Obiective:To investigate the effects of Xiaoxianxiong Tang on the proliferation,invasion,apoptosis,and other cell biological behaviors of non-small cell lung cancer A549 cells.Methods:Human lung adenocarcinoma A549 cells were cultured in vitro,and the IC50 concentration and effective time of administration of Xiaoxianxiong Tang were determined by the CCK-8 assay to detect the inhibitory effect of Xiaoxianxiong Tang on A549 cells proliferation.The effect of the Xiaoxianxiong Tang on apoptosis was determined by flow cytometry;the apoptosis-related protein was detected via Western blot;the metastasis-related protein mRNA was detected by RT-PCR.Results:Xiaoxianxiong Tang significantly inhibited the proliferation viability,the invasive ability,and the clonogenic ability of A549 cells compared with the control group(P<0.001).Moreover,Xiaoxianxiong Tang significantly promoted the apoptosis of A549 cells(P<0.001).Xiaoxianxiong Tang significantly up-regulated Bax and down-regulated Bcl2 expression in A549 cells compared with the control group(P<0.01).The mRNA expression of MMP2 and MMP9 was significantly down-regulated by Xiaoxianxiong Tang compared with the control group(P<0.05).Conclusion:Xiaoxianxiong Tang has the effect of regulating the biological behavior of A549 cells,and Xiaoxianxiong Tang significantly inhibites the proliferation viability,colony formation,and invasion ability of lung cancer A549 cells. 展开更多

The effects of trace yttrium(Y)element on the microstructure,mechanical properties,and corrosion resistance of Mg-2Zn-0.1Mn-0.3Ca-xY(x=0,0.1,0.2,0.3)biological magnesium alloys are investigated.Results show that grain size decreases from 310 to 144µm when Y content increases from 0wt%to 0.3wt%.At the same time,volume fraction of the second phase increases from 0.4%to 6.0%,yield strength of the alloy continues to increase,and ultimate tensile strength and elongation decrease initially and then increase.When the Y content increases to 0.3wt%,Mg_(3)Zn_(6)Y phase begins to precipitate in the alloy;thus,the alloy exhibits the most excellent mechanical property.At this time,its ultimate tensile strength,yield strength,and elongation are 119 MPa,69 MPa,and 9.1%,respectively.In addition,when the Y content is 0.3wt%,the alloy shows the best corrosion resistance in the simulated body fluid(SBF).This investigation has revealed that the improvement of mechanical properties and corrosion resistance is mainly attributed to the grain refinement and the precipitated Mg_(3)Zn_(6)Y phase. 展开更多

Reagents are optimized for the simultaneous determination of trace amounts of Cu^(2+), Cd^(2+) and Co^(2+) in zinc sulfate solution, which contains an extremely large excess of Zn^(2+). First, the reagents and their doses for the experiment are selected according to the characteristics of the zinc sulfate solution. Then, the reagent doses are optimized by analyzing the influence of reagent dose on the polarographic parameters(i.e. half-wave potential E_(1/2) and limiting diffusion current I_p). Finally, the optimization results are verified by simultaneously determining trace amounts of Cu^(2+), Cd^(2+) and Co^(2+) in the presence of an extremely large excess of Zn^(2+). The determination results indicate that the optimized reagents exhibit wide linearity, low detection limits, high accuracy and good precision for the simultaneous determination of trace amounts of Cu^(2+), Cd^(2+) and Co^(2+) in the presence of an extremely large excess of Zn^(2+). 展开更多

