Type 1 diabetes mellitus(T1DM) lacks insulin secretion due to autoimmune deficiency of pancreaticβ-cells.Protecting pancreatic islets and enhancing insulin secretion has been therapeutic approaches.Mannogalactoglucan is the main type of polysaccharide from natural mushroom, which has potential medicinal prospects.Nevertheless, the antidiabetic property of mannogalactoglucan in T1DM has not been fully elucidated. In this study,we obtained the neutral fraction of alkali-soluble Armillaria mellea polysaccharide(AAMP-N) with the structure of mannogalactoglucan from the fruiting body of A.mellea and investigated the potential therapeutic value of AAMP-N in T1DM.We demonstrated that AAMP-N lowered blood glucose and improved diabetes symptoms in T1DM mice.AAMP-N activated unfolded protein response(UPR) signaling pathway to maintain ER protein folding homeostasis and promote insulin secretion in vivo.Besides that,AAMP-N promoted insulin synthesis via upregulating the expression of transcription factors, increased Ca^(2+) signals to stimulate intracellular insulin secretory vesicle transport via activating calcium/calmodulin-dependent kinase Ⅱ(CamkⅡ) and cAMP/PKA signals, and enhanced insulin secretory vesicle fusion with the plasma membrane via vesicle-associated membrane protein 2(VAMP2).Collectively,these studies demonstrated that the therapeutic potential of AAMP-N on pancreatic islets function, indicating that mannogalactoglucan could be natural nutraceutical used for the treatment of T1DM. 展开更多

The effects of active modified atmosphere packaging (MAP) on the postharvest quality of shiitake mushrooms stored at cold temperature (4°C) were investigated. The gas components were 2% O2 +7% CO2 (MAP1),2% O2 +10% CO2 (MAP2) and 2% O2 +13% CO2 (MAP3),respectively.The results showed that active MAP could extend the shelf-life of shiitake mushrooms to 17 d and the concentration of carbon dioxide could influence the postharvest quality of shiitake mushrooms.MAP2 treatment inhibited the increase in respiration rate and malondinaldehyde (MDA) contents, delayed the decrease in firmness, soluble sugar and vitamin C,and obviously reduced the activity of polyphenol oxidase (PPO) and the degree of browning, therefore maintaining better quality. 展开更多

The heavy metal bioaccumulation levels of 13 species of mushrooms were studied. 13 different species of wild mushrooms growing in China were analyzed for Cu, Zn, Pb, Cd, and As. Contents of Cu, Zn, Cd and Pb in mushroom and Cu, Zn in Soil were tested by atomic absorption spectrometer (AAS), and As in samples by atomic fluorescent light (AFL). Contents of Cd and Pb in soil were analyzed by graphite furnace AAS. The results showed that contents of Cu, Pb, Cd, and As in Termitomyces microcarpus were the highest in the tested mushroom samples, which were 135.00, 13.28, 65.30, and 1.60 mg·kg^-1 (dry biomass) respectively, and the corresponding bioconcentration factor (BCF) for Cu and Cd was 57 and 1 674 separately, which was the highest data in 13 mushroom species. The capability ofBoletus griseus for Cd accumulation was very strong with BCF of 300. when people consume the wild edible mushrooms, contents of Cd and Pb should be greatly considered. 展开更多

The application of molecular tools for the identification of strains, populations and species has become a common practice in a variety of applied and basic investigations in many parts of the world. However, standardization of such applications varies widely among organisms and scientific fields. In this mini-review,the author provides a brief introduction to one of the most prominent effort for species identification-the international barcode of life(iBOL) project, discusses the features of fungal diversity including the proposed fungal barcode DNA fragment-the intergenic spacer regions(ITS) of the nuclear ribosomal RNA gene clusters, and illustrates the potential promises and problems of using ITS for barcoding and for analyzing the phylogeographic pattern of the wild gourmet mushroom Tricholoma matsutake species complex. The analyses show that identification to species level is often insufficient for practical applications and that sequences from multiple genes in combination with critical morphological and physiological evaluations are needed to identify strains, populations and species. 展开更多

