Half of the rats in each group received 1000 ppm aluminum in the diet. It was found that aluminum accumulated in the pituitary glands and testes when dietary copper levels were suboptimal. The ALP activity in testes was depressed by the added aluminum when the intake of zinc or copper was suboptimal. SDH, LDH, and LDH-X activities were inhibited and GRS and GGTP activities were elevated in rats fed either the suboptimal zinc or copper diet. However, the added aluminum in the diet reversed the changes to normal levels. The testosterone levels in plasma changed very little when the zinc or copper intake was suboptimal. An increase in plasma FSH was demonstrated in groups of both suboptimal zinc and copper intake. But the plasma LH was elevated only in the group receiving the suboptimal copper diet, and the added aluminum reversed plasma LH to control levels. A lower level of testosterone was demonstrated in the group given suboptimal copper with aluminum. It was concluded that dietary aluminum influenced the pituitary-testicular axis by interacting with certain essential trace metals, particularly zinc. 1990 Academic Press. Inc.展开更多

We previously developed radioimmunoassays (RIAs) for digitoxin and digoxin using antisera raised against digitoxin 3’-hemisuccinate-bovine serum albumin and digoxin 3’-hemisuccinate-bovine serum albumin conjugates, ...We previously developed radioimmunoassays (RIAs) for digitoxin and digoxin using antisera raised against digitoxin 3’-hemisuccinate-bovine serum albumin and digoxin 3’-hemisuccinate-bovine serum albumin conjugates, respectively. Very recently, we converted the RIA for digoxin to an enzyme immunoassay (EIA) system. Here, we aimed to convert the RIA for digitoxin to an EIA suitable for measuring serum digitoxin level in patients, using digitoxin 3’-hemisuccinate-β-D-galactosidase as an enzyme-labeled antigen. The developed EIA showed a quantification range of 1 to 70 ng/mL and exhibited high specificity for digitoxin, with low cross-reactivity to digitoxin metabolites. Compared with a commercial anti-digitoxin antiserum clinically used to monitor serum digitoxin level in patients, our antiserum showed much higher specificity for intact digitoxin. Intra- and inter-assay variations were less than 10.0% and 8.5%, respectively. Recovery was within the range of 93.7% - 107.5%. Mean digitoxin concentrations measured in serum samples (n = 26) from digitoxin-treated patients by EIA using our new antiserum and the commercial anti-digitoxin antiserum were 11.0 and 13.8 ng/mL, respectively. The present EIA, which is superior to RIA in terms of convenience and disposal of waste materials, is expected to be practically useful for clinical monitoring of intact digitoxin in serum.展开更多

It was found that the test values of 7 kinds of cancers with PHA or LCA were significantly higher than that of the normal (P<0.01); the values of 4 kinds of non-malignant diseases with PHA were not higher (P>0.05); the values of the postoperative cancer with PHA were obviously lower than that of the preoperative (P<0.05). The results showed that the serum glycoconjugates which can bind to PHA seemed related to the cancerous existence in human bodies. The significance of the findings was discussed.展开更多

Treatments were control (no CHM supplementation) and three formulations of CHM (CHM-A, CHM-B and CHM-C) that were arranged as completely randomized design. The calves were fed in indoor individual feeding barn with total mixed ration for 90 d. The CHM-A, CHM-B, and CHM-C was incorporated into the diet at the level of 20 g kg-1 DM (dry matter) of concentrate. The cattle were weighted at the beginning, at the end and every 15 d between to determine growth rate. Blood sample was taken every 30 d to analyze activities of total superoxide dismutase (T-SOD) and glutathione peroxidase (GSH-PX) and concentration of malondialdehyde (MDA). The cattle were slaughtered at the end of experiment and chymus samples from various section of the post-ruminal digestive tract were collected to analyze enzymes activities. All cattle had similar average daily gain, but cattle supplemented with CHM-A tended faster growth (P=0.08) compared to the control and other CHM treatments. Supplementation of CHM did not affect pH of chymus in any section of the post-ruminal digestive tract. Activities of pepsin, trypsin, chymotrypsin, and lipase in chymus samples were enhanced (P0.05) to various extents by supplementation of CHM-A, CHM-B, and CHM-C depending on the enzymes and the site of the digestive tract. Amylase activity was increased only by CHM-A and CHM-C from chymus sample collected at the ileum. Supplementation of CHM-A and CHM-B reduced (P0.05) concentration of MDA, but did not affect GSH-PX activity in the serum of cattle throughout the entire experimental period. Cattle supplemented with CHM-B also had higher (P0.05) T-SOD activity in the serum at day 30 and 60 as compared with the control and the other two CHM treatments. Chinese herbal mixtures supplemented at the levels of 20 g kg-1 DM of concentrate increased post-ruminal digestive enzymes activity and enhanced serum antioxidant status. Inclusion of CHM-A and CHM-B in the diet may be favourable for ruminant production.展开更多