Various biological indices were used to observe the effects of Tween-80 in combination with hyperthermia at different temperature (39-43℃) for different period of time (20-100 minutes) on human stomach cell line BGC-823. The results showed that Tween-80obviously reduced the activation energy of BGC-823 cells. Synergistic effect was observed if applied with heat at 39℃ with the increase in temperature and time,the inhibitory effect on the cancer cells was gradually intensified. The lethal rate of BGC-823 cells treated by heat at 41℃ in combination with Tween-80 was around 5.2 times as that treated by hyperthermia alone. The synergistic effect of heat at 41℃ for 100 min. in combination with Tween-80 was equivalent to the effect of 43℃ for 100 min.In other words, the critical temperature for BGC-823 cells was hereby reduced about 2℃. The measurement of membrane mobility, SDH activity etc.also showed that at 41℃ the synergistic effect of hyperthermia in combination with Tween-80 was the best, it exceeding the single effect of these factors in combination and showing effect of multiplication. The synergistic effect of heat at 41℃ in combination with Tween-80 was higher than that of heat at 41℃ in combination with MMC. It also demonstrated that the specific and sustained action on the inhibition of cancer cells did exist. These studies suggested that the synergistic mechanism of Tween-80 and hyperthermia probably was that both of them acted on the cell membrane system. Dehydroepiandrosterone (DHEA) is a weak androgen and is shown to have anti-cancer, anti-atherogenic, anti-adipogenic and anti-inflammatory effects on mouse, rat and rabbit models. However, human clinical trials data did not support animal findings and were inconclusive. These systemic differences in biological actions between rodents and humans were attributed to the low level of DHEA in rodents. In order to further understand the differences in biological functions between rodents and humans, we resorted to an in-vitroapproach involving mouse, rat and human cell lines to assess DHEA biological and anabolic functions separately and independently without systemic influence. Results indicated that DHEA was effective on mouse and rat cell lines but not on human cell lines, as observed in in-vivo studies. In addition, our in-vitrostudy showed that DHEA was able to induce myogenesis in mouse mesenchymal cells revealing its anabolic function, even though DHEA was considered as a weak androgen. This observation lent credence to the ban on DHEA by IOC medical commission, citing DHEA as an anabolic steroid. These in-vitro experiments suggested that the differences in biological actions of DHEA between rodents and humans existed not only in-vivo at the systemic level, but also in-vitro at the cellular level and thus paving the way to study the mechanism responsible for these differences at the cellular level itself. 展开更多

Moderate round cells were predominant an... A cell line designated as Ca 761-86 has been established from the solid mouse mammary cancer (Ca 761) by suspension culture. It has been passaged for more than 212 generations. Moderate round cells were predominant and most of them were mononuclear. Some characteristics of malignant cells and A-type viral-like particles were observed by electron microscopy. The results of cytochemical studies (DNA, RNA, SDH, 5' AMPase, ACP etc.) were comparable to the ultramicroscopic results. It multiplied approximately 27.4 fold on day 5 with mitotic index reaching 1.8% on day 3. This cell line was a hyperdiploid with karyotype of 45 or 45, -2X, tril2, tri17, +M1-5. Cell agglutination was observed when treated with ConA (≥7 fig, ml). Spontaneous agglutination might also take place without adding any ConA. After 5×106 cells of Ca 761-86 suspension were transplanted into the normal inbred 615 mice by different ways (subcutan eous, intrafoot-pad or intraperitoneal), the transplan lability rate reached 100%. Spontaneous remission was never observed and its metastatic ability reserved. PPLO were not detected. Ca 761-86 may be of value for practical purposes. 展开更多

Experimental results showed that type A and type C virus particles were observed in the cytoplasm of infected NIH/ 3T3 cells by electron-microscopy, X-C assay and reverse transcriptase were both positive. Free proviral-DN A of the leukemia vims was found in the infected NIH/3T3 cell by Southern blot hybridization.Morphologically, the NIH/3T3 cells infected in vitro appeared spherical and formed monolayer cell colonies of various sizes after 24 - 48 hours. When the cultured cells were Inoculated into nude mice (BALB/ c, nu nu ) subcutaneously, flbrosarcoma was Induced, 100%. Inoculation of the cell-free extract of the same cultured cells into Inbred SW-1 newborn mice, resulted in the production of lymphocytic leukemia and lymphoma in 52. 3% within 191 days. 展开更多

