Nerve cell metabolism in post brain ischemia depends on increased microcirculation perfusion and transport function of microvascular endothelial cells. In the present study, a rat model of middle cerebral artery occlusion was established to investigate the influence of electroacupuncture (EA) on hippocampal CA1 cerebral blood flow and glucose transporter 1 (GLUT1) expression in the microvascular endothelial cells. Following EA at Neiguan (PC 6), the cerebral blood flow in the ischemic hippocampal CA1 region was significantly elevated, the number and microvascular integrated absorbance of the GLUTl-positive cells were significantly increased, nerve cell damage was ameliorated, and GLUT1 protein expression in the ischemic hippocampus was significantly increased. Results demonstrate that EA increased the cerebral blood flow of the hippocampal CA1 region and improved the glucose transport function, thereby attenuating neuronal injuries. 展开更多

The changes of blood perfusion and oxygen transport in tumors during tumor vascular normalization are studied with 3-dimensional mathematical modeling and numerical simulation. The models of tumor angiogenesis and vascular-disrupting are used to simulate "un-normalized" and "normalized" vasculatures. A new model combining tumor hemodynamics and oxygen transport is developed. In this model, the intravasculartransvascular-interstitial flow with red blood cell(RBC) delivery is tightly coupled, and the oxygen resource is produced by heterogeneous distribution of hematocrit from the flow simulation. The results show that both tumor blood perfusion and hematocrit in the vessels increase, and the hypoxia microenvironment in the tumor center is greatly improved during vascular normalization. The total oxygen content inside the tumor tissue increases by about 67%, 51%, and 95% for the three approaches of vascular normalization,respectively. The elevation of oxygen concentration in tumors can improve its metabolic environment, and consequently reduce malignancy of tumor cells. It can also enhance radiation and chemotherapeutics to tumors. 展开更多

The influence of Reynolds number and mass flow ratio etc. parameters on the velocity field and mass transport were calculated. The computational results predict that the blood flow factors affect the macromolecular distribution and the transport across the wall, it shows that hemodynamic play an important role in the process of atherosclerosis. The LDL and Albumin concentration on the wall varies most greatly in flow bifurcation area where the wall shear stress varies greatly at the branching vessel and the atherosclerosis often appears there. 展开更多

The barriers present in the interfaces between the blood and the central nervous system form a major hurdle for the pharmacological treatment of central nervous system injuries and diseases.The family of ATP-binding cassette(ABC)transporters has been widely studied regarding efflux of medications at blood-central nervous system barriers.These efflux transporters include P-glycoprotein(abcb1),‘breast cancer resistance protein'(abcg2)and the various‘multidrug resistance-associated proteins'(abccs).Understanding which efflux transporters are present at the blood-spinal cord,blood-cerebrospinal fluid and cerebrospinal fluid-spinal cord barriers is necessary to determine their involvement in limiting drug transfer from blood to the spinal cord tissue.Recent developments in the blood-brain barrier field have shown that barrier systems are dynamic and the profile of barrier defenses can alter due to conditions such as age,disease and environmental challenge.This means that a true understanding of ABC efflux transporter expression and localization should not be one static value but instead a range that represents the complex patient subpopulations that exist.In the present review,the blood-central nervous system barrier literature is discussed with a focus on the impact of ABC efflux transporters on:(i)protecting the spinal cord from adverse effects of systemically directed drugs,and(ii)limiting centrally directed drugs from accessing their active sites within the spinal cord. 展开更多

The computational results predict that the vortex of the secondary flow is time-dependent in the aortic arch. The concentration of macromolecule concentrates at the region of sharp curve, and the wall concentration at the outer part is higher than that at the inner part. Atherosclerosis and thrombosis are prone to develop in such regions with sharp flow. 展开更多

This paper formulates an efficient numerical method for solving the convection diffusion solute transport equations coupled to blood flow equations in vessel networks.The reduced coupled model describes the variations of vessel cross-sectional area,radially averaged blood momentum and solute concentration in large vessel networks.For the discretization of the reduced transport equation,we combine an interior penalty discontinuous Galerkin method in space with a novel locally implicit time stepping scheme.The stability and the convergence are proved.Numerical results show the impact of the choice for the steady-state axial velocity profile on the numerical solutions in a fifty-five vessel network with physiological boundary data. 展开更多