The red tilapia were fed five diets supplemented with different levels of E. prolifera ultrafine powder as well as a control diet containing no E. prolifera for seven weeks(Diets 0–6 contained 0(control), 10, 20, 30, 40 and 50 g/kg of E. prolifera ultrafine powder, respectively). The results showed that diets supplemented with E. prolifera ultrafine powder generally improved growth, immunity and digestive enzyme activities of the red tilapia. In particular, the fish fed the diet incorporated 50 g/kg (5%) E. prolifera ultrafine powder(Diet 5) achieved the highest percentage weight gain, specific growth rate and the condition factor(increased by 15.4%, 8.0% and 5.7%, respectively when compared to the control). Feeding the diet also led to significantly increases( P <0.05) in serum nonspecific immune responses, including total superoxide dismutase, acid phosphatase, alkaline phosphatase, lysozyme activities and serum total protein(increased by 19.4%, 48.1%, 29.5%, 30.3% and 8.7%, respectively) as well as digestive enzyme activities of erepsin, gastric amylase, gastric lipase, pepsin, intestinal amylase and gastric lipase(increased by 15.7%, 33.3%, 16.3%, 21.3%, 52.3% and 28.2%, respectively) than those of the control. Based on these results, it is recommended that the inclusion level of E. prolifera ultrafine powder in the diet of the red tilapia should be 50 g/kg(or 5%).展开更多
BACKGROUND:Early recurrence of hepatocellular carcinoma(HCC)is associated with worse prognosis after liver resection This study aimed to investigate the prognostic value of com mon liver enzyme markers in HCC early...BACKGROUND:Early recurrence of hepatocellular carcinoma(HCC)is associated with worse prognosis after liver resection This study aimed to investigate the prognostic value of com mon liver enzyme markers in HCC early recurrence after cu rative hepatectomy and to establish a simple predictive model for HCC early recurrence.METHODS: A total of 200 patients who had undergone curative resection for HCC were retrospectively analyzed. The patients were divided into early recurrence (within 2 years) and non-early recurrence groups. Demographical characteristics, preopera- tive liver function parameters, surgical factors and tumor related factors of the patients were assessed by univariate analysis to identify potential significant predictors for early recurrence after resection of HCC. Parameters with statisti- cal significance were entered into a Cox proportional hazard model to find independent risk factors. Receiver operating characteristic analysis was done to determine optimal cut-off values and the number of combined factors in multi-factor predictive model. RESULTS: Of 13 potential risk factors for early recurrence identified by univariate analysis, high lactate dehydrogenase (LDH〉206 U/L, HR=1.711, P=0.006), high aspartate aminotransferase (AST)/alanine aminotransferase (ALT) ratio (AST/ ALT〉0.96, HR=1.769, P=0.006), elevated alpha-fetoprotein (AFP〉8.6 ng/mL, HR=2.079, P=0.007), small resection margin (〈1 cm, HR=2.354, P〈0.001) and advanced TNM stage (TNM III-IV, HR=2.164, P〈0.001) were independent risk factors for early recurrence of HCC shown by multivariate analysis. Patients with three or more concurrent independent risk factors had significantly higher risk for early recurrence than those with low risk factors. The sensitivity and specificity of this predictive model are 53.6% and 80.7%, respectively (area under curve=0.741, 95% CI 0.674-0.800, P〈0.0001). CONCLUSIONS: Preoperative common fiver enzyme markers, LDH and AST/ALT ratio, were independently associated with early recurrence of HCC. The combination of serum liver enzyme markers with AFP, resection margin and TNM stage bet- ter predicted early recurrence of HCC after curative resection in a simple multi-factor model.展开更多

Among the enzyme that produces is the catalase which is very useful in differentiating staphylococci from streptococci [1]. Catalase is nearly ubiquitous among some of organisms that can grow in the presence of oxygen (air). It promotes the conversion of hydrogen peroxide, a powerful and potentially harmful oxidizing agent, to water and molecular oxygen;so the major function of catalase within cells is to prevent the accumulation of toxic levels of hydrogen peroxide formed as a by-product of metabolic processes—primarily that of the electron transport pathway. Objectives: The main aim of this study is to prove that human WBCs can produce H2O2. This H2O2 when reacting with catalase producing S. aureus can easily be degraded to H2O + O2. Methodology: In this study a total of 40 subjects were included. Aliquots of 2.5 ml of venous blood were collected by venous puncture after disinfecting the site of collection with 70% alcohol and the collected blood was drawn into EDITA containers (20 subject) and anticoagulant free containers (other 20 subject), centrifugation for 5 minute at 1500 RPM. The separated sera and plasma were converted to new sterile eppendrof tubes and freezing until used (we leaved the eppendrof tubes that contained sera and plasma at room temperature before using it for DE freezing). Standard catalase producing S. aureus were used by taking 1 colony from Macconkey media by using applicator wooden stick, and inserted in eppendrof tube, then air bubbles would appear to indicate occurrence of the reactions. Results: According to this study, it was proved that WBCs in human plasma or serum can produce H2O2;this H2O2 was reacted with catalase enzyme produce from colony of S. aureus to produce air bubbles and water. There were no differences between using H2O2 or human plasma/serum that contains WBCs to detect and identify S. aureus by both techniques. Conclusion: Based on the results of this study, we can use WBCs that are found in human plasma or serum to identify catalase producing S. aureus.展开更多

