应激性胃溃疡大鼠前爪不同区域黑色素变化的ghrelin-POMC通路研究 被引量:8

目的探讨应激性胃溃疡大鼠前爪手掌不同区域黑色素变化及其机制。方法采用随机数字表法将20只SD雄性大鼠分为空白对照组、模型组,每组各10只,利用无水乙醇灌胃复制应激性胃溃疡模型后,利用多巴氧化酶检测两组大鼠前爪皮肤黑色素变化;用E... 目的探讨应激性胃溃疡大鼠前爪手掌不同区域黑色素变化及其机制。方法采用随机数字表法将20只SD雄性大鼠分为空白对照组、模型组,每组各10只,利用无水乙醇灌胃复制应激性胃溃疡模型后,利用多巴氧化酶检测两组大鼠前爪皮肤黑色素变化;用ELISA法检测两组大鼠血清ghrelin水平;用RT-PCR法检测两组大鼠胃黏膜组织ghrelin mRNA、垂体和前爪皮肤组织POMC mRNA水平。结果模型组大鼠5区的黑色素细胞明显增多;与空白对照大鼠比较,模型组大鼠的血清ghrelin水平及ghrelin mRNA降低,且差异有统计学差异(P<0.05);模型组大鼠的垂体和前爪皮肤组织POMC mRNA水平显著高于空白对照组(P<0.05)。结论应激性胃溃疡大鼠的病理变化可以在手掌特定区域通过黑色素的变化反映出来,初步推测其内在机制可能是通过作用于ghrelin-POMC通路实现的。 展开更多

目的探讨生理、病理状态下大鼠上肢手掌不同区域血含量和上肢手掌皮肤黑色素变化,为气色形态手诊乃至中医学的"司外揣内"提供合理的研究方法和实验依据。方法随机将SD雄性大鼠20只分为正常组和模型组,每组各10只。利用无水乙... 目的探讨生理、病理状态下大鼠上肢手掌不同区域血含量和上肢手掌皮肤黑色素变化,为气色形态手诊乃至中医学的"司外揣内"提供合理的研究方法和实验依据。方法随机将SD雄性大鼠20只分为正常组和模型组,每组各10只。利用无水乙醇灌胃复制急性胃损伤模型后,两组分别从大鼠尾静脉中给予一定浓度的伊文斯兰(EB)生理盐溶液。通过测量手掌不同部位组织内EB的吸光度(A)值大小,间接反映手掌各区域血含量的变化情况。利用多巴氧化酶检测两组大鼠前爪皮肤黑色素变化。结果 1模型组大鼠前爪5区的A值(0.96±0.46)显著高于正常组(0.35±0.17),差异有高度统计学意义(P<0.01)。2模型组大鼠5区的黑色素细胞增多。结论应激性胃溃疡的大鼠的病理变化可以在手掌特定区域反映出,初步推断这种变化是通过血流量的变化和手掌皮肤的黑色素变化实现的。 展开更多

Human amniotic epithelial cell transplantation for the repair of injured brachial plexus nerve: evaluation of nerve viscoelastic properties 被引量:13

The transplantation of embryonic stem cells can effectively improve the creeping strength of nerves near an injury site in animals. Amniotic epithelial cells have similar biological properties as em-bryonic stem cells; therefore, we hypothesized that transplantation of amniotic epithelial cells can repair peripheral nerve injury and recover the creeping strength of the brachial plexus nerve. In the present study, a brachial plexus injury model was established in rabbits using the C6root avulsion method. A suspension of human amniotic epithelial cells was repeatedly injected over an area 4.0 mm lateral to the cephal and caudal ends of the C6 brachial plexus injury site (1 × 106 cells/mL, 3μL/injection, 25 injections) immediately after the injury. The results showed that the decrease in stress and increase in strain at 7,200 seconds in the injured rabbit C6 brachial plexus nerve were mitigated by the cell transplantation, restoring the viscoelastic stress relaxation and creep properties of the brachial plexus nerve. The forepaw functions were also signiifcantly improved at 26 weeks after injury. These data indicate that transplantation of human amniotic epithelial cells can effec-tively restore the mechanical properties of the brachial plexus nerve after injury in rabbits and that viscoelasticity may be an important index for the evaluation of brachial plexus injury in animals. 展开更多

A brachial plexus injury model was established in rabbits by stretching the C6 nerve root. Imme- diately after the stretching, a suspension of human amniotic epithelial cells was injected into the injured brachial plexus. The results of tensile mechanical testing of the brachial plexus showed that the tensile elastic limit strain, elastic limit stress, maximum stress, and maximum strain of the injured brachial plexuses were significantly increased at 24 weeks after the injection. The treatment clearly improved the pathological morphology of the injured brachial plexus nerve, as seen by hematoxylin eosin staining, and the functions of the rabbit forepaw were restored. These data indicate that the injection of human amniotic epithelial cells contributed to the repair of brachial plexus injury, and that this technique may transform into current clinical treatment strategies. 展开更多

We present a catheter-based optical diffusion and fluorescence(ODF)probe to study the functional changes of the brain in vivo.This ODF probe enables the simultaneous detection ofthe multi-wavelength absorbance and fluorescence emission from the living rat brain.Our previous studies,including a transient stroke experiment of the rat brain as well as the brainresponse to cocaine,have established the feasibility of simultaneously determining changes incerebral blood volume(CBV),tissue oxygenation(StO2)and intracellular calcium([Ca^(2+)]i,using the fluorescence indicator Rhod2).Here,we present our preliminary results of somatosensory response to electrical forepaw stimulation obtained from the rat cortical brain by using theODF probe,which indicate that the probe could track brain activation by directly detecting[Ca^(2+)]i along with separately distinguishing CBV and StO2 in real time.The changes of CBV,StO2 and[Ca^(2+)]i are comparable with the blood-oxygen-level-dependent(BOLD)response tothe stimulation obtained using functional magnetic resonance imaging(fMRI).However,thehigh temporal resolution of the optical methodology is advanced,thus providing a new modalityfor brain functional studies to understand the hemodynamic changes that underlie the neuronalactivity. 展开更多
