The 1: 4 dilution of normal skin had no (?) on mitochondrial functions. There is a widegap between the malonaldehyde (MDA) contents of these two kinds of extracts. An apparent posi-tive correlation was found between MDA content and degree of inhibition. However, on close ex-amination, it was found that the MDA content is not the direct cause of the decrease of therespiratory control ratio (RCR). The increase of MDA content is probably related to the destruc-tion of the lipid bilayer of the mitochondria. Data from the energization experiment show that theenergization process is compromised when the destruction of the membrane has proceeded to a cer-tain degrce. The inhibition of mitochondrial respiratory function becomes evident when encrgizationfunction is still normal. Therefore at least two mechanisms are operating in the inhibition process.Further work is needed to elucidate the problem.展开更多

The point of change was at 1 mg/ml of D1 concentration.The ratio of FCCP-stimulated respiration rate/state 4 respiration rate was decreased when D1 dosage was increased.The results of experiments were similar whether glutamate+malate or succinate were used as the substrate. The point of change was also at 1 mg/ml of D1 concentration. The decrease of FCCP-stimulated rate/state 4rate implied that the respiratory chain was damaged.But complex Ⅳ-the terminal segment of the chain-remained intact. Thus, the site of damage must be somewhere in complexesⅠ ̄Ⅲ and/or complexes Ⅱ ̄Ⅲ. Since NADH dehydrogenase and succinate dehydrogenase-the main enzymes in complexesⅠand Ⅱ-were not affected by D1 but NADH: cytochrome c reductase and succinate:cytochrome c reductase markedly inhibited, the site of damage was most likely located in complex Ⅲ-the bc1 complex.展开更多

It was found that both D1 and D'1 were able to induce chemiluminescence in hepatocytic submitochondrial particles in a dosage-dependent manner and D1 was more potent than D'1 in this function when they were in the same dosage. When superoxide dismutase (SOD) and beta-carotene were in dosage smaller than 10μg/ml and 3. 2 mmol/L respectively, they were able to suppress the D1's capacity to induce chemiluminescence in the submitochondrial particles in a dosage-dependent manner , too. These findings suggest that there is the occurrence of free radicals in the interaction between D1' and submitochondrial particles and superoxide anions and siglet oxygen seem to be involved.展开更多

BACKGROUND Scrub typhus is a naturally occurring acute infectious disease that is primarily transmitted through the bites of chiggers or larval mites infected by Orientia tsutsugamushi(O.tsutsugamushi).Omadacycline,a ...BACKGROUND Scrub typhus is a naturally occurring acute infectious disease that is primarily transmitted through the bites of chiggers or larval mites infected by Orientia tsutsugamushi(O.tsutsugamushi).Omadacycline,a novel tetracycline,exhibits potent antibacterial efficacy against both typical bacteria and atypical pathogens.However,omadacycline application in the treatment of scrub typhus remains limited.CASE SUMMARY In the present work,we report several cases of scrub typhus,with the main clinical symptoms being fever,the formation of eschars or ulcers,local or systemic lymphadenopathy,headache,myalgia and rash.Blood samples were collected before omadacycline was administered,and O.tsutsugamushi infection was confirmed through targeted next-generation sequencing(tNGS).After two days of treatment,the patients’symptoms,including fever,were alleviated,with no adverse drug reactions.CONCLUSION tNGS is an effective method for diagnosing scrub typhus.Omadacycline can be considered an alternative option for antiinfective therapy in patients with O.tsutsugamushi infections.展开更多
