However, the genetic mechanism underlying FF is unclear. Here, a soft fruit melon(P5) and a hard fruit melon(P10) were crossed to generate F2, and the FF and fruit-related traits were recorded for two years. By performing quantitative trait locus(QTL) specificlocus amplified fragment(SLAF)(QTL-SLAF) sequencing and molecular marker-linkage analysis, 112 844 SLAF markers were identified, and 5 919 SNPs were used to construct a genetic linkage map with a total genetic distance of1 356.49 cM. Ten FF-and fruit-related QTLs were identified. Consistent QTLs were detected for fruit length(FL) and fruit diameter(FD) in both years, and QTLs for single fruit weight(SFW) were detected on two separate chromosomes in both years. For FF, the consistent major locus(ff2.1) was located in a 0.17-Mb candidate region on chromosome 2. Using 429 F2individuals derived from a cross between P5 and P10, we refined the ff2.1 locus to a 28.3-kb region harboring three functional genes. These results provide not only a new candidate QTL for melon FF breeding but also a theoretical foundation for research on the mechanism underlying melon gene function.展开更多

The study was conducted in a Sudano-Sahelian croppi...This study consisted to evaluate the effects of ecological practices on the yield performance of the JEWEL of orange-fleshed sweet potato (Ipomoea batatas (L.) Lam). The study was conducted in a Sudano-Sahelian cropping system (Lantargou, eastern region of Burkina Faso). Agro-ecological practices consisted of the inputs of 3.20 t/ha of compost + 2.45 t/ha of wood ash (CO + WA);4.90 t/ha of wood ash (WA);6.40 t/ha of compost (CO) were compared to control with no inputs (T<sub>0</sub>). Each treatment was repeated four times. The crop management consisted of plowing, harrowing, raising of ridges with 40 cm height, burying of treatments, transplanting of cuttings, two weeding’s, and using of biopesticide called PIOL for crop protection. Composite soil samples per treatment were also collected at tuber harvest and analyzed to determine the effects of treatments on residual soil fertility. Results showed that the plant heights and diameters under the CO were significantly (P < 0.001) improved by 16% and 12% compared to T<sub>0</sub>. The WA treatment significantly increased the number of large tubers by 43% (P ≤ 0.01) compared to T<sub>0</sub>. Total tuber numbers, large tuber numbers and tuber yields of sweet potato under CO + WA were significantly (P < 0.001) and respectively improved by 27%, 50% and 31% compared to T<sub>0</sub>. All treatments increased soil organic matter, N, P and K contents, and reduced soil acidity compared with those obtained under T<sub>0</sub>. Soil K content was improved by 39% under CO + WA, and soil N content by 34% under WA compared to T<sub>0</sub>. Soil C/N ratio under CO + WA was reduced by 20% compared others treatments. But, the CO + WA treatment outperformed by improving residual soil N content by 38%, and the WA treatment by increasing soil K content by 50% compared to T<sub>0</sub>. In addition, soil pHH<sub>2</sub>O increased by 1.2 units under WA treatment compared to T<sub>0</sub>. As conclusion, the application of 6.4 t/ha of compost performed well to improve the vegetative growth of orange-fleshed sweet potato while the inputs of 3.2 t/ha of compost + 2.45 t/ha of wood ash were efficacy to significantly increase the tuber yields and improve the residual fertility of soil.展开更多

The results showed t...The ultrastructure of the epidermis and flesh of apple ( Malus domestica Borkh cv. Red Fuji) fruit was systematically observed during the fruit development via transmission electron microscopy. The results showed that, in spite of the ultrastructural changes in many aspects of the developing fruit epidermal cells, it remained almost unchanged throughout the whole developmental process that the cytoplasm was filled with numerous endoplasmic reticula (ER). Most of these endoplasmic reticula were tube_like and rough_ER with enlarged cisterna from which many vesicles were produced. Some of the vesicles were shown to merge into vacuole. Some dynamic Golgi bodies were also found. All the ultrastructural characteristics showed that the epidermal cells have the features of excretory cells. The ultrastructure of the fruit flesh cells at the young fruit stage were shown to be metabolically active, characterized by the presence of numerous clustered plasmodesmata, cisterna enlarged_ and rough_ER filling the cytoplasm, plenty of vesicles and Golgi bodies, indicating their dynamic cellular transport function. Some giant_circular rough_ERs were found. All the ultrastructural features at this early developmental stage should be closely associated with the enlargement of the young fruit. At the rapid growing phase of the fruit the main changes were characterized by: the starch grain_filled amyloplasts, furcating of the single orifice of plasmodesmata, and the cytoplasm enrichment of both the Golgi body_formed vesicles and other vesicles. These features correspond well with those of a photoassimilate sink_cell. An ultrastructural degeneration phenomenon was observed at the fruit ripening stage, but the mitochondria and plasmalemma still remained intact, which might be related to the continuous development of fruit quality during the fruit ripening.展开更多

Jingchao) berry was observed via transmission electron microscopy. The results showed that during the early developmental stages of grape berry, numerous plasmodesmata were found in the phloem between sieve element (SE) and companion cell (CC), between SE/CC complexes, between SE/CC complex and phloem parenchyma cell and in between phloem parenchyma cells, which made the phloem a symplastic integration, facilitating sugar unloading from sieve elements into both companion cells and phloem parenchyma cells via a symplastic pathway. On the contrary, there was almost no plasmodesma between phloem and its surrounding flesh photoassimilate sink_cells, neither in between the flesh photoassimilate sink_cells giving rise to a symplastic isolation both between phloem and its surrounding flesh photoassimilate sink_cells, as well as among the flesh photoassimilate sink_cells. This indicated that both the sugar unloading from phloem and postphloem transport of sugars should be mainly via an apoplastic pathway. During the ripening stage, most of the plasmodesmata between SE/CC complex and the surrounding phloem parenchyma cells were shown to be blocked by the electron_opaque globules, and a phenomenon of plasmolysis was found in a number of companion cells, indicating a symplastic isolation between SE/CC complex and its surrounding parenchyma cells during this phase. The symplastic isolation between the whole phloem and its surrounding photoassimilate sink_cells during the early developmental stages shifted to a symplastic isolation within the phloem during the ripening phase, and thus the symplastic pathway of sugar unloading from SE/CC complex during the early development stages should be replaced by a dominant apoplastic unloading pathway from SE/CC complex in concordance.展开更多

