本试验旨在研究低蛋白日粮中不同比例氨基酸对藏羊背腰最长肌肉品质、氨基酸和脂肪酸组成以及维生素和矿物质含量的影响。选取90只体重相近[(15.60±0.10)kg],健康的高原型藏羊公羔,随机分为3组,每组30只,每组5个重复,每个重复6只... 本试验旨在研究低蛋白日粮中不同比例氨基酸对藏羊背腰最长肌肉品质、氨基酸和脂肪酸组成以及维生素和矿物质含量的影响。选取90只体重相近[(15.60±0.10)kg],健康的高原型藏羊公羔,随机分为3组,每组30只,每组5个重复,每个重复6只羊。日粮精料中蛋白水平为12%,3组分别为LP-L组(赖氨酸与蛋氨酸为1∶1);LP-M组(赖氨酸与蛋氨酸为2∶1);LP-H组(赖氨酸与蛋氨酸为3∶1)。试验期共97 d,其中7 d预饲期,90 d正试期。结果表明:1)解冻损失指标LP-L组显著低于LP-H组(P<0.05),蒸煮损失和剪切力指标LP-L组显著低于LP-M和LPH组(P<0.05);2)添加不同氨基酸比例对各组肌肉中氨基酸的组成没有显著影响(P>0.05);3)在不饱和脂肪酸中,反式亚油酸、二十二碳三烯酸和十五碳烯酸含量LP-L组显著高于LP-M组(P<0.05);十七碳烯酸、亚油酸和α-亚麻酸LP-L组显著高于LP-M和LP-H组(P<0.05);4)α-维生素E、γ-维生素E和维生素E指标LP-L组显著高于LP-M组(P<0.05),矿物质元素钙LP-L组显著高于LP-M组(P<0.05)。综上所述,当日粮中赖氨酸与蛋氨酸比例为1∶1时能提高不饱和脂肪酸含量以及肉质中维生素和矿物质元素含量;且对氨基酸组成没有不良影响,并能有效改善肉品质。 展开更多

土鳖虫是我国传统中药,分布于河南、河北、江苏、湖南等地,具有破血逐瘀、续筋接骨的功效。土鳖虫中化学成分种类丰富,主要包括蛋白质及多肽类、氨基酸类、脂肪酸类、生物碱、无机元素、核苷类等。现代药理研究表明,土鳖虫具备抗凝血并... 土鳖虫是我国传统中药,分布于河南、河北、江苏、湖南等地,具有破血逐瘀、续筋接骨的功效。土鳖虫中化学成分种类丰富,主要包括蛋白质及多肽类、氨基酸类、脂肪酸类、生物碱、无机元素、核苷类等。现代药理研究表明,土鳖虫具备抗凝血并防止血栓形成、调节血脂的作用,此外,还具有抗肿瘤、抗炎、增强免疫力、抗氧化等作用。近年来,土鳖虫等动物类中药在临床应用方面备受关注。本文对土鳖虫化学成分和药理作用的研究现状进行总结,在此基础上分析功效作用与化学成分之间的联系,发现蛋白质及多肽类成分与抗凝血、调血脂作用关联紧密,生物碱类成分与抗菌抗炎作用相关。并从传统功效、现代药理、化学成分可测性、中药配伍等角度预测分析其质量标志物(quality marker,Q-Marker),初步确定土鳖虫活性肽组分、纤溶活性蛋白、脂肪酸类成分、生物碱、核苷类成分等可作为其质量标志物。中药质量标志物的研究为鉴定土鳖虫的真假优劣提供依据,也为后续开发土鳖虫新剂型、制定质量标准提供理论基础。 展开更多