These enzymes normally circulate within the bloodstream in definite concentration ranges. There is a lack of available information on the interrelationships between the enzymatic activities in pancreatic juice and blood serum in poultry. The study presented was aimed at the determination of possible correlation between the activities of the digestive enzymes in pancreatic juice or pancreatic tissue and in blood serum in chicken. The study was performed on Leghorn cockerels with chronically fistulated main pancreatic duct, and broiler chicks at different ages (14, 24, 28, and 35 days). Activities of all digestive enzymes in pancreatic juice in cockerels were found to grow in 1 hr after the feeding: amylase by 1.8 times, lipase 1.5 times, and proteases 1.3 times compared to basal level (after starving). Activity of trypsin in serum significantly increased by 67.4% in 1 hr after the feeding (P 0.001) while activities of amylase and lipase remained at their respective preprandial levels. The activities of pancreatic enzymes in the pancreas and blood serum in broilers at different ages were positively correlated: r = 0.54 for amylase, r = 0.96 for lipase, and r = 0.99 for trypsin. The strong positive correlation between tryptic activities in the pancreas or pancreatic juice and in blood serum in chicken can be a starting point for further research on the functions of circulatory trypsin which can include the regulation of exocrine pancreatic activity and other vital functions.展开更多

The study determined effect of chronic alcohol intake on serum enzymes (alanine aminotransferase (ALT), aspartate aminotransferase (AST) and γ-glutamyl transferase (GGT)) in HIV-infected patients on d4T/3TC/NVP treatment regimen in Uganda using the WHO alcohol use disorders’ identification test (AUDIT) tool and chronic alcohol use biomarkers (ALT, AST, GGT, AST/ALT ≥ 2.0 and mean corpuscular volume (MCV)). A case control study using repeated measure design with serial measurements model was used. Alcohol use biomarkers were used to standardize the gender differences in alcohol use. A total of 41 patients (21 alcohol group and 20 control group) were followed up for 9 months with blood sampling done at 3 month intervals. The serum enzymes’ levels were determined by using the Cobas Intergra 400 Plus analyzer system. The mean GGT levels were higher in chronic alcohol use group as compared to control group in both groups. The levels were above reference ranges during 6 month and three times higher during 9-month follow-up period for both chronic alcohol use self reporting WHO AUDIT tool and biomarkers’ groups. Generally, the mean AST, ALT and AST/ALT levels were slightly higher in alcohol use group as compared to control group and were slightly higher in both groups as compared to reference ranges during the 9 month follow-up period. Chronic alcohol consumption by HIV-infected patients on d4T/3TC/NVP drug regimen increased GGT and AST/ALT serum enzyme levels and hence was used as chronic alcohol use biomarkers.展开更多

Clinical signs are unspecific. Routine biochemical tests don't have specificity and it is better defined through biopsy. The objective was evaluated the biochemical hepatic profile of red footed tortoises (Chelonoidis carbonaria) with the histopathological findings of lipidosis by laparoscopic biopsy. Samples of blood were collected for biochemistry of thirty-six animals from two different groups, twenty animals from a zoo and sixteen from a breeder. The animals were submitted to liver biopsy by videolaparoscopy. Three fragments were collected, two of them were processed by histology and one went to electron microscopy. Nineteen tortoises from the zoo had lipidosis in laparoscopy and macrovesicular degeneration. Fifteen tortoises from the breeder had microvesicular degeneration. The diagnosis of steatosis was confirmed by electron microscopy. There was no statistical difference of aspartate aminotransferase levels between the two groups; however there was significant difference of triglycerides levels. The enzymatic activity of transaminases is not related to the histological grading of lipidosis in tortoises, however triglycerides levels are higher in animals with more severe gradation.展开更多

Anti-erlotinib serum was obtained by immunizing mice with an antigen conjugated with bovine serum albumin and 3,4-bis(2-methoxyethoxy)benzoic acid using the N-succinimidyl ester method. Enzyme labeling of erlotinib with horseradish peroxidase was similarly performed using 3,4-bis(2-methoxyethoxy)benzoic acid. A simple competitive ELISA for erlotinib was developed using the principle of direct competition between erlotinib and the enzyme marker for anti-erlotinib antibody, which had been immobilized on the plastic surface of a microtiter plate. Serum erlotinib concentrations lower than 40 ng/mL were reproducibly measurable using the ELISA. This ELISA was specific to erlotinib and showed very slight cross-reactivity(6.7%) with a major metabolite, O-desmethyl erlotinib. Using this assay, drug levels were easily measured in the blood of mice after oral administration of erlotinib at a single dose of 30 mg/kg. ELISA should be used as a valuable tool for therapeutic drug monitoring and in pharmacokinetic studies of erlotinib.展开更多

Methods: A total of 48 patients with Parkinson's disease who were treated in this hospital between December 2014 and October 2017 were selected as Parkinson's disease group, and 50 healthy volunteers who received physical examination in this hospital during the same period were selected as normal control group. The differences in serum contents of Aβ1-42, inflammatory factors and receptors as well as antioxidant enzymes were compared between the two groups, and Pearson test was used to assess the correlation between serum Aβ1-42 content and illness in patients with Parkinson's disease. Results: Serum Aβ1-42 content of Parkinson's disease group was lower than that of normal control group;serum inflammatory factors and receptors IL-2, sIL-2R, IL-6 and sIL-6R contents were higher than those of normal control group;serum antioxidant enzymes SOD, GSH-Px, CAT and TPX contents were lower than those of control group. Pearson test showed that serum Aβ1-42 content of patients with Parkinson's disease was directly correlated with the contents of inflammatory factors and receptors as well as antioxidant enzymes. Conclusions: Serum Aβ1-42 content decreases in patient with Parkinson's disease, and the specific content is directly correlated with the condition of Parkinson's disease, and can be used as an important auxiliary indicator for diagnosis and judgment of Parkinson's disease.展开更多