[Method] The biochemical analytic method was used to detect the ratio of flesh content and biochemical compositions in muscle of Pachyhynobius shangchengensis. [Result] The ratio of flesh contents of female and male Pachyhynobius shangchengensis were 54.93% and 55.84% respectively. The contents of protein, total saccharides and fatty acid of female Pachyhynobius shangchengensis were 15.44% ,1.70% and 0.68% respectively,while the same contents of male Pachyhynobius shangchengensis were 15.67%, 1.89% and 0.53% respectively. The total content of 18 kinds of amino acids was 12.72% (the proportion in fresh weight), among which the content of 8 kinds of essential amino acids was 4.68%. The essential amino acids index(EAAI) was 72.68 and limiting amino acids was valine. There were 11 kinds of fatty acids in muscles of Pachyhynobius shangchengensis, among which the content of unsaturated fatty acids (80.02%) was higher than that of saturated fatty acids( 19.98% ). [ Conclusion] Pachyhynobius shangchengensis was a kind of valuable natural resource belonging to the nutritious type of low fat content and high protein content.展开更多

A total of 138 FI individuals were selected as experimental materi- als for construction of color around the stone gene pool (B1) and non-color around the stone gene pool (B2) by using bulked segregant analysis (BSA) method, molec- ular markers linked to the flesh color around the stone of P. persica (L.) Batsch were selected with SSR molecular marker technology. [Result] After selection with 256 pairs of SSR primers, three pairs of molecular markers linked to the gene con- trolling flesh color around the stone of P. persica (L.) Batsch were selected (UDP96- 003, ch04g09 and UDP97-402). In addition, genetic distances between the three molecular markers and the gene controlling flesh color around the stone of P. persi- ca (L.) Batsch were calculated, which were 16.7, 10.1 and 17.0 cM, respectively. The basal diet(FO) was formulated to contain 66.5% fis...We investigated the effect of the replacement of dietary fish oil with vegetable oils on the growth and flesh quality of large yellow croaker(Larmichthys crocea). The basal diet(FO) was formulated to contain 66.5% fish meal and 6.4% menhaden fish oil; whereas the other 3 experimental diets were formulated by replacing the fish oil with 50% soybean oil(SO50), 100% soybean oil(SO100) and 100% palm oil(PO100), respectively. The 4 diets were randomly assigned to 4 floating sea cages(3.0 m × 3.0 m × 3.0 m), and each was stocked with 250 fish individuals with an initial average weight of 245.29 g ± 7.45 g. The fish were fed to apparent satiation twice a day at 5:00 and 17:00, respectively, for 12 weeks. Experimental analysis showed that the specific growth rate of fish fed SO50 or PO100 were significantly higher than that of fish fed FO or SO100(P<0.05), and crude lipid contents of ventral muscle and viscera were significantly lower in fish fed FO than in those fed the other 3 diets(P<0.05). No significant differences in condition factor, viscerosomatic index, hepatosomatic index, gutted yield and colorimetric values of fish among the dietary treatments were observed(P>0.05). Compared to FO diet, SO50, SO100 and PO100 diets led to substantial decreases in the liquid loss and water loss from fresh fillets(1 d, 4℃)(P<0.05). Similarly, thiobarbituric acid reactive substance(TBARS) values of fillets under different storage conditions(1 d, 4℃; 7 d, 4℃; 4 weeks,-20℃; 8 weeks,-20℃) decreased significantly after partial or complete replacement of fish oil with vegetable oils. These findings indicated that the growth performance and selected flesh quality properties(liquid holding capacity and TBARS value) of large yellow croaker were substantially improved by replacing dietary fish oil with vegetable oils.展开更多