Background Goose, descendants of migratory ancestors, have undergone extensive selective breeding, resulting in their remarkable ability to accumulate fat in the liver and exhibit a high tolerance for significant energy intake. As a result, goose offers an excellent model for studying obesity, metabolic disorders, and liver diseases in mammals. Although the impact of the three-dimensional arrangement of chromatin within the cell nucleus on gene expression and transcriptional regulation is widely acknowledged, the precise functions of chromatin architecture reorganization during fat deposition in goose liver tissues still need to be fully comprehended.Results In this study, geese exhibited more pronounced changes in the liver index and triglyceride(TG) content following the consumption of the high-fat diet(HFD) than mice without significant signs of inflammation. Additionally, we performed comprehensive analyses on 10 goose liver tissues(5 HFD, 5 normal), including generating highresolution maps of chromatin architecture, conducting whole-genome gene expression profiling, and identifying H3K27ac peaks in the livers of geese and mice subjected to the HFD. Our results unveiled a multiscale restructuring of chromatin architecture, encompassing Compartment A/B, topologically associated domains, and interactions between promoters and enhancers. The dynamism of the three-dimensional genome architecture, prompted by the HFD, assumed a pivotal role in the transcriptional regulation of crucial genes. Furthermore, we identified genes that regulate chromatin conformation changes, contributing to the metabolic adaptation process of lipid deposition and hepatic fat changes in geese in response to excessive energy intake. Moreover, we conducted a cross-species analysis comparing geese and mice exposed to the HFD, revealing unique characteristics specific to the goose liver compared to a mouse. These chromatin conformation changes help elucidate the observed characteristics of fat deposition and hepatic fat regulation in geese under conditions of excessive energy intake.Conclusions We examined the dynamic modifications in three-dimensional chromatin architecture and gene expression induced by an HFD in goose liver tissues. We conducted a cross-species analysis comparing that of mice. Our results contribute significant insights into the chromatin architecture of goose liver tissues, offering a novel perspective for investigating mammal liver diseases. 展开更多

理中汤加减治疗非酒精性脂肪性肝病临床研究

目的:观察理中汤加减治疗非酒精性脂肪性肝病的临床疗效。方法:将110例非酒精性脂肪性肝病患者按照随机数字表法分为观察组和对照组,每组各55例。对照组给予多烯磷脂酰胆碱治疗,观察组在对照组治疗的基础上给予理中汤加减。比较两组患... 目的:观察理中汤加减治疗非酒精性脂肪性肝病的临床疗效。方法:将110例非酒精性脂肪性肝病患者按照随机数字表法分为观察组和对照组,每组各55例。对照组给予多烯磷脂酰胆碱治疗,观察组在对照组治疗的基础上给予理中汤加减。比较两组患者治疗前后肝功能指标[丙氨酸氨基转移酶(alamine aminotransferase, ALT)、门冬氨酸氨基转移酶(aspartate aminotransferase, AST)、谷氨酰胺转肽酶(glutamine transpeptidase, GGT)]、血脂指标[总胆固醇(total cholesterol, CHOL)、三酰甘油(triglyceride, TG)、低密度脂蛋白(low density lipoprotein, LDL-C)]、血糖指标[空腹血糖(fasting blood-glucose, FBG)、空腹胰岛素(fasting insulin, FINS)、糖化血红蛋白A1c(hemoglobin A1c, HbA1c)、胰岛素抵抗指数(homa-insulin resistance index, HOMA-IR)]、肝脏硬度值(liver stiffness measurement, LSM)、受控衰减参数值(controlled attenuation parameter, CAP)及中医证候积分变化情况。结果:观察组有效率为85.45%,高于对照组的54.55%(P<0.05)。两组患者治疗后ALT、AST、GGT水平低于本组治疗前,治疗后观察组低于对照组(P<0.05)。两组患者治疗后FBG、HOMA-IR、FINS、HbA1c低于本组治疗前,且治疗后观察组FBG、HOMA-IR低于对照组(P<0.05),治疗后观察组FINS、HbA1c水平与对照组比较,差异无统计学意义(P>0.05)。两组患者治疗后CHOL、TG、LDL-C水平低于本组治疗前,治疗后观察组低于对照组(P<0.05)。两组患者治疗后LSM、CAP水平低于本组治疗前,且治疗后观察组CAP水平低于对照组(P<0.05),治疗后观察组LSM水平与对照组比较,差异无统计学意义(P>0.05)。两组患者治疗后中医证候积分低于本组治疗前,治疗后观察组低于对照组(P<0.05)。结论:理中汤加减治疗非酒精性脂肪性肝病,可有效改善患者肝功能指标、血脂水平及胰岛素抵抗情况,缓解临床症状,提高生活质量。 展开更多

