[Methods] Comparative methods were applied to study the effects of plant growth substance on the growth and development of D. officinale Kimura et Migo. [Results] Plant growth regulators play an important role in the process of tissue culture rapid propagation of D. officinale Kimura et Migo,and the main regulatory pathways are as follows.( i) Plant growth regulators promote the growth and development of D. officinale Kimura et Migo through regulating the changes of its endogenous hormones.( ii) Plant growth regulators affect the accumulation of polysaccharide content in D. officinale Kimura et Migo through regulating the changes in chlorophyll content and enzymes involved in sucrose metabolism.( iii) Plant growth regulators can increase the activity of antioxidant enzymes of D. officinale Kimura et Migo,to increase its stress resistance. [Conclusions]Using molecular biology techniques,combined with the changes of plant hormones in D. officinale Kimura et Migo,and analysis on key enzyme in the synthesis of plant hormones and gene expression,it is feasible to study the rules o changes in the content of active content of D. officinale Kimura et Migo.展开更多

AIM:To investigate the mechanism of gastric mucosal demage induced by water immersion restraint stress(WRS) and its prevention by growth hormone releasing peptide-6(GHRP-6).METHODS:Male Wistar rats were subjected to c...AIM:To investigate the mechanism of gastric mucosal demage induced by water immersion restraint stress(WRS) and its prevention by growth hormone releasing peptide-6(GHRP-6).METHODS:Male Wistar rats were subjected to conscious or unconscious(anesthetized) WRS,simple restraint(SR),free swimming(FS),non-water fluid immersion,immersion without water contact,or rats were placed in a cage surrounded by sand.To explore the sensitivity structures that influence the stress reaction besides skin stimuli,a group the rats had their eyes occluded.Cervical bilateral trunk vagotomy or atropine injection was performed in some rats to assess the parasympathetic role in mucosal damage.Gastric mucosal lesions,acid output and heart rate variability were measured.Plasma renin,endothelin-1 and thromboxane B2 and gastric heat shock protein 70 were also assayed.GHRP-6 was injected [intraperitoneal(IP) or intracerebroventricular(ICV)] 2 h before the onset of stress to observe its potential prevention of the mucosal lesion.RESULTS:WRS for 6 h induced serious gastric mucosal lesion [lesion area,WRS 81.8 ± 6.4 mm 2 vs normal control 0.0 ± 0.0 mm 2,P < 0.01],decreased the heart rate,and increased the heart rate variability and gastric acid secretion,suggesting an increase in vagal nervecarrying stimuli.The mucosal injury was inversely correlated with water temperature(lesion area,WRS at 35 ℃ 56.4 ± 5.2 mm 2 vs WRS at 23 ℃ 81.8 ± 6.4 mm 2,P < 0.01) and was consciousness-dependent.The injury could not be prevented by eye occlusion,but could be prevented by avoiding contact of the rat body with the water by dressing it in an impermeable plastic suit.When water was replaced by vegetable oil or liquid paraffin,there were gastric lesions in the same grade of water immersion.When rat were placed in a cage surrounded by sand,there were no gastric lesions.All these data point to a remarkable importance of cutenuous information transmitted to the high neural center that by vagal nerves reaching the gastric mucosa.FS alone also induced serious gastric injury,but SR could not induce gastric injury.Bilateral vagotomy or atropine prevented the WRS-induced mucosal lesion,indicating that increased outflow from the vagal center is a decisive factor in WRS-induced gastric injury.The mucosal lesions were prevented by prior injection of GHRP-6 via IP did,but not via ICV,suggesting that the protection is peripheral,although a sudden injection is not equivalent to a physiological release and uptake,which eventually may affect the vagal center.CONCLUSION:From the central nervous system,vagal nerves carry the cutaneous stimuli brought about by the immersion restraint,an experimental model for inducing acute gastric erosions.GHRP-6 prevents the occurrence of these lesions.展开更多

Primed seeds from each treatment group and non-primed seeds were sown In a field In which NaCI salinity of 15 dS/m was developed. Priming of seeds with CaCl2, followed by priming with KCI and NaCI, was found to be effective In alleviating the adverse effects of salt stress on both wheat cultivars In terms of shoot fresh and dry weights and grain yield. Priming with CaCl2 alleviated the adverse effects of salt stress on hormonal balance In plants of both cultlvars. In MH-97 plants, CaCl2 pretreatment considerably reduced leaf absclslc acid (ABA) concentrations and Increased leaf free salicylic acid (SA) concentrations under both saline and non-saline conditions. Methods: The β TCP/rhBMP 2 composite was constructed by combining β Tricalcium phosphat (β TCP) that was prepared by the authors with recombinant human morphogenetic protein 2 (rhBMP 2) and was implanted into the muscle pouches in the thigh of mice. β TCP alone was implanted on the opposite side as controls. At intervals of 1,3,7,14 and 28 days after the implantation, the specimens were obtained, and histologic study and alkaline phosphatase assay (7,14,28 days) were performed. Results: There was a large amount of cartilage and bone formation within the composite, increasing with time; whereas there was no new bone formation where β TCP alone was implanted. Besides, the levels of alkaline phosphatase in the β TCP/rhBMP 2 implants also were increasing with time and were higher than those in controls. Conclusions: The results indicate that β TCP/rhBMP 2 composite possesses heterotopic osteoinductive potential.展开更多

Methods: Seventy three normal SD rats w...Objective: To evaluate in vivo the effect of the recombinant human glial growth factor 2 (rhGGF 2) on the recovery of motor function of rat sciatic nerve following crush injury. Methods: Seventy three normal SD rats were randomly divided into three groups. A sham operation was performed in Group Ⅰ (n=5), and a 5 mm segment of the sciatic nerve was subjected to a 100 g crush load for 2 hours of duration in Groups Ⅱ (n=34) and Ⅲ (n=34). Group Ⅲ was treated with rhGGF 2 (1 mg/kg by subcutaneous injection after nerve crush, and then injected daily for the subsequent 4 days), and Group Ⅱ was given vehicle of the same volume. The motor functional recovery of never was assessed by calculating the sciatic functional index (SFI) and measuring the tetanic contractile strength of the extensor digitorum longus (EDL) of the hind limb. Results: Nerve function in Group Ⅲ began to recover at day 11 whereas the nerves in Group Ⅱ were still paralyzed after crush injury. The tetanic contractile strength of EDL was generally stronger in Group Ⅲ than that in Group Ⅱ, with a significant difference at 70 and 100 Hz stimulus frequencies from day 4 to day 14. Histological sections revealed less axonal degeneration and earlier regeneration of nerve fibers in Group Ⅲ. Conclusions: It suggests that rhGGF 2 is effective in promoting nerve regeneration and can significantly improve the functional recovery of rat sciatic nerve following crush injury.展开更多