Having been passed for 160 generations, a cell linedesignated as H22-F25/L was established from a murine tumorlymphatic metastatlc model H22-F25 which had been set up in our college. The cell line was in suspension culture with a rapid proliferation and stable growth. The peak tune of cell division and proliferation was 48 and 96 hours after culture. In a week, the cell number was Increased by 25 tunes. H22-F25/L still keeps the features of a poorly differentiated cancer. Its tumor inducing rate (in vivo)was 100% in 615 mice. Lymph node metastasis rate was 50% and pulmonary metastasis rate 10%. H22- F25/ L Is a population of heterogenetlc tumor cells Including 2 stem cell lines (the model number of chromosomes being 43 in 40% tumor cells and 86 in 32%) and some side lines. 目的:观察骶髂关节错位患者在整复手法复位后腰背肌肌张力和对称性,骨盆、下肢生物力学的变化。方法:选取2022年5月至2023年3月期间就诊于福建中医药大学附属第二人民医院的骶髂关节错位患者120例,先理筋手法,再行整复手法,前错位60例,... 目的:观察骶髂关节错位患者在整复手法复位后腰背肌肌张力和对称性,骨盆、下肢生物力学的变化。方法:选取2022年5月至2023年3月期间就诊于福建中医药大学附属第二人民医院的骶髂关节错位患者120例,先理筋手法,再行整复手法,前错位60例,采用屈髋屈膝复位法,后错位60例,采用后伸扳法,隔日治疗1次,共治疗7次。对比治疗前后VAS评分、后表链肌张力K值、后表链肌张力对称值、骨盆倾斜、扭转、水平旋转情况,以及下肢支撑相、摆动相、足偏角的变化。结果:治疗后,骶髂关节前、后错位患者的VAS评分,后表链肌张力系数K值、肌张力对称性差值均较治疗前降低(P<0.05);治疗后,骶髂关节前、后错位患者的骨盆的倾斜、扭转、水平旋转角度均较治疗前减小(P<0.05);治疗后,骶髂关节前、后错位患者的步态周期中的支撑相较治疗前延长(P<0.05),摆动相较治疗前缩短(P<0.05),骶髂关节前错位患者较治疗前足偏角变大(P<0.05),后错位患者足偏角较治疗前变小(P<0.05)。结论:整复手法可以降低骶髂关节错位患者腰背肌肌张力,使两侧腰背肌趋于平衡,且能纠正骨盆的偏歪,改善下肢步态。 展开更多

哺乳动物被毛微量元素和重金属元素含量能够反映其生活状态和栖息地状况,是环境监测中重要的生物化学指示物。对北京南海子和浙江临安两地麋鹿(Elaphurus davidianus)种群被毛中的10种重金属和34种微量元素进行测定,以期探讨麋鹿被毛微... 哺乳动物被毛微量元素和重金属元素含量能够反映其生活状态和栖息地状况,是环境监测中重要的生物化学指示物。对北京南海子和浙江临安两地麋鹿(Elaphurus davidianus)种群被毛中的10种重金属和34种微量元素进行测定,以期探讨麋鹿被毛微量元素和重金属含量在栖息环境监测中的可行性。结果表明:除Re元素外,其余43种元素在麋鹿被毛中均被检出,统计分析表明Li、Be、Sc、V、Co、Ga、Rb、Ir、Mo、Cd、Cs、Ba、La、Ce、Pr、Nd、Sm、Eu、Gd、Tb、Dy、Ho、Er、Tm、Yb、Lu、Tl、Bi、Th、U、Nb、Ta、Zr和Hf 34种元素为麋鹿被毛的特征元素;与北京南海子麋鹿种群相比,浙江临安麋鹿种群被毛中的微量元素Sr、In、Sb、W、Bi、U和重金属元素Zn、Cu、Cr、Pb、Cd含量更少,其余32种元素含量均高于北京南海子麋鹿种群;独立样本t检验分析表明,麋鹿被毛微量元素和重金属元素含量存在地域差异性,仅Cr(p=0.064)、Ni(p=0.769)、Mo(p=0.129)、Tl(p=0.065)、Zr(p=0.138)和Hf(p=0.057)6种元素差异不显著,其余37种元素差异均显著;北京南海子和浙江临安麋鹿种群的栖息环境和食物均未受到Zn、Cu、Cr、Pb、Cd、V、Co、Ni、Sb和Tl 10种重金属元素污染的威胁。研究表明,麋鹿被毛可作为栖息环境重金属污染的非损伤监测指示材料。 展开更多