[Objectives]This study was conducted to investigate the effects of probiotics and enzyme preparation on cholesterol,triglyceride and serum alanine aminotransferase (ALT) and peroxidase (POD) in egg yolks.[Methods]Fort...[Objectives]This study was conducted to investigate the effects of probiotics and enzyme preparation on cholesterol,triglyceride and serum alanine aminotransferase (ALT) and peroxidase (POD) in egg yolks.[Methods]Forty eight 450-day-old Hyline brown hens were randomly divided into 4 groups with 3 replicates in each group and 4 individuals in each replicate.Group 1 as the control check (CK) group was fed the basal diet;group 2 was fed the basal diet with the addition of 150 g/t of probiotics;group 3 was the basal diet +200 g/t lipase group;and group 4 was fed the basal diet with the addition of 150 g/t of probiotics and 200 g/t of lipase.The preliminary trial period was 10 d,and the trial period was 30 d.[Results]The contents of cholesterol and triglyceride in the egg yolks of the test groups supplemented with the probiotics and enzyme preparation were higher than those of the CK.On the 14^th day of treatment,the cholesterol contents in the egg yolks of group 2 and group 4 were significantly higher than that of group 1 ( P <0.05).On the 28^th day of treatment,the contents of cholesterol and triglyceride in the egg yolks of the three test groups were significantly higher than those in group 1 ( P <0.05).There were no significant differences in serum ALT and POD between various groups ( P >0.05).[Conclusions]The addition of probiotics and enzyme preparations to the feed of laying hens can increase the contents of cholesterol and triglyceride in egg yolks,without significantly affecting serum ALT and POD.展开更多

Alkaline phosphatase was found to be increased up to 119.9% at the 6<sup>th</sup> hr of the toxin injection in comparison to control. This elevation may be due to cytolysis. Maximum increase i.e., 153.33% level of glutamate pyruvate transaminase (GPT) was found at 6 hrs of 40% of 24-h LD<sub>50</sub> treatment while it was found to be 151.1% at 6 hrs of 24 hr 80% of LD<sub>50</sub>, venom injection. A significant elevation was observed in LDH activity in serum, liver, and muscles, while the activity of AchE was decreased in serum, liver, and gastrocnemius muscles of albino mice after injecting the sub-lethal dose of Ropalidia marginata venom. This increase in the activity of LDH produces liver damage, massive disintegration and necrosis of hepatic cells. This elevation in LDH level led to a significant increase in the glucose catabolism and elevated oxidative stress in muscle and liver cells. It also displays insufficient oxygen supply and consequently leads to cell death. In experimental animals, venom toxin treatment decreased AchE level, and animals showed muscular paralysis. When mice were treated with 40% and 80% of 24-h LD<sub>50</sub> of purified venom caused a significant (p < 0.05) elevation in the level of ACP, GOT, GPT, and LDH while the reduction in ALP and AChE level. Present study will be useful in the development of prototypes for study of pharmacological and therapeutic effects of various venom toxins. For this purpose structure activity relationship of enzyme and venom toxin, its due interaction to various metabolic enzymes and receptors must be explored.展开更多

文摘A chemiluminescence enzyme immunoassay based on magnetic microparticles (MmPs-CLEIA) was developed to evaluate serum a-fetoprotein (AFP) in parallel with traditional colorimetric enzyme-linked immunosorbent assay (ELISA).A systematic comparison between the MmPs-CLEIA and colorimetric ELISA concluded that the MPs-CLEIA exhibited fewer dosages of immunoreagents,less total assay time,and better linearity,recovery,precision,sensitivity and validity.AFP was detected in forty human serum samples by the proposed MPs-CLEIA and ELISA,and the results were compared with commercial electrochemiluminescence immunoassay (ECLIA) kit.The correlation coefficient between MPs-CLEIA and ELISA was obtained with R 2 0.6703;however,the correlation between MPs-CLEIA and ECLIA (R 2 0.9582) was obviously better than that between colorimetric ELISA and ECLIA (R 2 0.6866).

文摘Rats were exposed to1 or 10 μg/mL bisphenol A (BPA) in water during pregnancy and lactation. Offspring rats were given normal water and a standard diet from weaning to postnatal day (PND) 50. Perinatal exposure to BPA resulted in significantly increased body weight, visceral adipose tissue, abnormal serum lipids, and lower adiponectin (ADP) levels in both female and male offspring rats. Liver adipose triglyceride lipase (Atgl) mRNA levels and ADP protein in visceral adipose tissue were significantly decreased in BPA-exposed offspring rats. In both female or male offspring rats, obesity and dyslipidemia induced by perinatal exposure to BPA were associated with down regulation of Atgl mRNA in liver and ADP protein in visceral adipose tissue.
文摘Sprague-Dawley male rats were divided into groups and maintained on a semipurified diet containing either 5 or 40 ppm of zinc or 2 or 8 ppm of copper. Half of the rats in each group received 1000 ppm aluminum in the diet. It was found that aluminum accumulated in the pituitary glands and testes when dietary copper levels were suboptimal. The ALP activity in testes was depressed by the added aluminum when the intake of zinc or copper was suboptimal. SDH, LDH, and LDH-X activities were inhibited and GRS and GGTP activities were elevated in rats fed either the suboptimal zinc or copper diet. However, the added aluminum in the diet reversed the changes to normal levels. The testosterone levels in plasma changed very little when the zinc or copper intake was suboptimal. An increase in plasma FSH was demonstrated in groups of both suboptimal zinc and copper intake. But the plasma LH was elevated only in the group receiving the suboptimal copper diet, and the added aluminum reversed plasma LH to control levels. A lower level of testosterone was demonstrated in the group given suboptimal copper with aluminum. It was concluded that dietary aluminum influenced the pituitary-testicular axis by interacting with certain essential trace metals, particularly zinc. 1990 Academic Press. Inc.

