Aquaporin-4-IgG-seropositive neuromyelitis optica spectrum disorders:progress of experimental models based on disease pathogenesis

Neuromyelitis optica spectrum disorders are neuroinflammatory demyelinating disorders that lead to permanent visual loss and motor dysfunction.To date,no effective treatment exists as the exact causative mechanism remains unknown.Therefore,experimental models of neuromyelitis optica spectrum disorders are essential for exploring its pathogenesis and in screening for therapeutic targets.Since most patients with neuromyelitis optica spectrum disorders are seropositive for IgG autoantibodies against aquaporin-4,which is highly expressed on the membrane of astrocyte endfeet,most current experimental models are based on aquaporin-4-IgG that initially targets astrocytes.These experimental models have successfully simulated many pathological features of neuromyelitis optica spectrum disorders,such as aquaporin-4 loss,astrocytopathy,granulocyte and macrophage infiltration,complement activation,demyelination,and neuronal loss;however,they do not fully capture the pathological process of human neuromyelitis optica spectrum disorders.In this review,we summarize the currently known pathogenic mechanisms and the development of associated experimental models in vitro,ex vivo,and in vivo for neuromyelitis optica spectrum disorders,suggest potential pathogenic mechanisms for further investigation,and provide guidance on experimental model choices.In addition,this review summarizes the latest information on pathologies and therapies for neuromyelitis optica spectrum disorders based on experimental models of aquaporin-4-IgG-seropositive neuromyelitis optica spectrum disorders,offering further therapeutic targets and a theoretical basis for clinical trials. 展开更多

[Objective] This study aimed to investigate the treatment efficacy of tradi- tional Chinese veterinary medicine mycoplasma gatlisepticum infection spray against experimental mycoplasma gallisepticum infection. [Method] Experimental chickens in- fected with mycoplasma gallisepticum were used as models to investigate the treat- ment efficacy of different doses of traditional Chinese veterinary medicine mycoplas- ma gallisepticum infection spray. [Result] Medication with traditional Chinese veteri- nary medicine mycoplasma gallisepticum infection spray for continuous 5 d showed certain treatment efficacy against mycoplasma gallisepticum infection. The relative weight gain rate of 0.5 and 1.0 g/m3 dose groups was significantly higher than the infection control group (P〈0.01); the cure rate of mycoplasma gallisepticum infection spray groups was significantly higher than the infection control group (P〈0.01) and showed no significant difference compared with Taimiaoling control group (P〉0.05); the mortality rate showed no significant difference among various groups (P〉0.05), indicating that traditional Chinese veterinary medicine mycoplasma gallisepticum in- fection spray has relatively ideal treatment efficacy against mycoplasma gallisepticum infection. Prostate cancer(PCa)is one of the most common malignant tumors in the male genitourinary system,ranking second in incidence worldwide.Traditional Chinese medicine(TCM),as an important component of complementary and alternative medicine,shows unique advantages in cancer treatment.Chinese herbal medicine is usually composed of multiple ingredients and involves multiple signaling pathways,which showed function of inducing apoptosis of cancer cells,arresting the cell cycle,inhibiting invasion and metastasis,reducing drug resistance,and regulating immune function.Physical therapy is also an important treatment of TCM.Currently,Physical therapy such as acupuncture or Tai Chi and Qigong are gaining increased recognition in the management of PCa,particularly in addressing issues like urinary incontinence and bone metastasis-related pain.This article reviews the TCM treatment and therapy of PCa,in order to provide new research avenues and treatment options for the treatment of PCa with TCM and improve the quality of life of patients. 展开更多