[Objectives]To understand the content changes of fatty acid compositions and seedling growths during seed germinations of Cassia obtusifolia L.[Methods]The fatty acid compositions of germinations and seedling growths of C.obtusifolia seeds treated with different concentrations of gibberellin(GA3)and naphthalene acetic acid(NAA)were analyzed using gas chromatograph-mass spectrometer(GC-MS),and the germination rates,root lengths,plant heights and fresh weights of their seedlings were analyzed.[Results]The contents of eicosenoic acid,α-linoleic acid,arachidic acid,eicosadienoic acid and eicosapentaenoic acid of C.obtusifolia seeds treated with different concentrations of GA3 and NAA decreased with germination time.There were significant differences between the ratios of germinations of C.obtusifolia seeds treated with different concentrations of GA3 and NAA.GA3 and NAA of different ratio treatments could promote seedling heights and fresh weights of C.obtusifolia,but increases of seedling heights and fresh weights were different.Eicosenoic acid,α-linoleic acid,arachidic acid,eicosadienoic acid and eicosapentaenoic acid were remarkably negative correlation with seedling heights and fresh weights.[Conclusions]The fatty acid components of C.obtusifolia seed germination and growth of their seedlings were affected.It indicated that GA3 and NAA treatments can affect fatty acid metabolism,the germination rates,root lengths,plant heights and fresh weights of C.obtusifolia.It is intended to provide a theoretical basis and technological reference for standardized cultivated techniques of C.obtusifolia. 展开更多

The pharmaceutical effects of n-3 polyunsaturated fatty acids(n-3 PUFAs) as dietary nutrients on human health and diseases have gained much attention and are investigated for decades. Docosahexaenoic acid(DHA), eicosapentaenoic acid(EPA) and docosapentaenoic acid(DPA) are the three major n-3 PUFAs enriched in marine organisms, such as fish, shrimp, algae, and so on. It has been well known that n-3 PUFAs, especially DHA and EPA, are beneficial in reducing the risk of cardiovascular and cerebrovascular diseases. Accumulating evidence suggests that n-3 PUFAs might cure inflammatory diseases through several mechanisms, such as plasma membrane remodeling of lymphocytes, down-regulating pro-inflammatory cytokines, and alternating adhesion molecule expressions. Several molecular targets of n-3 PUFAs on immune-regulation have also been identified, such as GPR120(FFA4), protein kinase C(PKC), and PPAR-γ. However, it remains inconclusive if dietary n-3 PUFAs function the same both in vitro and in vivo based on cohort studies. This review will focus on the molecular targets and mechanisms of anti-inflammatory and immunomodulatory effects of n-3 PUFAs on human health and diseases, such as obesity, tumor, diabetes, and autoimmune diseases. 展开更多

Non-alcoholic fatty liver disease(NAFLD) is a progressive disease of increasing public health concern.In western populations the disease has an estimated prevalence of 20%-40%,rising to 70%-90% in obese and type Ⅱ diabetic individuals.Simplistically,NAFLD is the macroscopic accumulation of lipid in the liver,and is viewed as the hepatic manifestation of the metabolic syndrome.However,the molecular mechanisms mediating both the initial development of steatosis and its progression through non-alcoholic steatohepatitis to debilitating and potentially fatal fibrosis and cirrhosis are only partially understood.Despite increased research in this field,the development of non-invasive clinical diagnostic tools and the discovery of novel therapeutic targets has been frustratingly slow.We note that,to date,NAFLD research has been dominated by in vivo experiments in animal models and human clinical studies.Systems biology tools and novel computational simulation techniques allow the study of large-scale metabolic networks and the impact of their dysregulation on health.Here we review current systems biology tools and discuss the benefits to their application to the study of NAFLD.We propose that a systems approach utilising novel in silico modelling and simulation techniques is key to a more comprehensive,better targeted NAFLD research strategy.Such an approach will accelerate the progress of research and vital translation into clinic. 展开更多

Our understanding of postharvest needle abscission physiology in conifers has greatly improved in the last decade. Abscission is initially triggered by root detachment, which begins a cascade of changes such as decreased water uptake, water potential, and auxins and increased membrane injury, ethylene, abscisic acid, volatile terpenes, and catalytic enzymes. Needle abscission is also affected by environmental factors. For example, a period of cold acclimation generally delays postharvest abscission. The aforementioned pieces of evidence, along with previous studies, strongly points to a role for plant lipids and fatty acids. Studies from other species have pointed out key roles in abscission and stress responses for a variety of phospholipids and galactolipids, which has not been studied in balsam fir. It is imperative to have an understanding of the role of plant lipids and fatty acids to further our overall understanding of the physiological mechanisms of postharvest abscission and needle abscission resistance. This review is an overview of membrane lipids and fatty acids, changes that occur postharvest and the interaction that lipids may have in the phenomenon of postharvest abscission. 展开更多

