The location of the toxin gene of B. thuringiensis subsp. galleriae (H5ab) on the Mr-130Mdplasmid is determined by molecular cloning. Double digestion fragments (BamHⅠ and SalⅠ)and PstⅠ restriction fragments as well, from the 130 Md plasmid of B. thuringiensis subsp.galleriae, are ligated with the cloning vector pAT 153 respectively and transformed into E.coli strain HB 101. Out of 208 transformants, three colonies (FG2, FG9, FG19) give posi-tive hybridization reaction using the HD-1 delta-endotoxin gene as a probe. They are presum-ed to contain the delta-endotoxin gene of B. thuringiensis subsp. galleriae. Western bolt assaysindicate that Mr-130 kDal and 68 kDal, crystal proteins produced by clone FG 2 react withanticrystal protein antibody. The protein extracts of clone FG2 are lethal to Ostrinia furna-calis (Guenee). This is the first report with regard to the cloning and expression of the B. thuringiensissubsp. galleriae (H5ab) delta-endotoxin gene. 展开更多

目的对精制马抗H5N1禽流感病毒免疫球蛋白疏水层析方法进行优化。方法将胃蛋白酶直接加入制备好的高效价马抗H5N1禽流感病毒血清进行消化裂解,再加入饱和度为26%的硫酸铵溶液,盐析沉淀离心一次后,直接进行疏水层析进一步纯化F(ab′)2抗... 目的对精制马抗H5N1禽流感病毒免疫球蛋白疏水层析方法进行优化。方法将胃蛋白酶直接加入制备好的高效价马抗H5N1禽流感病毒血清进行消化裂解,再加入饱和度为26%的硫酸铵溶液,盐析沉淀离心一次后,直接进行疏水层析进一步纯化F(ab′)2抗体。结果连续洗脱依次出现6个蛋白峰,50min左右,洗脱峰3开始出现,其中含大部分F(ab′)2抗体,此时硫酸铵摩尔浓度约为1.0~1.1mol/L。阶段梯度洗脱,当硫酸铵摩尔浓度降至1.0mol/L时,大量F(ab′)2片段被洗脱下来,纯度达90%以上。结论优化了精制马抗H5N1禽流感病毒免疫球蛋白的疏水层析方法,降低了硫酸铵用量,减少了离心次数,生产周期也大大缩短。 展开更多

The HeI photoelectron (PE) spectra of both 2(5H) furanone and its trans-chair-dimenc-compound (t-c-DFN) are reported.The assignment of the PES bands is made on the basis of band shapes,the PES results of the molecules which have the similar atomic groups,and the restricted Hartree-Fock (RHF) calculations for the molecules studied.From the results of both PES experimental and theoretical calculations,it is proved that the ionization potential (IPs) of the HOMO for the dimenc-compound is lower than that of the HOMO for the monomer.And the total energy computed for the t-c-DFN is the lowest in the four possible configurations of dimeric-compounds of 2(5H) furanone Therefore the synthesis of t-c-DFN is also the easiest. 展开更多

The stability constants of complex anion In(tiron)^-in mixed solvents of ethanol-water with weight percentages of ethanol being 0,5,15,25,35 and 50 have been measured spectrophotometrically at 293 K,using NaClO_4 as supporting electrolyte with ionic strength ranging from 0.1 to 3.3mol·dm^(-3). The thermodynamic stability constants of In(tiron)^-in various mixed solvents have been obtained by a polynomial approximation equation based on Pitzer's theory.As a comparison,the traditional Debye-Hückel equation has also been used to treat the same data.The results show that this polynomial approximate method is obviously superior to the old one.It was discovered that there is a linear rela- tion between the thermodynamic stability constants of In(tiron)^-and the mole fraction of organic component in mixed solvents or the reciprocal of dielectric constants of mixed solvents.The medium effect and the standard free energy of transfer,ΔG_(tr)~0,for the coordination reaction:In^(3+)+tiron^(4-)= In(tiron)^-,have also been discussed. 展开更多