Temperature Management of Red Blood Cell Solution Transported by Car for Transfusion at Home

However, there are no regulations ... <strong>Objective:</strong> In Japan, red blood cell (RBC) solution is usually transported by car from a medical institution to the patient’s house for home transfusion. However, there are no regulations for transporting blood by car in the medical setting. Therefore, we assessed and compared the methods (containers) used for transporting the RBC solution by car. <strong>Materials and Methods:</strong> Irradiated RBC solution samples (280 mL) supplied by the Japan Red Cross Society were each divided into two bags. The quality of blood transported by car (1 - 2 hours) in an active transport refrigerator (ATR) (control group) was compared with that transported in a cooler, or styrofoam box (study group). We tested the hemolytic effects of transportation by car, storage, and filtration through a transfusion set on the lactate dehydrogenase (LD) levels in the RBC solutions. <strong>Results:</strong> Post-filtered LD levels were significantly higher in the RBC solutions transported in a cooler-box with inadequate temperature control when compared to those transported in an ATR with optimal temperature control. However, under conditions of optimal temperature control, the post-filtered LD levels were comparable in the control and study (both cooler and styrofoam boxes) group RBC solutions. <strong>Conclusion:</strong> Temperature management is critical for the maintenance of the quality of the RBC solution transported by car. 展开更多

达格列净对2型糖尿病非增殖期视网膜病变的影响

目的:探讨达格列净对2型糖尿病非增殖期视网膜病变(non-proliferative diabetic retinopathy,NPDR)的影响。方法:选取2021年10月—2023年6月湖南师范大学附属湘东医院收治的88例2型糖尿病NPDR患者。随机将其分为观察组及对照组,各44例... 目的:探讨达格列净对2型糖尿病非增殖期视网膜病变(non-proliferative diabetic retinopathy,NPDR)的影响。方法:选取2021年10月—2023年6月湖南师范大学附属湘东医院收治的88例2型糖尿病NPDR患者。随机将其分为观察组及对照组,各44例。两组均给予常规降糖治疗,观察组加用达格列净治疗。比较两组治疗前后血糖指标及相关指标。分析血管内皮生长因子(vascular endothelial growth factor,VEGF)与血糖波动指标的相关性。结果:治疗后,两组空腹血糖(fasting blood glucose,FBG)、餐后2 h血糖(2 h postprandial blood glucose,2 h PBG)、糖化血红蛋白(glycosylated hemoglobin,HbA1c)、血糖水平标准差(standard deviation of blood glucose,SDBG)、餐后血糖波动幅度(postprandial glucose excursion,PPGE)、最大血糖波动幅度(large amplitude glycemic excursion,LAGE)均下降,观察组SDBG、PPGE、LAGE均低于对照组,差异有统计学意义(P<0.05)。治疗后,两组VEGF、黄斑中心凹厚度均下降,最佳矫正视力(best-corrected visual acuity,BCVA)升高,观察组VEGF、黄斑中心凹厚度均低于对照组,BCVA高于对照组,差异有统计学意义(P<0.05)。Pearson相关性分析结果显示:VEGF与SDBG、PPGE、LAGE均呈正相关(P<0.05)。结论:达格列净能够改善血糖波动,降低VEGF水平,对2型糖尿病NPDR有保护作用。 展开更多

