Electrophoresis of creatine kinase isoforms:a highly sensitive fluorescence scanning method

Objective To develop an agarose electrophoretic method for creatine kinase (CK) isoforms, using highly sensitive fluorescence scanning.Methods A discontinuous buffer system was used. Electrophoresis on agarose gel was performed under constant current and low voltage. CK isoforms were separated within 30 minutes and detected by fluorescence scanning.Results There were no significant differences when the activities of CK MM were between 853.0 U/L and 14.0 U/L and those of CK MB between 152.0 U/L and 2.4 U/L. The detection limits of stain method for CK MM and CK MB isoforms were 36.0 U/L and 12.3 U/L, respectively; while those of fluorescence method were 12.0 U/L and 2.1 U/L. The experimental results showed good precision for CK MM isoforms, as well for CK MB isoforms and isoenzymes.Conclusion An agarose electrophoretic method has been developed to measure CK isoenzymes and isoforms clinically. This method is rapid, simple, sensitive, highly reproducible and inexpensive. It is suitable for general laboratories. 展开更多

By laser scanning fluorescence microscopy for quan-titative measurement of fluorescence intensity changes on egg surface stained with fluorescein isothiocyanate duxing cleavage furrow extending forward, it was found that in area of presumptive cleavage furrow the scanning curve became ∨ shape, indicating dark stripe appeared in that place. Then the fluorescence intensity increased at the place where the botton of ∨ shape had located, and the scanning curve tuxned to ∧ shape, indicating single stripe was formed. While enhanced fluorescence appeared on the borders of ∧ shape, an M shape curve was found, show-ing double stripe occurred. During the distance between two borders of M shape incresing from 50 μm to 100μm,a fluorescence peak came to sight in the middle of the M shape, which being the cleavge furrow bottom. The two lateral sides of furrow bottom with decreasing fluorescence were nascent membrane. At that time the curve became W shape. By the sides of cleavage furrow the the stress folds became conspicous after double stripe stage, showing the stretching of the egg surface being increased. With our[31, 33] and others[32] reports that polylysine could induce the appearance of nascent membrane and phyto-hemagglutinins could decrease or prevent the appearance of nascent membrane, we believed the idea of Schroeder[25] that increasing mechanical stress could initiate nascent membrane formation and thought that the stress lay to the outsides of cleavage furrow. 展开更多

X-ray fluorescence scanning with synchrotron radiation was performed to study sediment core records of floods in Amur Bay,Sea of Japan.Interlayers of 3–8 mm with abnormally low bromine content were formed by the Razdolnaya River discharge to the central part of the bay during extreme floods,accompanied by severe storms at sea.Such conditions in the region are typical for periods of deep tropical cyclones(typhoons),to which the distinguished interlayers were compared on the timescale.This approach was made possible thanks to the high rate of sedimentation in the bay(3–5 mm/a) and low bioturbation of sediments under anoxic conditions. 展开更多

A sensitive analytical method, constant-energy synchronous fluorimetry, has been used to determine benzo[a]pyrene (BaP) selectively in airborne particulates (AP). A constant-energy difference of 1 400 cm^-1 was selected for obtaining single spectral peak. The method could easily identify BaP in an 18-component polycylic aromatic hydrocarbons(PAHs) mixtures. Calibration plots in methanol solution and in sodium dodecylsulfate (SDS) micellar media show a good linearity (R〉0.999) in the BaP concentration range generally fotund in the environment. The method gives a detection limit of 0.40 nmol/L in SDS micellar medium and 1.34 nmol/L in methanol solution. 展开更多
