As intracellular fatty acid(FA) carriers,FA-binding proteins(FABPs) widely participate in the absorption,transport,and metabolism of FAs.It is a key protein in insect lipid metabolism and plays an important role in various physiological activities of insects.An FABP gene(HvFABP) was cloned from the transcriptional library of Heortia vitessoides Moore(Lepidoptera:Crambidae),and its expression patterns were determined using reverse transcription quantitative PCR(RTqPCR).Stage-and tissue-specific expression profiles indicated that HvFABP highly expressed from prepupal to adult stages and in larval midgut and adult wings.HvFABP expression may be induced through starvation,mRNA expression was downregulated at 24 and 48 h and upregulated at 72 h after starvation.Furthermore,20-hydroxyecdysone can induce the upregulation of its expression.RNA interference-mediated silencing of Hv FABP significantly inhibited HvFABP expression,resulting in delayed development,abnormal molting or lethal phenotypes,and a significantly reduced survival rate.These results indicate that HvFABP plays a key role in the molting of H.vitessoides. 展开更多

[ Result] The intron 3 of porcine H-FABP gene was amplified successfully. Its whole sequence was 1 350 bp in length and had been submitted to GenBank (Accession no. : DQ 002993). Role of Heart-Type Fatty Acid Binding Protein in Early Detection of Acute Myocardial Infarction in Comparison with cTnI, CK-MB and Myoglobin 被引量:33

Heart fatty acid-binding protein (H-FABP) is supposed to be the most sensitive biomarker of early acute myocardial infarction (AMI). To evaluate the diagnostic value of H-FABP for AMI in the early stage, the plasma levels of H-FABP were measured by sandwich ELISA in 93 patients with suspected AMI at admission within 6 h after onset of chest pain and 69 normal healthy subjects. The plasma concentrations of cardiac troponin-I (cTnI), creatine kinase-MB (CK-MB) and myoglobin (Mb) were assayed at the same time by using corpuscle chemiluminescence for those patients. The patients were classified as AMI group (n=32) and non-AMI group (n=61) retrospectively. The diagnostic validity was evaluated in terms of sensitivity, specificity and receiver operating characteristic (ROC) curve analysis. The results showed the cutoff value of H-FABP for AMI was 16.8 ng/ml, and its diagnostic sensitivity for AMI was 64.29 % within 3 h and 84.38 % within 6 h after onset of chest pain, and the diagnostic specificity for non-AMI was 100 % within 3 h and 91.8 % within 6 h. H-FABP had higher sensitivity than that of cTnI and CK-MB at all time points (P<0.05), whereas there was no significant difference in specificity among the four markers. But the area under the ROC curve of H-FABP was significantly greater than that of cTnI, CK-MB and Mb within 3 h. These results revealed that H-FABP possessed high diagnostic sensitivity and specificity for AMI in early stage, especially within 3 h after onset of persistent angina pectoris. In conclusion, H-FABP can be used as a sensitive marker for AMI in the early stage. 展开更多

Suberoylanilide hydroxamic acid(SAHA) is a histone deacetylase inhibitor that shows marked efficacy against many types of cancers and is approved to treat severe metastatic cutaneous T-cell lymphomas. In addition to its anticancer activity,SAHA has significant effects on the growth of many viruses. The effect of SAHA on replication of human cytomegalovirus(HCMV) has not, however, been investigated. Here, we showed that the replication of HCMV was significantly suppressed by treatment with SAHA at concentrations that did not show appreciable cytotoxicity. SAHA reduced transcription and protein levels of HCMV immediate early genes, showing that SAHA acts at an early stage in the viral life-cycle. RNAsequencing data mining showed that numerous pathways and molecules were affected by SAHA. Interferon-mediated immunity was one of the most relevant pathways in the RNA-sequencing data, and we confirmed that SAHA inhibits HCMV-induced IFN-mediated immune responses using quantitative Real-time PCR(qRT-PCR). Fatty acid-binding protein 4(FABP4), which plays a role in lipid metabolism, was identified by RNA-sequencing. We found that FABP4 expression was reduced by HCMV infection but increased by treatment with SAHA. We then showed that knockdown of FABP4 partially rescued the effect of SAHA on HCMV replication. Our data suggest that FABP4 contributes to the inhibitory effect of SAHA on HCMV replication. 展开更多