To compare the flesh quality of large yellow croaker cultured with three different modes, enclosure culture(EC), cage culture with feeding trash fish(CCF) and cage culture with feeding formula diet(CCD), thirty six fi...To compare the flesh quality of large yellow croaker cultured with three different modes, enclosure culture(EC), cage culture with feeding trash fish(CCF) and cage culture with feeding formula diet(CCD), thirty six fish individuals of EC(484.6 ± 79.8 g), CCF(432.7 ± 87.9 g) and CCD(416.9 ± 49.5 g) were sampled to measure body color, flesh water holding capacity, flesh proximate composition, amino acids and fatty acids composition, and collagen and inosinic acid contents. The analysis showed that there was no significant difference in the yellowness of skin and the contents of crude protein and inosinic acid between CCD and EC(P > 0.05), and the two modes showed higher values than CCF(P < 0.05). Most of the amino acids(AA) in flesh were higher in EC than in CCF(P < 0.05), but CCD showed no difference in AA composition from the other two modes(P > 0.05). EC and CCD had higher contents of collagen, free AA and lower drip loss than CC(P < 0.05). In polyunsaturated fatty acids, EC showed significantly higher levels of C20:5 n-3 and C22:6 n-3 than CCF and CCD(P < 0.05), but CCD had significantly higher C18:2 n-6 than CCF and EC(P < 0.05). In conclusion, the flesh quality of CCD fish is similar to EC fish, and these two modes have better flesh quality than CCF.展开更多
Background:Muscle is the complex and heterogeneous tissue,which comprises the primary edible part of the trunk of fish and mammals.Previous studies have shown that dietary isoleucine(Ile)exerts beneficial effects on g...Background:Muscle is the complex and heterogeneous tissue,which comprises the primary edible part of the trunk of fish and mammals.Previous studies have shown that dietary isoleucine(Ile)exerts beneficial effects on growth in aquatic animals.However,there were limited studies regarding the benefits of Ile on fish muscle and their effects on flesh quality and muscle growth.Thus,this study was conducted to explore whether dietary Ile had affected flesh quality and muscle growth in hybrid bagrid catfish(Pelteobagrus vachelli♀×Leiocassis longirostris♂).Methods:A total of 630 hybrid fish,with an initial average body weight of 33.11±0.09 g,were randomly allotted into seven experimental groups with three replicates each,and respectively fed seven diets with 5.0,7.5,10.0,12.5,15.0,17.5,and 20.0 g Ile/kg diets for 8 weeks.Results:In the present study,we demonstrated that Ile significantly:(1)increased muscle protein and lipid contents and the frequency distribution of myofibers with≤20μm and≥50μm of diameter;(2)improved pH value,shear force,cathepsin B and L activities,hydroxyproline content,resilience,cohesiveness,and decreased cooking loss,lactate content,hardness,springiness,gumminess,and chewiness;(3)decreased reactive oxygen species(ROS),malondialdehyde(MDA),and protein carbonyl(PC)contents,GCLC and Keap1 mRNA levels,and up-regulated CuZnSOD,CAT,GPX1a,GST,and Nrf2 mRNA levels;(4)up-regulated the insulin-like growth factor 1,2(IGF-1,IGF-2),insulin-like growth factor 1 receptor(IGF-1R),proliferating cell nuclear antigen(PCNA),Myf5,Myod,Myog,Mrf4,and MyHC mRNA levels,and decreased MSTN mRNAlevel;(5)increased muscle protein deposition by activating AKT-TOR-S6K1 and AKT-FOXO3a signaling pathways.Conclusion:These results revealed that dietary Ile improved flesh quality,which might be due to increasing nutritional content,physicochemical,texture parameters,and antioxidant ability;promoting muscle growth by affecting myocytes hyperplasia and hypertrophy,and muscle protein deposition associated with protein synthesis and degradation signaling pathways.Finally,the quadratic regression analysis of chewiness,ROS,and protein contents against dietary Ile levels suggested that the optimal dietary Ile levels for hybrid bagrid catfish was estimated to be 14.19,12.36,and 12.78 g/kg diet,corresponding to 36.59,31.87,and 32.96 g/kg dietary protein,respectively.展开更多

Lam] were studied ...Variation laws of anthocyanin content in root during the development and among the varieties, and their relationships with major economic traits in purple-fleshed sweetpotato [Ipomoea batatas (L.) Lam] were studied in the present article. The dynamics of 20 economic traits in 13 purple-fleshed sweetpotato varieties at 20, 40, 60, 80, 100, 120, and 140 d after their transplanting were investigated, and these traits included anthocyanin content in root, length of the longest vine, number of base branches, root number, dry matter contents in stem, foliage and root, fresh/dry weight of root, fresh/dry weight of stem, flesh/dry weight of foliage, flesh/dry weight of stem and foliage, flesh/dry weight of whole plant, and rations of photosynthate to root, stem, and foliage. The correlations between the variations of anthocyanin content and the other 19 economic traits among varieties and during the whole developing stages, and the correlations of daily increase of anthocyanin content with other l0 kinds of yields were analyzed. The results showed that: (1) During the whole development, the anthocyanin content had three variation types, i.e. a slow-increase type, a fluctuating-change type, and a deviousrising type, and had different responses to the growth of length of the longest vine, number of base branches, flesh/dry yield of root, and photosynthate allotments. (2) The anthocyanin contents among 13 varieties began to have significant difference after 20 d, and showed completed differentiation during 40-100 d, which had significantly negative correlationships with the number of base branches, fresh/dry yield of root, photosynthate allotment ratio to root, and had significant positive correlationships with dry matter content of root, length of the longest vine, fresh/dry yield of stem, dry yield of whole plant and photosynthate allotment ratio to foliage. (3) Because of the significantly negative correlation between daily increase of anthocyanin content and dry matter weight of root, the anthocyanin accumulation competed with dry matter accumulation for photosynthate in root, and the competitive relation was resolved in different ways in different purple-fleshed sweetpotato (PFSP) varieties. So, there had three variation types of anthocyanin content among PFSP varieties during their development, and had different correlations between these variations of anthocyanin content and the major economic traits.展开更多

The apple cultivar ‘May’ represents an excellent model for the study of anthocyanin metabolism due to the spatial color pattern of its flesh, which transitions sequentially from red to white to red again,from the outside to the inside of the apple fruit. To screen the transcription factors involved in the anthocyanin pathway, transcriptomes from differently colored sections of ‘May’ fruits were analyzed by RNA sequencing. Kyoto Encyclopedia of Genes and Genomes(KEGG) database was used to analyse the gene expression data. The results suggested differentially expressed genes(DEGs) that are related to phytohormones are involved in regulating anthocyanin biosynthesis. K-means clustering analysis revealed 167 common DEGs between different sections of fruit with the same expression pattern as candidates for regulating anthocyanin synthesis. Further analysis showed that nine of the 167 DEGs were annotated as transcription factors and quantitative real time PCR(qRT-PCR) confirmed that their expression was obviously higher in red regions of the fruit, consistent with their roles as hub genes that regulate anthocyanin synthesis. This study provides valuable results for future studies of anthocyanin synthesis in apple fruit.展开更多