试验旨在研究道路运输对羔羊体重、血液生化指标和临床指标的影响,为生产中有效缓解羔羊运输应激提供理论依据。选取平均体重(21.52±1.22)kg健康4月龄羔羊10只进行8 h的长途运输,分别在运输前后进行称重、静脉采血,并测定体温、呼... 试验旨在研究道路运输对羔羊体重、血液生化指标和临床指标的影响,为生产中有效缓解羔羊运输应激提供理论依据。选取平均体重(21.52±1.22)kg健康4月龄羔羊10只进行8 h的长途运输,分别在运输前后进行称重、静脉采血,并测定体温、呼吸频率、脉搏等临床指标。结果表明:①道路运输使羔羊体温、呼吸和脉搏频率极显著升高(P<0.01),体重降低了15.31%(P<0.05)。②道路运输极显著升高了羔羊血清中促肾上腺皮质激素(ACTH)和乳酸脱氢酶(LDH)水平(P<0.01),显著升高了皮质醇(COR)和肌酸激酶(CK)水平(P<0.05),血清中谷丙转氨酶(ALT)水平无显著变化(P>0.05)。③道路运输显著降低了羔羊血清中免疫球蛋白A(IgA)、免疫球蛋白G(IgG)水平(P<0.05),显著升高了白介素-6(IL-6)水平(P<0.05),血清中免疫球蛋白M(IgM)、干扰素-γ(IFN-γ)、肿瘤坏死因子α(TNF-α)、白介素-1β(IL-1β)水平与运输前差异不显著(P>0.05)。④道路运输显著降低了羔羊血清中葡萄糖(GLU)、总蛋白(TP)、白蛋白(ALB)水平(P<0.05)。⑤道路运输显著降低了羔羊血清中红细胞总数(RBC)、红细胞压积(HCT)、血红蛋白含量(MCH)(P<0.05),红细胞平均体积(MCV)极显著降低(P<0.01),白细胞总数(WBC)显著升高(P<0.05)。综上所述,羔羊经过8 h道路运输后发生了明显的应激反应,导致羔羊体重下降,机体代谢增强,炎症反应增强,免疫系统受到了一定程度损伤。 展开更多

DCE-MRI定量参数鉴别诊断肺部肿瘤良恶性的价值

目的:探究动态增强磁共振成像(DCE-MRI)定量参数鉴别诊断肺部肿瘤良恶性的价值。方法:选取中国人民解放军陆军第七十三集团军医院2022年2月—2023年4月收治的100例肺部肿瘤患者,所有患者入院后均接受常规磁共振成像(MRI)平扫、DCE-MRI... 目的:探究动态增强磁共振成像(DCE-MRI)定量参数鉴别诊断肺部肿瘤良恶性的价值。方法:选取中国人民解放军陆军第七十三集团军医院2022年2月—2023年4月收治的100例肺部肿瘤患者,所有患者入院后均接受常规磁共振成像(MRI)平扫、DCE-MRI扫描及病理检查。比较良恶性肺部肿瘤DCE-MRI定量渗透性参数[血液回流常数(K_(ep))、容积转运常数(K^(trans))、血浆容积分数(V_(p))、血管外细胞外间隙容积分数(V_(e))],分析DCE-MRI定量参数鉴别诊断肺部肿瘤良恶性的价值。结果:100例肺部肿瘤患者中,经病理检查确诊为41例良性肿瘤,59例恶性肿瘤。恶性肿瘤组的K^(trans)、K_(ep)均比良性肿瘤组高(P<0.05)。两组V_(e)、V_(p)比较,差异均无统计学意义(P>0.05)。绘制受试者操作特征(ROC)曲线,结果显示,K^(trans)、K_(ep)单独及联合诊断肺部肿瘤良恶性的曲线下面积(AUC)为0.835、0.872、0.945,均有一定诊断价值。当K^(trans)、K_(ep)分别为0.525 min^(-1)、0.920 min^(-1)时,可获得最佳诊断效能,且二者联合诊断的价值较高。结论:DCE-MRI定量参数鉴别诊断肺部肿瘤良恶性的价值较高,可通过检测其K^(trans)、K_(ep)值来判断肺部肿瘤的良恶性。 展开更多