Objective:This study aims to investigate whether the heart fatty acid-binding protein(HFABP)in the cerebrospinal fluid(CSF)was a potential predictive biomarker for Alzheimer’s disease(AD).Methods:We evaluated the associations of CSF HFABP levels with core biomarkers,cognition,and brain structure in a sample population(n=302)from the Alzheimer’s Disease Neuroimaging Initiative(ADNI)database.Multiple linear regression and mixed-effects models were employed in the analyses.AD progression was assessed using the Kaplan–Meier survival analysis.Results:CSF HFABP was higher in patients with mild cognitive impairment and AD than the normal controls(p<0.001)and was particularly higher in those with amyloid-β(Aβ)pathologic features.CSF HFABP was associated with higher baseline CSF t-tau(p<0.001),CSF p-tau(p<0.001),and CSF t-tau/Aβ42 and CSF p-tau/Aβ42(p<0.01).Moreover,CSF HFABP was found to play predictive roles in hippocampal atrophy(p<0.01),cognitive decline(p<0.05),and the risk of AD(p<0.001).Conclusion:Our findings suggest that CSF HFABP can be a predictive biomarker of AD. 展开更多

目的:研究脊髓损伤后热休克蛋白27(HSP27)、表皮脂肪酸结合蛋白(FABPs)和金属蛋白酶组织抑制因子-1(TIMP-1)的基因表达及甲基强的松龙(MP)对其表达的影响。方法:SD大鼠30只。随机分为假手术组、单纯脊髓损伤组(损伤组)及脊髓损伤+大剂... 目的:研究脊髓损伤后热休克蛋白27(HSP27)、表皮脂肪酸结合蛋白(FABPs)和金属蛋白酶组织抑制因子-1(TIMP-1)的基因表达及甲基强的松龙(MP)对其表达的影响。方法:SD大鼠30只。随机分为假手术组、单纯脊髓损伤组(损伤组)及脊髓损伤+大剂量MP治疗组(MP组),每组10只。应用改良的Allen′s打击法致T8脊髓损伤。MP组大鼠伤后即刻从尾静脉内注射大剂量MP(30mg/kg)。损伤后24h切取损伤平面上下0.5cm的脊髓组织,进行RT-PCR反应,检测HSP27、FABPs和TIMP-1的基因表达。结果:术后24h假手术组HSP27、FABPs和TIMP-1的基因表达相对丰度分别为0.0643±0.0152、0.6413±0.1005和0.7091±0.0577;损伤组上述三个因子的表达升高,分别为1.0013±0.3861、1.2187±0.2851和0.8971±0.1092,与假手术组比较差异有显著性(P<0.01、P<0.05、P<0.05);MP组上述三个因子的表达继续升高,分别为1.2858±0.1384、1.7122±0.1766和1.2081±0.1093,与损伤组比较差异有显著性(P<0.05、P<0.01和P<0.01)。结论:脊髓损伤后,邻近损伤处的脊髓组织中HSP27、FABPs及TIMP-1的基因表达显著增高,大剂量MP能进一步促进三个因子表达,发挥组织保护作用。 展开更多

目的探讨细胞视黄酸结合蛋白(CRABP)Ⅱ、表皮型脂肪酸结合蛋白(E-FABP)及Ki-67在乳腺浸润性导管癌(IDC)组织中的表达及临床意义。方法收集2009年1月至2013年1月深圳市宝安区石岩人民医院收治的112例IDC患者的临床资料,患者均采用手术切... 目的探讨细胞视黄酸结合蛋白(CRABP)Ⅱ、表皮型脂肪酸结合蛋白(E-FABP)及Ki-67在乳腺浸润性导管癌(IDC)组织中的表达及临床意义。方法收集2009年1月至2013年1月深圳市宝安区石岩人民医院收治的112例IDC患者的临床资料,患者均采用手术切除,并对IDC组织进行免疫组织化学染色,观察IDC组织中CRABPⅡ、E-FABP及Ki-67的表达情况。结果 CRABPⅡ在Ki-67阴性表达组的阳性表达率高于Ki-67阳性表达组(67.6%比19.2%,χ2=24.761,P=0.000);E-FABP在K-i67阳性表达组中的阳性表达率高于Ki-67阴性表达组(84.6%比35.3%,χ2=27.245,P=0.000)。IDC组织中CRABPⅡ表达与Ki-67表达呈负相关(rs=-0.467,P=0.000),E-FABP表达与Ki-67表达呈正相关(rs=0.916,P=0.000)。E-FABP与Ki-67共同表达与肿瘤大小、组织学分级、IDC淋巴转移有关,与患者年龄、TNM分期均无关。结论 CRABPⅡ和E-FABP可能参与了肿瘤细胞的增殖调控,CRABPⅡ对肿瘤细胞的增殖具有抑制作用,E-FABP对肿瘤细胞的增殖具有促进作用,E-FABP和Ki-67对IDC的进展具有协同作用,两者的共同表达对IDC转移的判断具有一定的临床价值。 展开更多