Practical ...The study was conducted to demonstrate the effects of dietary protein and water temperature on growth and flesh quality of Songpu mirror carp (Cyprinus carpio) at an initial weight (165.24±5.08) g. Practical diets were formulated to contain five protein levels (29.12%, 31.46%, 34.49%, 38.17%, and 40.13%), and each diet was randomly assigned triplicate groups of 15 fishes at three temperatures (18℃, 22℃, and 26℃) in the recirculation system. Fishes were fed twice daily to apparent satiation for 56 days. Results indicated that fishes had higher weight gain rate at 22℃ and 26℃ than that at 18℃ (P〈0.05), but there were no significant differences between 22℃ and 26℃ (P〉0.05). Based on the weight gain rate, dietary protein level 29.12% could meet the requirement of the body at 18℃, 22℃, or 26℃. Crude protein, crude lipid, moisture and ash of muscle had no significant differences among those treatments (P〉0.05). pH (after 24 h) of muscle was the highest at 18℃ and the lowest at 22℃ (P〈0.05), but no differences were observed among different protein level groups at each temperature (P〉0.05). No significant differences on shear force, water holding capacity, collagen, glycogen and lactate among all the treatments were found (P〉0.05). It was concluded that when C. carpio fed to apparent satiation, the growth mainly depended upon temperature. Dietary protein could not significantly affect flesh quality, but temperature significantly affected pH of muscles. A dietary protein level 29.12% could meet the requirement of the body at 18℃, 22℃, or 26℃.展开更多
Sweet potato is a multifunctional root crop and a source of food with many essential nutrients and bioactive compounds.Variations in the flesh color of the diverse sweet potato varieties are attributed to the differen...Sweet potato is a multifunctional root crop and a source of food with many essential nutrients and bioactive compounds.Variations in the flesh color of the diverse sweet potato varieties are attributed to the different phytochemicals and natural pigments they produce.Among them,carotenoids and anthocyanins are the main pigments known for their antioxidant properties which provide a host of health benefits,hence,regarded as a major component of the human diet.In this review,we provide an overview of the major pigments in sweet potato with much emphasis on their biosynthesis,functions,and regulatory control.More-over,current findings on the molecular mechanisms underlying the biosynthesis and accumulation of carotenoids and anthocyanins in sweet potato are discussed.Insights into the composition,biosynthesis,and regulatory control of these major pigments will further advance the biofortification of sweet potato and provide a reference for breeding carotenoid-and anthocyanin-rich varieties.展开更多

The color of watermelon flesh is an important trait determined by a series of carotenoids.Herein,we used Cream of Saskatchewan(pale yellow flesh)and PI 186490(white flesh)as parental materials for an F2 segregation an...The color of watermelon flesh is an important trait determined by a series of carotenoids.Herein,we used Cream of Saskatchewan(pale yellow flesh)and PI 186490(white flesh)as parental materials for an F2 segregation and initial mapping using the bulked segregant analysis sequencing(BSA-seq)strategy.The BSA results revealed a flesh colorrelated QTL that spans approximately 2.45 Mb on chromosome 6.This region was preliminarily positioned in a 382-kb segment,and then narrowed down into a 66.8-kb segment with 1260 F2 individuals.A total of nine candidate genes were in the fine mapping interval,but only Cla007528(encoding chlorophyllase)had non-synonymous mutations and was significantly expressed between the parental materials throughout flesh development.We also checked the expression patterns of the carotenoid metabolic pathway genes based on RNA-seq data and qRT-PCR validation.Three genes in the xanthophyll cycle(ClCHYB,ClNCED-1 and ClNCED-7)exhibited differential expression patterns between the two parental lines at different flesh color formation stages.ClPSY1,ClPDS,ClZDS,ClCHXE,ClCRTISO and ClLCYB also exhibited clearly different expression patterns accompanied by carotenoid accumulation.展开更多