为筛选适宜在合浦县推广的木薯品系,本研究以主栽品种‘南植199’为对照,对9个木薯新品系的生物学特性进行了观察,并测定其农艺性状和经济性状,同时进行相关性分析。结果表明,9个品系分枝角度小、块根长度适中和主茎高度中等以上;‘RC4... 为筛选适宜在合浦县推广的木薯品系,本研究以主栽品种‘南植199’为对照,对9个木薯新品系的生物学特性进行了观察,并测定其农艺性状和经济性状,同时进行相关性分析。结果表明,9个品系分枝角度小、块根长度适中和主茎高度中等以上;‘RC4’的块根产量显著高于对照,其余8个品系与对照差异不显著;‘RC4’等8个品系的淀粉产量高于对照;9个品系的淀粉含量高于对照,其中‘RC3’等5个品系的淀粉含量显著或极显著高于对照;9个木薯品系的直链淀粉含量显著或极显著低于对照‘;RC6’等6个品系的氢氰酸含量低于50 mg/kg。相关性分析结果表明:块根产量与生长中期株高、成熟期株高、块根直径呈极显著正相关,与茎粗呈显著正相关;淀粉含量与生长中期株高、成熟期株高、茎粗呈显著正相关;淀粉产量与生长中期株高、成熟期株高、茎粗、块根直径、块根产量呈极显著正相关,与淀粉含量呈显著正相关。综合来看,这9个木薯新品系的综合表现良好,适合在合浦县进行推广。其中‘,RC3’、‘RC4’和‘RC7’可作为加工型品系;‘RC5’和‘RC10’可作为食用品系;‘RC2’、‘RC6’、‘RC11’和‘RC12’可作为鲜食加工兼用品系;‘RC2’和‘RC5’作为食用品系具有很好的推广前景。 展开更多

目的探讨紫草素介导Hippo信号通路对鼻咽癌(NPC)细胞株CNE2生物学功能的影响。方法于2021年1—12月使用含不同浓度紫草素培养液(0、2.0、4.0、8.0、16.0 mg/L)培养对数生长期人鼻咽癌细胞(CNE2细胞)48 h,细胞计数试剂盒-8(CCK-8)法检测... 目的探讨紫草素介导Hippo信号通路对鼻咽癌(NPC)细胞株CNE2生物学功能的影响。方法于2021年1—12月使用含不同浓度紫草素培养液(0、2.0、4.0、8.0、16.0 mg/L)培养对数生长期人鼻咽癌细胞(CNE2细胞)48 h,细胞计数试剂盒-8(CCK-8)法检测细胞存活率;流式细胞术、平板克隆、伤口愈合及Transwell实验分别检测CNE2细胞凋亡、集落形成、迁移及侵袭情况;实时荧光定量逆转录聚合酶链式反应(RT-qPCR)技术检测细胞yes相关蛋白1(YAP1)、具有PDZ结合基序的转录共激活子(TAZ)mRNA相对表达情况;蛋白质印迹法检测YAP1、yes相关蛋白1(p-YAP1)、TAZ、磷酸化具有PDZ结合基序的转录共激活子(p-TAZ)、N-钙黏蛋白(N-cad)、波形蛋白(Vim)及E-钙黏蛋白(E-cad)蛋白表达情况。结果与0 mg/L浓度紫草素CNE2细胞存活率(100%)、集落形成数(514.67±25.81)个、划痕愈合率(88.58±3.40)%、侵袭细胞数(233.67±15.01)个、YAP1与TAZ mRNA及蛋白(1.01±0.02、1.00±0.01、0.68±0.04、0.51±0.03)比较,2 mg/L浓度紫草素[(92.70±5.92)%、(452.33±22.72)个、(69.91±3.03)%、(195.33±18.15)个、0.93±0.02、0.91±0.05、0.56±0.03、0.44±0.02],4 mg/L浓度紫草素[(81.75±3.83)%、(308.33±22.12)个、(53.61±3.21)%、(153.33±10.02)个、0.81±0.03、0.76±0.04、0.45±0.03、0.30±0.03],8 mg/L浓度紫草素[(54.93±3.89)%、(173.67±13.65)个、(30.32±1.68)%、(92.67±6.66)个、0.65±0.03、0.54±0.04、0.31±0.03、0.24±0.02],16 mg/L浓度紫草素[(33.89±2.14)%、(85.33±13.05)个、(18.31±1.42)%、(52.33±6.03)个、0.41±0.02、0.30±0.02、0.18±0.02、0.16±0.02]降低(P<0.05),CNE2细胞凋亡率、p-YAP1与p-TAZ及E-cad蛋白相对表达水平均随紫草素作用浓度增大而升高(P<0.05);紫草素作用效果呈剂量依赖性(P<0.05)。结论紫草素可抑制NPC细胞株CNE2细胞恶性生物学功能,其作用机制可能与抑制Hippo信号通路核心下游信号因子YAP1、TAZ蛋白激活,阻止上皮间质转化(EMT)有关。 展开更多