To assess the prevalence of a panel of serologic markers that reflect gut barrier dysfunction in a mixed cohort of pediatric and adult primary sclerosing cholangitis (PSC) patients. METHODSSera of 67 PSC patients [median age (range): 32 (5-79) years, concomitant IBD: 67% and cirrhosis: 20%] were assayed for the presence of antibodies against to F-actin (AAA IgA/IgG) and gliadin (AGA IgA/IgG)] and for serum level of intestinal fatty acid-binding protein (I-FABP) by ELISA. Markers of lipopolysaccharide (LPS) exposure [LPS binding protein (LBP)] and various anti-microbial antibodies [anti-OMP Plus IgA and endotoxin core IgA antibody (EndoCAb)] were also determined. Poor disease outcome was defined as orthotopic liver transplantation and/or liver-related death during the follow-up [median: 99 (14-106) mo]. One hundred and fifty-three healthy subjects (HCONT) and 172 ulcerative colitis (UC) patients were the controls. RESULTSA total of 28.4%, 28.0%, 9% and 20.9% of PSC patients were positive for AAA IgA, AAA IgG, AGA IgA and AGA IgG, respectively. Frequencies of AAA IgA and AAA IgG (P < 0.001, for both) and AGA IgG (P = 0.01, for both) but not AGA IgA were significantly higher compared to both of the HCONT and the UC groups. In survival analysis, AAA IgA-positivity was revealed as an independent predictor of poor disease outcome after adjusting either for the presence of cirrhosis [HR = 5.15 (1.27-20.86), P = 0.022 or for the Mayo risk score (HR = 4.24 (0.99-18.21), P = 0.052]. AAA IgA-positivity was significantly associated with higher frequency of anti-microbial antibodies (P < 0.001 for EndoCab IgA and P = 0.012 for anti-OMP Plus IgA) and higher level of the enterocyte damage marker (median I-FABP<sub>AAA IgA pos</sub><sub>vs</sub><sub>neg</sub>: 365 vs 166 pg/mL, P = 0.011), but not with serum LBP level. CONCLUSIONPresence of IgA type AAA identified PSC patients with progressive disease. Moreover, it is associated with enhanced mucosal immune response to various microbial antigens and enterocyte damage further highlighting the importance of the gut-liver interaction in PSC. 展开更多

目的:探讨血浆表皮型脂肪酸结合蛋白(epidermal fatty acid binding protein,E-FABP)与脂肪酸合成酶(fatty acid syn-thase,FASN)蛋白水平与人股动脉粥样硬化(atherosclerosis,AS)及相关危险因素的关系。方法:对178例患者行股动脉彩超... 目的:探讨血浆表皮型脂肪酸结合蛋白(epidermal fatty acid binding protein,E-FABP)与脂肪酸合成酶(fatty acid syn-thase,FASN)蛋白水平与人股动脉粥样硬化(atherosclerosis,AS)及相关危险因素的关系。方法:对178例患者行股动脉彩超检查并测定股动脉内膜中层厚度(intimal medial thickness,IMT),根据股动脉IMT分为股AS组(128例)和对照组(50例),测定空腹血浆E-FABP、FASN蛋白水平,分析其与股AS及危险因素的关系。结果:与对照组比较,股AS患者血浆中E-FABP与FASN蛋白水平增高(P<0.05),血浆E-FABP与FASN水平呈正相关(P<0.05),血浆E-FABP水平与年龄、体质指数(body mass index,BMI)、甘油三酯(triglyceride,TG)、脂联素、C反应蛋白(C reactive protein,CRP)、IMT呈正相关(P<0.05),血浆FASN水平与年龄,BMI、空腹血糖(fasting blood-glucose,FPG)、稳态模型胰岛素抵抗指数(homeostasis model assessment-insulin resistance index,HOMA-IR)、TG、脂联素、CRP、IMT呈正相关(P<0.05),血浆E-FABP、FASN与高密度脂蛋白-胆固醇呈负相关(P<0.05);Logistic回归分析表明:年龄,FASN,E-FABP,HOMA-IR,CRP,TG是股AS的危险因素(P<0.05)。结论:股AS与血浆E-FABP、FASN水平相关联,可能成为新的诊断与评价人股AS的循环标志。 展开更多