文摘Flesh firmness(FF) is an important and complex trait for melon breeders and consumers. However, the genetic mechanism underlying FF is unclear. Here, a soft fruit melon(P5) and a hard fruit melon(P10) were crossed to generate F2, and the FF and fruit-related traits were recorded for two years. By performing quantitative trait locus(QTL) specificlocus amplified fragment(SLAF)(QTL-SLAF) sequencing and molecular marker-linkage analysis, 112 844 SLAF markers were identified, and 5 919 SNPs were used to construct a genetic linkage map with a total genetic distance of1 356.49 cM. Ten FF-and fruit-related QTLs were identified. Consistent QTLs were detected for fruit length(FL) and fruit diameter(FD) in both years, and QTLs for single fruit weight(SFW) were detected on two separate chromosomes in both years. For FF, the consistent major locus(ff2.1) was located in a 0.17-Mb candidate region on chromosome 2. Using 429 F2individuals derived from a cross between P5 and P10, we refined the ff2.1 locus to a 28.3-kb region harboring three functional genes. These results provide not only a new candidate QTL for melon FF breeding but also a theoretical foundation for research on the mechanism underlying melon gene function.
文摘This study consisted to evaluate the effects of ecological practices on the yield performance of the JEWEL of orange-fleshed sweet potato (Ipomoea batatas (L.) Lam). The study was conducted in a Sudano-Sahelian cropping system (Lantargou, eastern region of Burkina Faso). Agro-ecological practices consisted of the inputs of 3.20 t/ha of compost + 2.45 t/ha of wood ash (CO + WA);4.90 t/ha of wood ash (WA);6.40 t/ha of compost (CO) were compared to control with no inputs (T<sub>0</sub>). Each treatment was repeated four times. The crop management consisted of plowing, harrowing, raising of ridges with 40 cm height, burying of treatments, transplanting of cuttings, two weeding’s, and using of biopesticide called PIOL for crop protection. Composite soil samples per treatment were also collected at tuber harvest and analyzed to determine the effects of treatments on residual soil fertility. Results showed that the plant heights and diameters under the CO were significantly (P < 0.001) improved by 16% and 12% compared to T<sub>0</sub>. The WA treatment significantly increased the number of large tubers by 43% (P ≤ 0.01) compared to T<sub>0</sub>. Total tuber numbers, large tuber numbers and tuber yields of sweet potato under CO + WA were significantly (P < 0.001) and respectively improved by 27%, 50% and 31% compared to T<sub>0</sub>. All treatments increased soil organic matter, N, P and K contents, and reduced soil acidity compared with those obtained under T<sub>0</sub>. Soil K content was improved by 39% under CO + WA, and soil N content by 34% under WA compared to T<sub>0</sub>. Soil C/N ratio under CO + WA was reduced by 20% compared others treatments. But, the CO + WA treatment outperformed by improving residual soil N content by 38%, and the WA treatment by increasing soil K content by 50% compared to T<sub>0</sub>. In addition, soil pHH<sub>2</sub>O increased by 1.2 units under WA treatment compared to T<sub>0</sub>. As conclusion, the application of 6.4 t/ha of compost performed well to improve the vegetative growth of orange-fleshed sweet potato while the inputs of 3.2 t/ha of compost + 2.45 t/ha of wood ash were efficacy to significantly increase the tuber yields and improve the residual fertility of soil.
文摘The ultrastructure of the epidermis and flesh of apple ( Malus domestica Borkh cv. Red Fuji) fruit was systematically observed during the fruit development via transmission electron microscopy. The results showed that, in spite of the ultrastructural changes in many aspects of the developing fruit epidermal cells, it remained almost unchanged throughout the whole developmental process that the cytoplasm was filled with numerous endoplasmic reticula (ER). Most of these endoplasmic reticula were tube_like and rough_ER with enlarged cisterna from which many vesicles were produced. Some of the vesicles were shown to merge into vacuole. Some dynamic Golgi bodies were also found. All the ultrastructural characteristics showed that the epidermal cells have the features of excretory cells. The ultrastructure of the fruit flesh cells at the young fruit stage were shown to be metabolically active, characterized by the presence of numerous clustered plasmodesmata, cisterna enlarged_ and rough_ER filling the cytoplasm, plenty of vesicles and Golgi bodies, indicating their dynamic cellular transport function. Some giant_circular rough_ERs were found. All the ultrastructural features at this early developmental stage should be closely associated with the enlargement of the young fruit. At the rapid growing phase of the fruit the main changes were characterized by: the starch grain_filled amyloplasts, furcating of the single orifice of plasmodesmata, and the cytoplasm enrichment of both the Golgi body_formed vesicles and other vesicles. These features correspond well with those of a photoassimilate sink_cell. An ultrastructural degeneration phenomenon was observed at the fruit ripening stage, but the mitochondria and plasmalemma still remained intact, which might be related to the continuous development of fruit quality during the fruit ripening.
文摘The ultrastructure and intercellular connection of the sugar unloading zone (i.e. the phloem in the dorsal vascular bundle and the phloem_surrounding the assimilate sink_cells) of grape ( Vitis vinifera× V. labrusca cv. Jingchao) berry was observed via transmission electron microscopy. The results showed that during the early developmental stages of grape berry, numerous plasmodesmata were found in the phloem between sieve element (SE) and companion cell (CC), between SE/CC complexes, between SE/CC complex and phloem parenchyma cell and in between phloem parenchyma cells, which made the phloem a symplastic integration, facilitating sugar unloading from sieve elements into both companion cells and phloem parenchyma cells via a symplastic pathway. On the contrary, there was almost no plasmodesma between phloem and its surrounding flesh photoassimilate sink_cells, neither in between the flesh photoassimilate sink_cells giving rise to a symplastic isolation both between phloem and its surrounding flesh photoassimilate sink_cells, as well as among the flesh photoassimilate sink_cells. This indicated that both the sugar unloading from phloem and postphloem transport of sugars should be mainly via an apoplastic pathway. During the ripening stage, most of the plasmodesmata between SE/CC complex and the surrounding phloem parenchyma cells were shown to be blocked by the electron_opaque globules, and a phenomenon of plasmolysis was found in a number of companion cells, indicating a symplastic isolation between SE/CC complex and its surrounding parenchyma cells during this phase. The symplastic isolation between the whole phloem and its surrounding photoassimilate sink_cells during the early developmental stages shifted to a symplastic isolation within the phloem during the ripening phase, and thus the symplastic pathway of sugar unloading from SE/CC complex during the early development stages should be replaced by a dominant apoplastic unloading pathway from SE/CC complex in concordance.

文摘[Objective] The ratio of flesh content and biochemical compositions in muscles of Pachyhynobius shangchengensis were researched in this experiment to provide basic referrences for studying nutritive physiological ecology of Pachyhynobius shangchengensis. [Method] The biochemical analytic method was used to detect the ratio of flesh content and biochemical compositions in muscle of Pachyhynobius shangchengensis. [Result] The ratio of flesh contents of female and male Pachyhynobius shangchengensis were 54.93% and 55.84% respectively. The contents of protein, total saccharides and fatty acid of female Pachyhynobius shangchengensis were 15.44% ,1.70% and 0.68% respectively,while the same contents of male Pachyhynobius shangchengensis were 15.67%, 1.89% and 0.53% respectively. The total content of 18 kinds of amino acids was 12.72% (the proportion in fresh weight), among which the content of 8 kinds of essential amino acids was 4.68%. The essential amino acids index(EAAI) was 72.68 and limiting amino acids was valine. There were 11 kinds of fatty acids in muscles of Pachyhynobius shangchengensis, among which the content of unsaturated fatty acids (80.02%) was higher than that of saturated fatty acids( 19.98% ). [ Conclusion] Pachyhynobius shangchengensis was a kind of valuable natural resource belonging to the nutritious type of low fat content and high protein content.