The mushrooms are highly regarded as one of the most nutritious foods across the globe but also recognized bio-accumulators of heavy metals. The nature and level of industrial activities are continually changing and affecting the environment adversely. The mushrooms are not an exception and may inevitably have heavy metal contaminations. In this vein, this study aimed to determine heavy metal (Cu, Ni, Co, Zn, Pb, and Cd) uptake levels in wild edible mushrooms from the sites with different economic activities. The wild mushrooms considered for this study included Tente (Amanita Zambiana), Ichikolowa (Termitomyces Titaniscus), and Kabansa (Lactarius Tataniscus). The analysis of heavy metal concentration was carried out using atomic absorption spectrometry (AAS). For the selected mushrooms, concentration ranges (mg·kg-1 total dry weight (dw)) of 46.90 - 141.80 for Cu, 0.10 - 6.60 for Cd, 1.10 - 2.00 for Pb, 19.00 - 38.90 for Zn, 1.00 - 3.40 for Ni, and 44.80 - 79.70 for Co were obtained. However, for the respective soil samples, concentration ranges (mg·kg-1·dw) of 51.00 - 279.40 for Cu, 1.00 - 99.50 for Cd, 8.00 - 10.00 for Pb, 22.80 - 209.10 for Zn, 9.00 - 33.70 for Ni, and 60.00 - 111.90 for Co were obtained. To a certain degree, the concentrations reflected the impact of diversity in the surrounding activities. This study discovered that for the selected mushrooms, the contamination level of cadmium, cobalt, nickel, and copper exceeded the World Health Organization (WHO)/FAO (Food and Agriculture Organization) recommended limits. Although some minor aberrations from the prescribed limits were also observed in the case of copper and nickel. Further, the data established that the heavy metal concentrations in respective soils are not the sole determinant of concentrations in mushrooms. Thus, these findings merit attention as, in some cases, the extent of contamination has exceeded the WHO permissible limit, and it may pose a health risk to consumers. 展开更多

Auricularis polytricha, Lentinus edodes, Agrocybe sp and Pleurotus flabellatus were grown under submerged culture and screened for endopolysaccharides. The fermentation was done in 250 ml working volume Erlenmeyer flask and the fermentation curves for all species were established. Pleurotus flabellatus has the highest rate of biomass production at the rate of 0.180 g/L/day, at 10 days hence chosen for further investigation. Two additional media, viz. Mushroom Complete Media (MCM) and Yeast Malt (YM) were selected to be compared with potato extract(PE) media used initially. MCM media produced the highest biomass productivity at the rate of 0.311 g/L/day. Pleurotus flabellatus biomass was extracted using modified Mizuno method and the endopolysaccharide obtained was tested for β-glucan. The yield of β-glucan was 7.70 ± 1.11 g/100g. The polysaccharides were purified using column chromatography to yield four fractions. The fourth fraction F4, gave the highest molecular weight at 3.058 × 106 Dalton (11.8%) and 1.282 × 104 Dalton (88.2%). The mushroom, P. flabbelatus was cultured using air-lift bioreactor, and the highest productivity was obtained at air-flowrate 2 L/min, yielding 2.25 g/L/day. The yield of biomass against substrate used (glucose consumption) Yb/s was 0.78 g/g. 展开更多

The molecular mechanisms of energy status related to the umami taste of postharvest shiitake mushrooms during spore release remain poorly understood. In this study, the variations of energy status and umami taste of mushrooms were measured at 25℃.At 24 h storage, slight spore prints of mushrooms were first pictured, respiration peaked.Significant ATP decrease and ADP increase were also observed as the initiation of postharvest senescence(P<0.05).Meanwhile,the activities of phosphohexose isomerase, succinate dehydrogenase,glucose-6-phosphate dehydrogenase and cytochrome c oxidase and the contents of umami nucleotides and amino acids were maintained at higher levels in mushrooms with spore release.Notably, the equivalent umami concentration(EUC)was strongly correlated with energy levels(R=0.80).Fifteen related gene expression levels in the energy metabolism pathway were downregulated.LecpdP1 and LeAK were significantly expressed in the conversion of ATP into AMP and played key roles in connecting the energy state and umami level. These results provided valuable insights on the umami taste associated with energy metabolism mechanism during postharvest mushroom spore release. 展开更多

Every year mushroom wastes are produced in big volumes from edible mushroom production industries. Vermicomposting technology using earthworms as versatile natural bioreactors for effective recycling of organic wastes to the soil is an environmentally acceptable means of converting waste into nutritious composts for crop production. This study was undertaken to use earthworms (eisenia foetida) in vermicomposting on edible mushrooms wastes. The Randomized block design with three treatments in Kimia, Sabalan and Artashahr mushroom production industries were selected and four replications were applied for treatments. The total biomass of earthworms was estimated by counting the number of adults, juveniles and cocoons from each replication. The number of adult and mature worms, egg capsules and wet weight of earthworms were recorded in one month intervals (totally in five months). Vermicomposting was carried out by these worms in four months in spite of delay in their establishment in mushroom wastes. The worms wet weight, their number and the number of capsules were increased up to fourth month and then decreased. The produced vermicompost had suitable properties for amending soil. 展开更多

Eleven different wild-edible mushroom species growing in the Black Sea region of Turkey were analysed for their metal content. Specimens of mushrooms were gathered in Trabzon, Giresun and Ordu and analyzed for 31 minerals, four of which (Be, Sb, Te and Ti) were not detected. Whereas some minerals including Ag, As, Cd, La, Mo, Pb, Se, Y and Zr were detected in just a few mushroom species, another 18 minerals were found in all 11 species. All metal concentrations were expressed on a dry weight basis (d.w.). With regard to nutritionally important amounts of essential and trace minerals, the metal content (μg/g d.w.) of mushroom samples ranged from 21,800-39,800 for K, 2590-14,000 for P, 268-1600 for Ca, 561-1210 for Mg, 74-829 for Fe, 11.2-321 for Cu, 36.2-241 for Zn, 14.1-76.5 for Mn and 0.13-2.85 for Co. Small amounts of toxic metals such as As, Cd and Pb were found in all 11 mushroom species. L. laccata contained a large amount of As (145 μg/g d.w.). These results show that the investigated mushrooms can be a useful component for human diets because of their high content of many essential minerals and trace elements and low content of toxic metals. 展开更多