目的:探究多囊卵巢综合征(polycystic ovary syndrome,PCOS)患者血清表皮型脂肪酸结合蛋白(epidermal fatty acid binding protein,E-FABP)、血管生成素样蛋白2(angiopoietin-like protein,Angptl 2)、阿立新A(Orexin-A)水平与肥胖和糖... 目的:探究多囊卵巢综合征(polycystic ovary syndrome,PCOS)患者血清表皮型脂肪酸结合蛋白(epidermal fatty acid binding protein,E-FABP)、血管生成素样蛋白2(angiopoietin-like protein,Angptl 2)、阿立新A(Orexin-A)水平与肥胖和糖脂代谢的关系。方法:回顾性分析本院2019年11月-2020年11月收治的89例PCOS患者的临床资料,依据体质量指数(body mass index,BMI)将其分为PCOS肥胖组(49例,BMI≥25 kg/m2)和PCOS非肥胖组(40例,BMI<25 kg/m2),另取同期来本院进行健康体检的38例女性作为健康对照组,对比三组患者E-FABP、Angptl 2、Orexin-A、糖代谢指标[空腹胰岛素(fasting insulin,FINS)、2 h胰岛素(insulin 2 h,INS 2 h)、空腹血糖(fasting plasma glucose,FPG)、2 h血糖(glucose 2 h,GLU 2 h)]、脂代谢指标[甘油三酯(triglycerides,TG)、总胆固醇(total cholesterol,TC)、游离脂肪酸(free fatty acid,FFA)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)、高密度脂蛋白胆固醇(high density liptein cholesterol,HDL-C)]水平并分析其相关性。结果:(1)PCOS肥胖组患者E-FABP、Angptl 2水平高于PCOS非肥胖组与健康对照组,Orexin-A水平低于PCOS非肥胖组与健康对照组(P<0.05)。(2)PCOS肥胖组患者FINS、INS 2 h、FPG及GLU 2 h水平均高于PCOS非肥胖组与健康对照组(P<0.05)。(3)PCOS肥胖组患者TG、TC、FFA、LDL-C水平均高于PCOS非肥胖组与健康对照组,HDL-C水平低于PCOS非肥胖组与健康对照组(P<0.05)。(4)E-FABP水平与肥胖(r=0.514)、FINS(r=0.680)、INS 2 h(r=0.643)、FPG(r=0.509)、GLU 2 h(r=0.738)、TG(r=0.764)、TC(r=0.575)、FFA(r=0.630)、LDL-C(0.667)指标均呈显著正相关(P<0.05);Angptl 2水平与肥胖(r=0.468)、FINS(r=0.663)、INS 2 h(0.619)、FPG(r=0.622)、GLU 2 h(r=0.651)、TG(r=0.705)、TC(r=0.638)、FFA(r=0.623)、LDL-C(r=0.751)指标均呈显著正相关(P<0.05);Orexin-A水平与HDL-C(r=0.691)显著正相关,与肥胖(r=-0.720)、FINS(r=-0.419)、INS 2 h(r=-0.432)指标呈显著负相关(P<0.05)。结论:PCOS患者E-FABP、Angptl 2水平升高、Orexin-A水平降低均与肥胖及糖脂代谢水平有关。 展开更多

目的应用跨种属肿瘤关键基因筛选策略,从已报道的大量肝癌相关基因中筛选、定位出影响肝癌发生和发展的关键分子。方法结合本课题组的前期实验数据及国内外文献数据,建立肝癌差异表达分子的跨种属数据集,对首批筛选出的候选分子表皮... 目的应用跨种属肿瘤关键基因筛选策略,从已报道的大量肝癌相关基因中筛选、定位出影响肝癌发生和发展的关键分子。方法结合本课题组的前期实验数据及国内外文献数据,建立肝癌差异表达分子的跨种属数据集,对首批筛选出的候选分子表皮型脂肪酸结合蛋白(E—FABP),应用RT-PCR和Western blot技术,分别验证其在人、树鼩和大鼠的肝癌、癌旁及正常肝组织中mRNA和蛋白水平的差异表达情况。实验数据以均数±标准差(x^-±s)表示,采用单因素方差分析。结果初步建成跨种属肝癌差异表达数据集,RT-PCR检测结果显示E-FABP mRNA在人肝癌及其癌旁组织和正常肝组织中的相对表达量分别为0.87±0.14、0.64±0.12和0.67±0.07I在树鼩肝癌及其癌旁组织和正常肝组织中的相对表达量分别为0.87±0.25、0.73±0.19和0.68±0.19;在大鼠肝癌及其癌旁组织和正常肝组织中的相对表达量分别为0.97±0.22、0.78±0.16和0.80±0.13。人、树鼩和大鼠肝癌组织中的E—FABP mRNA表达水平均显著高于癌旁和正常肝组织,F值分别为20.910、3.807、4.482,P值均〈0.05,差异有统计学意义。Western blot检测结果显示E-FABP在人、树鼩和大鼠肝癌组织中的蛋白表达水平均较癌旁和正常肝组织显著上调。结论跨种属肿瘤关键基因筛选策略可能有助于发现肝癌关键基因,E—FABP有可能是影响肝癌发生和发展的重要分子之一。 展开更多