文摘[Objective] This study aimed to select SSR molecular markers linked to flesh color around the stone of Prunus persica (L.) Batsch. [Method] P. persica (L.) Batsch varieties Chongyanghong and Yanhong were used as parents to construct F1 orthogonal group. A total of 138 FI individuals were selected as experimental materi- als for construction of color around the stone gene pool (B1) and non-color around the stone gene pool (B2) by using bulked segregant analysis (BSA) method, molec- ular markers linked to the flesh color around the stone of P. persica (L.) Batsch were selected with SSR molecular marker technology. [Result] After selection with 256 pairs of SSR primers, three pairs of molecular markers linked to the gene con- trolling flesh color around the stone of P. persica (L.) Batsch were selected (UDP96- 003, ch04g09 and UDP97-402). In addition, genetic distances between the three molecular markers and the gene controlling flesh color around the stone of P. persi- ca (L.) Batsch were calculated, which were 16.7, 10.1 and 17.0 cM, respectively. [Conclusion] This study laid the foundation for further selection of co-dominant molecular markers with closer genetic distance.
文摘[Objective]The aim was to study the inheritance of flesh thickness,so as to provide a theoretical basis for breeding of thicker-flesh summer squash variety.[Method]Using mixed major genes and polygene inheritance model,a joint analysis of six generations from two crosses q-1×23-4G(Cross 1)and q-1×A-7(Cross 2)was conducted to investigate the inheritance of the flesh thickness of summer squash.[Result]The results showed that the flesh thickness was controlled by D-2 model(a pair of additive major genes plus additive-dominant polygene);genetic effect of flesh thickness in the two crosses was mainly the dominant effect of polygene.Heritability values of the genes of F2 generation were relatively low,while the environmental effect was great.[Conclusion]To flesh thickness,the method of individual selection on high separated generations was effective in the breeding of summer squash with thick flesh.

文摘We investigated the effect of the replacement of dietary fish oil with vegetable oils on the growth and flesh quality of large yellow croaker(Larmichthys crocea). The basal diet(FO) was formulated to contain 66.5% fish meal and 6.4% menhaden fish oil; whereas the other 3 experimental diets were formulated by replacing the fish oil with 50% soybean oil(SO50), 100% soybean oil(SO100) and 100% palm oil(PO100), respectively. The 4 diets were randomly assigned to 4 floating sea cages(3.0 m × 3.0 m × 3.0 m), and each was stocked with 250 fish individuals with an initial average weight of 245.29 g ± 7.45 g. The fish were fed to apparent satiation twice a day at 5:00 and 17:00, respectively, for 12 weeks. Experimental analysis showed that the specific growth rate of fish fed SO50 or PO100 were significantly higher than that of fish fed FO or SO100(P<0.05), and crude lipid contents of ventral muscle and viscera were significantly lower in fish fed FO than in those fed the other 3 diets(P<0.05). No significant differences in condition factor, viscerosomatic index, hepatosomatic index, gutted yield and colorimetric values of fish among the dietary treatments were observed(P>0.05). Compared to FO diet, SO50, SO100 and PO100 diets led to substantial decreases in the liquid loss and water loss from fresh fillets(1 d, 4℃)(P<0.05). Similarly, thiobarbituric acid reactive substance(TBARS) values of fillets under different storage conditions(1 d, 4℃; 7 d, 4℃; 4 weeks,-20℃; 8 weeks,-20℃) decreased significantly after partial or complete replacement of fish oil with vegetable oils. These findings indicated that the growth performance and selected flesh quality properties(liquid holding capacity and TBARS value) of large yellow croaker were substantially improved by replacing dietary fish oil with vegetable oils.

文摘Mesocarp color is an important agronomic trait of Cucurbita maxima and is determined mainly by the contents and compositions of the carotenoids.The two inbred lines with significant differences in fruit flesh color were used in the study,the orange’312-1’and white’98-2’.Changes in seven carotenoid contents and compositions in the flesh of fruit produced by inbred lines’312-1’and’98-2’were analyzed during fruit development.The expression of eight key carotenoid biosynthesis genes in the fruit flesh were investigated during five fruit development stage in two inbred lines.As the flesh color intensified,the orange flesh color of’312-1’was determined mainly by the increased contents of lutein,β-carotene and zeaxanthin and the lack of carotenoid accumulation led to the formation of white flesh in’98-2’fruit.The expression of the LCY-e and CHYb genes was significantly stronger in’312-1’than in’98-2’,and their expression was strongly correlated with lutein andβ-carotene contents during fruit development.This study provides a deep understanding of the molecular mechanism of carotenoid biosynthesis in fruit flesh and provides a basis for additional studies on the highly refined improvement of squash quality.

