[Objectives] The research aimed to study the impact on extraction effect of polysaccharide from Ganoderma lucidum( lingzhi) by different pretreatment methods. [Methods] The impacts on extraction of G. lucidum polysaccharide by soaking, microwave and air flow fine pulverization were contrasted, and the extraction effect of G. lucidum polysaccharide by combining the optimal pretreatment manner with hot water extraction, alcohol extraction, alkali extraction, ultrasonic binding enzyme extraction, and microwave extraction was compared. Finally, the property of G. lucidum polysaccharide obtained after air flow fine pulverization pretreatment was detected and analyzed by high performance liquid chromatography. [Results] The optimal pretreatment method was air flow fine pulverization. Compared with traditional method-direct extraction( coarse grinding combining hot water extraction),crude yield changed little, while polysaccharide content and yield were improved by 114% and 104%. The best combination manner was air flow fine pulverization pretreatment combining with alkali extraction. Compared with traditional method, crude yield, polysaccharide content and yield were improved by 76%,78% and 215% respectively. The property of G. lucidum polysaccharide obtained after air flow fine pulverization pretreatment was detected and analyzed by high performance liquid chromatography. It was found that the treatment method had little impact on the property of G. lucidum polysaccharide. [Conclusions]Air flow fine pulverization pretreatment could greatly improve extraction effect of G. lucidum polysaccharide, which mainly improved the content and yield of G. lucidum polysaccharide, and extraction was more complete, with less impact on the property of the extracted polysaccharide. It was speculated that air flow fine pulverization pretreatment mainly destroyed mechanical support wall membrane structure of G. lucidum fine powder, making that intracellular functional substances completely dissolved out of the cell, and the content would be studied in follow-up experiment. 展开更多

The analysis of the coupling mechanism of thermal-hydraulic-mechanical(THM)fields,and solid-liquidgas(SLG)phases during gas extraction process is of profound significance to explore its numerical application in the gas occurrence regularity and its effective extraction radius. In this study, the Hudi coal mine in Qinshui basin is taken as the research area, the influencing factors of gas occurrence were analyzed, the differences in overburden load for gas pressure distribution and the factors influencing the effective extraction radius were further discussed by using the COMSOL software. The results show that the derivation of mathematical model in gas extraction shows that the process is a process the THM fields restrict each other, and the SLG phases influence each other. The longer the extraction time, the larger the influencing range of borehole, and the better the extraction effect. The larger the diameter of borehole, the larger the effective extraction radius, and the influence on gas extraction effect is smaller in the early stage and larger in the late stage. The borehole arrangement should be flexibly arranged according to the actual extraction situation. The higher the porosity, the higher the permeability, the better the gas extraction effect. The larger the overburden load of reservoir, the stronger the effective stress, which will result in the more severe the strain, and the closure of pore and fracture, which in turn will lead to the decrease of permeability and slow down the gas extraction. The relationship among extraction time, borehole diameter, negative pressure of gas extraction, permeability with effective extraction radius is exponential. This study has important theoretical and practical significance for clarifying and summarizing the gas occurrence regularity and its engineering practice. 展开更多

Alternariol caused DNA single-strand breakage. Conversion of the closed circular double-stranded supercoiled DNA (pBR 322) to the nicked circular form and linear form was used to investigate the effect of extracts of some Chinese medical herbs on DNA nicking induced by alternariol. Some substances in the extracts of Rhizoma polygonati (RP) and Fructus lycii (FL) were shown to protect DNA from the attack by alternariol. Some substance in the RP may bind to plasmid DNA, and this binding reduces the electrophoretic mobility of DNA. These results indicate that substances from FL and RP may be used as DNA protectors. It is possible that they play an important role in preventing cancer. 展开更多

When evaluating ionic liquids (ILs) for extractive desulfurization (EDS) of fuel oils, the inevitable presence of water in the system may have a significant and in many cases strongly negative effect. However, few studies have considered this particular issue and a promoted water effect on EDS is scarcely reported. In this work, COSMO-RS was firstly employed to calculate the capacity and selectivity for EDS of various IL/H20 mixtures, which cover different IL characters and a wide water concentration range. Experiments were then conducted with a representative IL [C4MIM]IH2P04], whose stable and even promoted extraction performance with a small amount of water was suggested by COSMO-RS. Through analyses of the desulfurization ratio, the cross- solubility and the water content in the desulfurized fuel, the promoted effect of water within a certain range (〈 10 wt%) was experimentally demonstrated. Moreover, such effect of water was explained combining the viscosity, the solvent-solute interactions and the COSMO-RS based analysis. 展开更多