AIM: To investigate the efficacy of Magliasa, a traditional Iranian formula, on experimental colitis. METHODS: After botanical authentication of herbal ingredients, formulation of Magliasa, quantitative determination of total glucosinolates and total phenolic content, and analysis of the thin layer chromatography profile were performed. Colitis was then induced in male rats by instillation of 2,4,6-trinitrobenzenesulfonic acid (TNBS) in all groups, aside from the Sham group.The experimental groups consisted of: the Sham group that received only normal saline; the Mag-50, Mag-100 and Mag-200 groups, which received 50, 100 and 200 mg/kg per day of Magliasa, respectively; the control group, which received vehicle water orally; the infliximab group, which received infliximab (5 mg/kg per day, subcutaneously); and the Dexa group, which received dexamethasone (1 mg/kg per day, orally). After completing the treatment period (2 wk), the rats were sacrificed, the colon was removed, its macroscopic and microscopic changes were recorded, and tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1b), total antioxidant capacity, myeloperoxidase (MPO), and lipid peroxidation (LPO) were assessed in colon homogenate.RESULTS: The mean value of total glucosinolates in one gram of Magliasa was 19 ± 1 μmol. The mean value of the total phenolic content was 293.8 ± 17.6 mg gallic acid equivalents per 100 gram of Magliasa. Macroscopic scores were significantly decreased in Mag-100 (1.80 ± 0.58, P = 0.019) and Mag-200 (1.20 ± 0.20, P = 0.001) compared to the control group (3.40 ± 0.24), although some inflammation and hyperemia were evident. Treatment of rats by dexamethasone (0.33 ± 0.21, P < 0.001) and infliximab (0.83 ± 0.31, P < 0.001) remarkably attenuated scores where mild hyperemia was observed macroscopically. In comparison to the control group (4.00 ± 0.32), only Mag-200 (1.60 ± 0.40) showed a significant decrease in colonic histopathological scores (P = 0.005). Minimal mucosal inflammation was observed in the Dexa group (0.67 ± 0.21, P < 0.001). The levels of TNF-α, IL-1b and MPO were significantly lower in all groups compared to the controls (P < 0.05). A significant decrease in LPO was seen in the Mag-200 (3.27 ± 0.77, P = 0.01) and Dexa (3.44 ± 0.22, P = 0.011) groups in comparison to the control group (6.43 ± 0.61). Only dexamethasone caused a significant increase in antioxidant power in comparison to the control group (346.73 ± 9.9 vs 228.33 ± 2.75, P < 0.001). Infliximab and different doses of Magliasa did not show any remarkable increase in antioxidant capacity (P > 0.05). The effect of Magliasa in all of mentioned parameters, except antioxidant capacity, was dose dependent.CONCLUSION: The effects of Magliasa in TNBS-induced colitis are encouraging and warrant clinical trials for further confirmation. 展开更多

AIM:To investigate therapeutic effects of traditional Chinese medicine formulations,Hexuemingmu(HXMM)on laser-induced choroidal neovascularization(CNV)and followup effect in mice.METHODS:C57 BL/6 mice of 8-week-old were used and CNV was induced with 577 nm laser photocoagulation.Animals were randomly divided into groups and different doses of HXMM were administered daily.One,four,and eight weeks after the intervention,the electroretinogram(ERG),fundus fluorescence angiography,choroidal flat mount and immunofluorescence staining were preformed to evaluate the function and CNV formation.The expression levels of angiogenic proteins were determined by Western blotting and immunofluorescence staining.An analysis of variance and Kruskal-Wallis test were used to test the differences among the groups.RESULTS:The results showed that HXMM effectively increased amplitude of ERG of mice(P<0.05),alleviated fundus CNV leakage(P<0.05),and reduced the area of neovascularization and the expression of angiogenic proteins(P<0.05)after laser-induced CNV.CONCLUSION:HXMM can protect the retinal function of mice after laser-induced CNV,and inhibit the CNV development. 展开更多

This paper gives an overview of the Lee Carter method and reiterates the feasibility of using it to construct mortality forecast for the population data. In a first step, the model is fitted in a traditional way and used to extrapolate forecast of the time-varying mortality index. The observed pattern of the mortality rates shows a different variability at different ages, highlighting that the homoscedasticity hypothesis is quite unrealistic. Thus, in a second step, the paper aims to produce more reliable mortality forecasting, focusing on the errors in the estimation of the model parameters. The robustness of the estimated parameter is analysed throughout an experimental strategy which allows to assess the robustness of the Lee Carter model by inducing the errors to satisfy the homoscedasticity hypothesis. The graphical and numerical results are tested by means of a comparison in terms of prediction accuracy. 展开更多