针对当前印刷电路板上相邻微带线之间的串扰抑制问题,提出了一种在超宽带频率范围内利用周期性耦合枝节来抑制微带线远端串扰的电磁兼容设计。在串扰的形成原因分析基础上,该设计将沿传统微带线纵向方向加载的周期性开路枝节进一步与相... 针对当前印刷电路板上相邻微带线之间的串扰抑制问题,提出了一种在超宽带频率范围内利用周期性耦合枝节来抑制微带线远端串扰的电磁兼容设计。在串扰的形成原因分析基础上,该设计将沿传统微带线纵向方向加载的周期性开路枝节进一步与相邻两条微带线之间周期性加载的工字型耦合枝节相结合。微带线上的开路枝节以及线间的耦合枝节主要用于增加微带线等效自感和线间等效互容,并减小线间等效互感,由此显著降低远端串扰强度。同时,线间等效互感的减小能抑制因容性耦合增大而导致的近端串扰上升。全波结果表明,该设计在1~20 GHz范围内可使相邻微带线的远端串扰强度小于-48 dB,且物理结构中不涉及金属化通孔,具有几何结构简单、成本低和可布线空间大等特点。 展开更多

针对电力线路智能化巡检中难以准确辨识电力线触碰树木遗留痕迹,无法为事故防治和责任划分提供可靠依据的难题,提出一种基于前景区域提取与多特征决策融合的电力线放电痕迹识别方法。首先,搭建中压线路触树放电实验平台,采集导线表面放... 针对电力线路智能化巡检中难以准确辨识电力线触碰树木遗留痕迹,无法为事故防治和责任划分提供可靠依据的难题,提出一种基于前景区域提取与多特征决策融合的电力线放电痕迹识别方法。首先,搭建中压线路触树放电实验平台,采集导线表面放电遗留痕迹,构建图像集。然后,提出融合纹理稀疏性与对数变换的去阴影算法,消除阴影区干扰;进一步提出融合全局阈值和自适应阈值分割的目标初始框选取策略,解决了GrabCut算法中初始框无法自动确定的问题,实现复杂背景下导线痕迹的自动精准分割。最后,提取导线的纹理和颜色特征,通过多SVM决策融合实现导线痕迹识别。实验结果表明,所提方法平均识别准确率达到88.39%,证明了所提识别方法的有效性。 展开更多

近年来,国内外因树线故障导致森林火灾的事故频发,因外在社会因素引发森林火灾,从而导致输配电线路跳闸的事故也层出不穷。本文针对10 kV配电线路下方森林火灾事故,从社会因素引发森林火灾和树线故障引发森林火灾两个角度出发,考虑了纯... 近年来,国内外因树线故障导致森林火灾的事故频发,因外在社会因素引发森林火灾,从而导致输配电线路跳闸的事故也层出不穷。本文针对10 kV配电线路下方森林火灾事故,从社会因素引发森林火灾和树线故障引发森林火灾两个角度出发,考虑了纯火焰、树线放电及放电后引发火灾等多种极端环境对导线表面痕迹的影响,通过配电网故障模拟真型试验平台,对极端环境下导线表面形貌变化进行试验研究,得到了火烧痕迹、树线放电一次痕迹以及二次痕迹。基于宏观法、显微法和图像特征提取法分析了各类痕迹的特征,并找到了鉴别树线放电一次痕迹的方法。研究结果表明:放电痕迹一方面是树线电弧直接侵蚀导线,另一方面是电弧间接转移炭渣至导线表面形成炭黑痕迹;树线故障初期未引发森林火灾时,导线表面仅有一次放电痕迹,相同条件下,松树放电使导线质量变化最明显,可增重0.59 g;树线故障末期引发森林火灾后,火烧痕迹会覆盖放电痕迹形成二次痕迹,擦拭表面痕迹后发现,一、二次痕迹灰度曲面相似且分形维数之差小于0.03。 展开更多
