Determination of Tetracyclines and Their Epimers in Agricultural Soil Fertilized with Swine Manure by Ultra-High-Performance Liquid Chromatography Tandem Mass Spectrometry 被引量:9

A rapid, sensitive and specific ultra-high-performance liquid chromatography tandem mass spectrometry (UPLC-MS) method was developed for the analysis of tetracycline antibiotics, including tetracycline (TC), oxytetracycline (OTC), chlortetracycline (CTC) and their 4-epimers (4-epiTCs) in agricultural soil fertilized with swine manure. Soil samples were extracted and cleaned-up with 10 mL EDTA-McIlvaine buffer solution (pH 4.0), then cleaned-up and pre-concentrated using the Oasis MAX cartridge and then eluted with 1 mL solution by mixing formic acid, methanol and water at a ratio of 2:15:83 (v/v/v). The purified samples were separated by an ACQUITY UPLC BEH C18 column using acetonitrile and water containing 0.1% formic acid mobile phase and detected by a single quadrupole MS. The limits of detection for the soil extraction method (LODsoil) ranged from 0.6-2.5 lag kg-~ with recoveries from 23.3-159.2%. Finally, the method was applied to an agricultural field in an area with intensive pig-fattening farming. Tetracyclines were detected in soil from 2.8 to 42.4 μg kg-1 soil. These results demonstrate that soil from swine farms can become severely contaminated with tetracycline antibiotics and their metabolites. 展开更多

After recrystallization from ethanol, two compounds(A, prisms, m.p.127—129℃; B, needles, m.p.75—77℃) were obtaines.Their MS, IR spectral data and elemental analysis were consistent with the proposed structures. 为探讨氯雷他定对新型冠状病毒肺炎的潜在治疗作用。从基因表达谱数据库(Gene expression omnibus,GEO)获取氯雷他定数据,分别进行差异分析、基因本体学(GO)富集分析、京都基因与基因组百科全书(KEGG)富集分析、蛋白质相互作用(Protein-... 为探讨氯雷他定对新型冠状病毒肺炎的潜在治疗作用。从基因表达谱数据库(Gene expression omnibus,GEO)获取氯雷他定数据,分别进行差异分析、基因本体学(GO)富集分析、京都基因与基因组百科全书(KEGG)富集分析、蛋白质相互作用(Protein-protein interaction,PPI)网络分析并筛选关键基因,使用本课题组前期建立的表观精准治疗预测平台(Epigenomic Precision Medicine Prediction Platform,EpiMed)进行关联分析,筛选出负相关疾病、正相关药物及体内作用靶点,并在中、英文数据库中检索冠状病毒与关键基因作用的相关文献加以验证。共筛选出差异基因642个,其中上调230个,下调412个。其中,GO富集的基因大多参与炎症反应、激素作用和糖脂代谢等过程,KEGG通路富集主要和多种病毒感染、炎症介质、补体和凝血、消化道损伤、心律失常和氧化应激等过程或功能相关。结合EpiMed的预测结果,发现氯雷他定对全基因组表达谱的影响复杂,在PPI中,共得到642个蛋白,筛选出10个关键基因(Hub gene),主要与神经信号传导、磷酸化和炎症反应等功能相关,在EpiMed的结果中,预测出氯雷他定可能具有潜在治疗作用的疾病,包括严重急性呼吸综合征(SARS)、流行性感冒、社区获得性肺炎、严重脓毒症、呼吸道合胞病毒感染等,预测出对COVID-19可能具有治疗作用的药物,包括泛昔洛韦、奈韦拉平、氟伐他汀、磷酸氯喹、TNF-α抑制剂、金银花、虎杖、柴胡和知母等。通过文本挖掘,共检索到14篇冠状病毒与关键靶点作用相关的文献,验证了关键靶点的有效性。得出如下结论:氯雷他定对心脏调节,免疫反应,氧化应激和病毒感染等过程或通路具有调控作用,其可能在抗病毒的同时,还具有免疫调节和器官保护功能,对COVID-19危重症患者有潜在的治疗意义。 展开更多

目的通过生物信息学方法研究右美托咪定(DMED)药理作用的关键分子及其他药物与DMED联合应用发挥镇痛作用的潜在关键分子。方法收集公共基因表达数据库(GEO)中DMED给药前后的差异表达基因,应用功能富集分析、蛋白质相互作用研究DMED药理... 目的通过生物信息学方法研究右美托咪定(DMED)药理作用的关键分子及其他药物与DMED联合应用发挥镇痛作用的潜在关键分子。方法收集公共基因表达数据库(GEO)中DMED给药前后的差异表达基因,应用功能富集分析、蛋白质相互作用研究DMED药理作用,应用EpiMed平台进行多组学分析筛选DMED潜在相似药物。结果分析共筛选出差异表达基因(DEG)419个,其中219个表达上调、200个表达下调,其中映射至人类基因组的差异基因有271个,112个表达上调,159个表达下调。基因本体学(GO)富集分析结果显示,DEG主要定位在突触后膜、甲基转移酶复合体区域,主要参与调节MAP激酶活性、对钙离子转运的正向调节、对肿瘤细胞凋亡的反向调节等生物学过程。在分子功能方面,DEG主要与神经活性受体与配体的结合及与激素、Wnt蛋白的结合等相关。京都基因与基因组百科全书(KEGG)分析结果显示,DEG主要富集在神经活性的受体配体相互作用信号通路、Wnt信号通路,以及对胃癌、肺癌等恶性肿瘤的转录调节信号通路。蛋白互作网络(PPI)中共包含169个节点,其中CCND1、CCR5、TYROBP、CCL5、TLR7、SFRP1、NDC80、BDKRB1、FCER1G和MRC1处于核心位置。EpiMed多组学关联分析发现,右美沙芬、可乐定、吗啡等药物与右美托咪定呈正相关性。结论对来自公共基因表达数据库的DMED转录组数据分析发现,CCND1、CCR5、TYROBP、CCL5、TLR7、SFRP1、NDC80、BDKRB1、FCER1G和MRC1可能是DMED发挥药理学作用的潜在靶点,多组学关联分析表明LY75、RPL39、FMO5、FMO3、ORM1等可能是其他镇痛剂与DMED联合应用机制研究的潜在关键分子。 展开更多

Because many therapeutic agents are contaminated by epimeric impurities or form epimers as a result of metabolism, analytical tools capable of determining epimers are increasingly in demand. This article is a proof-of-principle report of a novel DMS–MS/MS method to separate and simultaneously quantify epimers, taking PGF2α and its 8-epimer, 8-iso-PGF2α, as an example. Good accuracy and precision were achieved in the range of 10–500 ng/m L with a run time of only 1.5 min. Isopropanol as organic modifier facilitated a good combination of sensitivity and separation. The method is the first example of the quantitation of epimers without chromatographic separation. 展开更多