Objective:To investigate the relationship between the fatty acid-binding protein 4(FABP4)and colorectal cancer(CRC).Methods:Using an enzyme-linked immunosorbent assay(ELISA),we measured the expression of FABP4 in plasma of50 patients who underwent surgery for CRC from October 2017 to May 2018 and 50 healthy controls.The content of the visceral fat area(VFA)as seen with abdominal computed tomography(CT)scanning was measured by ImageJ software.The expression levels of FABP4,E-cadherin,and Snail proteins in CRC and adjacent tissues were determined by immunohistochemistry.Results:The mean concentration of plasma FABP4 of CRC patients was higher than that of the control group(22.46 vs.9.82 ng/m L;P<0.05).The concentration of plasma FABP4 was related to the tumor,node,metastatis(TNM)stage and lymph node metastasis and was independent of age,body mass index(BMI),tumor size and location,and the degree of differentiation of CRC.The concentration of plasma FABP4 was positively correlated with high VFA and lipoprotein-a(LPA)(P<0.05);but it was not correlated with total cholesterol(TG),total triglyceride(TC),low-density lipoprotein(LDL),high-density lipoprotein(HDL),or apolipoprotein AI(Apo-AI).The expression of FABP4 protein in CRC tissues was positively correlated with the degree of CRC differentiation,tumor stage,and lymph node metastasis.The level of FABP4 protein was negatively correlated with E-cadherin protein(r=-0.3292,P=0.0196)and positively correlated with Snail protein(r=0.5856,P<0.0001).Conclusions:High LPA and VFA were risk factors for increased plasma FABP4 in CRC patients.FABP4 protein was highly expressed in CRC tissues and associated with TNM stage,differentiation,and lymph node metastasis of CRC.The level of FABP4 in CRC tissue was correlated with E-cadherin and Snail expression,suggesting that FABP4 may promote CRC progression related to epithelial-mesenchymal transition(EMT). 展开更多

目的探讨血清脂肪型脂肪酸结合蛋白(adipose fatty acid-binding protein,A-FABP)、表皮型脂肪酸结合蛋白(epidermal fatty acid-binding protein,E-FABP)与阻塞性睡眠呼吸暂停低通气综合征(OSAHS)之间的相关性。方法选取OSAHS患者60例... 目的探讨血清脂肪型脂肪酸结合蛋白(adipose fatty acid-binding protein,A-FABP)、表皮型脂肪酸结合蛋白(epidermal fatty acid-binding protein,E-FABP)与阻塞性睡眠呼吸暂停低通气综合征(OSAHS)之间的相关性。方法选取OSAHS患者60例作为实验组,健康体检者30名作为对照组。根据睡眠呼吸暂停低通气指数(AHI)的数值将实验组分为轻度组(5次/h≤AHI<15次/h)和中重度组(AHI≥15次/h)。测量两组AHI、最低血氧饱和度(LSaO2)、血清A-FABP及E-FABP等相关数值,测量实验组治疗后3个月AHI、血清A-FABP及E-FABP数值。结果①A-FABP与体重、体质量指数(BMI)、甘油三脂(TG)、AHI、E-FABP呈正相关(P均<0.05),与LSaO2呈负相关(P<0.05);②E-FABP与年龄、总胆固醇(TC)呈正相关(P均<0.05);③轻度组治疗后较治疗前A-FABP、E-FABP、AHI均降低(P均<0.05);④中重度组治疗后较治疗前A-FABP、AHI均降低(P均<0.05)。结论OSAHS患者血清A-FABP水平高于健康人群对照组,并且OSAHS病情越重,血清A-FABP水平越高;OSAHS患者AHI、血清A-FABP水平在通过有效治疗后均降低。 展开更多