目的探讨Mg^(2+)转运蛋白1(magnesium transporter protein 1,MagT1)在系统性红斑狼疮(systemic lupuserythematosus,SLE)患者外周血T淋巴细胞中的表达情况,并阐述其临床意义。方法选取西双版纳傣族自治州人民医院2019年1月~2021年6月... 目的探讨Mg^(2+)转运蛋白1(magnesium transporter protein 1,MagT1)在系统性红斑狼疮(systemic lupuserythematosus,SLE)患者外周血T淋巴细胞中的表达情况,并阐述其临床意义。方法选取西双版纳傣族自治州人民医院2019年1月~2021年6月收治的79例SLE患者作为研究对象,依据SLE疾病活动指数(SLE disease activityindex,SLEDAI)评分将SLE患者分为中重度组(SLEDAI≥10,n=32)和轻度组(SLEDAI<10,n=47),另选取同期体检健康者40例作为对照组。记录患者临床生化和血清免疫相关指标水平;采用实时定量PCR(quantitative realtimePCR,qRT-PCR)法和Western Blot法检测患者外周血T淋巴细胞中MagT1 mRNA和蛋白表达情况;Pearson相关性分析外周血T淋巴细胞MagT1蛋白与生化免疫相关指标表达的关系;绘制受试者工作特征(receiver operatingcharacteristic,ROC)曲线分析外周血T淋巴细胞MagT1蛋白表达对SLE及其严重程度的诊断价值。结果与对照组比较,轻度组和中重度组患者外周血T淋巴细胞中MagT1 mRNA[0.65(0.36,0.99),0.23(0.07,0.36)vs 1.20(0.83,1.37)]和MagT1蛋白[0.35(0.22,0.42),0.22(0.15,0.27)vs 0.53(0.40,0.63)]表达水平明显降低,差异具有统计学意义(Z=5.247,7.078;5.128,7.257,均P<0.05);与轻度组比较,中重度组患者外周血T淋巴细胞中MagT1 mRNA和蛋白表达水平明显降低,差异具有统计学意义(Z=5.169,3.599,均P<0.05)。Pearson相关性结果显示,外周血T淋巴细胞MagT1蛋白表达水平与血清血红蛋白(hemoglobin,HGB)及补体3(complement 3,C3)和补体4(complement 4,C4)水平呈正相关(r=0.496,0.637,0.588,均P<0.05);与红细胞沉降速率(erythrocyte sedimentation rate,ESR)、超敏C反应蛋白(hypersensitive-C reactive protein,hs-CRP)、免疫球蛋白(immunoglobulin G,IgG)水平和SLEDAI呈负相关(r=-0.598,-0.476,-0.646,-0.514,均P<0.05)。ROC曲线结果显示,外周血T淋巴细胞MagT1蛋白表达水平诊断SLE曲线下面积为0.893(95%CI:0.838~0.948),敏感度和特异度分别为90.0%,67.1%,诊断SLE严重程度曲线下面积为0.739(95%CI:0.631~0.848),敏感度和特异度分别为57.4%,84.4%。结论外周血T淋巴细胞MagT1在SLE患者中表达下调,且与SLE严重程度呈负相关,对SLE诊断和病情严重程度具有一定的诊断价值。 展开更多

目的探讨化痰祛瘀法对冠心病稳定型心绞痛患者外周血线粒体钙转运与动脉粥样硬化指数(AI)的影响。方法选取冠心病稳定型心绞痛患者100例,按随机数表法均分为对照组和观察组,对照组采用西医常规治疗,治疗组在西医常规治疗基础上+化痰祛... 目的探讨化痰祛瘀法对冠心病稳定型心绞痛患者外周血线粒体钙转运与动脉粥样硬化指数(AI)的影响。方法选取冠心病稳定型心绞痛患者100例,按随机数表法均分为对照组和观察组,对照组采用西医常规治疗,治疗组在西医常规治疗基础上+化痰祛瘀方治疗,连续治疗8周;分别于治疗前及治疗结束时,采用西雅图心绞痛量表(SAQ评分)评估两组患者的心绞痛程度,双试剂酶法检测两组患者甘油三酯(TG)、总胆固醇(TC)及低密度脂蛋白胆固醇水平(LDL-C),全自动血流变检测仪测定血液流变学指标,电化学发光法测定血清线粒体钙离子单向转运蛋白(MCU)水平,颈动脉超声检测两组患者AI,并比较两组患者的临床疗效。结果治疗结束后,两组患者躯体受限程度、心绞痛稳定状态、心绞痛发作情况、治疗满意程度及疾病认识程度等SAQ评分均较治疗前上升,且观察组均高于对照组(P<0.05);两组患者TG、TC、LDL-C治疗后水平均较治疗前下降,且观察组低于对照组(P<0.05);两组全血高切黏度、全血低切黏度、血浆黏度及纤维蛋白原治疗后水平均较治疗前下降,且观察组均低于对照组(P<0.05);两组患者治疗后MCU水平较治疗前上升、AI较治疗前下降,且治疗后观察组MCU水平高于对照组、AI水平低于对照组(P<0.05);治疗后,观察组临床总疗效高于对照组,差异有统计学意义(P<0.05)。结论化痰祛瘀法对冠心病稳定型心绞痛患者有较好疗效,其机制可能与改善外周血线粒体钙转运功能及AI有关。 展开更多