文摘We previously developed radioimmunoassays (RIAs) for digitoxin and digoxin using antisera raised against digitoxin 3’-hemisuccinate-bovine serum albumin and digoxin 3’-hemisuccinate-bovine serum albumin conjugates, respectively. Very recently, we converted the RIA for digoxin to an enzyme immunoassay (EIA) system. Here, we aimed to convert the RIA for digitoxin to an EIA suitable for measuring serum digitoxin level in patients, using digitoxin 3’-hemisuccinate-β-D-galactosidase as an enzyme-labeled antigen. The developed EIA showed a quantification range of 1 to 70 ng/mL and exhibited high specificity for digitoxin, with low cross-reactivity to digitoxin metabolites. Compared with a commercial anti-digitoxin antiserum clinically used to monitor serum digitoxin level in patients, our antiserum showed much higher specificity for intact digitoxin. Intra- and inter-assay variations were less than 10.0% and 8.5%, respectively. Recovery was within the range of 93.7% - 107.5%. Mean digitoxin concentrations measured in serum samples (n = 26) from digitoxin-treated patients by EIA using our new antiserum and the commercial anti-digitoxin antiserum were 11.0 and 13.8 ng/mL, respectively. The present EIA, which is superior to RIA in terms of convenience and disposal of waste materials, is expected to be practically useful for clinical monitoring of intact digitoxin in serum.
文摘After primary analyses on the serum glycocon-jugates of lung cancer and normal individul using the enzyme-linked lectin assay (ELLA) with 12 kinds of lectins, PHA and LCA were selected to further study in the sera of 8 kinds of cancers, 4 kinds of non-malignant disease and 1 kind of postoperative cancer. It was found that the test values of 7 kinds of cancers with PHA or LCA were significantly higher than that of the normal (P<0.01); the values of 4 kinds of non-malignant diseases with PHA were not higher (P>0.05); the values of the postoperative cancer with PHA were obviously lower than that of the preoperative (P<0.05). The results showed that the serum glycoconjugates which can bind to PHA seemed related to the cancerous existence in human bodies. The significance of the findings was discussed.

文摘Sixteen Limousin×Luxi crossbred beef cattle [(199.4±8.67) kg] were used to evaluate the effects of Chinese herbal mixtures (CHM) on growth, activities of post-ruminal digestive enzymes and serum antioxidant status of beef cattle. Treatments were control (no CHM supplementation) and three formulations of CHM (CHM-A, CHM-B and CHM-C) that were arranged as completely randomized design. The calves were fed in indoor individual feeding barn with total mixed ration for 90 d. The CHM-A, CHM-B, and CHM-C was incorporated into the diet at the level of 20 g kg-1 DM (dry matter) of concentrate. The cattle were weighted at the beginning, at the end and every 15 d between to determine growth rate. Blood sample was taken every 30 d to analyze activities of total superoxide dismutase (T-SOD) and glutathione peroxidase (GSH-PX) and concentration of malondialdehyde (MDA). The cattle were slaughtered at the end of experiment and chymus samples from various section of the post-ruminal digestive tract were collected to analyze enzymes activities. All cattle had similar average daily gain, but cattle supplemented with CHM-A tended faster growth (P=0.08) compared to the control and other CHM treatments. Supplementation of CHM did not affect pH of chymus in any section of the post-ruminal digestive tract. Activities of pepsin, trypsin, chymotrypsin, and lipase in chymus samples were enhanced (P0.05) to various extents by supplementation of CHM-A, CHM-B, and CHM-C depending on the enzymes and the site of the digestive tract. Amylase activity was increased only by CHM-A and CHM-C from chymus sample collected at the ileum. Supplementation of CHM-A and CHM-B reduced (P0.05) concentration of MDA, but did not affect GSH-PX activity in the serum of cattle throughout the entire experimental period. Cattle supplemented with CHM-B also had higher (P0.05) T-SOD activity in the serum at day 30 and 60 as compared with the control and the other two CHM treatments. Chinese herbal mixtures supplemented at the levels of 20 g kg-1 DM of concentrate increased post-ruminal digestive enzymes activity and enhanced serum antioxidant status. Inclusion of CHM-A and CHM-B in the diet may be favourable for ruminant production.