Among them, short-chain fatty acids (SCFAs), as important active products of gut bacteria, have been shown to exert anti-inflammatory and immunomodulatory effects and can play active roles as signaling molecules in the development of various intestinal and extraintestinal diseases, such as inflammatory bowel disease, colon cancer, multiple sclerosis, hypertension, allergic airway disease, obesity, diabetes, kidney disease, rheumatoid arthritis, etc. In this way, modulation of the intestinal microbiota and metabolism-active substances has gradually become a popular therapeutic method for many diseases of organs beyond the gut. The prevalence of obesity and related conditions like non-alcoholic fatty liver disease(NAFLD) is increasing worldwide and therapeutic options are limited.Alternative treatment options are therefore intensively sought after.An interesting candidate is the natural polyphenol resveratrol(RSV) that activates adenosinmonophosphate-activated protein kinase(AMPK) and silent information regulation-2 homolog 1(SIRT1).In addition,RSV has known anti-oxidant and anti-inflammatory effects.Here,we review the current evidence for RSVmediated effects on NAFLD and address the different aspects of NAFLD and non-alcoholic steatohepatitis(NASH) pathogenesis with respect to free fatty acid(FFA) flux from adipose tissue,hepatic de novo lipogenesis,inadequate FFA β-oxidation and additional intra- and extrahepatic inflammatory and oxidant hits.We review the in vivo evidence from animal studies and clinical trials.The abundance of animal studies reports a decrease in hepatic triglyceride accumulation,liver weight and a general improvement in histological fatty liver changes,along with a reduction in circulating insulin,glucose and lipid levels.Some studies document AMPK or SIRT1 activation,and modulation of relevant markers of hepatic lipogenesis,inflammation and oxidation status.However,AMPK/SIRT1-independent actions are also likely.Clinical trials are scarce and have primarily been performed with a focus on overweight/obese participants without a focus on NAFLD/NASH and histological liver changes.Future clinical studies with appropriate design are needed to clarify the true impact of RSV treatment in NAFLD/NASH patients. 展开更多

目的:观察吴门医派调脂颗粒治疗肥胖2型糖尿病(type 2 diabetes mellitus,T2DM)合并代谢相关脂肪性肝病(metabolic associated fatty liver disease,MAFLD)的临床疗效及对血清趋化素(Chemerin)水平的影响。方法:纳入55例肥胖T2DM合并MA... 目的:观察吴门医派调脂颗粒治疗肥胖2型糖尿病(type 2 diabetes mellitus,T2DM)合并代谢相关脂肪性肝病(metabolic associated fatty liver disease,MAFLD)的临床疗效及对血清趋化素(Chemerin)水平的影响。方法:纳入55例肥胖T2DM合并MAFLD患者,随机分为观察组(28例)和对照组(27例)。对照组采用常规西医治疗,观察组在此基础上加用调脂颗粒治疗。治疗12周后,观察两组体质指数(body mass index,BMI)、空腹血糖(fasting plasma glucose,FPG)、糖化血红蛋白(glucosylated hemoglobin,HbA1c)、胰岛素抵抗指数(homeostasis model assessment-insulin resistance,HOMA-IR)、血脂、肝功能指标及血清Chemerin水平。结果:治疗12周后,观察组BMI、FPG、HbA1c、谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)、血清总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)均较治疗前显著降低(P<0.01),血清Chemerin、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、白细胞介素8(interleukin-8,IL-8)水平亦较治疗前显著降低(P<0.01)。对照组BMI、FPG、HbA1c、ALT、AST、TC、TG、IL-8较治疗前下降(P<0.05),血清Chemerin、TNF-α水平较治疗前显著下降(P<0.01)。治疗后,观察组与对照组相比,BMI、FPG、TC、LDL-C、血清Chemerin、TNF-α、IL-8水平更低(P<0.05)。结论:吴门调脂颗粒治疗肥胖T2DM合并MAFLD能够减轻体质量、改善糖脂代谢,降低血清Chemerin水平,减轻炎症状态,提高临床疗效。 展开更多