Time dependent dispersion of nanoparticles in blood vessels

The dispersion of intravasculary injected nanoparticles can be efficiently described by introducing an effective diffusion coefficient Deff which quantifies the longitudinal mass transport in blood vessels. Here, the original work of Gill and Sankarasubramanian was modified and extended to include 1) the variati- on over time of Deff;2) the permeability of the blood vessels and 3) non-Newtonian rheology of blood. A general solution was provided for Deff depending on space (?), time (?), plug radius (?c) and a subset of permeability parameters. It was shown that increasing the vessel plug radius (thus hematocrit) or permeability leads to a reduction in Deff, limiting the transport of nanoparticles across those vessels. It was also shown that the asymptotic time beyond which the solution attains the steady state behaviour is always independent of the plug radius and wall permeability. The analysis presented can more accurately predict the transport of nanoparticles in blood vessels, compared to previously developed models. 展开更多

The purpose of this study was to evaluate the ability of aqueous extract of Aloe barbadensis Miller (Aloe vera) on oxidative damage and Anion Exchanger 1 (AE1, also known as Band 3) expression in human erythrocytes exposed to the water soluble free radical initiator 2.2’-azobis-2-amidinopropano dihydrochloride (AAPH). In addition, total phenolic compounds in the extracts were determined as catechin equivalent and the various antioxidant activities were compared to natural and synthetic standard antioxidants such as BHA and ascorbic acid. Since Aloe vera extract did not cause a consumption of the cytosolic antioxidant, glutathione (GSH) when it was direct incubated with GSH in basic aerated aqueous solution, this indicates that Aloe vera extract does not proceed auto oxidation at this experimental condition. Furthermore, Aloe vera extract prevent the consumption of GSH, in radical treated RBCs. It also inhibit consumption of GSH when it was direct incubated with AAPH. Aloe vera gel extract inhibits the generation of diphenyl-2-picrylhy-drazyl (DPPH) and the scavenging activity was increased in a dose dependent manner. Aloe vera extract was shown the similar reducing power than standards BHT and ascorbic acid. Biochemical analysis by SDS-PAGE and western blotting showed that AAPH-induced oxidative stress increased the susceptibility of AE1 to proteolytic degradation. Of note, our data evidenced that Aloe vera treatment was able to partially restore the normal RBC membrane protein profiles in a dose-dependent manner. These results clearly demonstrate the antioxidative activity of Aloe vera gel extract that might be ascribed to a synergistic action of the bioactive compounds contained therein. 展开更多

Objective To investigate the potential involvement of DMT1(IRE) protein in the brain vascular system in vivo during Pb exposure. Methods Three groups of male Sprague-Dawley rats were exposed to Pb in drinking water, among which two groups were concurrently administered by oral gavage once every other day as the low and high Fe treatment group, respectively, for 6 weeks. At the same time, the group only supplied with high Fe was also set as a reference. The animals were decapitated, then brain capillary-rich fraction was isolate from cerebral cortex. Western blot method was used to identify protein expression, and RT-PCR to detect the change of the m RNA. Results Pb exposure significantly increased Pb concentrations in cerebral cortex. Low Fe dose significantly reduced the cortex Pb levels, However, high Fe dose increased the cortex Pb levels. Interestingly, changes of DMT1(IRE) protein in brain capillary-rich fraction were highly related to the Pb level, but those of DMT1(IRE) m RNA were not significantly different. Moreover, the consistent changes in the levels of p-ERK1/2 or IRP1 with the changes in the levels of DMT1(IRE). Conclusion These results suggest that Pb is transported into the brain through DMT1(IRE), and the ERK MAPK pathway is involved in DMT1(IRE)-mediated transport regulation in brain vascular system in vivo. 展开更多