文摘The present study evaluated ef fects of ultrafine powder of the green macroalgae E nteromopha prolifera as dietary supplement on growth performance, digestive enzyme activities and serum nonspecific immune responses of the red tilapia(O reochromis mossambicus × Oreochromis niloticus). The red tilapia were fed five diets supplemented with different levels of E. prolifera ultrafine powder as well as a control diet containing no E. prolifera for seven weeks(Diets 0–6 contained 0(control), 10, 20, 30, 40 and 50 g/kg of E. prolifera ultrafine powder, respectively). The results showed that diets supplemented with E. prolifera ultrafine powder generally improved growth, immunity and digestive enzyme activities of the red tilapia. In particular, the fish fed the diet incorporated 50 g/kg (5%) E. prolifera ultrafine powder(Diet 5) achieved the highest percentage weight gain, specific growth rate and the condition factor(increased by 15.4%, 8.0% and 5.7%, respectively when compared to the control). Feeding the diet also led to significantly increases( P <0.05) in serum nonspecific immune responses, including total superoxide dismutase, acid phosphatase, alkaline phosphatase, lysozyme activities and serum total protein(increased by 19.4%, 48.1%, 29.5%, 30.3% and 8.7%, respectively) as well as digestive enzyme activities of erepsin, gastric amylase, gastric lipase, pepsin, intestinal amylase and gastric lipase(increased by 15.7%, 33.3%, 16.3%, 21.3%, 52.3% and 28.2%, respectively) than those of the control. Based on these results, it is recommended that the inclusion level of E. prolifera ultrafine powder in the diet of the red tilapia should be 50 g/kg(or 5%).

