A fast and sensitive method for determination of 8 diuretics (acetazolamide, bendroflumethiazide, bumetanide, chlorthalidone, furosemide, hydrochlorothiazide, metolazone, triamterene) and masking agent (probenecid) in human urine using gas-chromatography with mass spectrometric detection is described. The extraction of the substances as function of the nature of organic solvent, mixing time and pH of aqueous phase was studied. The tandem mass spectrometry was used to increase selectivity of diuretics determination due to elimination of background interferences. Fragmentation reactions were studied for each compound and their collision energies were optimized to obtain the best selectivity. The results of method’s validation demonstrate its suitability in routine analysis for confirmation purposes. 展开更多

In Veda, cow’urine was compared to the nectar. In Susrut, several medicinal properties of cow’ urine have been mentioned and are known to cause weight loss, reversal of certain cardiac and kidney problems, indigestion, stomach ache, edema, etc. However, the literature and scripture did not mention the antigenotoxic properties of cow’urine. Methods In the present investigation, the antigenotoxic/ antioxidant properties of cow’ urine distillate and redistillate were studied in vitro. The antioxidant status and volatile fatty acid levels were determined. Actinomycin-D (0.1ol/L) and hydrogen peroxide (150 mol/L) were used for inducing DNA strand break with 0.1% DMSO as negative control. Dose for the antigenotoxic effect of cow’ urine was chosen from the dose response study carried out earlier. Results Both actinomycin-D and H2O2 caused statistically significant DNA unwinding of 80% & 75% respectively (P<0.001) as revealed by fluorimetric analysis of DNA unwinding (FADU), and the damage could be protected with the redistilled cow urine distillate (1, 50 & 100 ) in simultaneous treatment with genotoxic chemicals. Conclusion The redistillate of cowurine was found to possess total antioxidant status of around 2.6 mmol, contributed mainly by volatile fatty acids (1500 mg/L) as revealed by the GC-MS studies. These fatty acids and other antioxidants might cause the observed protective effects. 展开更多

A simple and rapid dispersive liquid–liquid microextraction(DLLME)technique coupled with gas chromatography–ion trap mass spectrometry(GC–MS)was developed for the extraction and analysis of methamphetamine(MA),pethidine(PD),ketamine(KT)and tramadol(TD)from human urine.In this study,different parameters affecting the extraction process such as the type and volume of extraction solvent,type and volume of disperser solvent,extraction time and pH value and salt effect were studied and optimized.Under optimized conditions,the enrichment factor ranged from 185 to 226 and the average recovery ranged from 80.45%to 95.55%.The linear range was 10.0–1000.0 mg/L,the limit of detection and quantitation were in the range 0.43–1.96 mg/L and 1.44–6.53 mg/L,respectively.The relative standard deviations were in the range 1.98%–3.90%(n=7).The obtained results show that DLLME combined with GC–MS is a fast and simple method for the determination of MA,PD,KT and TD in human urine. 展开更多

Source separation sanitation systems have attracted more and more attention recently.However, separate urine collection and treatment could induce odor issues, especially in large scale application. In order to avoid such issues, it is necessary to monitor the odor related compounds that might be generated during urine storage. This study investigated the odorous compounds that emitted from source-separated human urine under different hydrolysis conditions. Batch experiments were conducted to investigate the effect of temperature, stale/fresh urine ratio and urine dilution on odor emissions. It was found that ammonia, dimethyl disulfide, allyl methyl sulfide and 4-heptanone were the main odorous compounds generated from human urine, with headspace concentrations hundreds of times higher than their respective odor thresholds. Furthermore, the high temperature accelerated urine hydrolysis and liquid–gas mass transfer, resulting a remarkable increase of odor emissions from the urine solution. The addition of stale urine enhanced urine hydrolysis and expedited odor emissions. On the contrary, diluted urine emitted less odorous compounds ascribed to reduced concentrations of odorant precursors. In addition,this study quantified the odor emissions and revealed the constraints of urine source separation in real-world applications. To address the odor issue, several control strategies are recommended for odor mitigation or elimination from an engineering perspective. 展开更多