Alzheimer’s disease is a complex neurodegenerative ailment accompanied by cognitive problems and progressive memory loss.The main causes of the disease are indistinguishable even though aging is said to be the key risk factor,along with the hypotheses such asβ-amyloid plaques,tau-proteins,neuroinflammation,and oxidative stress.Currently,Alzheimer’s disease is not curable,and the available medicines are only used to provide symptom relief.Scientists and researchers have developed numerous models to study Alzheimer’s disease,such as in silico methods,in vitro models of cultured rat cortical neurons,in vivo rodent models involving mice and rats,and non-rodent models as well.This review provides an insight into the modality of Alzheimer’s disease treatment using medicinal plants that are natural sources with abundant bioactive substances,and traditional formulations such as an extract from Notoginseng Radix et Rhizoma,Chinese herbal formulations(i.e.,traditional Chinese medicine kidney-nourishing formula,supplements such as coenzyme Q10,alpha-lipoic acid,Ginkgo Biloba,omega-3’s,and acetyl-L-carnitine).In addition,we have discussed the evaluation parameters for experimental Alzheimer’s disease,which are used to assess the mechanism of memory and pharmacological effects of molecules for drug discovery and development.The common experimental analysis methods for Alzheimer’s disease include behavioral,biochemical,and histological studies.This work will allow researchers to access an informative reference of traditional medicines along with experimental analysis methods in one place for Alzheimer’s disease research. 展开更多

In order to improve the intensive cultivation and management technology of Olea europaea in the high density orchard,through an analysis on the 3 a growth,yield and orchard construction cost for different varieties and different densities in Gansu Wudu ordinary O.europaea orchard and close O.europaea planting orchard,the appropriate conditions,varieties and density for intensive orchard construction were initially selected.The results showed that the tree height growth in the ordinary orchard was 119%of the tree height growth in the high density orchard,while the ground diameter growth in the ordinary orchard was 112%of the ground diameter growth in the high density orchard,but the yield in the ordinary orchard was only 62%of the yield in the high density orchard;of the 8 new varieties introduced from Spain,"Arbequina","Arbosana"and"Koroneiki"were the appropriate varieties for the intensive orchard construction;the ratio of the ordinary orchard construction cost to the high density orchard construction cost was 1∶6.24.Under high water and fertilizer conditions,the ratio of the ordinary orchard yield to the high density orchard yield was 1∶4.481.Therefore,in the flat land area,using the appropriate intensive cultivars,and integrating the high-tech achievements and advanced management tools,can increase output per unit area,and greatly save manpower and land costs,to achieve the purpose of high quality,high yield and stable production.In the production practice,it is necessary to vigorously promote the intensive cultivation technology of O.europaea. 展开更多

Experimental Study of the Anti-human Papilloma Virus Mechanisms by a Traditional Chinese Medicine “Keyouling”

To investigate the anti-human papilloma virus (HPV) effect of the external liniment of traditional Chinese medicine “Keyouling”, the efficacy of this drug acting upon the proliferation and growth of the normal epidermis cells of rabbits′ penis prepuce in different concentrations was observed. An experimental model of pathology of histocytes infected with HPV in vitro was established by using the human HPV infected prepuce epidermis cells as virus carrier to infect the sub-cultured epidermis cells of rabbits′ penis prepuce. The direct killing effect and the blocking effect of infections of epidermis cells in rabbits′ penis prepuce induced by HPV with different concentrations of “Keyouling” were determined by 3 kinds of culture methods: (1) the infected epidermis cells of rabbits′ penis prepuce were challenged by co-culture with HPV suspension and with culture medium containing drug; (2) the co-culture with epidermis cells of rabbits′ penis prepuce and HPV suspension was challenged with drug; (3) co-culture with culture medium containing drug and epidermis cells of rabbits′ penis prepuce was challenged with HPV suspension. It was found that “Keyouling” had little effect on the proliferation and growth of the normal epidermis cells of rabbits′ penis prepuce, but it had direct killing effect on HPV, and blocked effectively the infection of the epidermis cells of rabbits′ penis prepuce by HPV. It concludes that “Keyouling” has significant killing effect to HPV, but not normal histocytes in vitro , indicating that it is safe for clinical use. Its blocking effect on the HPV infection of rabbits′ penis prepuce epidermis cells suggested “Keyouling” may be clinically used to prevent the development of tumors or to inhibit the further progression or metastasis of tumors. 展开更多