文摘BACKGROUND:Early recurrence of hepatocellular carcinoma(HCC)is associated with worse prognosis after liver resection This study aimed to investigate the prognostic value of com mon liver enzyme markers in HCC early recurrence after cu rative hepatectomy and to establish a simple predictive model for HCC early recurrence.METHODS: A total of 200 patients who had undergone curative resection for HCC were retrospectively analyzed. The patients were divided into early recurrence (within 2 years) and non-early recurrence groups. Demographical characteristics, preopera- tive liver function parameters, surgical factors and tumor related factors of the patients were assessed by univariate analysis to identify potential significant predictors for early recurrence after resection of HCC. Parameters with statisti- cal significance were entered into a Cox proportional hazard model to find independent risk factors. Receiver operating characteristic analysis was done to determine optimal cut-off values and the number of combined factors in multi-factor predictive model. RESULTS: Of 13 potential risk factors for early recurrence identified by univariate analysis, high lactate dehydrogenase (LDH〉206 U/L, HR=1.711, P=0.006), high aspartate aminotransferase (AST)/alanine aminotransferase (ALT) ratio (AST/ ALT〉0.96, HR=1.769, P=0.006), elevated alpha-fetoprotein (AFP〉8.6 ng/mL, HR=2.079, P=0.007), small resection margin (〈1 cm, HR=2.354, P〈0.001) and advanced TNM stage (TNM III-IV, HR=2.164, P〈0.001) were independent risk factors for early recurrence of HCC shown by multivariate analysis. Patients with three or more concurrent independent risk factors had significantly higher risk for early recurrence than those with low risk factors. The sensitivity and specificity of this predictive model are 53.6% and 80.7%, respectively (area under curve=0.741, 95% CI 0.674-0.800, P〈0.0001). CONCLUSIONS: Preoperative common fiver enzyme markers, LDH and AST/ALT ratio, were independently associated with early recurrence of HCC. The combination of serum liver enzyme markers with AFP, resection margin and TNM stage bet- ter predicted early recurrence of HCC after curative resection in a simple multi-factor model.
文摘Background: Staphylococcus aureus is one of the most virulent gram positive bacteria. It produces a lot of toxins and enzymes, most of which are virulent factors. Among the enzyme that produces is the catalase which is very useful in differentiating staphylococci from streptococci [1]. Catalase is nearly ubiquitous among some of organisms that can grow in the presence of oxygen (air). It promotes the conversion of hydrogen peroxide, a powerful and potentially harmful oxidizing agent, to water and molecular oxygen;so the major function of catalase within cells is to prevent the accumulation of toxic levels of hydrogen peroxide formed as a by-product of metabolic processes—primarily that of the electron transport pathway. Objectives: The main aim of this study is to prove that human WBCs can produce H2O2. This H2O2 when reacting with catalase producing S. aureus can easily be degraded to H2O + O2. Methodology: In this study a total of 40 subjects were included. Aliquots of 2.5 ml of venous blood were collected by venous puncture after disinfecting the site of collection with 70% alcohol and the collected blood was drawn into EDITA containers (20 subject) and anticoagulant free containers (other 20 subject), centrifugation for 5 minute at 1500 RPM. The separated sera and plasma were converted to new sterile eppendrof tubes and freezing until used (we leaved the eppendrof tubes that contained sera and plasma at room temperature before using it for DE freezing). Standard catalase producing S. aureus were used by taking 1 colony from Macconkey media by using applicator wooden stick, and inserted in eppendrof tube, then air bubbles would appear to indicate occurrence of the reactions. Results: According to this study, it was proved that WBCs in human plasma or serum can produce H2O2;this H2O2 was reacted with catalase enzyme produce from colony of S. aureus to produce air bubbles and water. There were no differences between using H2O2 or human plasma/serum that contains WBCs to detect and identify S. aureus by both techniques. Conclusion: Based on the results of this study, we can use WBCs that are found in human plasma or serum to identify catalase producing S. aureus.
文摘It is presently an established fact that pancreatic enzymes (amylase, lipase, and trypsin) can enter the bloodstream as well as the intestine. These enzymes normally circulate within the bloodstream in definite concentration ranges. There is a lack of available information on the interrelationships between the enzymatic activities in pancreatic juice and blood serum in poultry. The study presented was aimed at the determination of possible correlation between the activities of the digestive enzymes in pancreatic juice or pancreatic tissue and in blood serum in chicken. The study was performed on Leghorn cockerels with chronically fistulated main pancreatic duct, and broiler chicks at different ages (14, 24, 28, and 35 days). Activities of all digestive enzymes in pancreatic juice in cockerels were found to grow in 1 hr after the feeding: amylase by 1.8 times, lipase 1.5 times, and proteases 1.3 times compared to basal level (after starving). Activity of trypsin in serum significantly increased by 67.4% in 1 hr after the feeding (P 0.001) while activities of amylase and lipase remained at their respective preprandial levels. The activities of pancreatic enzymes in the pancreas and blood serum in broilers at different ages were positively correlated: r = 0.54 for amylase, r = 0.96 for lipase, and r = 0.99 for trypsin. The strong positive correlation between tryptic activities in the pancreas or pancreatic juice and in blood serum in chicken can be a starting point for further research on the functions of circulatory trypsin which can include the regulation of exocrine pancreatic activity and other vital functions.
文摘Chronic alcohol use is a common problem globally among the HIV-infected patients on ARV treatment regimens, leading to severe liver damage and increase in serum enzymes. The study determined effect of chronic alcohol intake on serum enzymes (alanine aminotransferase (ALT), aspartate aminotransferase (AST) and γ-glutamyl transferase (GGT)) in HIV-infected patients on d4T/3TC/NVP treatment regimen in Uganda using the WHO alcohol use disorders’ identification test (AUDIT) tool and chronic alcohol use biomarkers (ALT, AST, GGT, AST/ALT ≥ 2.0 and mean corpuscular volume (MCV)). A case control study using repeated measure design with serial measurements model was used. Alcohol use biomarkers were used to standardize the gender differences in alcohol use. A total of 41 patients (21 alcohol group and 20 control group) were followed up for 9 months with blood sampling done at 3 month intervals. The serum enzymes’ levels were determined by using the Cobas Intergra 400 Plus analyzer system. The mean GGT levels were higher in chronic alcohol use group as compared to control group in both groups. The levels were above reference ranges during 6 month and three times higher during 9-month follow-up period for both chronic alcohol use self reporting WHO AUDIT tool and biomarkers’ groups. Generally, the mean AST, ALT and AST/ALT levels were slightly higher in alcohol use group as compared to control group and were slightly higher in both groups as compared to reference ranges during the 9 month follow-up period. Chronic alcohol consumption by HIV-infected patients on d4T/3TC/NVP drug regimen increased GGT and AST/ALT serum enzyme levels and hence was used as chronic alcohol use biomarkers.
文摘Hepatic lipidosis is a common lesion in chelonians and may be related with vitellogenesis, hyperparathyroidism, follicular stasis, fatty diets or lacking nutrients, sedentary life and obesity, bacterial toxins in liver and starvation. Clinical signs are unspecific. Routine biochemical tests don't have specificity and it is better defined through biopsy. The objective was evaluated the biochemical hepatic profile of red footed tortoises (Chelonoidis carbonaria) with the histopathological findings of lipidosis by laparoscopic biopsy. Samples of blood were collected for biochemistry of thirty-six animals from two different groups, twenty animals from a zoo and sixteen from a breeder. The animals were submitted to liver biopsy by videolaparoscopy. Three fragments were collected, two of them were processed by histology and one went to electron microscopy. Nineteen tortoises from the zoo had lipidosis in laparoscopy and macrovesicular degeneration. Fifteen tortoises from the breeder had microvesicular degeneration. The diagnosis of steatosis was confirmed by electron microscopy. There was no statistical difference of aspartate aminotransferase levels between the two groups; however there was significant difference of triglycerides levels. The enzymatic activity of transaminases is not related to the histological grading of lipidosis in tortoises, however triglycerides levels are higher in animals with more severe gradation.
文摘A selective and sensitive competitive enzyme-linked immunosorbent assay(ELISA) method was developed and validated for the quantification of erlotinib in 50 mL of samples of human serum. Anti-erlotinib serum was obtained by immunizing mice with an antigen conjugated with bovine serum albumin and 3,4-bis(2-methoxyethoxy)benzoic acid using the N-succinimidyl ester method. Enzyme labeling of erlotinib with horseradish peroxidase was similarly performed using 3,4-bis(2-methoxyethoxy)benzoic acid. A simple competitive ELISA for erlotinib was developed using the principle of direct competition between erlotinib and the enzyme marker for anti-erlotinib antibody, which had been immobilized on the plastic surface of a microtiter plate. Serum erlotinib concentrations lower than 40 ng/mL were reproducibly measurable using the ELISA. This ELISA was specific to erlotinib and showed very slight cross-reactivity(6.7%) with a major metabolite, O-desmethyl erlotinib. Using this assay, drug levels were easily measured in the blood of mice after oral administration of erlotinib at a single dose of 30 mg/kg. ELISA should be used as a valuable tool for therapeutic drug monitoring and in pharmacokinetic studies of erlotinib.
文摘Objective: To investigate the correlation of serum β-amyloid 1-42 (Aβ1-42) content with inflammatory factors and receptors as well as antioxidant enzymes in patients with Parkinson's Disease. Methods: A total of 48 patients with Parkinson's disease who were treated in this hospital between December 2014 and October 2017 were selected as Parkinson's disease group, and 50 healthy volunteers who received physical examination in this hospital during the same period were selected as normal control group. The differences in serum contents of Aβ1-42, inflammatory factors and receptors as well as antioxidant enzymes were compared between the two groups, and Pearson test was used to assess the correlation between serum Aβ1-42 content and illness in patients with Parkinson's disease. Results: Serum Aβ1-42 content of Parkinson's disease group was lower than that of normal control group;serum inflammatory factors and receptors IL-2, sIL-2R, IL-6 and sIL-6R contents were higher than those of normal control group;serum antioxidant enzymes SOD, GSH-Px, CAT and TPX contents were lower than those of control group. Pearson test showed that serum Aβ1-42 content of patients with Parkinson's disease was directly correlated with the contents of inflammatory factors and receptors as well as antioxidant enzymes. Conclusions: Serum Aβ1-42 content decreases in patient with Parkinson's disease, and the specific content is directly correlated with the condition of Parkinson's disease, and can be used as an important auxiliary indicator for diagnosis and judgment of Parkinson's disease.
文摘Objective To explore the effect of Daicong solu tion(DCS)on aged rats dementia model.Method The experiment used22-month old rats whose basal nuclei was destroyed by bilateral electrolytic method as model for aged rats dem entia.They were treated with DCS for a month.Then the level of superox ide dismutase(SOD),lipid peroxide(LPO),monoamine oxidase(MAO)and cholinesterase(CHE)in the serum was measured.Result Compared with the control,the activ ity of SOD increased markedly(P <0.01),the content of LOP and activity MAO and CHE were decreased(P <0.01).Conclusion DCS is effective in treating the aged dementia.
文摘In this study,the effects of three different concentrations(0.5,0.25 and 0.125μl/L)of dichlorvos solution on phenoloxidase(PO)activity,hemolysin activity,peroxidase(POD)activity and antibacterial activity in the serum of Macrobrachium nipponense during four days were investigated.The results indicated that phenoloxidase activity,hemolysin activity,peroxidase activity and antibacterial activity in the serum of Macrobrachium nipponense were improved under the stress of dichlorvos during a short time.With the extension of stress duration and increase of dichlorvos concentration,the activities of various immunological indices were inhibited due to the cumulative effect of dichlorvos in vivo;overall,the reduction increased gradually with the extension of stress duration.