The investigation used (1,1-diphenyl-2-picrylhydrazyl radical) DPPH as a substrate to determine radical scavenging abilities. Further quantitative analyses for carotenoids, flavonoid, total phenolic compounds and vitamin C content were done using spectrophotometric assay at 515 nm. Comparatively, fresh mushroom had higher vitamin C and Lycopene contents than their dry counterparts with β-carotene. Flavonoid and phenolic compounds were higher in dry forms than those in their fresh counterparts. Radical scavenging ability also varied with fresh forms depicting higher activity compared to dry forms. The variation was low in Afrocantharellus species (0.8% and 0.99%) while it was higher in Cantharellus species (24.12% and 7.52%). The highest ability was observed in fresh forms of Cantharellus pseudocibarius (EC50 50 = 0.4 mg/ml) was from dry form of Cantharellus rufopunctatus. This shows that studied mushrooms are rich in antioxidants and their quality and quantities are affected differently in both forms. Both dry and fresh mushrooms are potential sources of natural antioxidants. The portrayed high scavenging ability by these mushrooms envisages promoting them into potential nutraceutics. 展开更多

In Guizhou Province, the area of rocky desertification land is 3.023 8 million hm^2, accounting for 17.16% of total area of land in the province. Rocky desertification is the most important ecological problem that restricts the economic and social development in Guizhou, so the primary task of ecological construction in Guizhou Province is to curb rocky desertification. How to effectively, efficiently and persistently use rocky desertification land is not only the key to the improvement of ecological environment, but also an urgent problem solved for local people's survival and anti-poverty. Due to having developed roots, growing fast, high yield of grass, strong resistance to drought, and high regeneration capacity, Pennisetum sinese Roxb has become a new method to control rocky desertification. How to use P. sinese Roxb efficiently becomes the key to the ecological control of rocky desertification by P. sinese Roxb. In this study, the ecological agriculture development model of "P. sinese Roxb-rocky desertification control-edible mushrooms" was studied, and the effect of cultivating Pleurotus eryngii with P. sinese Roxb in rocky desertification land was analyzed. Moreover, an outlook for the application of P. sinese Roxb in rocky desertification areas of Guizhou was given. 展开更多

To examine the potential ability of edible mushrooms to act as biocatalysts, 19 basidiomycete strains were screened. Modified media (PG, O, and PGO medium) for liquid cultivation of these basidiomycete strains were designed and tested. Wet cells (>10 g) of 4 basidiomycete strains (Pleurotus salmoneostramineus H7, P. salmoneostramineus H13, Ganoderma lucidum NBRC31863, Flammulina velutipes NBRC31862) were harvested from PGO medium for 7 days. The stereoselective reduction of α-keto esters using the 4 strains was tested. It was found that each of these strains had a reducing activity toward 6 aliphatic α-keto esters. In the presence of L-alanine as an additive, the reduction of ethyl 2-oxobutanoate and ethyl 2-oxopentanoete by P. salmoneostramineus H7 produced the corresponding alcohol with a high conversion ratio and with excellent enantiomeric excess (>99% e.e. (R)). Furthermore, ethyl pyruvate, ethyl 2-oxobutanoate, and ethyl 2-oxopentanoate were predominantly reduced to the corresponding (R)-hydroxy ester (>99% e.e.) by G. lucidum. Thus, we found that these edible mushrooms have great potential to be used as biocatalysts for the stereoselective reduction of carbonyl compounds. 展开更多

A comparative study of the number of taxa and fungal biomass consumed by the Pygmies and the Bantus of Gabon has been realized in two Gabon north regions (Ogooue-Ivindo and the Woleu-Ntem). This study has allowed not only to establish the list of consumed fungal sorts by those populations, but also to estimate the daily consumed fungal biomasses by each ethnical group. The study has revealed a significant difference between a numbers of taxa and the quantity of mushrooms consumed by various ethnical groups: the Pygmies of the area who live especially by hunting and fishing consumed 96% of the recorded taxa with some high mushrooms quantities (around 3 kg/day/family);the Bantus who live nearby Pygmies also consumed some high mushrooms quantities (around 2 kg/day/family) but a low taxa number (56% of taxa counted by the Fang;69% by the Kota;39% by the Kwele);on the other hand, the Bantus living nearest the Pygmies consumed a high taxa number as much as Pygmies (around 90% of taxa counted) but eat a lower mushrooms quantities than theirs distant congeners Pygmies (around 800 g/day/family). 展开更多