Objective: To study the efficacy and mechanism of AT-1 a new compound (an aqueous agent extracted from a Traditional Chinese herb with antiplatelet efficacy) on antitumor metastases in mice. Methods:AT-1, a compound extracted from a Traditional Chinese herb, was prepared by our lab. C57BL/6 female mice, which were transplanted H 22 liver cancer cells, received the agent. The tumor nodules, the pulmonary metastasis rate and tumor cell proliferation activity were tested. The tumor cells were exposed to different concentrations of AT-1, and then the condition of the tumor proliferations were tested. Results: High and middle dosages of AT-1 inhibited both the lymphatic and blood systemic tumor pulmonary metastases significantly, but had no effect on inhibition of the tumor cell proliferation in vitro. Conclusion:The major antitumor actions of AT-1 may be on antitumor metastases, and the compounnd may be a new candidate for antitumor metastases agent. More study of the mechanism and action of AT-1 on antitumor metastases and the actions are still being done deeply. 展开更多

Solid oxide fuel cells(SOFCs)have attracted a great deal of interest because they have the highest efficiency without using any noble metal as catalysts among all the fuel cell technologies.However,traditional SOFCs suffer from having a higher volume,current leakage,complex connections,and difficulty in gas sealing.To solve these problems,Rolls-Royce has fabricated a simple design by stacking cells in series on an insulating porous support,resulting in the tubular segmented-in-series solid oxide fuel cells(SIS-SOFCs),which achieved higher output voltage.This work systematically reviews recent advances in the structures,preparation methods,perform-ances,and stability of tubular SIS-SOFCs in experimental and numerical studies.Finally,the challenges and future development of tubular SIS-SOFCs are also discussed.The findings of this work can help guide the direction and inspire innovation of future development in this field. 展开更多

BACKGROUND Currently,traditional Chinese medicine(TCM)formulas are commonly being used as adjunctive therapy for ulcerative colitis in China.Network meta-analysis,a quantitative and comprehensive analytical method,can systematically compare the effects of different adjunctive treatment options for ulcerative colitis,providing scientific evidence for clinical decision-making.AIM To evaluate the clinical efficacy and safety of commonly used TCM for the treatment of ulcerative colitis(UC)in clinical practice through a network metaanalysis.METHODS Clinical randomized controlled trials of these TCM formulas used for the adjuvant treatment of UC were searched from the establishment of the databases to July 1,2022.Studies that met the inclusion criteria were screened and evaluated for literature quality and risk of bias according to the Cochrane 5.1 standard.The methodological quality of the studies was assessed using ReviewManager(RevMan)5.4,and a funnel plot was constructed to test for publication bias.ADDIS 1.16 statistical software was used to perform statistical analysis of the treatment measures and derive the network relationship and ranking diagrams of the various intervention measures.RESULTS A total of 64 randomized controlled trials involving 5456 patients with UC were included in this study.The adjuvant treatment of UC using five TCM formulations was able to improve the clinical outcome of the patients.Adjuvant treatment with Baitouweng decoction(BTWT)showed a significant effect[mean difference=36.22,95%confidence interval(CI):7.63 to 65.76].For the reduction of tumor necrosis factor in patients with UC,adjunctive therapy with BTWT(mean difference=−9.55,95%CI:−17.89 to−1.41),Shenlingbaizhu powder[SLBZS;odds ratio(OR)=0.19,95%CI:0.08 to 0.39],and Shaoyao decoction(OR=−23.02,95%CI:−33.64 to−13.14)was effective.Shaoyao decoction was more effective than BTWT(OR=0.12,95%CI:0.03 to 0.39),SLBZS(OR=0.19,95%CI:0.08 to 0.39),and Xi Lei powder(OR=0.34,95%CI:0.13 to 0.81)in reducing tumor necrosis factor and the recurrence rate of UC.CONCLUSION TCM combined with mesalazine is more effective than mesalazine alone in the treatment of UC. 展开更多

This paper introduces a systems theory-driven framework to integration artificial intelligence(AI)into traditional Chinese medicine(TCM)research,enhancing the understanding of TCM’s holistic material basis while adhering to evidence-based principles.Utilizing the System Function Decoding Model(SFDM),the research progresses through define,quantify,infer,and validate phases to systematically explore TCM’s material basis.It employs a dual analytical approach that combines top-down,systems theory-guided perspectives with bottom-up,elements-structure-function methodologies,provides comprehensive insights into TCM’s holistic material basis.Moreover,the research examines AI’s role in quantitative assessment and predictive analysis of TCM’s material components,proposing two specific AIdriven technical applications.This interdisciplinary effort underscores AI’s potential to enhance our understanding of TCM’s holistic material basis and establishes a foundation for future research at the intersection of traditional wisdom and modern technology. 展开更多