Objective:This study has been done to evaluate the interaction between water extracts of Psidium guajava, Rosmarinus officinalis,Salvia fruticosa,Majorana syriaca,Ocimum basilucum,Syzygium aromaticum,Laurus nobilis, and Rosa damascena alone and then synergy testing of these extracts with known antimicrobial agents including oxytetracycline HCl,gentamicin sulfate, penicillin G,cephalexin and enrofloxacin. This study was conducted against five S.aureus isolates;one is Methicillin -resistant Staphylococcus aureus(MRSA) and 4 Methicillin - sensitive Staphylococcus aureus(MSSA).Methods:Evaluation of the interaction between plant extracts and different antimicrobial agents has been done using well - diffusion and microdilution methods. Results:The results of the conducted experiments using well - diffusion method demonstrate that these plants showed in vitro interactions between antimicrobial agents and plant extracts were additive, while using microdilution method showed synergistic effects with significant reduction in the MICs of the test antibiotics against these strains of S.aureus.This change in MIC was noticed in all plant extracts against test antibiotics including these plants showed weak antibacterial activity by well diffusion method.Synergism effect was occurred in both sensitive and resistant strains but the magnitude of minimum fold reduction of inhibitory concentration in resistant strains especially MRSA strain was higher than the sensitive strains.Coclusion:This study probably suggests the possibility of concurrent use of these antimicrobial drugs and plant extracts in combination in treating infections caused by S.aureus strains or at least the concomitant administration may not impair the antimicrobial activity of these antibiotics. 展开更多

Lead (Pb) is ubiquitous in the environment, and low-level Pb exposure can cause neurotoxicity and irreversible damage to children's cognition, learning and memory ability. Nutritional intervention is an effective method to prevent Pb poisoning. Mul- berry is rich in anthocyanins, possessing protective effects for nerves. This study investigated the neuroprotective effects of mulberry extract (ME) against Pb-induced learning and memory deficits in mice. The results showed that the learning and memory abilities of mice, assessed using the Morris test, improved significantly after treatment with ME at a dose of 100 mg/kg body weight. The level of Pb in the brains of mice in the three ME intervention groups decreased significantly, while NO production and anti-oxidant enzymes were significantly restored. It is suggested that ME inhibits Pb-induced neurotoxicity by reversing Pb-induced alterations in the aspect of neurotoxic effects and improving learning and memory. 展开更多

Peanut (Arachis hypogaea L.) leaf aqueous extracts (PLAE) has been reputed to be a type of sleep-aid in China. To investigate the sedative effects and effect pathways of PLAE, rats (n = 31) were employed in two experiments and intragastrically administrated of (1) distilled water, PLAE (500 mg/kg body weight (BW)) and peanut stem aqueous extracts (PSAE, 500 mg/kg BW);(2) 0, 100 or 500 mg/kg BW of PLAE, respectively for at least 14 days. Six relevant neurotransmitters were measured finally. Experiment-1 (n = 16) results showed that the brain Lactate were significantly elevated (p < 0.05) in rat cerebrums after PLAE administrations, compared with Control and PSAE groups. In respect of brain energy system, significant degradations of the brain adenosine triphos- phate (ATP) (p < 0.05) were observed in the brainstems and even the whole brains of rats though PLAE treatments. Moreover, we found that the brain Adenosine monophosphate (AMP) were clearly decreased (p < 0.05) in rat cerebrum and brainstem regions, while the brain Adenosine revealed an increasing propensity (p = 0.076) in the cerebrums of freely behaving rats. After experiment-2 (n = 15), the γ-aminobutyric acid (GABA) concentrations were statistically (p < 0.05) enhanced and the ratios of Glutamate/GABA were simultaneously reduced (p < 0.05) in rat brainstems, no matter which one dose (100 or 500 mg/kg BW) of PLAE were used. Results indicated that PLAE could influence the target neurotransmitters that related to rat circadian rhythms in the specific brain regions, possessing the potentialities as a sedative or sleep-aid for hypnic therapy purposes. 展开更多

Female Wister rats aged 8 weeks were randomly divided into sham operation group, ovariectomized (OVX) control group, and 20VX groups fed with soy isoflavone crude extract supplementation. The rats had free access to high fat diet and water for 9 weeks. No significant difference was found in body weight (BW), total abdominal fat, food intake and food utilization rate between OVX control group and 20VX groups. However, the fasting blood glucose and blood lipid levels were significantly higher in 20VX groups than in OVX control group (P〈0.05). Intraperitoneal glucose tolerance test (IGTI") showed that the area under AUC was smaller in 20VX groups than in OVX control group (P〈0.05). These findings showed that soy isoflavone crude extract supplementation can improve glucose tolerance and prevent high fat diet-induced insulin resistance in ovariectomized rats. 展开更多

