs的定量线性检测范围为5 nmol/L~2μmol/L,其中,美他环素(MTC)、四环素(TC)、土霉素(OTC)的相关系数R^(2)分别为0.939,0.997和0.999,检出限(LOD)分别为3.6,2.7和4.8 nmol/L,均远低于中国和欧盟的限量标准。本方法对MTC、TC、OTC表现出很高的特异性,与常见的其他抗生素均不产生干扰。在实际的牛奶检测中,该体系对TCs的加标回收率为91.64%~104.05%,相对标准偏差为1.20%~10.62%。本方法可适用于酸性体系中残留TCs的分析,为水体和食品中残留TCs检测提供了一种新的依据和思路。 展开更多

背景甲状腺自身抗体是诊断桥本甲状腺炎(HT)的标志,B淋巴细胞在HT的发病机制中发挥重要作用。Zeste同源物2增强子(EZH2)是一种表观遗传学蛋白,在淋巴细胞的发育与功能调控中扮演重要角色。目的本研究探讨EZH2在HT甲状腺组浆母细胞及浆... 背景甲状腺自身抗体是诊断桥本甲状腺炎(HT)的标志,B淋巴细胞在HT的发病机制中发挥重要作用。Zeste同源物2增强子(EZH2)是一种表观遗传学蛋白,在淋巴细胞的发育与功能调控中扮演重要角色。目的本研究探讨EZH2在HT甲状腺组浆母细胞及浆细胞中的表达,进一步探讨EZH2抑制剂在实验性自身免疫甲状腺炎(EAT)模型中的治疗作用。方法收集北京大学第一医院2010—2020年6例行甲状腺手术的患者,取肿瘤对侧的甲状腺组织(HT及正常甲状腺组织各3例),通过RNA-seq筛选B淋巴细胞相关基因的表达情况;收集16例HT患者的甲状腺组织和8例健康对照(HD)甲状腺组织,分别利用免疫组化及免疫荧光验证EZH2在HT甲状腺组织中B淋巴细胞中的表达;收集25例HT甲状腺细针穿刺液(FNA)、19例HT外周血以及12例健康人外周血样本应用流式细胞分析检测EZH2在浆母细胞及浆细胞中的表达改变。将15只7周龄NOD.H-2^(h4)小鼠EAT模型分为对照组(n=5)、EAT无注射(n=5)或注射EZH2抑制剂GSK126处理组(10 mg/kg,腹腔注射3次/周,n=5),8周后观察甲状腺炎症程度及甲状腺球蛋白抗体(TgAb)水平的改变。结果RNA-seq结果显示,相较于正常甲状腺组织,HT甲状腺组织中EZH2水平上调,一些与B淋巴细胞表型相关的基因例如CD19、CD27、CD38、CD52相应增加。免疫组化结果显示,16例HT甲状腺组织标本中EZH2免疫组化染色均可在生发中心(GC)见到阳性细胞,呈强阳性,8例正常甲状腺组织染色中未观察到阳性细胞。HT甲状腺组织中EZH2染色高表达在GC区,EZH2特异性地表达在CD_(19)^(+)B淋巴细胞中。流式细胞术检测结果显示HT FNA样本中CD_(19)^(+)B淋巴细胞、浆母细胞及浆细胞比例均高于HD外周血、HT外周血样本(P<0.01),HT FNA样本中EZH2在CD_(19)^(+)B淋巴细胞、浆母细胞中的阳性比例高于HT外周血(P<0.005)。小鼠实验中,EAT组甲状腺的淋巴细胞浸润较对照组增加。GSK126处理组炎症程度评分和TgAb水平高于对照组,低于EAT组(P<0.001)。结论EZH2在HT甲状腺组织CD_(19)^(+)B淋巴细胞中表达异常升高,可能促进了B淋巴细胞分化成浆细胞进而促进自身抗体生成破坏甲状腺,EZH2抑制剂可以减缓EAT模型甲状腺炎症程度。浆母细胞中EZH2表达增加可能参与了HT的发病机制,EZH2可能成为治疗HT的新靶点,相关机制需要进一步深入研究。 展开更多