文摘The red flesh in apple fruit is a desired trait by consumers and it is associated to the anthocyanin content,which is mainly controlled by MdMYB10 with a R6 promoter.In this study,a high-density linkage group was constructed using the‘Fuji’x‘Red3’population which contained homozygous alleles R1R1 and R6R6,respectively.The linkage group consists of 7630 SNPs along 17 linkage groups,spanning 2270.21 cM,with an average density of 0.30 cM permarker.The cyanidin-3-galactoside concentration was used as the phenotypic data in QTL analysis.Moreover,one QTL peak which was flaked by two markers,marker2187260 to marker2173766,with LOD scores of 4.49 was detected.This QTL ranged from 0 to 40.79 cM on the top of linkage group(LG16).In addition one candidate molecular marker(marker2175442)in this QTL was identified,which was significant correlated with the flesh cyanidin-3-galactoside concentration.These genetic findings enrich the breeding basis of fruit flesh coloration in apple.

文摘To compare the flesh quality of large yellow croaker cultured with three different modes, enclosure culture(EC), cage culture with feeding trash fish(CCF) and cage culture with feeding formula diet(CCD), thirty six fish individuals of EC(484.6 ± 79.8 g), CCF(432.7 ± 87.9 g) and CCD(416.9 ± 49.5 g) were sampled to measure body color, flesh water holding capacity, flesh proximate composition, amino acids and fatty acids composition, and collagen and inosinic acid contents. The analysis showed that there was no significant difference in the yellowness of skin and the contents of crude protein and inosinic acid between CCD and EC(P > 0.05), and the two modes showed higher values than CCF(P < 0.05). Most of the amino acids(AA) in flesh were higher in EC than in CCF(P < 0.05), but CCD showed no difference in AA composition from the other two modes(P > 0.05). EC and CCD had higher contents of collagen, free AA and lower drip loss than CC(P < 0.05). In polyunsaturated fatty acids, EC showed significantly higher levels of C20:5 n-3 and C22:6 n-3 than CCF and CCD(P < 0.05), but CCD had significantly higher C18:2 n-6 than CCF and EC(P < 0.05). In conclusion, the flesh quality of CCD fish is similar to EC fish, and these two modes have better flesh quality than CCF.

文摘Background:Muscle is the complex and heterogeneous tissue,which comprises the primary edible part of the trunk of fish and mammals.Previous studies have shown that dietary isoleucine(Ile)exerts beneficial effects on growth in aquatic animals.However,there were limited studies regarding the benefits of Ile on fish muscle and their effects on flesh quality and muscle growth.Thus,this study was conducted to explore whether dietary Ile had affected flesh quality and muscle growth in hybrid bagrid catfish(Pelteobagrus vachelli♀×Leiocassis longirostris♂).Methods:A total of 630 hybrid fish,with an initial average body weight of 33.11±0.09 g,were randomly allotted into seven experimental groups with three replicates each,and respectively fed seven diets with 5.0,7.5,10.0,12.5,15.0,17.5,and 20.0 g Ile/kg diets for 8 weeks.Results:In the present study,we demonstrated that Ile significantly:(1)increased muscle protein and lipid contents and the frequency distribution of myofibers with≤20μm and≥50μm of diameter;(2)improved pH value,shear force,cathepsin B and L activities,hydroxyproline content,resilience,cohesiveness,and decreased cooking loss,lactate content,hardness,springiness,gumminess,and chewiness;(3)decreased reactive oxygen species(ROS),malondialdehyde(MDA),and protein carbonyl(PC)contents,GCLC and Keap1 mRNA levels,and up-regulated CuZnSOD,CAT,GPX1a,GST,and Nrf2 mRNA levels;(4)up-regulated the insulin-like growth factor 1,2(IGF-1,IGF-2),insulin-like growth factor 1 receptor(IGF-1R),proliferating cell nuclear antigen(PCNA),Myf5,Myod,Myog,Mrf4,and MyHC mRNA levels,and decreased MSTN mRNAlevel;(5)increased muscle protein deposition by activating AKT-TOR-S6K1 and AKT-FOXO3a signaling pathways.Conclusion:These results revealed that dietary Ile improved flesh quality,which might be due to increasing nutritional content,physicochemical,texture parameters,and antioxidant ability;promoting muscle growth by affecting myocytes hyperplasia and hypertrophy,and muscle protein deposition associated with protein synthesis and degradation signaling pathways.Finally,the quadratic regression analysis of chewiness,ROS,and protein contents against dietary Ile levels suggested that the optimal dietary Ile levels for hybrid bagrid catfish was estimated to be 14.19,12.36,and 12.78 g/kg diet,corresponding to 36.59,31.87,and 32.96 g/kg dietary protein,respectively.

文摘Variation laws of anthocyanin content in root during the development and among the varieties, and their relationships with major economic traits in purple-fleshed sweetpotato [Ipomoea batatas (L.) Lam] were studied in the present article. The dynamics of 20 economic traits in 13 purple-fleshed sweetpotato varieties at 20, 40, 60, 80, 100, 120, and 140 d after their transplanting were investigated, and these traits included anthocyanin content in root, length of the longest vine, number of base branches, root number, dry matter contents in stem, foliage and root, fresh/dry weight of root, fresh/dry weight of stem, flesh/dry weight of foliage, flesh/dry weight of stem and foliage, flesh/dry weight of whole plant, and rations of photosynthate to root, stem, and foliage. The correlations between the variations of anthocyanin content and the other 19 economic traits among varieties and during the whole developing stages, and the correlations of daily increase of anthocyanin content with other l0 kinds of yields were analyzed. The results showed that: (1) During the whole development, the anthocyanin content had three variation types, i.e. a slow-increase type, a fluctuating-change type, and a deviousrising type, and had different responses to the growth of length of the longest vine, number of base branches, flesh/dry yield of root, and photosynthate allotments. (2) The anthocyanin contents among 13 varieties began to have significant difference after 20 d, and showed completed differentiation during 40-100 d, which had significantly negative correlationships with the number of base branches, fresh/dry yield of root, photosynthate allotment ratio to root, and had significant positive correlationships with dry matter content of root, length of the longest vine, fresh/dry yield of stem, dry yield of whole plant and photosynthate allotment ratio to foliage. (3) Because of the significantly negative correlation between daily increase of anthocyanin content and dry matter weight of root, the anthocyanin accumulation competed with dry matter accumulation for photosynthate in root, and the competitive relation was resolved in different ways in different purple-fleshed sweetpotato (PFSP) varieties. So, there had three variation types of anthocyanin content among PFSP varieties during their development, and had different correlations between these variations of anthocyanin content and the major economic traits.