建立了尿中除草剂莠去津(ATRZ)代谢物去乙基莠去津(DEA)、去异丙基莠去津(DIA)及去乙基去异丙基莠去津(DDA)的分析方法。尿样加入内标2-氨基-4-甲氧基石-甲基-1,3,5-三氮嗪(AMMT),碱化后用高极性GDXS01大孔树脂吸附、乙酸... 建立了尿中除草剂莠去津(ATRZ)代谢物去乙基莠去津(DEA)、去异丙基莠去津(DIA)及去乙基去异丙基莠去津(DDA)的分析方法。尿样加入内标2-氨基-4-甲氧基石-甲基-1,3,5-三氮嗪(AMMT),碱化后用高极性GDXS01大孔树脂吸附、乙酸乙酯洗脱进行固相萃取,萃取物在乙腈溶剂中用N-甲基-N-特丁二甲硅烷基三氟乙酰胺(MTBSTFA)进行衍生化,衍生物进行选择离子监测气相色谱-质谱联用法分析。ATRZ不发生特丁二甲硅烷基(TBDMS)衍生化,则以非衍生化形式同时进行分析。尿中DEA、DIA、DDA、ATRZ的萃取率分别为80%、78%、71%和85%,检出限分别为0.2、0.2、0.1、1μg/L。尿样中ATRZ的3种代谢物质量浓度在0.5~100μg/L范围内,ATRZ在5~1000μg/L范围内线性关系良好。在1、50μg/L加标水平下3种代谢物的回收率分别为97%~103%和98%~101%,相对标准偏差分别为9.2%~11.7%和5.2%~8.9%;空白尿中添加10μg/L和500μg/L ATRZ的回收率分别为101%和99%,相对标准偏差分别为12.7%和6.7%。该方法灵敏度高,可以分析莠去津中毒者和职业接触莠去津者尿中的莠去津及其代谢物。 展开更多

Pesticides can remain in the environment for decades and contaminate surface water that is used for irrigation of produce. This study examined pesticide residues in some surface waters and foods in Louisiana. Samples of 8 foods (tomato, corn, rice, blueberry, cucumber, cabbage, wheat and melon) and 35 surface waters were studied using a QuEChERS extraction method for food samples and liquid-liquid extraction method for the water samples. Gas chromatography-mass spectrometry was used to analyze water and food samples. Nine pesticides were detected in the surface water samples and 5 in the food samples. Pesticides detected in foods were below FDA tolerance limit but 0.18 ppm cypermethrin found in tomato was within 90% of the FDA limit (0.2 ppm). Four water samples had atrazine levels that were above the FDA limit for potable water. This study suggests the need to intermittently monitor pesticide contamination in our food and water. 展开更多

Alleviation of Pesticide Residue in Surface Water

Zeolite is a natural mineral capable of removing certain chemical contaminants from water. This study was carried out to test the effect of zeolite treatment on pesticide residue alleviation in surface water. Ten surface water samples were treated with natural zeolite by filtering through. An EPA method was used to extract pesticide residue from the water samples and the surfactant used to modify the net charge on the zeolite was hexadecyltrimethylammonium chloride (HDTMA-Cl). Gas chromatography-mass spectrometry was used to analyze water samples. Alleviation was achieved in all the 10 water samples that were filtered through zeolite. The highest removal of pesticides from water with zeolite included 100% of bifenthrin in sample CLC, atrazine in BPH, CDG and LBT;metolachlor in CLC, LBT, BCH, TRH2 and BPI;acetolachlor in BBH and BCH;azoxystrobin in BBH;desethylatrazine in BCH and BPI;metribuzin in BCH, TRH2 and BPI;and both clomazone and bromacil in sample BDC. A minimum reduction of 10.9% was found for metolachlor in sample BRH. Further reduction of pesticide residues up to 50% was recorded in the SMZ treatment as the concentrations of 4 out of 8 pesticide residues were reduced. This study confirms the potential of both the natural zeolite-Clinoptilolite, and SMZ of alleviating pesticide residues in water. 展开更多