文摘[Objectives]This study was conducted to investigate the effects of probiotics and enzyme preparation on cholesterol,triglyceride and serum alanine aminotransferase (ALT) and peroxidase (POD) in egg yolks.[Methods]Forty eight 450-day-old Hyline brown hens were randomly divided into 4 groups with 3 replicates in each group and 4 individuals in each replicate.Group 1 as the control check (CK) group was fed the basal diet;group 2 was fed the basal diet with the addition of 150 g/t of probiotics;group 3 was the basal diet +200 g/t lipase group;and group 4 was fed the basal diet with the addition of 150 g/t of probiotics and 200 g/t of lipase.The preliminary trial period was 10 d,and the trial period was 30 d.[Results]The contents of cholesterol and triglyceride in the egg yolks of the test groups supplemented with the probiotics and enzyme preparation were higher than those of the CK.On the 14^th day of treatment,the cholesterol contents in the egg yolks of group 2 and group 4 were significantly higher than that of group 1 ( P <0.05).On the 28^th day of treatment,the contents of cholesterol and triglyceride in the egg yolks of the three test groups were significantly higher than those in group 1 ( P <0.05).There were no significant differences in serum ALT and POD between various groups ( P >0.05).[Conclusions]The addition of probiotics and enzyme preparations to the feed of laying hens can increase the contents of cholesterol and triglyceride in egg yolks,without significantly affecting serum ALT and POD.

文摘In the present, investigation effects of sub-lethal dose of purified paper wasp Ropalidia marginata venom toxins were evaluated on important metabolic enzymes i.e. ALP ACP, GPT, GOT, LDH, and AchE enzyme activity in serum, liver, and gastrocnemius muscles of albino mice. Alkaline phosphatase was found to be increased up to 119.9% at the 6<sup>th</sup> hr of the toxin injection in comparison to control. This elevation may be due to cytolysis. Maximum increase i.e., 153.33% level of glutamate pyruvate transaminase (GPT) was found at 6 hrs of 40% of 24-h LD<sub>50</sub> treatment while it was found to be 151.1% at 6 hrs of 24 hr 80% of LD<sub>50</sub>, venom injection. A significant elevation was observed in LDH activity in serum, liver, and muscles, while the activity of AchE was decreased in serum, liver, and gastrocnemius muscles of albino mice after injecting the sub-lethal dose of Ropalidia marginata venom. This increase in the activity of LDH produces liver damage, massive disintegration and necrosis of hepatic cells. This elevation in LDH level led to a significant increase in the glucose catabolism and elevated oxidative stress in muscle and liver cells. It also displays insufficient oxygen supply and consequently leads to cell death. In experimental animals, venom toxin treatment decreased AchE level, and animals showed muscular paralysis. When mice were treated with 40% and 80% of 24-h LD<sub>50</sub> of purified venom caused a significant (p < 0.05) elevation in the level of ACP, GOT, GPT, and LDH while the reduction in ALP and AChE level. Present study will be useful in the development of prototypes for study of pharmacological and therapeutic effects of various venom toxins. For this purpose structure activity relationship of enzyme and venom toxin, its due interaction to various metabolic enzymes and receptors must be explored.