Hypoglycemic effects were investigated in the ethanolic extract of fruits of Phyllanthus emblica Linn. with long evans rats (100-120 g). Residual gut sucrose content, gut perfusion and disaccharidase activity studies were carried out to assess these activities. The residual gut sucrose content study was carried out to reveal the amount of sucrose remaining in the gastro intestinal tract (GIT) at six different segments. The ethanolic fruit extract of Phyllanthus emblica Linn. significantly increases the residual sucrose content throughout the gut after sucrose ingestion. Study on intestinal enzyme activity showed that the extract caused a significant (p 0.05), dose dependent inhibition of intestinal disaccharidase enzyme activity. Evaluation of the effect of the plant extract on intestinal enzyme activity showed significant inhibition of intestinal disaccharidase activity, suggesting that reduction of sucrose absorption may be partly related to the inhibition of disaccharidase activity in the gut. The gut perfusion analysis showed that the extract significantly reduced intestinal glucose absorption. The results of the present study suggest that ethanolic extract of Phyllanthus emblica Linn. has significant antidiabetic effects in a dose dependent manner and this may be effective in the treatment of diabetes. 展开更多

This study aimed to evaluate the effect of water extracts from sweet potato [Ipomoea batatas (L.) Lamarck] on seed germination of Ageratum conyzoides L. under controlled conditions. The aqueous was produced from plant parts i.e., roots, stems, and leaves of sweet potato at concentrations of 0.025, 0.050, 0.075, and 0.100 g·mL-1. The results showed that the plant parts of sweet potato all contained allelopathic substances, which showed high-concentration inhibition and low-concentration promotion of seed germination of A. conyzoides. When the aqueous extract concentrations were 0.050, 0.075, and 0.100 g·mL-1, the germination of A. conyzoides seeds was inhibited, while the germination was promoted at a concentration of 0.025 g·mL-1. This shows that when the planting density of sweet potato is large, it can form an obvious prevention and control effect on A. conyzoides, and thus improve herbicide resistance management. 展开更多

Four kinds of assays were used to study the effect of a fat-soluble extract of spinach powder (SPFE) on the proliferation of human gastric adenocareinoma cell line (SGC-7901) in vitro. These studies included: (Ⅰ) cell growth assay, (Ⅱ) colony forming assay, (Ⅲ) MTT colorimetric assay, and (Ⅳ) 3H-TdR incorporation assay. The concentrations of SPFE expressed as the level of β-carotene in the medium were 2×10-8, 2×10-7 and 2×10-6 mol/L β-carotene in assays (Ⅰ)~(Ⅲ), but 4×10- 8, 4×10-7 and 4×10-6 mol/L β-caretene in assay (Ⅳ) respectively. The results indicated that SPFE inhibited the prolifendion and colony forming ability of SGC-7901 cells. And in MTT assay, SPFE inhibited the viability of SGC7901 cells, but no inhibitory effect of SPFE was observed on the viability of lymphocytes in peripheral blood of healthy people. Finally, in the 3H-TdR incorporation test, both SPFE and β-carotene showed significant inhibitory effects on DNA synthesis in SGC-7901 cells, but SPFE was more effective than β-carotene. 展开更多

[Objective] This study aimed to investigate the allelopathic effects of Torreya fargesii aril. [Method] By indoor bioassay, the effects of different con-centrations (10, 20,40, 80 g/L) of aqueous extract of T. fargesii aril on seed germination rate, seedling root length, seedling height, fresh weight, chlorophyll content, malondialdehyde (MDA) content and anti oxidative enzyme activities of radish, mustard and cabbage were analyzed and compared. [ Result] Different concentrations of aqueous extract of T. fargesii aril inhibited seed germination and seedling growth of three crops, and the inhibitory effects were enhanced with the increase of concentration. Seedling height and fresh weight of three crops were improved by low concentrations of aqueous extract of T. fargesii aril and inhibited by high concentrations of aqueous extract. Overall, aqueous extract of T. fargesii aril exhibited allelopathic inhibitory effects on three crops, and the level of allelopath-ic inhibitory effects demonstrated a descending order of radish 〉 mustard 〉 cabbage. Compared with the control group, 80 g/L aqueous extract of T. fargesii aril almost significantly reduced seed germination rate, seedling root length, seedling height, fresh weight and chlorophyll content, and significantly improved MDA con-tent and antioxidative enzyme (SOD, CAT, POD) activities of radish, mustard and cabbage (P 〈 0 .0 5 ) . [ Conclusion] Aqueous extract of T. fargesii aril exhibited remarkable allelopathic inhibitory effects on seed germination of radish, mustard and cabbage, which indicated that there might be dormancy-associated germination in-hibiting allelochemicals in T. fargesii aril. This study provided theoretical basis for subsequent clarification of the mechanism of seed dormancy of T. fargesii. 展开更多

The apparent and thermodynamic extraction equilibrium constants and the solubility parameters of the extraction complexes were obtained. The relationship between apparent equilibrium constants Kex and the physical parameters of solvents can be derived. The results indicated that the different solvents affect the apparent extraction equilibrium constants. The solubility parameters of the extraction complexes are nearly the same and not sensitive to rare earth ions. 展开更多