文摘Anthocyanins, a class of flavonoid compounds that provide pigmentation to plant organs, are beneficial components of the human diet and there is an interest in understanding the regulation of their formation in crop plants. The apple cultivar ‘May’ represents an excellent model for the study of anthocyanin metabolism due to the spatial color pattern of its flesh, which transitions sequentially from red to white to red again,from the outside to the inside of the apple fruit. To screen the transcription factors involved in the anthocyanin pathway, transcriptomes from differently colored sections of ‘May’ fruits were analyzed by RNA sequencing. Kyoto Encyclopedia of Genes and Genomes(KEGG) database was used to analyse the gene expression data. The results suggested differentially expressed genes(DEGs) that are related to phytohormones are involved in regulating anthocyanin biosynthesis. K-means clustering analysis revealed 167 common DEGs between different sections of fruit with the same expression pattern as candidates for regulating anthocyanin synthesis. Further analysis showed that nine of the 167 DEGs were annotated as transcription factors and quantitative real time PCR(qRT-PCR) confirmed that their expression was obviously higher in red regions of the fruit, consistent with their roles as hub genes that regulate anthocyanin synthesis. This study provides valuable results for future studies of anthocyanin synthesis in apple fruit.

文摘The study was conducted to demonstrate the effects of dietary protein and water temperature on growth and flesh quality of Songpu mirror carp (Cyprinus carpio) at an initial weight (165.24±5.08) g. Practical diets were formulated to contain five protein levels (29.12%, 31.46%, 34.49%, 38.17%, and 40.13%), and each diet was randomly assigned triplicate groups of 15 fishes at three temperatures (18℃, 22℃, and 26℃) in the recirculation system. Fishes were fed twice daily to apparent satiation for 56 days. Results indicated that fishes had higher weight gain rate at 22℃ and 26℃ than that at 18℃ (P〈0.05), but there were no significant differences between 22℃ and 26℃ (P〉0.05). Based on the weight gain rate, dietary protein level 29.12% could meet the requirement of the body at 18℃, 22℃, or 26℃. Crude protein, crude lipid, moisture and ash of muscle had no significant differences among those treatments (P〉0.05). pH (after 24 h) of muscle was the highest at 18℃ and the lowest at 22℃ (P〈0.05), but no differences were observed among different protein level groups at each temperature (P〉0.05). No significant differences on shear force, water holding capacity, collagen, glycogen and lactate among all the treatments were found (P〉0.05). It was concluded that when C. carpio fed to apparent satiation, the growth mainly depended upon temperature. Dietary protein could not significantly affect flesh quality, but temperature significantly affected pH of muscles. A dietary protein level 29.12% could meet the requirement of the body at 18℃, 22℃, or 26℃.

文摘Sweet potato is a multifunctional root crop and a source of food with many essential nutrients and bioactive compounds.Variations in the flesh color of the diverse sweet potato varieties are attributed to the different phytochemicals and natural pigments they produce.Among them,carotenoids and anthocyanins are the main pigments known for their antioxidant properties which provide a host of health benefits,hence,regarded as a major component of the human diet.In this review,we provide an overview of the major pigments in sweet potato with much emphasis on their biosynthesis,functions,and regulatory control.More-over,current findings on the molecular mechanisms underlying the biosynthesis and accumulation of carotenoids and anthocyanins in sweet potato are discussed.Insights into the composition,biosynthesis,and regulatory control of these major pigments will further advance the biofortification of sweet potato and provide a reference for breeding carotenoid-and anthocyanin-rich varieties.

文摘The color of watermelon flesh is an important trait determined by a series of carotenoids.Herein,we used Cream of Saskatchewan(pale yellow flesh)and PI 186490(white flesh)as parental materials for an F2 segregation and initial mapping using the bulked segregant analysis sequencing(BSA-seq)strategy.The BSA results revealed a flesh colorrelated QTL that spans approximately 2.45 Mb on chromosome 6.This region was preliminarily positioned in a 382-kb segment,and then narrowed down into a 66.8-kb segment with 1260 F2 individuals.A total of nine candidate genes were in the fine mapping interval,but only Cla007528(encoding chlorophyllase)had non-synonymous mutations and was significantly expressed between the parental materials throughout flesh development.We also checked the expression patterns of the carotenoid metabolic pathway genes based on RNA-seq data and qRT-PCR validation.Three genes in the xanthophyll cycle(ClCHYB,ClNCED-1 and ClNCED-7)exhibited differential expression patterns between the two parental lines at different flesh color formation stages.ClPSY1,ClPDS,ClZDS,ClCHXE,ClCRTISO and ClLCYB also exhibited clearly different expression patterns accompanied by carotenoid accumulation.

文摘In order to screen out the small watermelon varieties with red flesh suitable for soilless cultivation and to meet people's requirements of high-quality watermelons,comparative tests were conducted on 6 small watermelon varieties with red flesh under soilless culture conditions in 2018.Based on the data of field traits,yield traits,quality traits and commercial traits,the integrated results showed that ‘Chuanqi 2’ had good quality and high yield,and the overall performance was the best.
